Expression of Pseudorabies Virus (PRV) Glycoproteins gB, gC and gD using Bacterial Expression System

  • The Pseudorabies (PR), also called Aujeszky's disease (AD), is an infectious viral disease caused by an alpha herpes virus and has domestic and wild pigs, as well as a wide range of domestic and wild animals, as the natural host. Pseudorabies virus (PRV) virions contain several envelope glycoproteins. Among them, gB, gC and gD are regarded as the major immunogenic proteins. We expressed these glycoproteins using the bacterial expression system and analyzed recombinant proteins. Expression of glycoproteins gC and gD were observed on SDS-PAGE or Western blot analysis, but gB was not. Optimal concentration of IPTG and inducing time were determined as 1.0 mM and 4 h, respectively, for the expression of both gC and gD in E. coli. A sodium dodecyl sulfate (SDS) was the most efficient detergent in solubilizing insoluble recombinant protein.

  • C3 glomerulopathy (C3G) is a recently defined pathological entity characterized by C3 accumulation with absent or scant immunoglobulin deposition, leading to variable glomerular inflammation. The clinical presentation of patients with C3G is highly variable, as they may present with symptoms ranging from microscopic or mild proteinuria to full-blown nephrotic syndrome, with or without renal impairment. However, there is no consensus recommendation for specific treatment in children with C3G. Recently, new therapies have been suggested to target complement pathways, owing to an improvement in the understanding of the pathogenesis of C3G. C3G complement blockade with eculizumab, a monoclonal antibody targeted against complement C5, inhibits activation of the alternative complement pathway. We could not use eculizumab owing to its high price; thus, we administered oral prednisolone and mycophenolate mofetil (MMF). MMF was replaced with cyclosporine because proteinuria persisted, with a consistently low serum C3 level; we tapered off the prednisolone because of a Cushingoid appearance and amenorrhea. Thereafter, proteinuria improved, and the serum C3 level returned to normal. Thus, we report the effectiveness of cyclosporine in a patient with C3G and an inadequate response to prednisolone and MMF, who was detected via school urinary screening.

Effect of Transglutaminase on the Rheological Properties of Fried Surimi Gel (Transglutaminase 첨가가 튀긴 어묵의 레올로지 특성에 미치는 영향)

  • Transglutaminase(TGase) 첨가가 튀긴 어묵의 레올로지 특성에 미치는 영향이 평가되었다. 최적의 TGase 함량과 setting시간은 저온$(25^{\circ}C)$ setting에서는 0.1%와 180분이었고, 고온$(40^{\circ}C)$ setting에서는 0.3%와 80분으로 각각 결정되었다. 각각의 최적조건에서의 겔 강도는 $25^{\circ}C$에서는 TGase를 첨가한 겔이 첨가하지 않은 겔보다 최대 750(g cm)도 증가했으며, $40^{\circ}C$에서는 약 800(g cm)정도의 증가를 보여주었다. 동적 점탄성의 setting 측정 결과로부터 TGase 첨가 시료는 $25^{\circ}C$인 경우 G#과 G@ 값이 각각 1.0 kPa과 0.5 kPa 정도 증가하는 것으로 나타났으며, $40^{\circ}C$ 경우는 G#과 G@ 값이 각각 15 kPa과 2.2 kPa 정도 증가함을 보여 주었다. 동적 점탄성의 가열-냉각 측정 결과에서도 $25^{\circ}C$$40^{\circ}C$ setting 모두에서 G#, G@ 값이 $5{\sim}15\;kPa$ 정도 증가함을 나타났다.

Nucleotide Insertion Fidelity of Human Hepatitis B Viral Polymerase

  • The hepadnaviruses replicate their nucleic acid through a reverse transcription step. The MBP-fused HBV polymerase was expressed in E. coli and purified by using amylase affinity column chromatography. The purified protein represented DNA-dependent DNA polymerase activity. In this report, the MBP-HBV polymerase was shown to lack 3'$\rightarrow$5' exonuclease activity, like other retroviral RTs. The ratio of the insertion efficiency for the wrong versus right vase pairs indicates the misinsertion frequency (f). The nucleotide insertion fidelity (1/f), observed with the MBP-HBV polymerase and HIV-1 RT, was between 60 and 54,000, and between 50 and 73,000, respectively, showing that they are in close range. A relatively efficient nucleotide incorporation by the MBP-HBV polymerase was observed with a specificity of three groups: (1) A : T, T : A>C : G, G : C (matched pairs), (2) A : C, C : A>G: T, T : G (purine-pyrimidine and pyrimidine-purine mispairs), and (3) C : C, A : A, G : G, T : T>T : C, C : T>A : G, G : A (purine-purine or pyrimidine-pyrimidine mispairs), and their order is (1)>(2)>(3). The data from the nucleotide insertion fidelity by the MBP-HBV polymerase suggest that the HBV polymerase may be as error-prone as HIV-1 RT.

Simultaneous Determination of Flavanone Glycosides in the Fruit of Citrus paradisi and C. grandis by HPLC-PDA

  • An HPLC (high-performance liquid chromatography)-PDA (photodiode array) detection method was established for the determination of naringin, hesperidin and neohesperidin in the fruit of Citrus paradisi and C. grandis. The flavonoids were separated in less than 20 min using an YMC RP 18 column with isocratic elution using acetonitrile and water (23 : 77, v/v) at a flow rate of 1 ml/min, and a PDA detector. The levels of naringin, hesperidin and neohesperidin were 1345.92, 950.62, and 2078.82 ${\mu}g/g$, respectively, in the peel, and 102.43, 59.13, and 86.68 ${\mu}g/g$, respectively, in the flesh of C. paradisi. In C. grandis, the levels of naringin, hesperidin and neohesperidin were 3530.56, 80.00, and 5.26 ${\mu}g/g$, respectively, in the peel, and 59.59, 7.43, and 38.41 ${\mu}g/g$, respectively, in the flesh. The total content was highest in the peel, reaching 0.44% and 0.36% in C. paradisi and C. grandis, respectively, while the flesh contained only 0.025% and 0.011%, respectively. Therefore, the peels of C. paradisi and C. grandis are necessary for the processing and utilization of flavonoids.

  • Nano-sized cathode materials for high power lithium ion polymer battery are easily and economically prepared using united eutectic self-mixing method without any artificial mixing procedures of reactants and ultra-miniaturization of products. While the micro-sized $LiNi_{0.7}Co_{0.3}O_2$ exhibits the discharge capacities of 167.8 mAh/g at 0.1C and 142.5 mAh/g at 3.0C, those of the nano-sized $LiNi_{0.7}Co_{0.3}O_2$ are 170.8 mAh/g at 0.1C and 159.3 mAh/g at 3.0C. In the case of $LiCoO_2$, the micro-sized $LiCoO_2$ exhibits the discharge capacities of 134.8 mAh/g at 0.1C and 118.6 mAh/g at 5.0C. Differently, the nano-sized $LiCoO_2$ exhibits the discharge capacities of 137.2 mAh/g at 0.1C and 131.7 mAh/g at 5.0C.

  • 이중 전송율(5.3 과 6.3kbit/s)을 제공하는 G.723.1 음성 코더는 공중망을 통한 H.324 POTS 영상 회의 규격의 음성 코더로 채택된 것으로, MPMLQ, ACELP 알고리즘에 근거한다. 본 논문에서는 Annex A를 포함한 G.723.1 음성 코더 알고리즘을 C&S Technology에서 개발한 음성 신호 처리를 위한 범용 DSP인 CSD17C00 칩을 이용하여 실시간 응용이 가능하도록 구현하였다. G.723.1 에 대한 양방향 평가가 Codec loopback을 통해 수행되었으며, ITU에서 제공한 테스트 절차에 따라 평가되었다. 또한, 본 논문에서 구현된 G.723.1 음성 코더는 27MIPS의 계산 속도를 갖으며, 프로그램 ROM의 크기는 8.85K Words이고, 10K 데이터 ROM과 4K 데이터 RAM을 필요로 하고 있다. 경쟁 제품과의 MOS 측정 음질 평가를 실시한 결과, CSD17C00에서의 음질 성능이 더 우수함을 입증 함으로써, 본 논문에서 보여준 CSD17C00을 기반으로 구현된 G.723.1 알고리즘의 실시간 구현기술의 타당성을 검증하게 되었다.

Cultural Conditions of Exopolysaccharide KS-1 Produced by Bacillus polymyxa KS-1 (Bacillus polymyxa KS-1에 의한 다당류 KS-1 생산의 발효 조건)

  • Optimized fermentation medium and cultural conditions for the production or exopolysaccharide KS-1 with Bacillus polymyxa KS-1 was following as; 30g g1ucose, 2.59g yeast extract, $2.5g KH_2PO_4, 0.5g NaCl, 0.3g MgSO_4.7H_20, 0.1g CaC0_3 0.05g, FeSO_4.7H_2O, and 0.05g MnS0_4 . 4H_20in 1 liter distilled water. The exopolysaccharide production was influenced by the by the temperature and pH, the optimal conditions for the production of exopolysaccharide KS-1 seemed to be $30^{\circ}C$ and pH 7.0, respectively. About $10.3g/\ell$ of maximum exopolysaccharide was obtained al the initial pH 7.0, $30^{\circ}C$, 2vvm of aeration rate and 400 rpm of impeller speed in a jar fermentor.

Food Composition of Crab(Charybdis japonica) Preserved in Brine (민꽃게(Charybdis japonica)장의 식품 성분)

  • Pickled crabs made using Charybdis japonica are becoming part of our dietary habits. Yet research on this food is disappointingly scarce. It is generally accepted that the original food composition of pickled crabs defies scrutiny because various seasonings are added to soy for the production of pickled crabs in most regions. Accordingly, this study attempted to measure the food composition of the Charybdis japonica preserved in brine according to different storage days (10, 15, 20, 25, and 30 days) and different temperatures ($5{^{\circ}C}$, $15{^{\circ}C}$, and ambient temperature). It was found that Charybdis japonica is comprised of moisture (77.5%), ash (2.3%), crude protein (19.7%), and crude lipid (0.5%). The content of crude protein and crude lipid during the storage period tended to decrease over time.

Chemical Compositions and Antioxidative Activity of Leek (Allium tuberusum) Seeds (부추(Allium tuberusum) 씨의 이화학적 특성과 항산화 활성)

  • The chemical compositions as amino acids, minerals, fatty acids, and total polyphenolic compounds of the seeds of leek (Allium tuberusum) were analyzed. The antioxidative activity of water soluble extract from leek seeds was also tested in DPPH ($\alpha$, $\alpha$ - diphenyl-$\beta$ -picrylhydrazyl) method. The chemical compositions of leek seeds were moisture 4.4%, curde protein 25.7%, crude fat 16.6%, and crude ash 2.9%. Major amino acid compositions were proline 11 g, glutamic acid 4.9 g, arginine 2.1g, aspartic acid 1.6g, leucine 1.3g, valine 1.2 g, and methionine 1.1 g as per 100g. Mineral contents were K 215 ppm, Ca 142 ppm, Fe 124 ppm, and Mg 100 ppm. Major fatty acid compositions were linoleic acid 71.9%, oleic acid 12.7%, palmitic acid 8.6%, and stearic acid 1.4%. The changes of contents in polyphenolic compound from leek seeds caused by heat treatment were also listed in the following order; $20^{\circ}C$(364mg/100g), $40^{\circ}C$(462 mg/100g), and $60^{\circ}C$(551 mg/100g). Antioxidative activity as electron donating ability showed in the following order; 0.05% BHT(butylated hydroxytoluene)(45.6%)>0.05% water-extract(31.3%)>0.1% water extract(30.3%). On the basis of chemical analysis, the leek seedsshowed to have relatively high contents of nutrients as amino acids, minerals, fatty acids.
