Microtubule Acetylation-Specific Inhibitors Induce Cell Death and Mitotic Arrest via JNK/AP-1 Activation in Triple-Negative Breast Cancer Cells

  • Microtubule acetylation has been proposed as a marker of highly heterogeneous and aggressive triple-negative breast cancer (TNBC). The novel microtubule acetylation inhibitors GM-90257 and GM-90631 (GM compounds) cause TNBC cancer cell death but the underlying mechanisms are currently unknown. In this study, we demonstrated that GM compounds function as anti-TNBC agents through activation of the JNK/AP-1 pathway. RNA-seq and biochemical analyses of GM compound-treated cells revealed that c-Jun N-terminal kinase (JNK) and members of its downstream signaling pathway are potential targets for GM compounds. Mechanistically, JNK activation by GM compounds induced an increase in c-Jun phosphorylation and c-Fos protein levels, thereby activating the activator protein-1 (AP-1) transcription factor. Notably, direct suppression of JNK with a pharmacological inhibitor alleviated Bcl2 reduction and cell death caused by GM compounds. TNBC cell death and mitotic arrest were induced by GM compounds through AP-1 activation in vitro. These results were reproduced in vivo, validating the significance of microtubule acetylation/JNK/AP-1 axis activation in the anti-cancer activity of GM compounds. Moreover, GM compounds significantly attenuated tumor growth, metastasis, and cancer-related death in mice, demonstrating strong potential as therapeutic agents for TNBC.

효소면역측정법을 이용한 두부 중의 유전자 재조합 대두단백질 분석 (Quantification of Genetically Modified Soy Proteins in Fresh Soybean Curd by Antigen-coated Plate ELISA)

  • 본 연구에서는 유전자 재조합 되지 않은(non-GM) 대두와 유전자 재조합된(GM) 대두가 혼입되어 제조된 두부에서 효소 면역 측정법을 이용하여 non-GM 대두의 혼입량을 추정하고자 하였다. 두부의 SDS-PAGE 실행 결과 non-GM 두부에서만 나타나는 특이 단백질 non-GM 113kDa 밴드와 non-GM과 GM 두부에서 모두 나타나는 non-GM 24kDa 밴드를 선별하고 이들을 토끼에 면역하여 항체생성 여부를 ELISA한 결과 non-GM 113kDa과 non-GM 24kDa 단백질 모두 항체가 형성됨을 확인하였고 $10^{-1}-10^{-6}$의 단백질 희석배수에서 두부를 이들 항체에 대하여 ELISA함으로써 원료대두의 GM여부를 확인할 수 있었다. 이들 중, 보다 감도가 높았던 non-GM 113kDa 단백질을 $10^{-7}-10^{-6}$의 배수로 희석하여 ELISA 흡광도와 non-GM 단백질의 관계를 나타내는 표준곡선을 작성하였고, 임의로 non-GM 대두와 GM 대두를 혼합하여 제조한 두부의 ELISA 흡광도를 이 표준곡선과 비교하여 non-GM 원료와 GM 원료 작물의 혼입율을 측정한 결과, 높은 정확도를 보였다.

Preliminary Studies on the Effects of Dietary Genetically Modified Soya and Corn on Growth Performance and Body Composition of Juvenile Olive Flounder(Paralichthys olivaceus) and Rockfish (Sebastes schlegeli)

  • Two feeding experiments were conducted to investigate the effects of dietary genetically modified (GM) soya and com on growth performance, feed utilization and body composition of juvenile olive flounder, Paralichthys olivaceus and rockfish, Sebastes schlegelii. For each fish species, four isonitrogenous (50% crude protein) and isocaloric (4.1 kcal/g) diets (designated as nGM soya, GM soya, nGM com and GM com) were formulated to contain 20% non-GM (nGM) and GM soya and com. Thirty olive flounder (initial body weight, 15.4${\pm}$0.4 g) and fifty rockfish (initial body weight, 3.1${\pm}$0.02 g) were distributed in each 400 L tank (200 L water) in a flow through system. Each experimental diet was fed to triplicate groups of fish to visual satiation, twice a day (9:00 hand 17:00 h) for 6 weeks. Growth performance was measured every three weeks. No effects of GM feedstuffs on survival were observed. Dietary inclusion of GM feedstuffs did not affect growth performance and feed utilization of fishes, except for rockfish fed GM com. Rockfish fed the GM com diet showed higher weight gain, daily feed intake and daily protein intake than did fish fed the nGM com diet, but no significant differences were observed in final body weight between the dietary treatments. Condition factor, hepatosomatic index, visceral somatic index and body composition were not altered by the inclusion of GM feedstuffs. These results indicate that dietary inclusion of GM soya and com could have no effects on growth performance and feed utilization of juvenile flounder and rockfish. Lower weight gain and feed intake in flounder and rockfish fed the diets containing 20% soya were likely due to anti-nutritional factors, rather than transgenic factors in the feedstuffs. Dietary inclusion of GM soya and com at the level tested did not alter the body composition of fishes. Further studies to investigate the effects of GM feedstuffs on health conditions and the development of fishes, as well as those of residue of transgenic fragments in ambient environments and in animals are necessary for safe use of the ingredients in aquaculture.

해충저항성 GM감자와 non-GM감자의 Housekeeping gene 발현 분석 (Analysis of Housekeeping Genes in Mice Feeding on GM and non-GM Potatoes)

  • GM 및 non-GM 감자의 인체 위해성 여부를 판단하기 위하여 마우스 비장세포에서 인터루킨과 housekeeping gene의 발현을 RT-PCR로 비교 분석하였다. 유전자변형 감자의 잎과 이를 섭취한 마우스의 비장세포에서 발현되는 housekeeping gene의 발현을 수행한 결과, APRT, $\beta$-tubulin, Actin, Hsp 20.2, Cyclophilin, 18S RNA and Efla, and Tbp, GAPDH, $\beta$-actin, Tuba2, Hprt, Cyclophlin A, Tfrc, and RPL13A의 발현에는 유전자 변형 감자와 그렇지 않은 감자와의 차이를 발견할 수 없었다. GM작물의 안전성에 대한 연구는 많은 표본을 가지고, 보다 장기적인 관점에서 다양한 모니터링 시스템을 도입이 필요하다고 판단되며 이의 향후 연구에 많은 관심이 필요하다.

선박의 운항 상태를 고려한 선종별 안정적인 GM 운용에 관한 연구 (A Study on Securing a Stable GM for Each Ship Type Considering the Ship's Operating Status)

  • 최근 선박의 전복사고는 무게중심의 부적절한 관리, 기상악화 또는 과도한 조타사용 등으로 인한 선체운동이 발생하면서 화물 또는 중량물이 이동하여 복원력 저하 또는 상실 등이 주된 원인으로 분석되고 있다. 따라서 전복사고를 예방하기 위해서는 위험한 횡경사가 발생하더라도 직립 상태로 되돌아오려는 복원성 확보가 이루어져야 한다. GM은 복원성을 판단하는 중요한 평가요소이며, 항해사는 GM을 복원성 검토에 좋은 척도로 활용한다. 본 연구에서는 5년간 운항 중인 선박으로부터 수집한 복원성 자료를 바탕으로 운항 상태에 따른 선종별 GM을 선박 길이, 폭, 총톤수 등 선박제원으로 분석하였다. 선박 길이에 따른 GM 분포의 특징을 확인하였고, 선박 폭과 GM의 상관분석을 수행한 후 선폭대비 GM의 비율을 산출한 후 과거의 비율과 비교하여 최근 경향을 검토하였다. 또한 선박 대형화 추세를 반영하기 위하여 총톤수(GT)/선폭(B)을 이용하여 GM을 회귀분석하여 선종별 GM 추정을 위한 간이근사식 및 최소 GM을 제안하였다. 연구의 결과는 선박에서 항해사가 안정적인 GM 확보의 검토를 위한 자료로 이용되길 기대된다.

Comparison of myofibrillar protein degradation, antioxidant profile, fatty acids, metmyoglobin reducing activity, physicochemical properties and sensory attributes of gluteus medius and infraspinatus muscles in goats

  • Background: The functionality of myofibrillar proteins is a major factor influencing the quality attributes of muscle foods. Nonetheless, the relationships between muscle type and oxidative changes in chevon during ageing are meagrely elucidated. Postmortem changes in antioxidant status and physicochemical properties of glycolytic gluteus medius (GM) and oxidative infraspinatus (IS) muscles in goats were compared. Methods: Twenty Boer bucks (9-10 months old, body weight of $36.9{\pm}0.725kg$) were slaughtered and the carcasses were subjected to chill storage ($4{\pm}0.5^{\circ}C$). Analyses were conducted on GM and IS muscles sampled on 0, 1, 4 and 7 d postmortem. Results: Chill storage did not affect the antioxidant enzyme activities in both muscles. The IS had greater (P < 0.05) superoxide dismutase and catalase activities than GM. Carotenoid and tocopherol contents did not differ between muscles but decreased (P < 0.05) over storage. The IS had higher (P < 0.05) glycogen and ultimate pH and lower (P < 0.05) shear force and cooking loss than GM. The carbonyl content, % metmyoglobin, drip loss and TBARS increased (P <0.05) while free thiol, metmyoglobin reducing activity (MRA), shear force and myoglobin decreased (P < 0.05) over storage. Muscle type had no effect (P > 0.05) on free thiol, MRA and TBARS. The GM had lower (P < 0.05) redness on d 0 and 1 than IS while the IS had greater carbonyl, % metmyoglobin and drip loss than GM on d 7. The reflective density of slow myosin heavy chain (MHC) was higher (P < 0.05) while the density of fast MHC and actin was lower (P < 0.05) in IS than GM. Regardless of muscle type, the density of MHC decreased (P < 0.05) while that of actin was stable over storage. Nonetheless, the degradation of fast and slow MHC was greater (P < 0.05) in IS than GM. Muscle type had no effect (P > 0.05) on consumer preference for flavour, juiciness and overall acceptability. However, IS had higher (P < 0.05) tenderness score than GM on d 1 and 4 postmortem. Intramuscular fat was higher (P< 0.05) in IS compared with GM. Fatty acid composition did not differ between the muscles. However, GM had lower (P < 0.05) n-6/n-3 ratio than IS. The n-3 and n-6 PUFA declined (P < 0.05) while the SFA increased (P < 0.05) over storage. Conclusion: The changes in myofibrillar proteins and physicochemical properties of goat meat during postmortem chill storage are muscle-dependent.

Determinants for the Social Acceptance of New Emerging Science and Technology: The Case of Genetically Modified Foods

  • This study identifies the structural determinants of the social acceptance of genetically modified (GM) foods across European countries. Toward this end, we suggest an integrated theoretical model to explain the social acceptance of GM foods by including both perception factors (perceived benefit, perceived risk, feelings, trust, and knowledge) and value factors (ethical concerns, science optimism, religiosity, and ideology). This model is then tested by analyzing survey data collected from 18,634 Europeans in 32 countries. The results indicate that first, not only perception factors but also value factors significantly contribute to explaining the acceptance of GM foods. Second, perceived benefits, perceived risk, feelings, and ethical concerns tend to be the four biggest determinants for acceptance. Third, this two-factor model could be generalized even with variation across countries. Finally, ethical concerns and scientific optimism play a moderating role between predictors and outcomes in the acceptance of GM foods.

효소면역측정법을 이용한 유전자재조합 콩과 콩나물의 분석 (Analysis of Genetically-Modified Soybean and Soybean Sprout by Enzyme-Linked Immunosorbent Assay (ELISA))

  • CP4 EPSPS에 대한 특이항체(다클론 및 단클론)을 이용하여 확립한 전보의 sandwich ELISA 분석법을 원료 콩의 GMO 분석에 적용가능한 가를 검토하였다. 원료 콩은 수입산 47점, 국산 20점을 분석하였다. 그 결과, 수입산 콩의 CP4 EPSPS 분석치는 크게 두 군으로 나뉘어 높은 군$(39.1{\pm}13.5{\mu}g/g,\;n=33)$과 낮은 군$(2.6{\pm}1.2{\mu}g/g,\;n=14)$으로 나타났다. 수입산 콩의 GMO 비율은 약 70.2%로 나타났다. 한편, 국산 콩의 CP4 EPSPS의 함량 $(0.9{\pm}0.5{\mu}g/g,\;n=20)$은 아주 낮은 것으로 나타났고 이는 non-GMO인 것으로 판단되었다. 콩나물의 경우에 GM과 non-GM 간 CP4 EPSPS의 차이를 보이고 있었고 GM 콩나물의 자엽부에서 뿌리보다 높은 CP4 EPSPS의 함량을 보여주고 있다. 국산 콩으로 만든 콩나물의 경우 CP4 EPSPS의 함량은 아주 낮은 것으로 나타나 GM과 non-GM 콩간의 구분이 가능하였다. 또한 PCR에 의해 sandwich ELISA가 정확하다는 것을 보여주었다. 이상과 같이 sandwich ELISA 방법은 신속, 간편하게 많은 콩 시료의 GMO 여부를 판단할 수 있음을 보여주었다.

Path Space Approach for Planning 2D Shortest Path Based on Elliptic Workspace Geometry Mapping

  • A new algorithm for planning a collision-free path based on algebraic curve is developed and the concept of collision-free Path Space (PS) is introduced. This paper presents a Geometry Mapping (GM) based on two straight curves in which the intermediate connection point is organized in elliptic locus ($\delta$, $\theta$). The GM produces two-dimensional PS that is used to create the shortest collision-free path. The elliptic locus of intermediate connection point has a special property in that the total distance between the focus points through a point on ellipse is the same regardless of the location of the intermediate connection point on the ellipse. Since the radial distance, a, represents the total length of the path, the collision-free path can be found as the GM proceeds from $\delta$=0 (the direct path) to $\delta$=$\delta$$\_$max/(the longest path) resulting in the minimum time search. The GM of elliptic workspace (EWS) requires calculation of interference in circumferential direction only. The procedure for GM includes categorization of obstacles to .educe necessary calculation. A GM based on rectangular workspace (RWS) using Cartesian coordinate is also considered to show yet another possible GM. The transformations of PS among Circular Workspace Geometry Mapping (CWS GM) , Elliptic Workspace Geometry Mapping (EWS GM) , and Rectangular Workspace Geometry Mapping (RWS GM), are also considered. The simulations for the EWS GM on various computer systems are carried out to measure performance of algorithm and the results are presented.
