신경전달물질 조절효소의 절제 및 특성 연구

  • 포유 동물의 중추 신경계에서 주 억제성 신경전달물질로 알려진 4-aminobutyrate(GABA) 대사에 관여하는 효소인 GABA transaminase (GhBA-T)와 Succinic semialdehyde reductase(SSR)를 각각 여러 가지 크로마토그래픽 방법을 사용하여 순수 분리 정제하였다 GABA-T는 subunit 분자량 50kDa을 가진 dimer이며 화학 변형(chemical modification), 흡광, 형광스펙트럼 연구에 의하면 cysteinyl잔기가 활성 부위에 존재하고 있음을 보여주었다 SSR은 2-mercaptoethanol이 존재하든 안하든 o-pthalaldehyde(OPA)에 의하여 효소 활성도가 상실되었으며 흡광 및 형광스펙트럼에 의하면 OPA가 효소의 활성 부위의 lysyl잔기와 반응하여 cysteinyl잔기와 함께 intermolecular cross-linking을 이루는 것을 알 수 있었다. 한편 SSR은 몇가지 anticonvulsant drug에 의하여 부분적인 저해 효과를 나타내었다.

Anticonvulsant, Antioxidant Effect of Cheongsinhwadam-Jeon(청신화담전) (청신화담전 투여가 경련방생시 뇌조직 중 경련 및 산화 관련물질에 미치는 영향)

  • Objective and Methods : This study was performed to evaluate the anticonvulsant, antioxidant effect of modified formulas Korean traditional medicine Cheongsinhwadam-Jeon(CSHDJ). The extract of CSHDJ was administered (p.o.) to mice for 14 days in anticonvulsant and antioxidant tests. Results : The pretreatment of CSHDJ extract prohibited pentylenetrazol (PTZ)-induced convulsion. In PTZ-induced convulsion, lowered level of brain ${\gamma}$-aminobutyric acid(GABA) was restored normal state by the pretreatment of CSHDJ. Increased level of brain glutamic acid was lowered to normal state by CSHDJ, and increased activity of brain ${\gamma}-aminobutyric$ acid transaminase(GABA-T) was reduced to normal state by CSHDJ. In PTZ-induced convulsion, increased level of brain lipid peroxide was lowered to normal state by the pretreatment of CSHDJ. Increased activity of brain xanthine oxidase(XOD) was lowered by CSHDJ, and increased activity of brain aldehyde oxidase lowered to normal state by CSHDJ. In PTZ-induced convulsion, increased activities of superoxide dismutase(SOD) and catalase in brain were lowered by the pretreatment of CSHDJ, whereas increased level of glutathione and increased activity of gluthathione peroxidase in brain were not changed significantly. Conclusions : Above results suggest that CSHDJ has anticonvulsant, antioxidant effect. That seems to be strongly related with the levels of GABA, glutamate, lipid peroxide and the activities of GABA-T, XOD, aldehyde oxidase, SOD, catalase in brain tissue. From these results, CSHDJ could be applied to various convulsive disorders.

Effect of GABA on the growth performance, nutrient digestibility, and backfat thickness in growing-finishing pigs

  • A total of 100 growing pigs (25.66 ± 1.55 kg) were used in a 16-week feeding trial. Pigs were randomly distributed into two treatment groups on the basis of body weight and sex. There were ten replicate pens per treatment, with five pigs (three barrows and two gilts) per pen. The dietary treatments for this trial were as follows: 1) Basal diet (CON) and 2) T1 (γ-aminobutyric acid, GABA), CON + 100 mg·kg-1 GABA. During weeks 0 to 6, the feed efficiency (G : F) in the T1 group was higher (p < 0.05) than that in the CON group, but no improvements in the average daily feed intake (ADFI) and average daily gain (ADG) were noted (p > 0.05). During the experimental period, the final body weight (BW) was improved (p < 0.05). However, no differences (p > 0.05) were noted in the apparent total tract digestibility (ATTD) of dry matter (DM) nitrogen (N). Meanwhile, there were no significant differences (p > 0.05) observed in the backfat thickness among the treatment groups. These results therefore indicate that supplementation with GABA may improve the overall growth performance but may not lead to differences in the nutrient digestibility and backfat thickness in growing-finishing pigs.

An experimental study on the anticonvulsive effects of Buthus extract (전갈(全蝎) 추출물(抽出物)의 항전간효과(抗癲癎效果)에 관(關)한 연구(硏究))

  • In convulsion state by PTZ in rat, anticonvulsive effect and some of ${\gamma}-aminobutyric$ acid(GABA)-related mechanisms of Buthus extract in brain was experimented. It was inhibited GABA-T activity, lipid peroxide generation and xanthine oxidase activity as scheduled administration in vitro and vivo. And the content of brain glutathione was increased as scheduled administration in rat. In convulsion state by PTZ of previously managed rat by Buthus extract, onset time and duration were non-specific changes but recovery time and severity was remarkably reduced. In conclusion speculated that Buthus extract inhibits convulsion by control of GABA content In brain.

Specific Effects on Monocular OKN Directional Asymmetry of Unilateral Microinjections of GABA Antagonist into the Mesencephalic Structures in the Chicken (OKN을 유발하는 단축 Mesencephalic 구조에 GABA Antagonist를 미량 주입할 때의 닭의 OKN 방향적 불균형성에 관한 특수효과)

  • The SR 95531, a GABA antagonist was microinjected into either the pretectum nuclei (nucleus Superficialis Synencephali nSS) or the nBOR (nucleus Ectomammilaris nEM) of chickens. Monocular optokinetic nystagmus (01(N) was reorded by the search coil technique before and after unilateral intracerebral drug administration. Unilateral microinjections of SR 95531 into either the nSS or nEM induce a reversible increase of gain in OKN directed by contralateral eye for both directions of stimulation. The administration into the nSS increased directional asymmetry by increasing the T~ component velocity gain more strongly than the N-T component velocity gain. On the other hand, the unilateral administration of the drug into the nEM suppressed the diretional O1(N asymmetry by increasing the N-T component velocity gain more strongly than the T-N component velocity gain. The nSS seems especially involved in monocular OKN in response to a T-N stimulation, while the nEM seems more involved in the OKN response to N-T stimulation. These results indicate that the drug suppresses GABAergic inhibition at the mesencephalic level. The increase in gain of OKN directed by the ipsilateral eye to microinjeded nuclei could account for the strong interactions existing between these two mesencephalic structures responsible for horizontal OKN.

Effect of fermented sarco oyster extract on age induced sarcopenia muscle repair by modulating regulatory T cells

  • Sarcopenia is an age-related, progressive skeletal muscle disorder involving the loss of muscle mass and strength. Previous studies have shown that γ-aminobutyric acid (GABA) from fermented oysters aids in regulatory T cells (Tregs) cell expansion and function by enhancing autophagy, and concomitantly mediate muscle regeneration by modulating muscle inflammation and satellite cell function. The fermentation process of oysters not only increases the GABA content but also enhances the content of branched amino acids and free amino acids that aid the level of protein absorption and muscle strength, mass, and repair. In this study, the effect of GABA-enriched fermented sarco oyster extract (FSO) on reduced muscle mass and functions via Treg modulation and enhanced autophagy in aged mice was investigated. Results showed that FSO enhanced the expression of autophagy markers (autophagy-related gene 5 [ATG5] and GABA receptor-associated protein [GABARAP]), forkhead box protein 3 (FoxP3) expression, and levels of anti-inflammatory cytokines (interleukin [IL]-10 and transforming growth factor [TGF]-β) secreted by Tregs while reducing pro-inflammatory cytokine levels (IL-17A and interferon [IFN]-γ). Furthermore, FSO increased the expression of IL-33 and its receptor IL-1 receptor-like 1 (ST2); well-known signaling pathways that increase amphiregulin (Areg) secretion and expression of myogenesis markers (myogenic factor 5, myoblast determination protein 1, and myogenin). Muscle mass and function were also enhanced via FSO. Overall, the current study suggests that FSO increased autophagy, which enhanced Treg accumulation and function, decreased muscle inflammation, and increased satellite cell function for muscle regeneration and therefore could decrease the loss of muscle mass and function with aging.

A Comparative Study of GABA, Glutamate Contents, Acetylcholinesterase Inhibition and Antiradical Activity of the Methanolic Extracts from 10 Edible Plants (10가지 식용식물 추출물의 가바, 글루탐산 함량, 아세틸콜린에스테라제 억제 및 라디컬 소거능 비교)

  • In order to investigate the efficacies for cognitive function of edible plants, we measured the inhibitory effects of acetylcholinesterase(AChE) activity, radical scavenging activities, and the contents of GABA and glutamate in the plant extracts. Among the plant extracts, Schizandra chinensis contained the highest GABA 14.8 mg/g and the extracts of Cnidium officinale and Polygonum multiflorum also had a relatively high GABA. On the other hand, plant extracts except, Acorus gramineus, showed similar glutamate contents. S. chinensis, Hovenia dulcis, Thuja orientalis, and Eleutherococcus senticosus exhibited high inhibition against AChE activities at about 18-33% at 1 mg/mL. In addition, strong radical scavenging activities were also detected in those extracts which showed high AChE inhibition. Taken together, H. dulcis, T. orientalis, E. senticosus, and S. chinensis could be effective resources for enhancing cognitive function. Further, it was suggested that the AChE inhibitory activities of plant extracts might be related to antioxidative activity.

  • The action of neuronally released ${\gamma}$-aminobutyric acid (GABA) is terminated by uptake into the neurons by GABA transporters (GATs). The mechanism underlying the stabilization and regulation of GAT2 has not yet been elucidated. We used the yeast two-hybrid system to identify proteins that interact with and, thereby, regulate betaine-${\gamma}$-aminobutyric acid transporter 1 (BGT-1/mGAT2). We found an interaction between BGT-1/mGAT2 and Munc-18-interacting proteins (Mints). The "T-H-L" motif at the C-terminal end of BGT-1/mGAT2 was essential for the interaction with Mint2 in the yeast two-hybrid assay. Mint2 bound to the tail region of BGT-1/mGAT2, but not to other GAT members. When co-expressed in HEK-293T cells, Mint2 was co-immunoprecipitated with BGT-1/mGAT2. In addition, we demonstrated the cellular co-localization of BGT-1/mGAT2 and Mint2 in the cells. These results suggest that Mint2 contributes to the regulation of BGT-1/mGAT2.

Effect of Air-Phase Germination with Anion Radiation and Water-Spraying on Germination Ratio, Sprout Growth, and GABA Contents of Germinated Brown Rice

  • The objective of this study was to investigate the effects of air-phase germination with water-spraying and anion stimuli on germination ratio, sprout growth and ${\gamma}$-aminobutyric acid (GABA) of brown rice. Air-phase germination method with intermittent spraying water improved germination ratio and sprout growth by about 100% compared with the conventional water-soaking method. Anion radiation was applied during the germination process and improved the germination ratio, sprout growth and color quality of the germinated brown rice. Germination ratio and sprout growth were improved up to 9% with anion radiation, and its brightness was higher than brown rice germinated with no anion radiation. The air-phase germination with water-spraying improved the GABA content of germinated brown rice by about 8-9 times compared with that of brown rice.
