Enterobacter intermedium 60- 2G의 유기산 생성과 불용성인의 가용화 (Organic acid production and phosphate solubilization by Enterobacter intermedium 60-2G)

  • 강한 인산 가용력을 가진 인산 용해 세균인 균주 60-2G를 잔디의 근권에서 분리하였다. GC-FAME구조와 탄소이용형태 및 16S rRNA의 부분 염기서열 분석을 통해 균주 60-2G는 Enterobacter intermedium으로 동정되었다. Hydroxyapatite를 첨가한 배지와 생장 시킨 균주 60-2G는 gluconic acid 와 2-ketogluconic acid 및 소량의 lactic acid를 생성하였다. 균주 60-2G의 생장 기간동안 배지의 pH는 3.8 까지 낮아지는 반면에 배지의 유효 인산 농도는 증가하였다. 배지의 낮은 pH와 유효인산농도의 증가는 역 상관관계이며, 이는 균주 60-2G가 생성하는 유기산에 의한 영향이다. E. intermedium 60-2G 균주는 유기산 생성에 관여하는 glucose dehydrogenase의 co-factor인 PQQ를 생성하였으며, pqq의 부분 염기서열 분석 결과 기존에 보고된 서열과 85% 이상의 상동성을 가지고 있었다.

Obesity-Associated Metabolic Signatures Correlate to Clinical and Inflammatory Profiles of Asthma: A Pilot Study

  • Purpose: Obesity is associated with metabolic dysregulation, but the underlying metabolic signatures involving clinical and inflammatory profiles of obese asthma are largely unexplored. We aimed at identifying the metabolic signatures of obese asthma. Methods: Eligible subjects with obese (n = 11) and lean (n = 22) asthma underwent body composition and clinical assessment, sputum induction, and blood sampling. Sputum supernatant was assessed for interleukin $(IL)-1{\beta}$, -4, -5, -6, -13, and tumor necrosis factor $(TNF)-{\alpha}$, and serum was detected for leptin, adiponectin and C-reactive protein. Untargeted gas chromatography time-of-flight mass spectrometry (GC-TOF-MS)-based metabolic profiles in sputum, serum and peripheral blood monocular cells (PBMCs) were analyzed by orthogonal projections to latent structures-discriminate analysis (OPLS-DA) and pathway topology enrichment analysis. The differential metabolites were further validated by correlation analysis with body composition, and clinical and inflammatory profiles. Results: Body composition, asthma control, and the levels of $IL-1{\beta}$, -4, -13, leptin and adiponectin in obese asthmatics were significantly different from those in lean asthmatics. OPLS-DA analysis revealed 28 differential metabolites that distinguished obese from lean asthmatic subjects. The validation analysis identified 18 potential metabolic signatures (11 in sputum, 4 in serum and 2 in PBMCs) of obese asthmatics. Pathway topology enrichment analysis revealed that cyanoamino acid metabolism, caffeine metabolism, alanine, aspartate and glutamate metabolism, phenylalanine, tyrosine and tryptophan biosynthesis, pentose phosphate pathway in sputum, and glyoxylate and dicarboxylate metabolism, glycerolipid metabolism and pentose phosphate pathway in serum are suggested to be significant pathways related to obese asthma. Conclusions: GC-TOF-MS-based metabolomics indicates obese asthma is characterized by a metabolic profile different from lean asthma. The potential metabolic signatures indicated novel immune-metabolic mechanisms in obese asthma with providing more phenotypic and therapeutic implications, which needs further replication and validation.

건조조건에 따른 고추의 색도와 향기 성분의 변화 (Effect of Drying Condition on the Colors and Flavors Change of Fresh pepper)

  • 고추의 품질향상과 품질평가척도 등을 제시하는 효율적인 건조조건을 구명(究明)하기 위하여 건조온도(40, 50, 70, $90^{\circ}C$)와 건조시간(12, 24, 36, 48시간)을 달리하여 고추를 건조한 다음, 건조조건별로 고추의 수분함량변화, 색도 및 향기 성분을 조사하였다. 1) 고추품질면에서 최적수분함량($11.85{\sim}11.05%$)을 고려할 때 $5^{\circ}C$에서 36시간 또는 48시간 건조하는 것이 양호한 것으로 나타났다. 2) 태양건조와 열풍건조를 비교한 결과 열풍으로 $50^{\circ}C$에서 48시간 건조하는 것이 고추의 색도가 가장 양호하였다. 3) 고추의 색도 특성은 hexane 추출물에서 284nm와 carotenoids($430{\sim}474nm$)의 전형적인 흡광도 값의 비가 $2.5{\sim}3.4$범위에 있는 것이 가장 좋았으며, 이것은 육안판정에서도 확인되었다. 4) 고추의 향기성분 분석에서 hexane 추출물이 benzene추출시보다 다양한 peak를 보였으며 BSTFA으로 silylatin하여 고추의 향기성분 조성 profile을 얻었으며 이 profile은 고추의 향기성분 및 정미성분의 품질척도로 사용할 수 있을 것으로 판단된다. 5) 고추의 향기성분으로 benzene dicarboxylic acid의 7종을 확인하였으며, 주요성분으로는 oleic acid와 palmitic acid이다. 6) 건조온도와 건조시간에 따라서 향기성분의 특징적인 성분차이는 발견할 수 없었으나 각 성분의 조성비율면에서는 차이가 있었다.

Streptomyces lividans Tk24에서 secY homolog의 클로닝과 분석 (Molecular Cloning and Characterization of the secY Homolog from Streptomyces lividans TK24)

  • 몇 가지 그람양성 세균에서 secY 유전자가 함유된 operon의 구성이 rplO(L15)-secY-adk라는 결과를 토대로 L15와 adk 유전자일부를 primer로 제작하여 Streptomyces lividans TK24에서 secY 유전자가 함유된 1.8kb 단편을 PCR로 증폭하여 얻은 후 secY 유전자를 cloning하였다. 전 단편을 sequencing하여 추론한 아미노산으로 상동성을 조사해 본 결과 Escherichia coli, Bacillus subtilis, Micrococcus luteus, Bacillus licheniformis, Staphylococcus carnosus, Brevibacterium flavum, Streptomyces scabies의 SecY와 각각 46%, 43%, 57%, 44%, 42%, 56%, 90%의 유사성을 보이는 것으로 나타났으며 SecY의 소수성 profile 또한 서로 유사하고 10개의 membrane spanning segment를 동일하게 가지는 것으로 나타났다. 유전자 구조도 다른 그람양성균에서와 같이 L15-SecY-Adk순 이였으며 secY 유전자의 종결코돈과 adk 유전자의 개시코돈이 한 염기를 공유하는 상태의 translational coupling 구조를 하고 있었다. 이와 같이 단백질분비에 관여하는 유전자와 ribosome 구성요소, 그리고 ATP 합성에 관여하는 요소는 세포성장에 상호 연관 작용이 있는 것으로 사료된다.

Fatty Acid Composition of Adipose Tissues in Obese Mice and SD Rats Fed with Isaria sinclairii Powder

  • Isaria sinclairii (Cicada Dongchunghacho) was studied as a potential crude natural food in powdered form. The role of tissue fatty acids in relation to the anti-obesity effects of I. sinclairii (IS) was examined by feeding the powder to SD rats ad libitum at 0, 1.25, 2.5, 5 and 10% (calculated about 8 g/kg) of the feed for a period of 3 months and 6 months. The fatty acid composition profile as indicated GC-MS, showed significantly slight dose-dependent increases in the levels of unsaturated fatty acids, particularly, arachidonic acid (C20: 4n6), oleic acid, linoleic acid, eicosadienoic acid, eicosapentaenoic acid (EPA) (C20: 5) concentration in the the ad libitum IS-fed groups compared to the control group in SD abdominal fat over 6 month period. Over viewing of the SD and Ob mice treated Isaria sinclairii powder; there were increases in the single (mono) unsaturated fatty acids ratio but decreases in polyunsaturated fatty acid. In IS-fed groups in proportion to the treatment period, this Dongchunghacho also induced an increase in the level of same result of unsaturated fatty acid in C57BL/6 obese (ob/ob) mice over a 6-month period treatment compared to those given 10% dry mulberry leaf powder (ML) or silkworm powder mixed with the standard diet.

Phytase를 생산하는 Enterobacter cloacae의 분리 및 효소 생산의 배지 최적화 (Isolation of Enterobacter Cloacae Producing Phytase and Medium Optimization of Its Production)

  • 본 연구는 phytic acid를 myo-inositol과 무기태인으로 분해시키는 효소인 phytase 생산균주의 분리 및 효소생산의 배지최적하에 관한 것이다. 1차로 calcium phytate를 기질로 함유한 phytase screening 배지를 이용하여 phytase 생산을 나타내는 균주 35가지를 이용하여 분리한 후, sodium phytate를 기질로 하여 재현성 있는 phytase 활성을 나타내는 균주 12가지를 선발하였다. 12개의 균주 중 BHI broth에서 배양한 조효소액의 phytase 활성이 가장 우수한 것으로 나타난 YH100을 선발하여 주사현미경 관찰, 16S rRNA sequence 분석, GC conten(mol%) 조성, 지방산 분석, API 20E kit를 이용한 test 결과 Enterobacter cloacae로 동정되어, 이 균주를 Enterobacter cloacae YH100이라 명명하였다. Enterobacter cloacae YH100에 의한 phytase 생산을 위한 최적 배지 조성을 파악한 결과 glucose 2.0%(w/v), peptone 1.0%(w/v), beef extract 1.0%(w/v), KCI 0.1%(w/v), sodium phytate 0.1%(w/v)로 나타났다.

Safety Profile Assessment and Identification of Volatile Compounds of Krill Eupausia superba Oil and Residues Using Different Extraction Methods

  • In this study, Krill Eupausia superba oil was extracted using different solvents and supercritical carbon dioxide (SC-$CO_2$). During SC-$CO_2$ extraction, the pressure was set at 40 MPa and temperatures ranged from $40^{\circ}C$ to $55^{\circ}C$. We examined the differences in volatile compounds and safety profiles among extraction methods. Volatile compounds were determined using the thermal desorption system integrated with gas chromatography-mass spectrometry (GC-MS). Heavy metal content was analyzed by inductively coupled plasma mass spectrometry (ICP-MS). According to our results 10 volatile compounds were identified in krill sample. After SC-$CO_2$ extraction of oil, the concentrations of volatile compounds decreased, but increased after solvent extraction. In krill, heavy metal concentrations remained within the permissible limit. Moreover, Zn and Fe which have health benefits were detected at high concentrations. During a 90 days storage period at different temperatures, microbial activity was found to be lowest in SC-$CO_2$ extracted residues. Thus, the quality of krill oil and the residues obtained using SC-$CO_2$ extraction was higher and the oil was safer than those obtained using conventional solvent extraction. These results can be applied to the food industry to maintain high quality krill products.

Chemical and Textural Properties in Commercial Fermented Soybean Curds of Sufu

  • A survey aiming to find out the chemical and textural properties of commercial fermented soy bean curd called sufu was conducted. Sixteen brands of plain sufu produced in the Northern or the Southern part of China were collected and examined for their crude protein, crude fat, texture profiles, free amino acids, and free fatty acid contents. Twenty-one free amino acids were extracted and derivatized using a commercial kit followed by separation and analyzed by the gas chromatography-mass spectrometry (GC-MS). Similarly, ten free fatty acids were extracted using alumina, eluted, separated and analyzed. The content ranges of crude fat and protein were 22~36% and 31~38%, respectively. In texture profile analysis, ranges of the texture parameters were 131~493 g (hardness), 0.4~0.5 (cohesiveness), -137 to -50 gs (adhesiveness), 0.6~1 (springiness), 47~220 g (gumminess) and 32~177 g (chewiness). Twenty-one different free amino acids, especially alanine, glycine, $\alpha$-aminobutyric acid, valine, leucine, allo-isoleucine, aspartic acid, glutamic acid and lysine in large amount, as well as ten fatty acids in total, notably linoleic acid (9-octadecanoic acid), oleic acid (9,12-octadecadienoic acid), linolenic acid (9,12,15-octadecadienoic acid), hexadecanoic acid and octadecanoic acid were found. This information provides important quality reference ranges for product developers and manufacturers to optimize and produce the plain sufu.

흰쥐에서의 트리프로리딘의 대사 (Metabolism of Triprolidine in Rat)

  • The metabolic profile of triprolidine, 2-[1-(4-methylphenyl)-3-(1-pyrrolidinyl-1-propenyl)] pyridine, was determined in rat urine and bile. The free fractions of urinary and biliary extracts were obtained without hydrolysis, and the conjugated fractions of extracts were obtained with enzyme hydrolysis using ${\beta}-glucuronidase$ from Escherichia coli. The mixture of N-methyl-N-trimethylsilyltrifluoroacetamide/trimethylsilyl chloride (100 : 1, v/v) was used to derivatize the extracts and then analyzed by gas chromatography/mass spectrometry. Hydroxymethyltriprolidine, hydroxytriprolidine, triprolidine carboxylic acid, dihydroxytriprolidine 1, dihydroxytriprolidine 2, oxotriprolidine carboxylic acid and unchanged triprolidine were detected in rat urine and bile, which were obtained after oral treatment with triprolidine hydrochloride. The maximum urinary excretion rate of triprolidine and hydroxymethyltriprolidine which were extracted from free fraction was at 1 to 2 hours after drug administration. Hydroxymethyltriprolidine was detected in conjugated fraction, and the maximum urinary excretion rate of that metabolite was at 2 to 3 hours in rat. In rat bile analysis, triprolidine was detected only in free fraction and its biliary excretion rate showed the maximum within 30 minutes after drug administration and decreased continuously thereafter. The excretion percentage of triprolidine and hydroxymethyltriprolidine to the initial dose of the parent drug in bile and urine of rats were all low.

Relationship between sensory attributes and volatile compounds of polish dry-cured loin

  • Objective: The aim of this work was to determine the relationship between objective sensory descriptors and volatile flavour compound composition of Polish traditional dry-cured loin. Methods: The volatile compounds were investigated by using solid phase microextraction (SPME) and gas chromatography-mass spectrometry (GC-MS). For sensory assessment, the quantitative descriptive analysis (QDA) method was used. Results: A total of 50 volatile compounds were found and assigned to 17 chemical families. Most of the detected volatile compounds derived from smoking, lipid oxidative reactions and seasoning (46.8%, 21.7%, and 18.9%, respectively). The dominant compounds were: aromatic hydrocarbon (toluene); alkanes (hexane, heptane, and 2,2,4-trimethylpentane); aldehyde (hexanal); alcohol (2-furanmethanol); ketone (3-hydroxy-2-butanone); phenol (guaiacol); and terpenes (eucalyptol, cymene, ${\gamma}-terpinen$, and limonene). Correlation analysis showed that some compounds derived from smoking were positively correlated with the intensity of cured meat odour and flavour and negatively with the intensity of dried meat odour and flavour, while terpenes were strongly correlated with odour and flavour of added spices. Conclusion: The analysed dry-cured loins were characterized by specific and unique sensory profile. Odour and flavour of studied loins was mainly determined by volatile compounds originating from smoking, seasoning and lipid oxidation. Obtained results suggest that smoking process is a crucial stage during Polish traditional dry-cured loins production.
