Research of pesticide residue of domestic Lentinula edodes related with the positive list system (농약 허용물질목록 관리제도와 연계한 국내산 표고 잔류농약 실태 조사)

  • The study was conducted for the safety evaluation of 320 pesticide residues in 768 Lentinula edodes fruit body samples and 143 L. edodes media samples, which are distributed nationwide in South Korea. The monitoring method was the second of the multi-residue methods in the Korean Food Code. GC-ECD, GC-NPD, and GC-MSD were used as evaluation equipment for analysis. Single-analysis of the target pesticides was performed for mepiquat chloride. Through the analysis of collected L. edodes samples, pesticide residues were detected in total seven cases, including four L. edodes fruit body samples and three L. edodes media samples. The detected pesticide residues were carbendazim, diflubenzuron, fluopyram, and dinotefuran. In this study, carbendazim was detected in three L. edodes fruit body samples and one L. edodes media sample. The detected amount of carbendazim was 0.056, 0.17, 0.043, and 0.09 mg/kg, respectively. The amount of carbendazim in the collected L. edodes samples was detected below the MRLs (maximum residue level). The detected amounts of fluopyram and dinotefuran were 0.068 mg/kg and 0.06 mg/kg, respectively. Two pesticide residues were detected in the medium in one case. Mepiquat chloride was not detected in this study. These results suggested that residual pesticides were detected in a small number of collected L. edodes. However, the PLS for unregistered pesticides MRL was 0.01 ppm; therefore, we have to conduct research on preparing safety standards for mushrooms, including L. edodes.

Characteristics of Pesticide Residues in Leafy Vegetables Collected from Wholesale and Traditional Markets in Cheongju (청주지역 도매시장과 재래시장 유통 엽채류 중 농약의 잔류 특성)

  • This study was carried out to survey the residual characteristics of pesticides and assess their safety. Nineteen agricultural commodities, collected from wholesale and traditional markets in Cheongju. Nineteen agricultural commodities including perilla leaves were collected from the markets on October 29th, 2010. Total 240 pesticides which can be analyzed by multiresidue analysis method by GLC and HPLC were monitored and the pesticides detected were confirmed by GC-MSD and LC-MS. Five pesticides, alachlor, bifenthrin, endosulfan, procymidone and triflumizole, were detected from five samples, such as welsh onion, leek and celery in case of wholesale market and perilla leaves and welsh onion in case of traditional market. Detection rate of 13.2% was obtained as a result of pesticide analysis but 2.6% of the pesticides detected exceeded their maximum residue limits. The estimated daily intakes (EDIs) and maximum permissible intakes (MPIs) of the pesticides detected were less than 26% and 0.05% of their acceptable daily intakes (ADIs) respectively, representing that residue levels of the pesticides detected would be safe.

  • The extraction methods of PBBs from environmental samples by soxhlet extraction for solid phase and liquid-liquid extraction for liquid phase were compared. After extraction, silicagel and florisil as column clean up methods were used for the comparison of cleanup efficiency with different solvent system. Also, the analytical instruments were used GC/MSD. The 22 kinds of PBBs standards were used to establish the analytical methods to perform the experiments of recoveries and detection limits in water and soil. The detection limits of PBBs represented 5~10 ng/L for water and 0.5~3.5 ng/g for soil samples, respectively. The recoveries of water samples represented 96~107% in n-hexane, dichloromethane and toluene for water, and 60~80% in combined solvent with n-hexane and acetone. The recoveries of soil sample were surveyed 67~97% in soxhlet extraction and 64~76% in ultrasonic extraction with selected solvents.

Antimicrobial Activity of Essential Oils from Mentha arvensis L. var. piperascens Malivaud and Agastache rugosa O. Kuntze on Escherichia coli and Salmonella typhimurium (대장균과 살모넬라균에 대한 박하와 배초향 정유성분의 항균활성)

  • For developing natural presevatives, essential oils of Mentha arvensis L. var. piperascens Malivaud and Agastache rugosa O. Kuntze were analyzed the composition of two oils and experimented on microorganism survival. Main components of Mentha arvensis oil were isomenthol (26.84%) and menthol (25.48%), and those of Agastache rugosa oil were estragole (79.83%) and limonene (4.13%) from GC-MSD analysis. Inhibition activities of Mentha arvensis oil against growth of Escherichia coli O157 : H7 ATCC 43895 and Salmonella typhimurium ATCC 7988 were observed from their clear zone $(9{\sim}14 mm\;&\;9{\sim}13\;mm)$, and that of Agastache rugosa oil were done from the clear zone $(13{\sim}20 mm\;&\;10{\sim}18\;mm)$ by concentration-dependent manner, respectively. In the inhibition test on CFU/ml of the microorganisms, both of the plant essential oils at concentration of 5 and 10 mg showed potent growth inhibition activities from 9 hour of incubation. Analysis using transmission electron microscope on E. coli also showed antimicrobial activities of the oils as deformation of the cell and loss of the intracellular materials.

Content characteristics of persistent organic pollutants waste from paint, iron making and steel making process (국내 도료 및 제철·제강산업 발생 폐기물 중 잔류성 유기오염물질류의 함량 특성 -PCDD/DFs, PAHs, PCBs-)

  • This study has been carried out in order to effectively manage three groups of unregulated hazardous organic substances (PCDD/DFs, PAHs, PCBs) in South Korea. The investigated substances have been analyzed according to the test methods for hazardous substances in specified wastes provided by the National Institute of Environmental Research, Korea. Total contents of the organic compounds have been determined for hazardous wastes from three major industrial categories (paint, iron removal, steelmaking), such as waste organic solvent, waste paint or dust. By investigating the waste samples for 7 PAHs using GC/MSD, Naphthalene has been detected (N.D~1631.33 mg/kg). The highest Naphthalene concentration, which exceeded the korean marine dumping waste standard, was found in waste organic solvents and waste paints. Although a content analysis of 7 PCB isomers has been conducted, none of these compounds was detected. The analysis of PCDD/DFs has revealed that all samples meet the criterion for low POP content defined in the technical guidelines developed under the Basel Convention. The PCDD/DFs content in dust samples deriving from 10 manufacturing processes (billet, bloom) was determined to be in the range of N.D~5.66 ng I-TEQ/g waste.

Research on Odor Analysis Technology to Secure the Reliability of Air Quality Improvement in Air Conditioners (에어컨디셔너 공기질 개선의 신뢰도 확보를 위한 냄새 분석 기술 연구)

  • In this study, the odor of the parts and the odor of the surrounding environment were classified and verified. In order to increase the reliability of odor quantitative/qualitative analysis, the selection criteria for 5 sensory evaluators were established, and the n-Butanol control solution for each odor intensity was periodically trained to recognize the odor intensity before sensory evaluation. In addition, although various odor thresholds have been used through several studies, verification of whether the odor intensity value obtained through GC/MSD analysis is similar to the degree to which a person directly smells and feels it. It is important to select the odor threshold that has the best correlation with the odor intensity calculated by the person smelling the odor. Finally, sampling and measuring flowing airflow and temporary odors such as odor component analysis was experimentally difficult due to limited collection space and differences in concentration of generated components. In this study, a quantitative analysis was made possible by using the low temperature concentration (cooling) trap method. Through this, it was confirmed that the correlation with the actual odor intensity was not caused by the product itself, but by the environmental factor discharged from the product after creating the odor environment.

The Efficiency and Performance of Porous Film Containing Freshness Maintenance Ingredients (신선도 유지성분을 포함한 다공성 필름의 성능과 효능)

  • To identify effective food packaging compounds that could significantly affect the freshness of stored food, the efficiency and performance of porous polypropylene film containing mustard oil as a freshness maintenance ingredient was studied by GC-MS analysis and storage testing of bread. AITC (allyl-isothiocyanate)-emitting properties of films impregnated with mustard oil were evaluated by GC-MS. AITC was extracted from mustard oil, and used as a vapor as an effective antimicrobial agent. Films were prepared under four different conditions (the film types were abbreviated 25SF1, 25SF2, 50LF, and IAF) and the amounts of AITC inside vinyl packs constructed using the four films were measured. The results showed that the 25SF2 film (width 25 mm, length 20 cm) yielded a greater amount of AITC than did the 50LF film (width 50 mm, length 20 cm). We confirmed that the amount of gas emission showed better between layer and layer of the film side than the internal film. In storage testing using various films at $35^{\circ}C$ for 25 days, 25SF2 film provided excellent preservation of bread compared with 50LF film. This was in line with the fact that 25SF2 film yielded the highest amount of AITC. Emission capacities AITC of 2 cm film were measured using bottles various volumes (43 mL, 500 mL, 1000 mL) and both closed and open systems. The AITC content of the film in 43 mL bottle was much higher than that yielded by other films in the closed system, and AITC was rapidly emitted, with relatively low residual gas emission after 4 days in an open system. Mustard oil is a useful freshness maintenance ingredient hence, analysis of AITC emission kinetics from various films were helpful to develop films with optimal antimicrobial effects, and will allow application of such films in food packaging systems.

Monitoring and Risk Assessment of Pesticide Residues in Agricultural Products Collected from Wholesale and Traditional Markets in Cheongju (청주지역 유통 농산물 중 잔류농약 모니터링 및 안전성 평가)

  • In order to survey residual characteristics of pesticides in the agricultural products selling at markets and to assess their safety, a total of 120 agricultural products were collected from the wholesale and traditional markets in Cheongju and analyzed the pesticide residues in them by multiresidue analysis method using GLC, HPLC and GC-MSD. Three pesticides, procymidone, penconazole, and tetraconazole, were detected from 4 samples such as onion, leek, tomato, and green pepper. Fungicide penconazole was detected from the onion collected from wholesale market. Fungicide procymidone was detected from leek and tomato and fungicide tetraconazole was detected from green pepper. Pesticide residues were detected from 3.3% of the total samples. The estimated daily intakes (EDIs) of the pesticides detected were less than 0.1% of their acceptable daily intakes (ADIs), representing that residue levels of the pesticides detected were evaluated as safe.

Changes of Component on Green Discolored Garlic (녹변된 마늘의 성분 변화)

  • Effects of temperature, discoloration, and conditioning on garlic compositions before and after storage were examined. Proximate composition, soluble solids, pH, organic acids, and total sugars of all samples were not significantly different. Sugar contents were sucrose > fructose > glucose, and included such fructooligosaccharide as 1-kestose, 1-nystose, and 1-F-fructosyl nystose. Allicin contents in control, green discolored, and conditioned garlics were 647,9, 613.8, and 648.0 mg%, respectively. Thirty-two flavor compounds were identified by GC/MSD, including 11 sulfide compounds, 5 acids, and 3 aldehydes. Contents of sulfur compounds, including disulfide(di-propenyl), 2-thiophene methanethiol, thiophene, 3,4 dihydro-3-vinyl-1,2-dithiin, and 2-vinyl-4H-1,3-dithin, and water-soluble pectins in low temperature-stored garlic were higher than those in control, and decreased by conditioning or inducing green discoloration. These results show sulfur compounds and water-soluble pectins accumulated in low temperature stored-garlic, and are then converted into green pigments by alliinase in disintegrated garlic tissues.

Comparison of Volatile Flavor Compounds of Domestic Onions Harvested in Various Regions (지역별 생산 양파종의 휘발성 향기성분 비교분석)

  • To provide scientific information for processing of value-added products from onion, volatile flavor compounds were compared in onions harvested in 6 regions, such as Muan (Jeonnam), Buan (Jeonbuk), Andong (Gyeongbuk), and 3 regions of Gyeongnam around Changnyeong (Yueo, Jangma, Seongsan). A total of 51 compounds were detected in samples by solid phase microextraction (SPME)/GC/MSD and they consisted mainly of sulfur-containing compounds (20), aldehydes (4), ketones (5), esters (11), aromatic hydrocarbons (4), nitrogen containing compounds (2), and miscellaneous compounds (5). The sulfur-containing compounds were the major compounds with ranges of $62.6{\sim}80.3%$ of total volatiles, regardless of harvested regions. The amounts of 5 sulfur containing compounds known as having antioxidant activity (2,4-, 2,5-dimetylthiophene, 2-vinyl-1,3-dithiane, 5-methoxy thiazole, and 3,5-diethyl-1,2,4-trithiolane) were the highest in Andong and followed by Yueo, Jangma, Buan, Seongsan, and Muan. However, onions from Buan region had the highest amounts of 5 sulfur-containing compounds known as having anticarcinogenic activity ((Z)-, (E)-methyl propenyl disulfide, (Z)-, (E)-propenyl propyl disulfide, and di-2-propenyl disulfide), followed by Yueo, Andong, Jangma, Seongsan, and Muan.
