GRA를 활용한 무기체계 모듈화 우선순위선정 (Priority of Modularization in Weapon System by using Grey Relational Analysis)

  • 방위산업은 경제논리보다 안보논리가 우선시되며 고비용, 장기간의 연구개발이 이루어져왔다. 하지만, 최근 몇 년간 급변하는 기술의 발전과 안보상황을 통해 방위산업에서도 체계개발간에 있어 비용절감과 개발기간 단축의 필요성이 대두되었다. 이에 민수 분야의 부품 모듈화 전략을 방위산업에서 활용하기 위해 OO 사업 LRU(Line Replaceable Unit)들의 모듈화 우선순위를 도출하고자 한다. 프로젝트 평가기법 중 하나인 GRA(Grey Relational Analysis)를 사용하였으며, 문헌연구를 통해 선정된 6개의 평가지표 데이터를 수집하여 OO 사업의 11개 모듈에 대하여 우선순위를 선정하였다. 그 결과, M11(메인보드)과 M8(EMI모듈), M3(싱글보드컴퓨터)의 Grey 관계등급이 0.83, 0.81, 0.80 순으로 도출되었고, 우선순위에 따라 모듈화 하는 것이 비용 절감 및 기간 단축의 효과가 있을 것으로 판단된다. 본 연구는 유사 무기체계 개발이나 향후 성능개량 사업 등을 고려한 LRU 모듈화 설계 시, 모듈화 적용 판단 및 의사결정의 근거가 되는 기초 연구로 활용할 수 있을 것이다.

A Comparative Study of Estimation by Analogy using Data Mining Techniques

  • Software Estimations provide an inclusive set of directives for software project developers, project managers, and the management in order to produce more realistic estimates based on deficient, uncertain, and noisy data. A range of estimation models are being explored in the industry, as well as in academia, for research purposes but choosing the best model is quite intricate. Estimation by Analogy (EbA) is a form of case based reasoning, which uses fuzzy logic, grey system theory or machine-learning techniques, etc. for optimization. This research compares the estimation accuracy of some conventional data mining models with a hybrid model. Different data mining models are under consideration, including linear regression models like the ordinary least square and ridge regression, and nonlinear models like neural networks, support vector machines, and multivariate adaptive regression splines, etc. A precise and comprehensible predictive model based on the integration of GRA and regression has been introduced and compared. Empirical results have shown that regression when used with GRA gives outstanding results; indicating that the methodology has great potential and can be used as a candidate approach for software effort estimation.

Genetic characteristics of the Korean isolate KI -1 of Toxoplasma gondii

  • Toxoplasma gondii tachyzoites were isolated from an ocular patient in the Republic of Korea and maintained in the laboratory (designated KI-1). In the present study, its genotype was determined by analyzing dense granule antigen 6 (GRA6) gene and surface antigen 2 (SAG2) gene as typing markers. Digestion of the amplification products of GRA6 and of the 5' and 3' ends of SAG2, respectively, with Mse I, Sau3A I, and Hha I, revealed that KI-1 is included in the genotype I, which includes the worldwide virulent RH strain. In addition, when the whole sequences of the coding regions of SAG1, rhoptry antigen 1 (ROP1), and GRA8 genes of KI-1 were compared with those of RH, minor nucleotide polymorphisms and amino acid substitutions were identified. These results show that KI-1 is a new geographical strain of T. gondii that can be included in the genotype I.

Proteomic Analysis of Toxoplasma gondii KI-1 Tachyzoites

  • We studied on the proteomic characteristics of Toxoplasma gondii KI-1 tachyzoites which were originally isolated from a Korean patient, and compared with those of the well-known virulent RH strain using 2-dimensional electrophoresis (2-DE), mass spectrometry, and quantitative real-time PCR. Two-dimensional separation of the total proteins isolated from KI-1 tachyzoites revealed up to 150 spots, of which 121 were consistent with those of RH tachyzoites. Of the remaining 29 spots, 14 showed greater than 5-fold difference in density between the KI-1 and RH tachyzoites at a pH of 5.0-8.0. Among the 14 spots, 5 from the KI-1 isolate and 7 from the RH strain were identified using MALDI-TOF mass spectrometry and database searches. The spots from the KI-1 tachyzoties were dense granule proteins (GRA 2,3,6, and 7), hypoxanthine-guanine-xanthine phosphoribosyltransferase (HGRPTase), and uracil phosphoribosyltransferase (UPRTase). The spots from the RH strain were surface antigen 1 (SAG 1), L-lactate dehydrogenase (LDH), actin, chorismate synthase, peroximal catalase, hexokinase, bifunctional dihydrofolate reductase-thymidylate synthase (DHTR-TS), and nucleosidetriphosphatases (NTPases). Quantitative real-time PCR supported our mass spectrometric results by showing the elevated expression of the genes encoding GRA 2,3, and 6 and UPRTase in the KI-1 tachyzoites and those encoding GRA 7, SAG 1, NTPase, and chorismate synthase in the RH tachyzoites. These observations demonstrate that the protein compositions of KI-1 and RH tachyzoites are similar but differential protein expression is involved in virulence.

Prevalence and Genetic Characterization of Toxoplasma gondii in Pet Dogs in Central China

  • The prevalence and genotype of Toxoplasma gondii infection in dogs in Henan Province, Central China was investigated. A total of 125 blood samples were collected from pet dogs during April to June 2013, and all samples were examined by indirect hemagglutination antibody test (IHA) and nested PCR. The overall T. gondii prevalence in pet dogs was 24.0% (30/125), with 20.8% (26/125) in IHA and 10.4% (13/125) in PCR, respectively. No statistical associations were found between animal gender and age and the prevalence of T. gondii infection. Thirteen positive DNA samples were genotyped using 11 PCR-RFLP markers, including SAG1, (3'+5') SAG2, alt.SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico. Of these, only 2 samples were genotyped with complete data for all loci, and a novel genotype (type III at SAG3 and GRA6 loci, and type I at other loci) was identified. This is the first report of genetic characterization of T. gondii infection in dogs in China.

  • In this experiment well-know additives for Mn-Zn ferrites CaO and {{{{ { SiO}_{2 } }} were added at various content and their effects on microstructures and magnetic properties with sintering atmosphere were examined. By adding up to 0.04wt% {{{{ { SiO}_{2 } }} and and 0.06wt% CaO the sintered density was increased and then decreased gra-dually. Higher sintered density and initial permeability were observed by adding both CaO and {{{{ { SiO}_{2 } }} than by adding CaO or {{{{ { SiO}_{2 } }} alone. Without any additives initial permeability and tan$\delta$/${\mu}$ decreased at 1000kHz as oxidation degree(${\gamma}$) increased.

Genotyping of a Korean isolate of Toxoplasma gondii by multilocus PCR-RFLP and microsatellite analysis

  • Although the Korean isolate KI-1 of Toxoplasma gondii has been considered to be a virulent type I lineage because of its virulent clinical manifestations, its genotype is unclear. In the present study, genotyping of the KI-1 was performed by multilocus PCR-RFLP and microsatellite sequencing. For 9 genetic markers (c22-8, c29-2, L358, PK1, SAG2, SAG3, GRA6, BTUB, and Apico), the KI-1 and RH strains exhibited typical PCR-RFLP patterns identical to the type I strains. DNA sequencing of tandem repeats in 5 microsatellite markers (B17, B18, TUB2, W35, and TgM-A) of the KI-1 also revealed patterns characteristic of the type I. These results provide strong genetic evidence that KI-1 is a type I lineage of T. gondii.

Toxoplasmosis in a Pet Peach-Faced Lovebird (Agapornis roseicollis)

  • Toxoplasma gondii atypical type II genotype was diagnosed in a pet peach-faced lovebird (Agapornis roseicollis) based on histopathology, immunohistochemistry, and multilocus DNA typing. The bird presented with severe neurological signs, and hematology was suggestive of chronic granulomatous disease. Gross post-mortem examination revealed cerebral hemorrhage, splenomegaly, hepatitis, and thickening of the right ventricular free wall. Histologic sections of the most significant lesions in the brain revealed intralesional protozoan organisms associated with malacia, spongiform changes, and a mild histiocytic response, indicative of diffuse, non-suppurative encephalitis. Immunohistochemistry confirmed the causative organisms to be T. gondii. DNA isolated from the brain was used to confirm the presence of T. gondii DNA. Multilocus genotyping based on SAG1, altSAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico markers demonstrated the presence of ToxoDB PCR-RFLP genotype #3 and B1 gene as atypical T. gondii type II. The atypical type II strain has been previously documented in Australian wildlife, indicating an environmental transmission route.

Isolation and Genotyping of Toxoplasma gondii in Brazilian Dogs

  • Strains of Toxoplasma gondii in Brazil are highly genetically diverse compared to strains from North America and Europe. Dogs are epidemiologically important because they act as sentinels for T. gondii infections in humans and are good indicators of environmental contamination. The aim of this study was to isolate and genetically characterize T. gondii strains from tissues of naturally infected Brazilian dogs. For this study, 21 blood samples were collected from dogs at the Zoonosis Control Centers of $Ilh{\acute{e}}us$ and Itabuna cities, Bahia, Brazil. The sera were examined for T. gondii antibodies using the indirect hemagglutination test. Brains and hearts of seropositive dogs were bioassayed in mice to isolate and characterize T. gondii parasites by PCR-RFLP using 10 genetic markers (SAG1, newSAG2, SAG3, BTUB, c22-8, c29-2, GRA6, PK1, APICO, and L358). However, T. gondii was isolated from only 4 (57.1%) dogs, designated TgDgBr6, 13, 17, and 21. All strains were virulent, causing clinical changes (rough hair coat, lethargy, and abdominal distention) and the death of all mice within 8-20 days after inoculation. Genetic analysis of these 4 T. gondii isolates revealed 4 distinct genotypes with different clonal lineage combinations (types I, II, and III) and 2 atypical alleles. Using PCR-RFLP with several markers, this study contributes to evaluations of the genetic diversity of strains circulating in Brazil.

Isolation, characterization, and phylogenetic position of a new sulfur-oxidizing bacterium

  • A sulfer-oxidizing bacterium was isolated from mine wastewater and characterized. The isolate was gra-negative, rod (0.2 * 1.2-1.5.mu.m), nonmotiloe, catalase positive, and oxidase prositive. The opotimal pH and temperature for growth were 7.0 and 30.deg.C. respectively. The optimum thiosulfate concentration was 70 mM and the maximum growth rate was 0.081 hr. The major ubiquinone contained in the isolate was Q-8. The cellular fatty acid composition was $C_{16 : 0}$, $C_{18 : 1}$, $C_{17cyc}$,and $C_{19cyc}$ as nonpolar fatty acids, and 3-OH C10 : 0 and 3-OH $C_{12 : 0}$ as hydroxylated fatty acids. The isolate was a facultative chemolithoautotroph which can grow autotrophically on sodium thiosulfate and sodium sulfide and which can grow heterotrophically on yeast extract. It can also grow mixotrophically on sodium thiosulfate and yeast extract. Comparison of the 16S rRNA gene sequence of the isolate with that of Thiobacillus species and Paracoccus thiocyanatus revealed that it is closely related to T. caldus which belongs to the .betha.-subclass of the class Proteobacteria. However, the isolated could not grow at extremely low pH (pH 1-3.5). On the basis of the phenotypic, chemotaxonomic and phylogenetic data, the isolate was tentatively named Thiobacillus sp. strain C.ain C.
