Effects of Rapamycin on Cell Apoptosis in MCF-7 Human Breast Cancer Cells

  • Background: Rapamycin is an effective anti-angiogenic drug. However, the mode of its action remains unclear. Therefore, in this study, we aimed to elucidate the antitumor mechanism of rapamycin, hypothetically via apoptotic promotion, using MCF-7 breast cancer cells. Materials and Methods: MCF-7 cells were plated at a density of $1{\times}10^5$ cells/well in 6-well plates. After 24h, cells were treated with a series of concentrations of rapamycin while only adding DMEM medium with PEG for the control regiment and grown at $37^{\circ}C$, 5% $CO_2$ and 95% air for 72h. Trypan blue was used to determine the cell viability and proliferation. Untreated and rapamycin-treated MCF-7 cells were also examined for morphological changes with an inverted-phase contrast microscope. Alteration in cell morphology was ascertained, along with a stage in the cell cycle and proliferation. In addition, cytotoxicity testing was performed using normal mouse breast mammary pads. Results: Our results clearly showed that rapamycin exhibited inhibitory activity on MCF-7 cell lines. The $IC_{50}$ value of rapamycin on the MCF-7 cells was determined as $0.4{\mu}g/ml$ (p<0.05). Direct observation by inverted microscopy demonstrated that the MCF-7 cells treated with rapamycin showed characteristic features of apoptosis including cell shrinkage, vascularization and autophagy. Cells underwent early apoptosis up to 24% after 72h. Analysis of the cell cycle showed an increase in the G0G1 phase cell population and a corresponding decrease in the S and G2M phase populations, from 81.5% to 91.3% and 17.3% to 7.9%, respectively. Conclusions: This study demonstrated that rapamycin may potentially act as an anti-cancer agent via the inhibition of growth with some morphological changes of the MCF-7 cancer cells, arrest cell cycle progression at G0/G1 phase and induction of apoptosis in late stage of apoptosis. Further studies are needed to further characterize the mode of action of rapamycin as an anti-cancer agent.

  • The $Nd_{1+x}Ba_{2-x}Cu_3O_{7-{\delta}}$(Nd123) superconductor exhibits high performance in high magnetic field and high temperature. We have studied phase stability for Nd123 under reduced oxygen partial pressure and various heat-treatment conditions. The main phase is Nd123 and some samples contain small amounts of Nd422 depending on the temperature and oxygen partial pressure. The decomposition temperature decreases with decreasing oxygen partial pressure from $1052^{\circ}C(P(O_2)$=150 Torr) to about $845^{\circ}C(P(O_2)$=0.1 Torr). The liquidus line was steeper temperature with decreasing oxygen partial pressure. In same condition of oxygen partial pressure, the region of stable Nd123 phase was formed at slightly higher temperature than the region of stable YBCO phase.

Effects of yeast culture (Saccharomyces cerevisiae) supplementation on growth performance, fecal score, and nutrient digestibility of weaning pigs

  • Weaning pigs often face post-weaning challenges such as diarrhea, low feed intake, and body weight (BW) loss which affects the health and economic value of weaning pigs. Interestingly, the use of yeast cultures (YCs) as feed supplements for pigs has increased markedly in recent years. This study evaluated the effects of yeast cultures (Saccharomyces cerevisiae) on the growth performance, fecal score, and nutrient digestibility of weaning pigs. A total of 50 crossed healthy weaning pigs [(Yorkshire ${\times}$ Landrace) ${\times}$ Duroc] with an average BW of $7.46{\pm}1.60kg$ (28 day of age) were used in a 6-week experiment. The experiment was divided into 3 phases (Phase 1, 1 - 2 weeks; Phase 2, 2 - 4 weeks; Phase 3, 4 - 6 weeks). Dietary treatments were as follows: 1) CON: basal diet and 2) CON + 0.50% YC. During phase 1, the average daily gain (ADG) and average daily feed intake (ADFI) were significantly increased (p < 0.05) in the weaning pigs fed YC supplementation diets compared with the weaning pigs fed the CON diet. During phase 3 as well as overall, the gain/feed ratio (G/F) was significantly increased (p < 0.05) in the YC supplementation group compared with the pigs fed the CON diet. In conclusion, the supplementation of YCs in the diet positively affected the growth performance of weaning pigs during the first two weeks after weaning.

  • The phase sequence of codeposited Co-Si alloy and Co/si multilayer thin film was investigated by differential scanning calormetry(DSC) and X-ray diffraction (XRD) analysis, The phase sequence in codeposition and codeposited amorphous Co-Si alloy thin film were CoSilongrightarrow Co2Si and those in Co/Si multilayer thin film were CoSilongrightarrowCo2Silongrightarrow and CoSilongrightarrowCo2Si longrightarrowCoSilongrightarrowCoSi2 with the atomic concentration ration of Co to Si layer being 2:1 and 1:2 respectively. The observed phase sequence was analyzed by the effectvie heat of formatin . The phase determining factor (PDF) considering structural facotr in addition to the effectvie heat of formation was used to explain the difference in the first crystalline phase between codeposition, codeposited amorphous Co-Si alloy thin film and Co/Si multilayer thin film. The crystallinity of Co-silicide deposited by multitarget bias cosputter deposition (MBCD) wasinvestigated as a funcion of deposition temperature and substrate bias voltage by transmission electron microscopy (TEM) and epitaxial CoSi2 layer was grown at $200^{\circ}C$ . Parameters, Ear, $\alpha$(As), were calculate dto quantitatively explain the low temperature epitaxial grpwth of CoSi2 layer. The phase sequence and crystallinity had a stronger dependence on the substrate bias voltage than on the deposition temperature due to the collisional daxcade mixing, in-situ cleannin g, and increase in the number of nucleation sites by ion bombardment of growing surface.

  • In this study $H_2O$-NaCl mixture was selected as a cold thermal storage material and its phase change temperature($liquid{\Leftrightarrow}solid$) was controlled with the molar concentration of NaCl. Ion dipole interaction mechanism and the fusion and crystallization structure of $H_2O$-NaCl were visualized with the low and high concentration of NaCl in the heating and cooling processes. In this study, the original cause of the appearance of two steps phase change period in heating and cooing processes were found by the visualization of the ion dipole interaction mechanism of $H_2O$-NaCl, and the theoretical equation of the phase change temperature variation in the NaCl high molar concentration was rearranged.

A Separation of manganese (II) and cobalt (II) ions by D2EHPA/TBP-immobilized PolyHIPE membrane

  • The D2EHPA/TBP co-extractants immobilized PolyHIPE membrane can be used for the selective separation of Mn (II) from Co (II). By solvent-nonsolvent method, D2EHPA/TBP co-extractants can be effectively immobilized into PolyHIPE membrane. The pore structure of PolyHIPE membrane and the presence of TBP enhance the stability of immobilized co-extractants. The optimal operating conditions for the separation of Mn (II) and Co (II) are feeding phase at pH 5.5, sulfuric acid concentration in the stripping phase of about 50 g/L and stirring speed at 400 rpm. The D2EHPA/TBP co-extractants ratio of 5:1 shows synergetic effect on Mn/Co separation factor about 22.74. The removal rate and recovery rate of Mn (II) is about 98.4 and 97.1%, respectively, while for Co (II) the transport efficiency is insignificant. The kinetic study of Mn (II) transport shows that high initial flux, $J_f^o=80.1({\mu}mol/m^2s)$, and maxima stripping flux, $J_s^{max}=20.8({\mu}mol/m^2s)$, can be achieved with D2EHPA/TBP co-extractants immobilized PolyHIPE membrane. The stability and reusability study shows that the membrane can maintain a long term performance with high efficiency. High purity of Co (II) and Mn (II) can be recovered from the feeding phase and stripping phase, respectively.

Vapour Effect of Kresoxim-methyl on Powdery Mildew of Barley and Cucumber (보리와 오이 흰가루병에 대한 Kresoxim-methyl의 훈증 효과)

  • The protective effect of kresoxim-methyl was investigated on 6 important plant diseases, and the vapour effect of it did on barley and cucumber powdery mildews, respectively. With 2.0 ${\mu}g\;mL^{-1}$ of kresoxim-methyl, its high activities against wheat leaf rust and barley powdery mildew were showed such as 92 and 100%, while activities were very low against rice sheath blight, tomato gray mold, and tomato late blight. In vapour phase control activity of kresoxim-methyl against barley powdery mildew was positively correlated with the applied concentrations, except for azoxystrobin and metominostrobin. With 200 ${\mu}g\;L^{-1}$, its control value was 71.9%. When 1000 ${\mu}g\;m^{-3}$ of kresoxim-methyl in vapour phase was applied on 4 plants of cucumber in a vinyl chamber, 51.1% of control value on kresoxim-methyl-treated cucumber was showed 7 days after the application.

The Study of Muscle Activity for the Upper Extremity over Morto-Seoinage(Two-handed shoulder throw) in Judo (유도 양손 업어치기 시 상지근의 근 활성도에 관한 연구)

  • The purpose of this study is Muscle Activity for the Upper Extremity and Morto-Seoinage(Two-handed shoulder throw) according to performance levels and elite judo players (G1) and non-elite judo players, namely university players (G2). To achieve this purpose, three players in G1 whose main special skill was Morote-Seoinage and three university judo players(G2) were selected as comparative group. After analysing this study, conclusions were derived as follows. 1. Muscle activity of musculi membri superious to group of Kuzushi to Tsukuri was the highest in left biceps brachii and right biceps brachii in order. On the other hand, G2 was the highest in left biceps brachii and left flexer carpi randialis in order. 2. Intergral electromyogram of according to the phase showed right triceps 1P of left flexer carpi radialis. G1 and G2 of showed left biceps brachii was the highest in 1P and 2P. As a result, extra training of left flexer carpi radialis and left biceps brachii is needed.

  • Polycaprolactone (PCL) macromer containing terminal methacrylate group was synthesized by ring-opening polymerization. The number average molecular weight of PCL macromer was 11600 g/mole and polydispersity index was 1.09. The synthesized PCL macromer was copolymerized with styrene by stable free radical polymerization using 2,2,6,6-tetramethyl-1-piperidinyloxy (TEMPO), benzoyl peroxide, and well-defined poly(styrene-g-caprolactone)s were synthesized. The synthesized copolymers was characterized by $^1$H-NMR and gel permeation chromatography equipped with multiangle laser light scattering detector. Thermal properties of graft copolymers were investigated by DSC.
