Levels of Viral Glycoprotein Provide a Measure of Modulated Chemotherapeutic Effect

  • A chemosensitivity assay with small replicate Mm5mt/cl C3H mammary tumor cell cultures was developed to determine whether changes in viral antigen expression and release into culture fluids could be utilized as an in vitro measure of modulating drug effect. The 52,000 MW viral envelope glycoprotein (gp52) of the mouse mammary tumor virus (MMTV) was measured in culture fluids of control and drug-treated cultures while cell density was simultaneously determined by cell staining and OD 664 nm determination. While extra-cellular gp52 levels and cell density progressively increased over 72 hours for control cultures, declines in both parameters provided dual measures of effect for combination [N(phophonacetyl-L-aspartic acid)+5-fluorouracil], combination 〔N(phophonacetyl-L-aspartic acid )+5-fluoro-5'-deoxyuridine〕and single component treatment of this combination. At each treated time point, thesecombinations begin to produce a greater decline in both cell density and gp52 levels as compared to single drug treatments. These results indicate that N(phopho-nacetyl-L-aspartic acid) in combination can enhance the effectiveness of single drug.

Effects of Kaempferol, an Antioxidant, on the Bioavailability and Pharmacokinetics of Nimodipine in Rats

  • The aim of this study was to investigate the effects of kaempferol on the pharmacokinetics of nimodipine in rats. Nimodipine and kaempferol interact with cytochrome P450 (CYP) enzymes and P-glycoprotein (P-gp), and the increase in the use of health supplements may result in kaempferol being taken concomitantly with nimodipine as a combination therapy to treat orprevent cardiovascular disease. The effect of kaempferol on P-gp and CYP3A4 activity was evaluated and Pharmacokinetic parameters of nimodipine were determined in rats after an oral (12 mg/kg) and intravenous (3 mg/kg) administration of nimodipine to rats in the presence and absence of kaempferol (0.5, 2.5, and 10 mg/kg). Kaempferol inhibited CYP3A4 enzyme activity in a concentration-dependent manner with 50% inhibition concentration ($IC_{50}$) of $17.1{\mu}M$. In addition, kaempferol significantly enhanced the cellular accumulation of rhodamine-123 in MCF-7/ADR cells overexpressing P-gp. Compared to the oral control group, the area under the plasma concentration-time curve ($AUC_{0-\infty}$) and the peak plasma concentration ($C_{max}$) of nimodipine significantly increased, respectively. Consequently, the absolute bioavailability of nimodipine in the presence of kaempferol (2.5 and 10 mg/kg) was 29.1-33.3%, which was significantly enhanced compared to the oral control group (22.3%). Moreover, the relative bioavailability of nimodipine was 1.30- to 1.49-fold greater than that of the control group. The pharmacokinetics of intravenous nimodipine was not affected by kaempferol in contrast to those of oral nimodipine. Kaempferol significantly enhanced the oral bioavailability of nimodipine, which might be mainly due to inhibition of the CYP3A4-mediated metabolism of nimodipine in the small intestine and /or in the liver and to inhibition of the P-gp efflux transporter in the small intestine by kaempferol. The increase in oral bioavailability of nimodipine in the presence of kaempferol should be taken into consideration of potential drug interactions between nimodipine and kaempferol.

  • To improve the interfacial bonding of carbon fiber-nylon 6 composite, carbon fiber(CF) were oxidized by nitric acid treatment, and two types of graft polymer(GP) of nylon 6-g-polyacrylamide (PAAm) -water dispersable GP(WDGP) and m-cresol solu ble GP(CSGP) were treated as coupling agents. Introduction of polar groups such as -COOH, -OH, etc, on the surface of the oxidized CF was confirmed by IR spectra. The stem polymer of nylon 6 in the coupling agent (GP) could be compatible with'matrix nylon 5, and the grafted branch of PAAm on GP could react to the polar groups on the oxidized CF in composite. The interfacial strength was measured by the transverse tensile test to the fiber direction for single CF embedded nylon 6 film especially prepared and by the pull-out test method. The interfacial strength of the composite reinforced with oxidized CF is greater than that reinforced with unoxidized CF. The interfacial strength of the composite was increased by treatment of coupling agents(GPs) considerably, and the increasing tendency by the WDGP is greater than that by the CSGP. The optimum conditions of coupling agent treatment are as follows: the concentration, adsorption tlme of GP, and curing temperature are 2%, 20 minutes, and $170^{\circ}$, respectively.

Effect of molar ratios on strength, microstructure & embodied energy of metakaolin geopolymer

  • In this study, twenty-five geopolymer (GP) mixes were prepared by varying the alkaline solids to Metakaolin (MK) and sodium silicate to NaOH ratios from 0.1 to 0.5 and 0.2 to 1.0, respectively, thus giving a wide range of molar ratios of silica to alumina, sodium oxide to alumina and water to sodium oxide. The compressive strength of these GP mixes was determined for four curing schemes involving oven curing at 100℃ for 24 h and three ambient curing with the curing ages of 3, 14, and 28 days. The test results revealed that for the manufacture of GP binder for structural applications of strength up to 90 MPa, the molar ratio of silica to alumina should be greater than 2.3, sodium oxide to alumina should be between 0.6 to 1.2, and water to sodium oxide should not exceed 12. The compressive strength of ambient cured GP mortar gets stabilized at 28 days of ambient curing. Experimental findings were also corroborated by GP microstructure analysis. The embodied energy of MK-based GP mortars, especially of high strength, is significantly less than the cement mortar of equivalent strength.

Overexpression of a delayed early gene hlg1 of temperate mycobacteriophage L1 is lethal to both M. smegmatis and E. coli

  • Two genes of temperate mycobacteriophage L5, namely, gp63 and gp64, were hypothesized to be toxic to M. smegmatis. An identical L5 gp64 ortholog (designated hlg1) was cloned from homoimmune mycobacteriophage L1 and characterized at length here. As expected, hlg1 affected the growth of M. smegmatis when overexpressed from a resident plasmid. HLG1 (the protein encoded by hlg1) in fact caused growth retardation of M. smegmatis and the region encompassing its 57-114 C-terminal amino acid residues was found indispensable for its growthretardation activity. Both nucleic acid and protein biosynthesis were severely impaired in M. smegmatis expressing HLG1. Interestingly, HLG1 also affected E. coli almost similarly. This putative delayed early lipoprotein did not participate in the lytic growth of L1.

Quality and Sensory Characteristics of White Bread added with Various Ginseng Products (인삼제품의 첨가에 따른 제빵적성 및 관능평가)

  • This study investigated the effect of ginseng products on the baking properties of white breads. Flour was substituted by ginseng products (ginseng at levels 2, 4, 6, and 8%, all w/w, of flour levels). Both ginseng powder (GP) and red ginseng powder (RGP) were used. Similarly, red ginseng extract (RGE) was substituted at levels of 1, 2, 3, and 4% (all w/w) of flour. The physical properties of each dough were assessed using farinograms, extensograms, and amylograms. Water absorption increased as the proportions of ginseng products rose. The dough surface areas and R/E (resistance/extensibility) values decreased, as did peak viscosities, at the proportions of ginseng products increased. The pH values during fermentation decreased as the proportions of ginseng products increased. The fermentation power of dough with GP was lower than that of control, and higher than that of dough with RGP or RGE, but the addition of ginseng products beyond certain levels weakened the gas retention power: The specific loaf volumes of breads with 2% GP were the highest at 5.41 mL/g. In breads with RGE, the specificloaf volume increased from 5.52 mL/g to 5.82 mL/g as RGE levels rose from 0% to 4%. Hardness increased with rising GP and RGP levels in breads with GP and RGP, but decreased with RGE levels in breads with RGE. The moisture contents of breads during storage tended to be higher than control in breads with ginseng products. Lightness increased with addition of GP and decreased upon addition of RGP or RGE, while redness and yellowness increased after addition of any ginseng products. In sensory evaluation tests, the sensory scores for texture, color, mouth feel, and overall acceptability, were high, when any ginseng product (GP, RGP or RGE) was present at 2%. Of these breads, the bread with 2% RGE attained the highest sensory score.

Complete genome sequence of Microbulbifer agarilyticus GP101 possessing genes coding for diverse polysaccharide-degrading enzymes (다양한 다당류를 분해하는 세균 Microbulbifer agarilyticus GP101의 완전한 유전체 서열)

  • Microbulbifer agarilyticus GP101 was isolated from the gut of a marine invertebrate Turbo cornutus and capable of degrading polysaccharide such as agar, alginate, and ${\kappa}$-carrageenan constituting algal cell wall. To obtain genomic basis of polysaccharide-degrading activity, we sequenced genome of strain GP101. The genome consists of 4,255,625 bp, 3,458 coding sequences with 55.4% G + C contents. BLASTP search revealed the presence of seven agarases, five alginate lyases, ten glucanases, four chitinases, two xylanases, one ${\kappa}$-carrageenase, and one laminarinase. The genomic data of strain GP101 will provide potential uses in the bioconversion process of diverse polysaccharide into bioenergy and biochemicals.

Optimization of field Application Conditions of the Multistage Convergent Photographing Technique for the Measurement of Joint Orientation on Rock Slope (암반사면 절리의 방향성 측정을 위한 수렴다중촬영기법의 현장 적용성 연구)

  • One set of hardware system of guide point method (GP method), modified from the multistage convergent photographing technique, was developed to interpret the geometrical characteristics of Joints photogrammetrically on rock slope. Before the field application of the hardware system, the level of errors and constraints that ran be acceptable in the field measurement has been severely investigated in the laboratory and the optimum photographing scheme was analyzed. The range of the most suitable convergence angle between two cameras was $25^{\circ}{\sim}150^{\circ}$ and the photographing distance was about 5.5 m when using a 2 M pixel digital camera. An extended analyzing technique, which was newly developed in this study, was applied to the field measurement to magnify the benefits of GP method. This technique can be applied when survey for the wide range of rock surface is necessary. The global coordinates of ground control points for the neighbor photographing area ran be introduced without any preparation from the previous photographed area using this technique. It could reduce phographing time in the field.

Effects of Adamantyl Derivatives on Pharmacokinetic Behavior of Paclitaxel in Rats

  • Paclitaxel (PTX) is one of the most frequently used anticancer agent for treating refractory ovarian cancer, metastatic breast cancer and non-small cell lung cancer. However, its oral administration is impeded by very low bioavailability (<5%) due to the P-glycopprotein (P-gp) efflux pump effect. This study investigated in vitro and in vivo P-gp inhibitory effects of adamantyl derivatives AC-603 and AC-786 in rats. Two adamantyl derivatives tested in this study increased the cytotoxicity of daunomycin (DNM) in P-gp overexpressed cell line by inhibiting P-gp efflux function. Pharmacokinetics of PTX with orally co-administered P-gp inhibitors were assessed in rats to improve PTX absorption. The pharmacokinetic parameters of PTX were determined in rats after intravenous (2 mg/kg) or oral (25 mg/kg) administration in the presence or absence of verapamil (a positive control), AC-603 or AC-786 (0.5 mg/kg or 5 mg/kg). Compared to control group (PTX alone), experimental groups (PTX with AC-603 or AC-786) significantly increased the area under the plasma concentration-time curve of PTX following oral administration by 1.7-2.2 fold. The volume of distribution and total clearance of PTX were decreased, while other parameters were not significantly changed. In conclusion, co-administration of AC-603 or AC-786 enhanced the relative bioavailability of orally administered PTX as compared to control.

Leaf Damage Symptom of Grape (Vitis vinifera) by Tea Bagworm (Eumeta minuscula) and Control Efficacies of Several Insecticides against the Eumeta minuscula (차주머니나방(Eumeta minuscula)에 의한 포도 잎 피해증상 및 몇 가지 약제의 차주머니방에 대한 방제효과)

  • Damage of tea bagworms, Eumeta minuscula was observed for the first time from grape leaves of grape (varieties: Cambell Early and M.B.A.) from a vinery at Gumho-up, Gyeongsan, Gyeongbuk on 31 August, 2012. Activity of insecticides [Bacillus thuringiensis var. kurstaki (serotype IIIa, IIIb) WP, chlorantraniliprole 5% GP, methoxyfenozide 21% SC, spinetoram 5% GP, and spinosad 10% GP] was tested on fourth instar larvae of Eumeta minuscula in laboratory. Mortality of 4th instar of Eumeta minuscula was higher when treated with spinosad 10% WG (88.7%) and spinetoram 5% WG (67.7%) sprayed with field recommended concentration using a home sprayer in laboratory. Mortality decreased as the concentration decreased, however leaf damage rate was significantly different between the one-fourth rate and the control. Spinetoram 5% WG and spinosad 10% WG showed 63.7 and 55.7% respectively mortality of fourth instar Eumeta minuscula in field. Though these two insecticides could be used for the control of bagworms on grape, a more effective insecticide needs to be found.
