Inhibition of Trypsin-Induced Mast Cell Activation by Water Fraction of Lonicera japonica

  • Lonicera japonica Thunb.(Caprifoliaceae) has long been known as an anti-inflammatory. In the present study, the effect of water fraction of Lonicera japonica (LJ) on trypsin-induced mast cell activation was examined. HMC-1 cells were stimulated with trypsin (100 nM) in the presence or absence of LJ (10, 100, and 1000 $\mu$ g/mL). TNF-$\alpha$ and tryptase production were measured by enzyme-linked immunosorbent assay (ELISA) and reverse transcription-PCR. Extracellular signal-regulated kinase (ERK) phosphorylation was assessed by Western blot. Trypsin activity was measured by using Bz-DL-Arg-p-nitroanilide (BAPNA) as substrate. LJ (10, 100, and 1000 $\mu$g/mL) inhibited TNF-$\alpha$ secretion in a dose-dependent manner. LJ (10, 100, and 1000 $\mu$g/mL) also inhibited TNF-$\alpha$ and tryptase mRNA expression in trypsin-stimulated HMC-1. Furthermore, LJ inhibited trypsin-induced ERK phosphorylation. However, LJ did not affect the trypsin activity even 1000 $\mu$g/mL. These results indicate that LJ may inhibit trypsin-induced mast cell activation through the inhibition of ERK phosphorylation than the inhibition of trypsin activity.

Antioxidative Activity of Hot Water Extracts from Medicinal Plants (한약재 열수추출물의 항산화 활성)

  • The antioxidative activity and the related parameters of hot water extracts obtained from 16 medicinal plants were tested. The extraction yield was the highest in Salvia miltiorrhiza Bunge $(36.49\%)$ pH range was $4.00\~5.92$ in all samples. Absorbance at 280 nm was examined to determine aromatic compounds content. The absorbance of $250{\mu}g/mL$ sample was the highest in Prunella vulgaris Linne var. lilacina Nakai (2.872) and below 0.5 in Cirsium maackii, Crataegus pinnatifida Bunge and Zizyphus jujuba Miller. Also, absorbance at 420 nm was high in order of Prunella vulgaris Linne vu. lilacina Nakai (1.312), Zea mays Linne (0.917) and Inula japonica Thunberg (0.725) Total phenolic compounds contents was the highest in Prunella vulgaris Linne var. lilacina Nakai $(5.07{\pm}0.05\;mg/100g)$ and flavonoids contents was 2-fold higher in Salvia miltiorrhiza Bunge $(4.82{\pm}1.16\;mg/100g)$ than the other samples. Electron donating abilities of Zizyphus jujuba Miller, Cornus officinalis Siebold et Zuccarini and Salvia miltiorrhiza Bunge were over $90\%$ at $1000{\mu}g/mL$. Reducing power had similar tendency to electron donating ability while reducing power was significantly lower in samples compared to BHT. Two samples (Prunella vulgaris Linne val. lilacina Nakai and Inula japonica Thunberg) were found to have more than $50\%$ nitrite scavenging effect at $500{\mu}g/mL$ while 8 kinds samples (Zizyphus jujuba Miller, Cornus officinalis Siebold 81 Zuccarini, Chrysanthemum indicum Linne, Prunella vulgaris Linne var. lilacina Nakai, Inula japonica Thunberg, Acanthopanax sessiliflorum Seeman, Salvia miltiorrhiza Bunge and Curcuma longa Linne) showed more than $50\%$ nitrite scavenging effect at $1000{\mu}g/mL$. Prunella vulgaris Linne var. lilacina Nakai showed significantly stronger nitrite scavenging effect than other samples, and its activity was $59.62{\pm}1.573\%$ and $80.58{\pm}0.300\%$ at concentrations of 250 and $1000{\mu}g/mL$, respectively.

Vapour Effect of Kresoxim-methyl on Powdery Mildew of Barley and Cucumber (보리와 오이 흰가루병에 대한 Kresoxim-methyl의 훈증 효과)

  • The protective effect of kresoxim-methyl was investigated on 6 important plant diseases, and the vapour effect of it did on barley and cucumber powdery mildews, respectively. With 2.0 ${\mu}g\;mL^{-1}$ of kresoxim-methyl, its high activities against wheat leaf rust and barley powdery mildew were showed such as 92 and 100%, while activities were very low against rice sheath blight, tomato gray mold, and tomato late blight. In vapour phase control activity of kresoxim-methyl against barley powdery mildew was positively correlated with the applied concentrations, except for azoxystrobin and metominostrobin. With 200 ${\mu}g\;L^{-1}$, its control value was 71.9%. When 1000 ${\mu}g\;m^{-3}$ of kresoxim-methyl in vapour phase was applied on 4 plants of cucumber in a vinyl chamber, 51.1% of control value on kresoxim-methyl-treated cucumber was showed 7 days after the application.

Estimation of Dietary Fiber Intake of College Students (대학생의 식이섬유 섭취에 관한 연구)

  • Three-day dietary records data from 237 college students(male 65, female 172) were used as the basis to estimate total dietary fiber(DF) intake of Korean adults & to investigate the major food sources of fiber. Mean daily DF intake of the subjects was 15.2$\pm$3.7g range being 7.5-34g When related to energy intake women consume more DF(7.7$\pm$1.5/1000kcal) than men(6.8$\pm$1.7g/1000kcal) Mean daily crude fiber(CF) intake of the subjects was 6.2$\pm$2.4g The mean DF:CF ratio for the subjects was 2.5$\pm$0.5 indicating that DF intake is 2-3 times of CF intakes The intakes of the two measures of fiber were highly correlated(r=0.791) The major food sources for DF in Korean are vegetables cereals and fruits and 14.3% respectively In comparision with limited international data on the DF intake by adults DF intake by the subjects was similar to those of developed countries. Present study indicates that DF intakes of the subjects are considerably lower than recommended level and they need to consume more DF than the present level.

Anticancer Effect of Extracts from the Marine and Salted Fish Products. (수산물 및 수산 발효식품의 암세포 억제효과)

  • This study was performed to observe the cytotoxic effect of the various salted fish extracts against cancer cell line, human hepatocellular carcinoma (HepG2) using MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide) method. Urechis unicinctus was the strongest cytotoxic effects among any other traditional salted fish products. The growth inhibition ratio of Urechis unicinctus hot-water extracts was 94.5% at the concentration of 1000$\mu\textrm{g}$/$m\ell$. On the other hand, in case of salted fish methanol extracts, salt-fermented shad gizzard was showed the strongest cytotoxic effects. The growth inhibition ratio of salt-fermented shad gizzard methanol extracts was investigated 90% at the concentration of 1000$\mu\textrm{g}$/.$m\ell$.

  • 2020년대에는, 7조개의 스마트 디바이스의 급증, 70억의 인구, 새로운 고품질 매체에 대한 요구가 급증하면서, 새로운 이동통신 환경 및 비즈니스 모델의 변화에 대한 선제대응이 필요하고, 현재 셀률라의 1000배의 용량, 사용자당 Gbps 서비스, 그리고 사용자당 1000분의 1의 에너지효율을 높이면서도, 사용자와 인터액션과 감성을 만족시키는 서비스에 대한 선도적인 연구개발이 필요하다. 이를 통해서, 현재 이동 통신 사업자들의 투자 대비 수익 감소에 따른 불합리한 투자 및 수익 구조를 개선할 수 있을 것으로 예상한다. 이러한 기술을 바탕으로 5G 이동통신 네트워크는 Early Adpater 시장뿐 아니라, 보편적 모바일 핵심 인프라기반으로 미래의 사회 문제를 해결하고 지식의 양극화를 적은 비용으로 해소할 수 있어야 하며, 또한 창조지식 국가 실현을 위한 성장엔진으로, 새로운 혁신 산업 및 지식산업의 생태계를 창조함으로서, 산업 육성 패러다임의 역할을 할 수 있어야 한다. 본 고에서는 이동통신 환경 및 서비스 변화를 예측하면서, 5G 이동통신에서 실현해야 할 이동통신 기술의 비젼과 그 추진 전략에 대해서 살펴보고자 한다. 그 구체적인 추진 전략으로서 국가 정책 및 연구 개발 활성화 방안을 수립하고, 그 실행을 위한 민관 연합의 협력 기구인 5G 포럼의 구성과 역할에 대해서 살펴본다.

The effect of trypsin and chymotrypsin on the chemotaxis and activation of eosinophil (Trypsin과 chymotrypsin이 호산구 화학주성 및 활성화에 미치는 효과)

  • Background : Eosinophilic leukocytes are prominent cellular participants in the pathogenesis of allergic disease and asthma. Chemotaxis is still a very useful method in evaluating the response of human eosinophil to novel modulators. Degranulated mast cells and activated T lymphocytes are responsible for the pathophysiology of asthma and tryptase is one of most important proteases released after activation of mast cells. The purpose of this study was to investigate the actions of trypsin and chymotrypsin on eosinophils in terms of chemotaxis and activation. Method : Eosinophils were isolated by negative immunoselection from the peripheral blood of atopic donors. Chemotaxis was studied by using micro-Boyden chambers and ECP release was assayed by fluoroimmunoassay. Results : Eosinophil showed a chemotactic response to trypsin. Maximal chemotactic response was with $1000{\mu}g/ml$ trypsin ($56.52{\pm}14.50$/HPF) which was comparable to PAP. But chymotrypsin showed no significant chemotactic response to eosinophils. Trypsin at the concentration of 10, 100, $1000{\mu}g/ml$ induced secretion of ECP, which at the concentration of $10{\mu}g/ml$ represented about 2.7 times of the spontaneous rate of release. Soybean protease inhibitor reduced trypsin induced ECP release. Conclusion : Trypsin can induce chemotactic response to eosinophils and activation of eosinophils that can induce secretion of ECP. On the contrary, chymotrypsin showed no direct effect on eosinophils. We propose a role of trypsin on the chemotaxis and activation of eosinophils.

A Study of Clinical Feature of Premature of Cerebral Palsied Children at Kyoung-Nam${\cdot}$Pusan (부산${\cdot}$경남 일부 뇌성마비아들의 임상특성 연구)

  • The purpose of this study was to research the clinical of cerebral palsy taking physical therapy at the department of physical therapy of various clinics at Kyungnam${\cdot}$Pusan. Among the subjects that was born from January, 1985 to June, 2000, 226 parents was answered to questionary of this study. The results of the study were as follows: 1) During the embryonic period, the cerebral palsied children above 37weeks were 114 subjects(50.9%) and there was 51 subjects(22.8%) between 28weeks to 31weeks and 32weeks to 36weeks. The children below 28weeks were 8 subjects(3.6%) and showed the lowest rate. As compared to the delievery methods, the normal delievery, cesserian section delievery, and forceps delievery was 124 subjects(55.1%), 81(36.0%), 16(7.1%), nad 4(1.8%). Among them the mormal delievery indicated the highest percentage. 2) Compared to the weight during birth time, the above 2500g of 121 subjects(55.3%) showed the highest rate and the 28 subjects(12.8%) had the birth weight of 1000to 1499g. There was 4 subjects(1.8%) below the 1000g. 3) Compared to the birth weight of the pregnancy period, the weight of the cerebral palsied children below 28weeks were 1000g to 1499g and showed the highest rate of 4 subjects(50%). The children between 28weeks to 31weeks and 32weeks to 36weeks were 1500 to 2499g, each 23(47.9%), and 28(54.9%) subject. The weight of the children of the above 37weeks were above 2500g and 94 subjects(87.4%). Therefore, if the period of pregnancy is short, the weight birth would indicate the lower weight than the weight of the other times(p<0.05). 4) The spastic type of the pregnant period had the highest rate and the period was the below 28weeks to 31weeks. The cerebral palsied children of athetoid and mixed type were 6 subjects(13.3%) and 5 subjects(31.9%) between 28 and 31weeks. The mixed type of them was each 15 (31.9%) and 33 (30.6%) subjects between 32 to 36 weeks and the above 37weeks. The mixed type showed a slightly high rate (p<0.05). 5) The spastic type indicated the highest rate in the weight of birth time and especially showed the high rate in the case of 1000 to 1499g. The mixed type indicated a slightly high rate of 17 subjects (25.8%) and 32 subjects (29.1%) in case of 1500 to 2499g and the above 2500g (p<0.05).

Antioxidant, Antimutagenicity and Anticancer Activities of Extracts from Cirsium japonicum var. ussuriense KITAMURA (엉겅퀴 추출물의 항산화성, 항돌연변이원성 및 항암활성 효과)

  • Cirsium japonicum var. ussuriense were extracted with methanol and then fractionated with nhexane, EtOAc and BuOH to get active fractions. And their antioxidant and antimicrobial activities in each fraction were determined. Ethyl acetate and butanol fraction of Cirsium japonicum var. ussuriense showed strong antioxidant activities, but hexane fraction did not show any activities. But in the antimicrobial test, Ethyl acetate fraction showed strong antimicrobial activities except to Aspergillus awamori, Asperigillus niger. Especially, Ethyl acetate fraction showed the strongest activities against Bacillus subtilis. And aqueous fraction showed the strongest activities against Cladosporium herbarum, Hypocrea nigricans. This study was performed to determine the antimutagenic and cytotoxic effect of Cirsium japonicum var. ussuriense methanol extract on Salmonella typhimurium TA98, TA100 and cancer cell lines using Ames test and cytotoxicity assay, respectively. Cancer cell lines include human lung carcinoma(A549), human breast adenocarcinoma(MCF-7) and human hepatocellular carcinoma (Hep3B). Futher fractionations with hexane, ethyl acetate, butanol and water from methanol extract of Cirsium japonicum var. ussuriense were performed to obtain effective fraction, methanol extract showed 60.14% inhibition effect on the mutagenesis induced by MNNG against TA100, while 77% and 72.5% inhibition was observed on the mutagenesis induced by 4NQO against TA98 and TA100, respectively. and methanol extract showed 82.25% and 73.7% inhibitory effect on the mutagenesis induced by Trp-P-1 against TA98 and TA100, respectively. methanol extract showed the strongest effect against A549, MCF-7 and Hep3B at the same concentration compared to those of other fration.
