Brain GABA transaminase is inactivated by preincubation with antidepressant/antipanic drug pheneizine (${\beta}$ethylphenylhydrazine) (mixing molar ratio 10:1) at pH 7.4. The reaction of enzyme with phenelzine was monitored by absorption and fluorescence spectroscopic methods. The inactive enzyme was fully reconstituted by addition of cofactor pyridoxal-5-phosphate. This result implies that the blocking of 1 mol of pyridoxal-5-phosphate per enzyme dimer is needed for inactivation of the enzyme. The time course of the reaction is significantly affected by the substrate .alpha.-ketoglutarate, which afforded complete protection against the loss of catalytic activity. The kinetic studies shows that phenelzine reacts with the cofactor of enzyme with a second-order rate constant of $2.1{\times}10^3M^{-1}s^{-1}$. It is postulated that the antidepressant/antipanic drug phenelzine is able to elevate the neurotransmitter GABA levels in central nervous system by inhibitory action on GABA degradative enzyme GABA transaminase.
The effects of $Zn^{2+}$ on spontaneous glutamate and GABA release were tested in mechanically dissociated rat CA3 pyramidal neurons which retained functional presynaptic nerve terminals. The spontaneous miniature excitatory and inhibitory postsynaptic currents (mEPSCs and mIPSCs, respectively) were pharmacologically isolated and recorded using whole-cell patch clamp technique under voltage-clamp conditions. $Zn^{2+}$ at a lower concentration $(30{\mu}M)$ increased GABAergic mIPSC frequency without affecting mIPSC amplitude, but it decreased both mIPSC frequency and amplitude at higher concentrations $({\ge}300{\mu}M)$. In contrast, $Zn^{2+}$ (3 to $100{\mu}M$) did not affect glutamatergic mEPSCs, although it slightly decreased both mIPSC frequency and amplitude at $300{\mu}M$ concentration. Facilitatory effect of $Zn^{2+}$ on GABAergic mIPSC frequency was occluded either in $Ca^{2+}$-free external solution or in the presence of $100{\mu}M$ 4-aminopyridine, a non-selective $K^{+}$ channel blocker. The results suggest that $Zn^{2+}$ at lower concentrations depolarizes GABAergic nerve terminals by blocking $K^{+}$ channels and increases the probability of spontaneous GABA release. This $Zn^{2+}$-mediated modulation of spontaneous GABAergic transmission is likely to play an important role in the regulation of neuronal excitability within the hippocampal CA3 area.
ADHD (Attention Deficit/Hyperactivity Disorder)은 4-17세의 아동 및 청소년의 약 10%가 겪는 흔한 신경 발달 장애이지만 그 핵심 기전이 알려져 있지 않은 가운데 관련한 여러 단백질들이 보고되어왔다. 이중 GIT1 (G-protein coupled-receptor kinase interacting protein-1)은 중추신경계에서 dendritic spine formation와 growth에 영향을 미치는 multifunctional adaptor protein으로, GIT1이 제거된 생쥐는 과잉행동, 주의력결핍 그리고 충동성을 보이는 ADHD 증상을 보이게 된다. 이 논문에서는 GIT1 유전자 변형 생쥐를 이용하여 genotype별로 신경교세포의 전달물질(gliotransmitter)을 비교 분석하는 실험을 진행하였다. 그 결과 주요 흥분성 전달물질인 glutamate는 HE (hetero)와 KO (knock-out)의 세포 내에서 WT (wildtype)보다 더 높은 농도로 존재했다. 한편, 억제성 신경전달물질인 GABA와 glycine의 경우 전반적으로 HE에서 가장 많은 함유량을 보였지만 소뇌 세포내의 경우, KO이 WT보다 많은 양을 함유한 것에 비해 대뇌 세포 내에서는 KO보다 WT의 억제성 전달물질 함유량이 높았다. 또한, glutamate와 GABA를 기준으로 흥분성/억제성 비율(excitation/inhibition ratio)을 보았을 때, 소뇌 세포 내/외 모두에서 KO이 가장 높은 수치를 보였고, 대뇌에서는 세포 내/외 모두 HE에서 가장 높은 수치를 보였다. 억제성 신경전달물질인 GABA가 KO의 대뇌 세포 외에서 가장 많은 것으로 보아 GIT1 결손을 보완하기 위해 억제성 물질을 더 많이 분비하거나 또는 과도하게 분비된 GABA를 재흡수하지 못하는 것이라 사료된다. 이는 ADHD 병리기전으로써 기능할 가능성을 제시하며 후속 연구를 통해 해당 기전에 대한 규명이 필요할 것으로 보인다.
A γ-aminobutyric acid (GABA)-producing microorganism was isolated from galchi (hairtail fish, Trichiurus lepturus) jeotgal, a Korean salted and fermented seafood. The G144 isolate produced GABA excessively when incubated in MRS broth containing monosodium glutamate (MSG, 3%, w/v). G144 was identified as Lactobacillus brevis through 16S rRNA and recA gene sequencing. gadB and gadC encoding glutamate decarboxylase (GAD) and glutamate/GABA antiporter, respectively, were cloned and gadB was located downstream of gadC. The operon structure of gadCB was confirmed by reverse transcription (RT)-polymerase chain reaction. gadB was overexpressed in Escherichia coli and recombinant GAD was purified and its size was 54.4 kDa as evidenced by SDS-PAGE results. Maximum GAD activity was observed at pH 5.0 and 40℃ and the activity was dependent on pyridoxal 5'-phophate. The Km and Vmax of GAD were 8.6 mM and 0.01 mM/min, respectively.
본 연구는 저분자 해양성 콜라겐과 GABA 생성 L. brevis CFM20의 섭취 시 다양한 인체 장내 조건에 의한 유산균의 사멸과 콜라겐의 분해를 최소화하고자 진행되었다. 유산균과 저분자 해양성 콜라겐의 안정성을 향상하기 위하여 alginate와 chitosan을 이용하여 이중코팅캡슐을 제조하였다. L. brevis CFM20의 온도별 생육특성을 분석하여 캡슐화에 유리한 배양온도를 조사한 결과 $37^{\circ}C$에서 배양한 균의 대수기가 가장 빠르고 GABA양 또한 $400{\mu}g/mL$로 가장 높은 수치를 나타내어 생육속도에서 가장 유리한 것으로 나타났다. Calcium-alginate bead 법으로 캡슐을 제조한 결과 1~2 mm 사이의 일정한 구형의 캡슐을 제조하였으며, chitosan을 이용하여 이중코팅캡슐을 제조한 결과 캡슐의 크기가 감소하였고 외형적으로도 더욱 단단해짐을 확인하였다. 제조된 이중코팅캡슐을 CLSM과 SEM을 이용하여 관찰한 결과, chitosan 코팅을 한 이중코팅캡슐의 표면이 alginate와의 가교결합에 의해 더욱 치밀해짐을 확인하였다. 체내 위와 장내 조건에서 시간에 따른 이중코팅캡슐의 L. brevis CFM 20 생균수를 분석한 결과 캡슐화한 경우 균의 감소가 더디어 더 안정하다고 판단하였다. 이중코팅캡슐을 체내 위와 장내 조건에서 시간에 따른 GABA와 hydroxyproline의 용출량을 분석한 결과 두 경우 모두 위 조건에서는 낮은 수치를 나타내었고 장내 조건에서는 시간이 지남에 따라 급격하게 증가한 것을 확인하였다. 동물실험 결과 콜라겐 섭취군이 비섭취군에 비해 체중증가량, 혈청 지질 농도 등이 적은 것을 확인하였고, 혈액 내 hydroxyproline 함량, 진피의 피부조직 밀도가 더 높은 것으로 관찰되었다. 본 연구에서 개발된 이중코팅캡슐을 섭취하는 경우 위에서는 안정한 상태였다가 장에 도달한 후 붕해됨으로 인하여, 유산균, GABA, 콜라겐 및 키토산의 작용으로 인체에 유익한 효과를 기대할 수 있을 것으로 생각된다.
GABA 생성 젖산균으로 선발한 Lactobacillus acidophilus RMK567 균주를 멸균한 50 L의 MRS broth가 들어 있는 fermenter에 접종하여 40oC의 발효온도에서 24시간 배양 하였다. L. acidophilus RMK567 생균수는 $10.04{\pm}0.13$ Log CFU/mL이었으며, 이때 5.16시간의 유도기(L)를 거쳐 2.203시간의 세대시간(g)으로 증식하였으며, 생장률상수(k)는 0.454세대/h 이었다. 50 L의 MRS 배양액으로부터 회수한 cell paste의 총 중량은 487 g, 수분함량은 40.5%이었으며, 생균수는 12.48 Log CFU/g 이었다. Cell paste에 glycerol, glucose 및 polydextrose 등의 동결보호제를 첨가하여 $-45^{\circ}C$에서 deep frozen하고 84 mtorr의 진공 하에서 3시간 동결건조하여 총 408 g의 FD cell powder를 생산하였다. FD cell powder에 Streptococcus thermophilus를 혼합한 FD starter cultures의 수분함량은 4.25%, 3.90%, 및 4.08%이었으며, 생균수는 각각 12.42(FDA-GY), 12.60(FDB-GG)과 12.91(FDC-GP) Log CFU/g이었다. 모든 FD starter cultures는 $-18^{\circ}C$ 저장 초기부터 급속히 사멸하여 3.24% 이하의 생존률을 보였다. 각 동결보호제 처리구 간의 생균수 차이는 유의성을 보이지 않았으나, 저장기간에 따른 유의성은 p<0.01 수준에서 관찰되었다. 원유를 기초로 하는 요구르트 조제액에 MSG의 농도를 0.02%, 0.04% 및 0.05%을 첨가하고, FD starter culture로 배양한 요구르트의 GABA 함량은 각각 $155.16{\pm}8.53$ ppm, $243.82{\pm}4.27$ ppm 및 $198.64{\pm}23.46$ ppm 이었다. 이 결과는 L. acidophilus RMK567의 FD starter culture가 동결건조 공정에 의하여 GABA 생성력이 감소하지 않았으며, GABA 함량이 높은 요구르트 제조에 유용하다는 사실을 보여주는 것이었다.
Two lactic acid bacteria (LAB) with high ${\gamma}$-aminobutyric acid (GABA)-producing capacity were isolated from naturally aged cheese. Examination of the biochemical features using an API kit indicated that the two strains belonged to Lactobacillus. They were gram positive, rod-type bacteria, and fermented arabinose, melezitose, melibiose and xylose, but did not utilize cellobiose or trehalose. 16S rDNA sequencing analysis confirmed that they were Lactobacillus buchneri and Lactobacillus sp. They were accordingly named as Lactobacillus buchneri OPM-1 and Lactobacillus sp. OPM-2, and could produce GABA from MRS broth supplemented with 10 g/L of monosodium glutamate (MSG) at a productivity of 91.7 and 116.7 mg/L/hr, respectively. Cell extracts of L. buchneri OPM-1 and Lactobacillus sp. OPM-2 showed glutamate decarboxylase (GAD) activity, for which the optimum pH and temperature were 5.5 and $30^{\circ}C$, respectively.
To investigate propofol's effects on ionic currents induced by ${\gamma}-aminobutyric$ acid (GABA) and glycine as well as on those produced by the nicotinic acetylcholine- and glutamate-responsive channels, rat dorsal raphe neurons were acutely dissociated and the nystatin-perforated patch-clamp technique under voltage-clamp conditions was used to observe their responses to the administration of propofol. Propofol evoked ion currents in a dose-dependent manner, and propofol $(10^{-4}\;M)$ was used to elicit ion currents through the activation of $GABA_A,$ glycine, nicotinic acetylcholine and glutamate receptors. Propofol at a clinically relevant concentration $(10^{-5}\;M)$ potentiated $GABA_A-,$ glycine- and NMDA receptor-mediated currents. The potentiating action of propofol on $GABA_A-,$ glycine- and NMDA receptor-mediated responses involved neither opioid receptors nor G-proteins. Apparently, propofol modulates inhibitory and excitatory neurotransmitter-activated ion channels either by acting directly on the receptors or by potentiating the effects of the neurotransmitters, and this modulation appears to be responsible for the majority of the anaesthetic and/or adverse effects.

This study was investigated to know whether pachymic acid (PA), one of the predominant triterpenoids in Poria cocos (Hoelen) has the sedative-hypnotic effects, and underlying mechanisms are mediated via ${\gamma}$-aminobutyric acid (GABA)-ergic systems. Oral administration of PA markedly suppressed locomotion activity in mice. This compound also prolonged sleeping time, and reduced sleep latency showing synergic effects with muscimol (0.2 mg/kg) in shortening sleep onset and enhancing sleep time induced by pentobarbital, both at the hypnotic (40 mg/kg) and sub-hypnotic (28 mg/kg) doses. Additionally, PA elevated intracellular chloride levels in hypothalamic primary cultured neuronal cells of rats. Moreover, Western blotting quantitative results showed that PA increased the amount of protein level expression of $GAD_{65/67}$ over a broader range of doses. PA increased ${\alpha}$- and ${\beta}$-subunits protein levels, but decreased ${\gamma}$-subunit protein levels in $GABA_A$ receptors. The present experiment provides evidence for the hypnotic effects as PA enhanced pentobarbital-induced sleeping behaviors via $GABA_A$-ergic mechanisms in rodents. Taken together, it is proposed that PA may be useful for the treatment of sleep disturbed subjects with insomnia.

This study was carried out to determine the physicochemical, microbiological, and quality characteristics of a new type of fermented sausage manufactured by incorporating Baechu-kimchi powder and gamma-aminobutyric acid (GABA)-producing lactic acid bacteria (LAB). The LAB count was at the maximum level by day nine of ripening in inoculated sausages, accompanied by a rapid decrease in the pH. The addition of kimchi powder decreased the lightness ($L^*$) and increased the redness ($a^*$) and, yellowness ($b^*$) values, while also significantly increasing the hardness and chewiness of the sausage (p<0.05). Moreover, although the thiobarbituric acid reactive substances values increased in all samples during the study period, this increase was lower in the kimchi-treated samples, indicating a reduction in lipid oxidation. Overall, our results show that the addition of Baechu-kimchi powder to sausages reduced the off-flavor properties and improved the taste profile of the fermented sausage in sensory evaluations. The GABA content of all fermented sausages increased from 17.42-25.14 mg/kg on the third day of fermentation to 60.95-61.47 mg/kg on the thirtieth day. These results demonstrate that Baechu-kimchi powder and GABA-producing LAB could be functional materials in fermented sausage to improve quality characteristics.
