Effects of Vegetative Buffers on Reducing Soil Erosion and Nutrient Loss of Highland Field in Korea (고랭지밭의 토양침식 저감을 위한 완충식생대의 효과)

  • This study was carried out to investigate the effect of vegetative buffer to reduce runoff and soil and nutrient loss at highland agricultural area. The soil of experimental field was classified as Ungyo series (Fine, Humic Hapludults). An area of each field with lysimeter was $50m^2(width\;2.5m{\times}length\;20m)$ and was a gradient of 17%. Chinese cabbage (Brassica campestris L.) was cultivated by general management in each field. For establishing vegetative buffer, rye (Secalecereale L.), tall fescue (Festucaarundinacea Schreb) and orchard grass (Dactylis glomerata L.) were planted at the edge of field. Rye buffers were 1m, 2m and 4m wide. Both orchard grass and tall fescue buffers were 2m wide. Vegetative buffers were set up in September 2005 and chinese cabbage was planted in June 2006. Soil loss, runoff and nutrient loss were measured from June to August in 2006. Since the precipitation amount was heavy in July, amounts of runoff, soil erosion and nutrient loss were the highest in July during this study period. In comparison with control, vegetative buffers of rye 2m, orchard grass 2m and tall fescue 2m reduced runoff by 3%, 1% and 2%, respectively. In comparison among width of rye buffer, rye 1m, rye 2m, and rye 4m reduced by 1%, 4% and 13%, respectively. Vegetative buffers of rye 2m, orchard grass 2m and tall fescue 2m showed the reducing of soil loss by 59%, 46% and 28%, respectively. In comparison among width of rye buffer, the highest reducing effect of 88% was observed in 4m treatment. Additionally, vegetative buffer reduced N, P and K losses in runoff and eroded soil which were 10 to 54%, 7 to 24% and 11 to 21%, respectively. In different widths, wider vegetative buffer showed lower loss of N, P and K in runoff and eroded soil. As a result of this study, the vegetative buffer of rye was most effective for reducing runoff and soil loss in comparisons with other plants. In addition, wider range of buffers recommended for reducing runoff and soil loss, if possible.

Influence of Vetiver Grass (Vetiveria zizanioides) on Rhizosphere Chemistry in Long-term Contaminated Soils (중금속으로 오염된 토양에서 근권부의 화학적 특성에 미치는 vetiver grass (Vetiveria zizianioides)의 영향)

  • A detailed understanding and appreciation of the important mechanisms operating at the soil:root interface, commonly identified as the rhizosphere, is critical for evaluating the potential for particular plant species to be successfully used as part of a phytoremediation technique. For specific plants, mechanisms may exist to overcome the inherit limitation of the phytoremediation technique when poorly mobile soil metals are of interest. In the present study, the influence of root exudates on the rhizosphere chemistry of soil and consequential metal uptake were investigated following culture of vetiver grass (Vetiveria zizanioides), recognized as a promising plant for land stabilization, in three different long-term contaminated soils and one non-contaminated control soil. The soil solution pH increased (0.3-1.1 units) following vetiver grass culture and dissolved organic carbon (DOC) also significantly increased in all soils with the highest increase in PP02 (23 to $173mg\;L^{-1}$). Chemical changes are contributed to root exudation by vetiver grass when exposed to high concentration of heavy metals. Chemical changes, consequently, influenced metal (Cd, Cu, Pb, and Zn) solubility and speciation in the rhizosphere. The highest solubility was observed for soil Ko01 (eg. 2091 and $318{\mu}g\;L^{-1}$ for Cd and Pb, respectively). Initial heavy metal solubility in soils varied with soil and either increased or decreased following vetiver grass culture depending on the soil type. An increase in pH following plant culture generally resulted in a decrease in metal solubility, while elevated DOC due to root exudation resulted in an increase in metal solubility via the formation of metal-DOC complexes. Donnan speciation demonstrated a significant decrease in free Cd and Zn in the rhizosphere and the concentration of Cd, Pb, and Zn in vetiver grass shoot was highly correlated with soluble concentration rather than total soil metal concentration.

  • It was proven that rice straw can serve can serve as a good support medium for trickling filter with livestock wastewater treatment. The efficiency of removal of removal of environmentally undesirable components from the wastewater by the straw medium was no less inferior to other commonly used support materials. The treated straw was also proven to be a source of abundant plant nutrients. The growth of orchard grass was enhanced greatly by the application of large quantity of the support medium. A maximum yield of dry matter was obtained at the rate of 40 ton/ha. Inorganic NPK elements as background fertilizers were highly recommended for better development in an early stage of growth. The nutritional status of the soil was very much improved by the addition of the medium in terms of exchangeable cations(K, Ca, Mg and Na), available phosphate, and organic matter contents. Nutrient uptake by orchard grass such as N, P and cations was enhanced by rice straw medium treatment and that was proportional to the levels of treatment.

Differences in Soil Improvement Effects of Four Green Manure Crops in Greenhouse Cultivation (녹비작물 도입에 따른 토마토 시설재배지 연작장해 토양 개량 효과)

  • Four different green manure crops, hairy vetch, Italian ryegrass, crimson clover, orchard grass were cultivated to test the differences in their effects on soil improvement. After plowing to incorporate the green manure crops, tomato plants were grown as a succeeding crop to investigate the effects of green manure on the soil physical properties and plant growths. The experimental results showed that green manure could decrease the soil hardness. Italian ryegrass and hairy vetch as the green manure were especially effective in decreasing the soil hardness. Orchard grass and Italian ryegrass provided more void in soil matrix, resulting increased soil moisture contents. After two months, bulk density was decreased in the plots of Italian ryegrass and orchard grass. However, there were no significant differences between green manure plots and control after the cultivation of the succeeding crop. The results indicated that the growing Italian ryegrass and hairy vetch as green manure helped growing tomato plants. Orchard grass also improved the soil characteristics, comparable to Italian ryegrass in spite of their relatively small biomass.

Transcription Factor for Gene Function Analysis in Maize (옥수수 유전자 기능 분석을 위한 전사인자의 이해)

  • Transcription factors are essential for the regulation of gene expression in plant. They are binding to either enhancer or promoter region of DNA adjacent to the gene and are related to basal transcription regulation, differential enhancement of transcription, development, response to intercellular signals or environment, and cell cycle control. The mechanism in controlling gene expression of transcription can be understood through the assessment of the complete sequence for the maize genome. It is possible that the maize genome encodes 4,000 or more transcription factors because it has undergone whole duplication in the past. Previously, several transcription factors of maize have been characterized. In this review article, the transcription factors were selected using Pfam database, including many family members in comparison with other family and listed as follows: ABI3/VP1, AP2/EREBP, ARF, ARID, AS2, AUX/IAA, BES1, bHLH, bZIP, C2C2-CO-like, C2C2-Dof, C2C2-GATA, C2C2-YABBY, C2H2, E2F/DP, FHA, GARP-ARR-B, GeBP, GRAS, HMG, HSF, MADS, MYB, MYB-related, NAC, PHD, and WRKY family. For analyzing motifs, each amino acid sequence has been aligned with ClustalW and the conserved sequence was shown by sequence logo. This review article will contribute to further study of molecular biological analysis and breeding using the transcription factor of maize as a strategy for selecting target gene.

Optimization of Mannitol Fermentation by Leuconostoc mesenteroides sp. strain JFY (Leuconostoc mesenteroides sp. strain JFY 균주에 의한 만니톨 발효 조건의 최적화)

  • The production of functional foods providing health benefit is one of the fast growing fields in the food industry. Mannitol as GRAS (generally recognized as safe) is a functional food. Mannitol is about $70\%$ as sweet as sucrose and slowly and incompletely absorbed from the intestine, suppling only about one-half energy value of glucose. Commercially, the mannitol is synthesized by catalytic or electrochemical reduction of glucose. However, as strong demand for natural products increased, biological techniques have been developed for mannitol production. The object of this study was to determine the optimum conditions of mannitol fermentation by Leuconostoc mesenteroides sp. strain JFY isolated from fermented vegetables. The processes parameters such as pH, temperature, yeast extract concentration, and fructose concentration were optimized. The chosen ranges were 4.5 to 7.5 for pH, 22 to $34^{\circ}C$ for temperature, 0.05 to $2.0\%$ for yeast extract. and 5 to 350 g/L for fructose. The mineral medium used consisted of 3.0g $KH_2PO_4,\;0.01g\;FeSO_4{\cdot}H_2O,\;0.01g\;MnSO_4{\cdot}4H_2O,\;0.2g\; MgSO_4{\cdot}7H_2O,\;0.01g\;NaCl,\;and\;0.05g\;CaCl_2$ per 1 liter of deionized water. The optimum values of pH, temperature, yeast extract, and fructose concentration were obtained at about pH 6.5, temperature $28^{\circ}C$, yeast extract $0.5\%$ and fructose 30g/L. At optimum condition, the production of mannitol amounted to 31.6g/l. We hope that these findings are of particular importance for industrial application of mannitol production.

Ecological Studies on the Major Characters of Introduced Orchard grass Varieties (한국(韓國)에 도입(導入)된 Orchard grass(Pactylis glomerata L.) 중요형질(重要形質)에 관(關)한 육종학적(育種學的) 연구(硏究) (I))

  • This trial was conducted for the selection of drought registance strains and other main characters of orchard grass. Some preliminary results obtained are as follows : 1. The initial growth of orchard grass in sufficient soil moisture condition showed highly significant differences among the varieties. Turkey's D=0.84. However, in proceeding the growth, the grass height among the varieties showed non-significant. 2. Addition of nitrogen fertilizer before first flowering tended to increase the grass height occationally significantly, compared to the control. 3. The survived varieties on the drought soil moisture condition had more numbers of tillering than the wilted significantly except some varieties. However, in grass height between the survival and the wilted, almost varieties showed non-significant differences. 4. In the varierties of Metia, Phyllox and Dorise, all individuals were survived in drought soil moisture condition and showed almost the same growth responces with willted ones comparatively. The plant height and the numbers of tillering of these 3 varieties were as follows: Merita; 26.4 cm 24.2 ea, Phyllox; 31.4 cm 24.6 ea. Dorise; 26.8 cm 35.6 ea.

Fermentation characteristics of mulberry (Cudrania tricuspidata) fruit vinegar produced by acetic acid bacteria isolated from traditional fermented foods (전통발효식품으로부터 분리한 초산균을 이용한 꾸지뽕 열매 발효식초 제조 및 발효특성)

  • To raise the added value of the fruits of Cudrania tricuspidata, Cudrania tricuspidata vinegar was produced and examined for its fermentation conditions. Forty nine acetic acid bacteria with resistance against acetic acid, ethanol, and sulfide as high acetic acid producers were isolated from fermented foods and identified as Acetobacter indonesiensis, A. cerevisiae, A. orientalis, A. tropicalis, A. fabarum, A. pasteurianus, and A. syzygii based on the results of the analysis of the 16S rRNA gene sequences. Among them, two GRAS strains, A. pasteurianus SCMA5 and SCMA6, were finally selected for the production of acetic acid. Optimal vinegar productions were obtained from the medium containing 40% (v/v) fruit juice of Cudrania tricuspidata and 5% (v/v) ethanol at $25^{\circ}C$ for 72 hr. The sensory panel preferred the vinegar fermented with the SCMA06 to that with the SCM05 strain. The radical scavenger capacity of DPPH was 53% higher than that of the control in the vinegar fermented with the SCMA06 strain. The ${\alpha}$-glucosidase inhibitor activity as an index of the antidiabetic drug showed 91% inhibition, which is higher than that of acabose. This study will be helpful for the scale-up production of vinegar with the fruit of Cudrania tricuspidata.

Isolation and characterization of a protease deficient mutant of Aspergillus niger

  • Aspergillus niger has been used as a host system to express many heterologous proteins. It has various advantages over other expression systems in that it is a small eukaryotic GRAS (Generally Recognized aS Safe) organism with a capacity of secreting large amount of foreign proteins. However, it has been known that the presence of an abundant protease is a limiting factor to express a heterologous protein. The proteases deficient mutants of A. niger were obtained using UV -mutagenesis. A total of 1 ${\times}$ $10^5$ spores were irradiated with 10-20% survival dose of UV, 600J/M2 at 280nm, and the resulting spores were screened on the casein -gelatin plates. Ten putative protease deficient mutants were further analyzed on the starch plates to differentiate the pro from the secretory mutant. An endogenous extracellular enzyme, glucose oxidase, was also examined to confirm that the mutant phenotype was due to the proteases deficiency rather than the mutation in the secretory pathway. The reduced proteolytic activity was measured using SDS-fibrin zymography gel, casein degradation assay, and bio-activity of a supplemented hGM -CSF (human Granulocyte-Macrophage Colony Stimulating Factor). Comparing with the wild type strain, less than 30 % of proteolytic activity was observed in the culture filtrate of the protease deficient mutant (pro -20) without any notable changes in cell growth and secretion.

Development of Enhanced Yeast Expression System for GAP Promoter by Directed Evolution

  • Escherichia coli and Saccharomyces cerevisiae have been used as host for production of recombinant proteins. It is known that S. cerevisiae has advantages such as good folding and secretion capability, and safety as host over E. coli. But S. cerevisiae has shortcomings of low expression level which is just 20% of that of E. coli. To solve this problem, directed evolution method was tried to enhance the GAP promoter strength of S. cerevisiae in this study. As result, modified GAP promoter that has increased expression level of about 360% compared to that of wild type was selected.
