Iso-suillin Isolated from Suillus luteus, Induces G1 Phase Arrest and Apoptosis in Human Hepatoma SMMC-7721 Cells

  • Iso-suillin, a natural product isolated from Suillus luteus, has been shown to inhibit the growth of some cancer cell lines. However, the molecular mechanisms of action of this compound are poorly understood. The purpose of this study was to investigate how iso-suillin inhibits proliferation and induces apoptosis in a human hepatoma cell line (SMMC-7721). We demonstrated the effects of iso-suillin on cell proliferation and apoptosis in SMMC-7721 cells, with no apparent toxicity in normal human lymphocytes, using colony formation assays and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) analysis. Western blotting was used to examine the expression of $G_1$ phase-regulated and apoptosis-associated protein levels in iso-suillin treated SMMC-7721 cells. The results indicated that iso-suillin significantly decreased viability, induced $G_1$ phase arrest and triggered apoptosis in SMMC-7721cells. Taken together, these results suggest the potential of iso-suillin as a candidate for liver cancer treatment.

Novel Suspension-Phase Enzyme Reaction System Using Insoluble Extrusion Starch as Glycosyl Donor for Intermolecular Transglycosylation of L-Ascorbic Acid

  • A novel suspension-phase enzyme reaction system for the intermolecular transglycosylation of L-ascorbic acid into 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid supplementing extrusion starch as the glycosyl donor was developed using cyclodextrin glucanotransferase from Thermoanaerobacter sp. A high conversion yield compared to the conventional soluble-phase enzyme reaction system using cyclodextrins and soluble starch was achieved. The optimal reaction conditions were 2,000 units of cycIodextrin glucanotransferase, 20 g/l of L-ascorbic acid, and 50 g/l of extrusion starch at $50^{\circ}C$ for 24 h. The new suspension-phase enzyme reaction system also exhibited several distinct advantages other than a high conversion yield, including a lower accumulation of oligosaccharides and easily separable residual extrusion starch by centrifugation or filtration in the reaction mixture, which will facilitate the purification of 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid. The new suspension-phase enzyme reaction system seems to be potentially applicable as the industrial process for the production of thermally and oxidatively stable 2-O-${\alpha}$-D-glucopyranosyl L-ascorbic acid.

The Spatial Distribution of Harmful Chemical Substance in Sediment Around Busan Southern Port (부산 남항 해저퇴적물 중 유기오염물질 분포 특성에 관한 연구)

  • Located mostly inside the megacity of Busan, the Busan Southern Port is a multifunctional port with various nearby industry activities, including a joint fish market, ship repair facilities, and fishing boat facilities. If toxic chemicals generated by the industrial activities continue to flow into and accumulate in the sediment of the port, they can affect aquatic ecosystems and humans. Therefore, in this study, distribution levels and potential influent sources of organic pollutants, including polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), and butyltin compounds (BTs), in the sediment were investigated. The sediment samples were collected from eight sampling sites in November 2013 (first phase) and November 2014 (second phase). The mean concentrations of PAHs, PCBs, and BTs in the first and second sampling phages were 4174.0 ng/g-dry wt. and 1919.0 ng/g-dry wt., 166.3 ng/g-dry wt. and 21 ng/g-dry wt., and 50.9 ng/g-dry wt. and 30.8 ng/g-dry wt., respectively. The concentrations of the organic pollutants detected in the seabed sediments were lower in the second phase than in the first phase. In this study, the inflow sources of PAHs, PCBs, and BTs were found to be combustion, land, and municipal sewage or industrial wastewater, respectively.

Triacetin이 탄소복합 필터의 연기성분 흡착능에 미치는 영향

  • This study was conducted to evaluate the effect of triacetin(TA) treatment during manufacturing carbon dual filter for the adsorption of cigarette smoke components by activated carbons. The measurements were carried out by separation of activated carbon from carbon dual filter, and the specific surface area analyzed. The specific surface area of activated carbon from the domestic cigarette filter and from the foreign cigarette filter by degassing at 9$0^{\circ}C$ was 163$\pm$32$m^2$/g, and 16.6$\pm$1.9$m^2$/g, respectively. Those values were very lower than that of degassing at 35$0^{\circ}C$ (Domestic brand: 952$\pm$30$m^2$/g, and Foreign brand: 847$\pm$73$m^2$/g). By comparing the adsorption capacity of acetone and benzene with and without triacetin treated activated carbon, there was a 20% reduction of adsorption capacity by 5% triacetin treatment. Also, from the cilia toxicity test with carbon dual filter treated 0 % TA and 8 % TA, the cilia survival time was 706$\pm$74sec. and 603$\pm$64sec. for 0% TA and 8% TA, respectively. The delivery rate of vapour phase of cigarette smoke, which consists of main components of cilia toxicity, was higher at 8% TA filter than 0 % TA filter. Our results indicate that the treated TA covered the micro-pore of activated carbon, and then reduced specific surface area, finally, decreased the adsorption of vapour phase from cigarette smoke.

6-Aminonicotinamide Induces $G_1$ Arrest by Elevating $p27^{kip1}$ as well as Inhibiting cdk2, Cyclin E and p-Rb in IMR32 Neuroblastoma Cell Line

  • The effects of 6-aminonicotinamide (6-AN) on viability of IMR32 neuroblastoma cells in the presence of ATP or $NAD^+$ have been investigated. 6-AN caused marked reduction in cell viability and similar observations were also made with cells treated with 6-AN+ATP. However, cells treated with $6-AN+NAD^+$ showed cell viability similar to untreated cells. Morphologically, 6-AN and 6-AN+ATP treated cells showed loss of neurites, polyhedric shapes, shrinkage of cell bodies and formation of lysed cells, while $6-AN+NAD^+$ cells did not show any such changes. The flow cytometry analysis demonstrated that 6-AN increased cell population in $G_0/G_1$ phase and decreased cell population in Sand $G_2/M$ phase following a 72 h exposure. Western blot analysis showed that 6-AN stimulated a substantial increase in the level of the cdk inhibitor $p27^{kip1}$, but lowered the levels of cdk2, cyclin E and p-Rb. However, cdc25A and p53R2 were not significantly affected. Immunofluorscence staining of $p27^{kip1}$, cdk2, cyclin E and p-Rb revealed close correlation between the signal observed in the Western blot analysis. 6AN+ATP treated cells showed similar results obtained with 6-AN treated cells in expression of cdk2, cyclin E, p-Rb proteins and $p27^{kip1}$, $6-AN+NAD^+$ cells showed greater expression of cdk2, cyclin E and p-Rb than those in 6-AN and 6-AN+ATP treated cells. The results suggest that 6-AN induced the $G_0/G_1$ phase arrest in IMR32 neuroblastoma cell lines through the increase of $p27^{kip1}$ and the decrease of cdk2, cyclin E and p-Rb.

Growth Inhibition and G2/M Phase Cell Cycle Arrest by 3,4,5-Trimethoxy-4'-bromo-cis-stilbene in Human Colon Cancer Cells

  • Resveratrol (3,5,4’-trihydroxy-trans-stilbene), a naturally occurring phytoallexin abundant in grapes and several plants, has been shown to be active in inhibiting proliferation and inducing apoptosis in several human cancer cell lines. On the line of the biological activity of resveratrol, a variety of resveratrol analogs were synthesized and evaluated for their growth inhibitory effects against several human cancer cell lines. In the present study, we found that one of the resveratrol analogs, 3,4,5-trimethoxy-4’-bromo-cis-stilbene, markedly suppressed human colon cancer cell proliferation (EC$_{50}$ = 0.01 ${\mu}$g/ml), and the inhibitory activity was superior to its corresponding trans-isomer (EC$_{50}$ = 1.6 ${\mu}$g/ml) and resveratrol (EC$_{50}$ = 18.7 ${\mu}$g/ml). Prompted by the strong growth inhibitory activity in cultured human colon cancer cells (Col2), we investigated its mechanism of action. 3,4,5-Trimethoxy-4’-bromo-cis-stilbene induced arrest of cell cycle progression at G2/M phase and increased at sub-G1 phase DNA contents of the cell cycle in a time- and dose-dependent manner. Colony formation was also inhibited in a dose-dependent manner, indicating the inhibitory activity of the compound on cell proliferation. Moreover, the morphological changes and condensation of the cellular DNA by the treatment of the compound were well correlated with the induction of apoptosis. These data suggest the potential of 3,4,5-trimethoxy-4’-bromo-cis-stilbene might serve as a cancer chemotherapeutic or chemopreventive agent by virtue of arresting the cell cycle and inducing apoptosis for the human colon cancer cells.

Theoretical Studies on the Gas-Phase Alkylation of Delocalized Ambident Anions with Methyl Fluoride

  • Gas-phase alkylations of delocalized ambident anions, Y---CH---X- where X, Y=CH2, O, or S, have been investigated theoretically at the MP2/6-31+G*//MP2/6-31+G* and QCISD/6-31+G*//MP2/6-31+G* lev-els. O-and S-alkylations (X=O and S) are more favored kinetically by ΔE^≠ = 4.6 and 9.8 kcal mol-1 than the respective C-alkylations even though they are thermodynamically less favored by 22.4 and 6.0 kcal mol-1 respectively. It was found that the transition structures for the C-alkylations are imbalanced due to the endoergic rehybridi-zation of the carbon center from sp2 to sp3 which leads to premature bond contraction of the C-Y bond and delayed bond stretching of the C-X bond. In the O-, or S-alkylation, such endoergic process is not required since the σ-lone pair on O or S is involved in the initial stage of alkylation. The imbalanced TSs for the C-alkylation are accompanied by higher intrinsic barriers and deformation energies.

  • In case while modulation index (M) is more than 0.7, the spectrum of motor voltage and current of a conventional two-phase SRP scheme are not reduced considerably. To solve the problems of a conventional two-phase SRP, this paper proposes a two-phase SRP(DZSRP) with double zero vector mode which zero vector is selected as V(111) in case of $M\;\geqq\;0.7$, and zero vector is selected as V(000) if M < 0.7. For the validity of the proposed method, a 16 bit micro-controller C167 was used and the experiments were conducted with the 1.5kw induction motor under load condition. And the experimental results show that the switching noise spectrum for all the M are spread to a wide band area.

Cell Cycle Arrest Effects by Artemisia annua Linné in Hep3B Liver Cancer Cell (Hep3B 간암세포에서 개똥쑥 추출물에 의한 Cell Cycle Arrest 효과)

  • Cells proliferate via repeating process that growth and division. This process is G1, S, G2 and M four phases consists. Monitoring the progression of the cell cycle is a specific step that to be a continuous process is repeated to adjust the start of the next step. At this time, this process is called a Checkpoint. Currently, there are three known checkpoints that G1-S phase, G2-M phase, and the M phase. In this study, we confirmed that cell cycle arrest effects by ethanol extracts of Artemisia annua Linne (AAE) in Hep3B liver cancer cells. AAE was regulated proteins which involved in cell cycle such as pAkt, pMDM2, p53, p21, pCDK2 (T14/Y15). AAE induced cell cycle arrest in G1 checkpoint through phosphorylation of CDK2. Akt and p53 upstream is inhibited by AAE and p53 activated by non-activated pMDM2, p53 inhibitor. Thereby, activated p53 is transcript to p21 and activated p21 protein is combined with Cyclin E-pCDK2 complex. Therefore, we confirmed that AAE-induced cell cycle arrest was occurred by p21-Cyclin E-pCDK2 complex by inhibition of pAkt signal. Because of this cell cycle can't pass to S phase from G1 phase.

Effects of Complex Probiotic Supplementation on Growth Performance, Nutrient Digestibility, Blood Metabolites, Noxious Gas and Fecal Microflora in Weaning Pigs (사료 내 복합생균제 첨가가 이유자돈의 사양성적, 영양소 소화율, 혈액성상, 분내 유해가스 및 분 중 미생물에 미치는 영향)

  • This study was undertaken to determine the effects of diet supplementation with complex probiotics (CPB), on growth performance, nutrient digestibility, blood metabolites, noxious gas, and fecal microflora in weaning pigs. On the basis of body weight, a total of 234 weaned pigs (Landrace×Yorkshire×Duroc, 6.14±0.78kg) were randomly allotted to 3 treatments and 6 replicates (13 pigs per pen). The experimental diets were fed in a meal form for 28 days (days 0-14, PhaseI, and days 15-28, PhaseII). The dietary treatment groups were as follows: T1 (basal diet), T2 (T1+0.13% CPB) and T3 (T1+0.25% CPB). The CPB supplement contained Bacillus subtilis 1.0×106 CFU/g, Enterococcus faecium 1.0×106 CFU/g, Saccharomyces cerevisiae 1.0×106 CFU/g, Bacillus licheniformis 3.0×108 CFU/g, and Bacillus polyfermenticus 3.0×108 CFU/g. Pigs fed the T3 diet showed an increase (p<0.05) in the overall average daily gain and average daily feed intake, increased (p<0.05) crude protein digestibility in PhaseI, and greater (p<0.05) dry matter and gross energy digestibility in PhaseII. Supplementation of CPB had no effect on the blood profile. Furthermore, pigs fed the T3 diet had lower (p<0.05) NH3 emission and overall count of fecal Clostridium spp. In conclusion, we believe that CPB supplementation has a beneficial effect on growth performance, nutrient digestibility, noxious gas, and fecal microflora in weaning pigs.
