Anticancer Activity of Petroselinum sativum Seed Extracts on MCF-7 Human Breast Cancer Cells

  • Pharmacological and preventive properties of Petroselinum sativum seed extracts are well known, but the anticancer activity of alcoholic extracts and oil of Petroselinum sativum seeds on human breast cancer cells have not been explored so far. Therefore, the present study was designed to investigate the cytotoxic activities of these extracts against MCF-7 cells. Cells were exposed to 10 to $1000{\mu}g/ml$ of alcoholic seed extract (PSA) and seed oil (PSO) of Petroselinum sativum for 24 h. Post-treatment, percent cell viability was studied by 3-(4, 5-dimethylthiazol-2yl)-2, 5-biphenyl tetrazolium bromide (MTT) and neutral red uptake (NRU) assays, and cellular morphology by phase contrast inverted microscopy. The results showed that PSA and PSO significantly reduced cell viability, and altered the cellular morphology of MCF-7 cells in a concentration dependent manner. Concentrations of $50{\mu}g/ml$ and above of PSA and $100{\mu}g/ml$ and above of PSO were found to be cytotoxic in MCF-7 cells. Cell viability at 50, 100, 250, 500 and $1000{\mu}g/ml$ of PSA was recorded as 81%, 57%, 33%, 8% and 5%, respectively, whereas at 100, 250, 500, and $1000{\mu}g/ml$ of PSO values were 90%, 78%, 62%, and 8%, respectively by MTT assay. MCF-7 cells exposed to 250, 500 and $1000{\mu}g/ml$ of PSA and PSO lost their typical morphology and appeared smaller in size. The data revealed that the treatment with PSA and PSO of Petroselinum sativum induced cell death in MCF-7 cells.

  • A polysaccharide, G009, isolated from Ganoderma lucidum IY 009, was subjected to investigating on general pharmacology. This material at the large oral doses of 1000 and 2000 mg/kg in mice did not exhibit any abnormal behaviors and another effects on central nervous system. It also had no influences on hexobarbital-induced sleeping time, rotarod test and spontaneous activity test at each oral dose of 1000 mg/kg in mice. No effects on the body temperature and on acetic acid induced writhing syndrome in mice were observed with its oral administration at 1000 mg/kg, and the convulsions induced by strychnine and pentetrazole were not inhibited at its oral doses of 1000 mg/kg in mice. The solution of G009 as given intravenously at the doses of 30 and 60 mg/kg in rabbit had no influences on blood pressure and respiration rates and depth. In isolated organs of rat uterus and fundus muscles and guineapig ileum and trachea, it did not show any contraction or relaxation at the concentrations of 2$\times$10$^{-3}$ g/ml, and the contractive actions produced by oxytocin, acetylcholine, serotonin and histamine were not inhibited at the same doses. This material showed no effect on intestinal propulsion test in mice and gastric secretion in rats at the oral doses of 1000 mg/kg. However, it is interesting that the material exhibited potent inhibition of acidified aspirin induced gastric damage at the doses of 500 and 1000 mg/kg in rats.

Comparison of Food Consumption Pattern , Nutrient and Dietary Fiber Intakes between Female College Students and Middle Aged Women in Korea

  • One hundred twenty healthy college students and housewives from urban middle income household were investigated in terms of food consumption patterns, nutrient intake and dietary fiber intake. Total food intake was slightly lower in college students(1059.9g/d) than in housewives(1160.g/d) . However, the ration of animal food to total food was considerably higher in college students(23.7%) than in housewives(16.8%). College students tended to consume more dairly products, meat & products, and cereals & grain products but less vegetables , fruits, fish and shellfish than housewives. Although nutrient intake was not much different (except for total calories, fats, and ascorbic acid), dietary fiber intake was slightly and significantly lower in college students(14.9g/d, 8.3g/1000kcal) than in housewives(16.9g/d, 103g/1000kcal) . That is partially due to college students irregular food habits and the change in their meal patterns from traditional to western diets. There was a significantly positive correlation between dietary fiber intake and thiamin, riboflavin, niacin and crude fiber intake.

  • Ganoderma lucidum이 생산하는 polygalacturonase의 유용 활용 방안을 위한 최적 생산조건을 검토한 결과는 다음과 같다. Polygalacturonase 생산을 위한 최적 배양조건은 $30^{\circ}C$, pH5.5에서 14일이었다. Polygalacturonase 생산을 위한 최적 영양조건은 pectin 10 g, soluble starch 10 g, yeast extract 1 g, peptone 2 g, phenylalanine 1 g, $KH_2PO_4$ 2 g, $MgSO_4$ 0.2 g, $CaCI_2$ 50 mg, $thiamin{\cdot}HCI\;100{\mu}g$, D.W. 1000ml이었다.

톳 자숙액 에탄올 추출물의 항산화 활성 (Antioxidant Activity on Ethanol Extract from Boiled-water of Hizikia fusiformis)

  • Antioxidant activity of the ethanol extract from boiled-water of Hizikia fusiformis (EBH) were compared with those of BHA, L-ascorbic acid, gallic acid, caffeic acid and (-)-catechin. The free radical scavenging ability against DPPH (1,1-diphenyl-2-picrylhydrazyl), authentic peroxynitrite and reducing power were measured as indices of antioxidant activity. EBH showed the potent DPPH radical and peroxynitrite scavenging activities, showing 85.23 and 96.97% at final concentration of $1000{\mu}g/ml$, respectively. The reducing power increased with the increasing amount of EBH (final concentration of 1, 10, 100 and $1000{\mu}g/ml$). Total phenolic content of EBH was 588 mg (-)-catechin/g at the final concentration $1000{\mu}g/ml$. Total phenolic contents correlated with DPPH radical scavenging activity $(R^2=0.766)$ and reducing power $(R^2=0.944)$. These results suggested that EBH could be a natural antioxidative source containing antioxidative components.

간암 및 자궁암 세포주 증식에 미치는 오매 추출물의 영향 (The Effect of Prunus Mume Extracts on the Growth of HepG2 and HeLa Cell Lines)

  • 본 연구에서는 예로부터 민간과 한방에서 널리 이용되어 온 오매를 이용하여 오매 추출물이 간암 및 자궁암 세포주 증식에 미치는 효과를 알아보았다. 오매를 hexane. chloroform, ethylacetate, methanol 및 열수로 순차 추출하여 분획물을 농도별로 조제하였으며, MTT assay, cytotoxicity test 및 현미경 관찰 등을 통하여 암세포의 성장 억제 효과를 알아보았다. Hep G2의 경우 오매의 ethylacetate추출물이 가장 큰 성장 저해 효과를 나타내었으며 MTT assay결과 250 $\mu\textrm{g}$/ml 농도 이상에서부터 유의적인 증식 억제 효과를 보였다. 또한 cytotoxicity test결과 오매의 ethylacetate 추출물 1000$\mu\textrm{g}$/ml농도에서 72시간 배양시 75%의 세포 파괴율을 나타내었으며 현미경 관찰 결과 세포의 모양이 변화되고 죽은 세포가 배양액위에 떠있음을 관찰할 수 있었다. HeLa cell에서는 오매의 hexane 추출물이 가장 큰 저해 효과를 나타내었으며 MTT assay 결과 250 $\mu\textrm{g}$/ml 농도 이상에서부터 유의적인 증식 억제 효과를 보였다 또한 cytotoxicity test 결과 오매의 hexane추출물 1000 $\mu\textrm{g}$/ml농도에서 72시간 배양시 HeLa cell의 68%가 파괴되었으며 현미경 관찰 결과 세포 모양의 형태학적 변화가 일어났다. 정상세포주인 fibroblast에는 오매의 ethylacetate 및 hexane 추출물이 아무런 유의적 영향을 미치지 않았으며 현미경 관찰 시에도 변화가 일어나지 않았다

흰 쥐 모델에서 지연재식 시 dexamethasone과 OP-1의 표면처리가 치주조직 재생 및 항흡수 작용에 미치는 효과 (THE EFFECT OF PERIODONTAL REGENERATION AND ANTI-RESTORATION OF DEXAMETHASONE AND OP-1 FOLLOWING DELAYED REPLANTATION IN RAT MODEL)

  • The purpose of this study was to observe the effect of dexamethasone and osteogenic protein-1(BMP-7) on bone, cementum and periodontal tissue regeneration. A total of 60 Sprague-Dawley white female mice were selected and beta-APN was used for five days to extract the maxillary first molar a traumatically. After the extraction of the teeth, the mesiobuccal root canal was filled with Caviton$^{\circledR}$. The teeth were etched with citric acid for 1 min and coated with one of four different experimental solutions : DEX(500nM/ml), DEX(1000nM/ml), OP-1(100$\mu\textrm{g}$/ml) and OP-1(500$\mu\textrm{g}$/ml) for three minutes depending on the group. All teeth were then replanted under microscope. All replantation procedures were done within 30 minutes. Teeth that were replanted after 30 minutes of bench dry only was used as positive control. All animals were sacrificed at 3 weeks following replantation and histologic observtion was done. The results were as follows ; 1. Active root resorption rate was decreased by the order of OP-1(500$\mu\textrm{g}$/ml), DEX(1000nM/ml), OP-1(100$\mu\textrm{g}$/ml), and DEX(500nM/ml). There was statistically less root resorption in OP-1 (500$\mu\textrm{g}$/ml) and DEX(1000nM/ml) group(P<0.05). 2. The group with higher concentration of dexamethasone(1000nM/ml) had statistically more bone union compared to positive control group(P<0.05),but there were no significant differences among four experimental groups. 3. OP-1(500$\mu\textrm{g}$/ml) and DEX(1000nM/ml) groups showed less degree of inflammation compared to the OP-1(100$\mu\textrm{g}$/ml). DEX(500nM/ml), and positive control group (P<0.05). In conclusion, the group with higher concentration of OP-1 had the best results on root resorption, bone ankylosis and anti-inflammatory effects compared to the other experimental groups, but a long-term study is also necessary to evaluate the exact pharmacological effects of the drugs in the future.

  • 본 논문은 다양한 전기 자동차의 배터리 전압을 위해 150V-1000V의 넓은 출력전압을 만족하고 전기자동차의 수요자원화를 위해 양방향 제어가 가능한 DC-DC 컨버터를 제안한다. 제안하는 컨버터는 절연형 컨버터와 비절연형 컨버터가 결합된 2단 방식의 구조로 연결된 배터리의 전압에 따라 CC(Constant Current) 또는 CP(Constant Power)로 동작한다. 전체시스템은 20kW급으로 설계하여 시뮬레이션 및 시제품을 통해 출력특성 및 효율을 분석하였다.

Escherichia coli O157:H7균에 대한 녹차 추출물 및 분획물의 항균효과 (Antimicrobial Activities of Green Tea Extract and Fractions on the E. coli O157:H7)

  • 본 연구는 시판되는 증자 녹차의 에탄을 추출물과 에탄을 추출물로부터 분획한 분획물로부터 최근 병원성 식중독균으로서 문제시되고 있는 E. coli O157:H7균에 대한 항균성을 검색하였다. 에탄을 추출물로부터 그 유효성분을 조사하기 위하여 핵산, 에틸아세테이트, 클로로포름 및 물로서 분획한 후 MIC를 검색한 결과 에틸아세테이트 분획물이 $500{\mu}g/ml$의 농도로 가장 강한 활성을 나타내었으며, 헥산 분획물은 활성이 없었다. 가장 강한 항균력을 나타낸 에틸아세테이트 분회물의 농도별 항균력 및 pH에 대한 영향을 조사한 결과 균종에 따라 차이는 있지만 3균주 모두 $250-2000{\mu}g/disk$농도에서 생육이 억제되었다. 또한 E. coli O157:H7균은 산성조건에서는 생육이 가능하지만 pH 10 이상의 강한 알칼리 조건에서는 생육이 크게 억제되었다. $35^{\circ}C$에서 E. coli O157:H7 933과 932균에 대한 에틸아세테이트 분획물의 농도별 생육저해 효과를 검색한 결과 두 균주 모두 $250{\mu}g/ml,\;500{\mu}g/ml$의 농도에서는 12시간 이후부터 생육하기 시작하였으나 $1000{\mu}g/ml$의 농도에서는 생육이 크게 저해되었다.

  • Bisphenol A(BPA). a monomer used in the manufacturing epoxy resins and polycarbonates. has been reported to induce estrogenic activity, it has been considered as an environmental endocrine disruptor. But the immunomodulatory effects of BPA exposure have not been systemically evaluated. We investigated whether BPA effects on the ability of immunoglobulin(lg) production of mice. To initiate investigation of BPA-induced alterations of the immune system. BPA at dose of 100. 500,1000 mg/kg b.w./day with or without OVA-antigen for 30 days were orally administered to female ICR mice. Mice were sacrificed and serum was colleted on day 2 following administration of BPA for 30days. Total lgG1. total lgG2a. total lgE. OVA-specific lgG1. OVA-specific lgG2a. and OVA-specific lgE in serum were detetmined and compared with those of non-treated mice. In the groups of BPA with OVA antigen, total 1gG1, total lgG2a, total lgE. OVA-specific lgG1 and OVA-specific lgG2a were significantly decreased at dose of 500mg/kg/day. However, in mice treated with BPA alone, total lgG1, and lgG2 were not much altered and total lgE was significantly increased at dose of 1000mg/kg/day. These results demonstrated the BPA modulates the production of immunoglobulin.
