Anti-growth Effects of Imatinib and GNF5 via Regulation of Skp2 in Human Hepatocellular Carcinoma Cells

  • Background: Human hepatocellular carcinoma (HCC) is a common liver tumor and the main cause of cancer-related death. Tyrosine kinase inhibitors, such as imatinib and GNF5 which were developed to treat chronic myelogenous leukemia, regulate the progression of various cancers. The aim of this study was to confirm the anti-tumor activity of tyrosine kinase inhibitors through regulation of S-phase kinase-associated protein 2 (Skp2), an important oncogenic factor in various cancer cells, in human hepatocarcinoma SK-HEP1 cells. Methods: Cell viability and colony formation assays were conducted to evaluate the effects of imatinib, GNF5 and GNF2 on the growth of SK-HEP1 cells. Using immunoblot analysis, we assessed change of the activation of caspases, PARP, Akt, mitogen-activated protein kinases, and Skp2/p27/p21 pathway by imatinib and GNF5 in SK-HEP1 cells. Using sh-Skp2 HCC cells, the role of Skp2 in the effects of imatinib and GNF5 was evaluated. Results: Imatinib and GNF5 significantly inhibited the growth of SK-HEP1 cells. Treatment of imatinib and GNF5 decreased Skp2 expression and Akt phosphorylation, and increased the expression of p27, p21, and active-caspases in SK-HEP1 cells. In sh-Skp2 HCC cells, cell growth and the expression of Skp2 were inhibited by more than in the mock group treated with imatinib and GNF5. Conclusions: These results suggest that the anti-growth activity of tyrosine kinase inhibitors may be associated with the regulation of p27/p21 and caspases through Skp2 blockage in HCC cells.

Chemical kinomics: a powerful strategy for target deconvolution

  • Kinomics is an emerging and promising approach for deciphering kinomes. Chemical kinomics is a discipline of chemical genomics that is also referred to as "chemogenomics", which is derived from chemistry and biology. Chemical kinomics has become a powerful approach to decipher complicated phosphorylation-based cellular signaling networks with the aid of small molecules that modulate kinase functions. Moreover, chemical kinomics has played a pivotal role in the field of kinase drug discovery as it enables identification of new molecular targets of small molecule kinase modulators and/or exploitation of novel functions of known kinases and has also provided novel chemical entities as hit/lead compounds. In this short review, contemporary chemical kinomics technologies such as activity-based protein profiling, T7 kinasetagged phages, kinobeads, three-hybrid systems, fluorescenttagged kinase binding assays, and chemical genomic profiling are discussed along with a novel allosteric Bcr-Abl kinase inhibitor (GNF-2/GNF-5) as a successful application of chemical kinomics approaches.

카본 나노파이버가 도핑된 리튬이온전지의 전기화학적 특성 (Electrochemical Characteristics of Lithium-ion Battery with Doped Graphite Nanofiber)

  • 올리빈 구조의 $LiFePO_4$ 정극 활물질은 $650^{\circ}C$에서 고상법으로 제조되었다. $LiFePO_4$의 전자전도도를 향상시키기 위하여 graphite nanofiber(GNF)를 각각 3wt%, 5wt%, 7wt%, 9wt% 첨가하여 $LiFePO_4$-C를 제조하였다. 제조된 분말의 입자 형태를 확인하기 위하여 X-ray diffraction(XRD)과 File Electronic Scaning Electromicroscopy(FE-SEM)를 측정하였다. XRD결과로부터 제조된 분말은 모두 순수한 결정 구조를 나타내었고 입자의 크기는 약 200nm였다. 5wt% GNF를 첨가한 $LiFePO_4$-C는 기타 첨가량에 비해 방전용량이 가장 높았다. 첫 사이클의 용량은 151.73mAh/g 나타났고 50 사이클 뒤에도 92% 이상을 유지하고 있었다. 첨가하지 않은 것에 비해 43% 증가하였다. $LiFePO_4$-C(3wt%), $LiFePO_4$-C(7wt%), $LiFePO_4$-C(9wt%)의 첫 사이클 방전용량은 각각 147.94mAh/g, 136.64mAh/g, 121.07mAh/g 나타났다. $LiFePO_4$-C(5wt%)에 비해 용량은 떨어쪘지만 순수한 $LiFePO_4$보다 많이 높았다. 임피던스 결과를 보면 기타 첨가량에 비해 $LiFePO_4$-C(5wt%)의 저항 제일 낮았다. 이는 충방전 결과와 일치하였다. graphite nanofiber의 첨가로 인하여 $LiFePO_4$ 정극 활물질의 전자전도도가 높아지고, 따라서 전기화학적 특성도 크게 향상되었다.

KOH-activated graphite nanofibers as CO2 adsorbents

  • Porous carbons have attracted much attention for their novel application in gas storage. In this study, porous graphite nano-fiber (PGNFs)-based graphite nano fibers (GNFs) were prepared by KOH activation to act as adsorbents. The GNFs were activated with KOH by changing the GNF/KOH weight ratio from 0 through 5 at 900℃. The effects of the GNF/KOH weight ratios on the pore structures were also addressed with scanning electron microscope and N2 adsorption/desorption measurements. We found that the activated GNFs exhibited a gradual increase of CO2 adsorption capacity at CK-3 and then decreased to CK-5, as determined by CO2 adsorption isotherms. CK-3 had the narrowest micropore size distribution (0.6–0.78 nm) among the treated GNFs. Therefore, KOH activation was not only a significant method for developing a suitable pore-size distribution for gas adsorption, but also increased CO2 adsorption capacity as well. The study indicated that the sample prepared with a weight ratio of ‘3’ showed the best CO2 adsorption capacity (70.8 mg/g) as determined by CO2 adsorption isotherms at 298 K and 1 bar.

  • RSA 암호 시스템은 가장 널리 사용되는 공개키 암호 알고리즘 중 하나이며, RSA 암호 시스템의 안전성은 큰 수의 인수분해의 어려움에 기반을 둔다. 따라서 RSA 암호 시스템의 합성수 n을 인수분해하려는 시도는 계속 진행 중에 있다. General Number Field Sieve는 현재까지 알려진 가장 빠른 인수분해 방법이고, RSA-704를 인수분해 하는데 사용된 소프트웨어인 CADO-NFS도 GNFS를 기반으로 설계되어 있다. 그러나 CADO-NFS는 다항식 선택 과정에서 입력된 변수로부터 항상 최적의 다항식을 선택하지 못하는 문제점이 있다. 본 논문에서는 CADO-NFS의 다항식 선택 단계를 분석하고 중국인의 나머지 정리와 유클리드 거리를 사용하여 다항식을 선택하는 방법을 제안한다. 제안된 방법을 이용하면 기존의 방법보다 좋은 다항식이 매번 선택되며, RSA-1024를 인수분해 하는데 적용할 수 있을 것으로 기대한다.

Electro-catalytic Performance of PtRu Catalysts Supported on Urea-treated MWNTs for Methanol Oxidation

  • In this work, nitrogen and oxygen functionalities was introduced to the graphite nanofibers (GNFs) and their effect on electrocatalytic performance of the GNF supports for direct methanol fuel cells (DMFCs) was invesigated. The nitrogen and oxygen groups were introduced through the urea treatments and acid treatment, respectively. And, PtRu catalysts deposited on modified GNFs were prepared by a chemical reduction method. The catalysts were characterized by means of elemental analysis, nitrogen adsorption, and X-ray photoelectron spetroscopy (XPS). The structure and morphological characteristics of the catalysts were characterized by X-ray diffraction (XRD) and transmission electron microscopy (TEM). As a result, the Pt-Ru nanoparticles were impregnated on GNFs with good formation in 3-5 nm. And, the cyclic voltammograms for methanol oxidation revealed that the methanol oxidation peak varied depending on changes of surface functional groups. It was thus considered that the PtRu deposition was related to the reduction of PtRu and surface characteristics of the carbon supports. The changes of surface functional groups were related to PtRu reduction, significantly affect the methanol oxidation activity of anode electrocatalysts in DMFCs.

Towards a hierarchical global naming framework in network virtualization

  • Network virtualization enables autonomous and heterogeneous Virtual Networks (VNs) to co-exist on a shared physical substrate. In a Network Virtualization Environment (NVE), the fact that individual VNs are underpinned by diverse naming mechanisms brings about an obvious challenge for transparent communication across multiple VN domains due to the complexity of uniquely identifying users. Existing solutions were mainly proposed compatible to Internet paradigm with little consideration of their applications in a virtualized environment. This calls for a scalable and efficient naming framework to enable consistent communication across a large user population (fixed or mobile) hosted by multiple VNs. This paper highlights the underlying technical requirements and presents a scalable Global Naming Framework (GNF), which (1) enables transparent communication across multiple VNs owned by the same or different SPs; (2) supports communication in the presence of dynamics induced from both VN and end users; and (3) greatly reduces the network operational complexity (space and time). The suggested approach is assessed through extensive simulation experiments for a range of network scenarios. The numerical result clearly verifies its effectiveness and scalability which enables its application in a large-scale NVE without significant deployment and management hurdles.
