The present paper was carried out to clear up the polyploidal constitution and the banding pattern of three species in tetraploidal Allium(X=8) through the studies of meiosis, karyotype and G-bands. From the meiotic data and G-banding patterns obtained under this study, it is confirmed that A. tuberosum and A. chinense are autotetraploids, and A. senescens is allotetraploid. Some aneuploids out of the employed species were found; A. senescens is 2n=34, and A. chinense is 2n=33. The chromosome types of these species are meta-and submetacentrics except the sat-chromosomes and the f-chromosomes. G-bands of these species are generally located in the end of each arm. A. senescens is similar in the quantity of heterochromatin with A. chinense, but A. tuberosum has a little than the other species. The quantity of heterochromatin is higher in small groups of chromosome than large ones, and higher in short arms than long arms.
The comparative karyological analysis of the Korean treefrogs, Hyla japonica and Hyla suweonensis, were performed by C-banding method. Heteromorphic sex chromosomes, female heterogamety, has been identified in the 3rd chromosomes of H. suweonensis H. suweonensis seem to have sex chromosomes which are Zz/ZW type. The Z chromosomes contain large amount of constitutive heterochromatin, but little heterochromatin is located in the W chromosomes. This is in contrast to all previously known amphibian and most other vertebrate's W or Y chromosomes, except Gastrotheca oui'ern and G. walken (Schmid et al., 1988).
This study was examined chromosome structure of kidney, fin, blood cell, sexual organ that were differently examined chromosome of a good cell activity from organ, medium, staining methods, instruments with rainbow trout 100. And so we obtained following conclusion. 1. The difference from each organ and medium is that a good cell activity of fin, kidney, sexual organ were obtained in TC-199 medium and a good cell activity in TC-199 medium+PHA(5%). 2. In the difference by staining methods, G-banding and C-banding were analyzed by electron microscope or cytoscan. Among them, heterochromatin of centromere analyzed by C-banding. 3. The zygotene and the leptotene were analyzed in this study. 4. Karyotype, heterochromatin and each stage of cell division were clearly analyzed by cytoscan.

Mosaic trisomy 14 syndrome is a well-known but unusual chromosomal abnormality with a distinct and recognizable phenotype. Array comparative genomic hybridization (CGH) analysis has recently become a widely used method for detecting DNA copy number changes, in place of traditional karyotype analysis. However, the array CGH shows a limitation for detecting the low-level mosaicism. Here, we report the detailed clinical and cytogenetic findings of patient with low-frequency mosaic trisomy 14, initially considered normal based on usual cut-off levels of array CGH, but confirmed by G-banding karyotyping. Our patient had global developmental delay, short stature, congenital heart disease, craniofacial dysmorphic features, and dark skin patches over her whole body. Estimated mosaicism proportion was 23.3% by G-banding karyotyping and 18.0% by array CGH.
The chromosome analyses of blood culture were made of 11 Korean native and 53 crossbred males between the Korean native cattles(K) and Charolais(C), which consisted of $K\times$K, $C\times$K, $C\times$CK, CK$\times$CCK and Charolais synthetic males(CK$\times$CCK or CCK$\times$CK). 1. The diploid(2n=60, XY) Charolais synthetic male has the 29 pairs of acrocentric autosomes, a single large submetacentric X and a small metacentric Y chromosome. 2. The numbers of G-band of karyotype in these males were a few differences in the 8 pairs of autosomes(chromosome 2, 4, 5, 6, 9, 11, 19 and 26) compared to those of purebred Korean native ones. G-banding qualities were not matched in chromosome 16, 19 and 29 with the Korean native males and also in chromosome 14, 20 and 22 with other domestic cattles. 3. The G-banding pattern between chromosome 4-6-7 and 24-25-27 was alomost similar together and the possibilityof misidentification was greater in the G-banded preparations. 4. 1/29 Robertsonian translocation and other abnormalities were not observed among 11 Korean native and 53 crossbred males. This result is considered in relation to limited data and further investigation based on larger samples may be necessary for definite conclusion.

An enzyme analysis of the liver fluke, Clonorchis sinensis from Kimhae, Korea and from Shenyang, China was conducted using a horizontal. starch gel electrophoresis in order to elucidate their genetic relationships. A total of eight enzymes was employed from two different kinds of buffer systems. Two loci from each enzyme of aconitase and esterase (${\alpha}-Na{\;}and{\;}{\beta}-Na$) : and only one locus each from six enzymes, gluucose-6-phosphate dehydrogenase (G6PD), ${\alpha}-glycerophosphate$ dehydrogenase (GPD), 3-hydroxybutyrate dehydrogenase (HBDH), malate dehydrogenase (MDH), phosphoglucose isomerase (PGI), and phosphoglucomutase (PGM) were detected. Most of loci in two populations of C. sinensis showed homozygous monomorphic banding patterns and one of them, GPD was specific as genetic markers between two different populations. However, esterase (${\alpha}-Na$), GPD, HBDH and PGI loci showed polymorphic banding patterns. Two populations of C. sinensis were more closely clustered within the range of genetic identity value of 0.998-1.0. In summarizing the above results, two populations of C. sinensis employed in this study showed mostly monomorphic enzyme protein banding patterns, and genetic differences specific between two populations.
A technique is described for producing high resolution G- and R-banded chromosomes in human peripheral lymphocyte cultures. Cultured lymphocyte cells were exposed to ethidium bromide ($10{\mu}g/ml$) and colcemid ($0.02{\mu}g/ml$) each for 2.5h and 0.5h prior to harvest for high resolution G-banded chromosomes. High resolution R-band patterns were obtained by BrdU substitution which was revealed by the fluorochrome-photolysis-Giemsa staining technique. These methods are easy to perform and highly reliable. The data on relative length of chromosomes at the four mitotic stages are presented in units of percentage of haploid autosome length. The characteristic patterns of GTG-bands (G-bands after trypsin and Giemsa) and RBG bands (R-bands after BrdU and Giemsa) were analyzed.
Nucleolus Organizer Regions (NORs) are the specific chromosome sites where ribosomal genes are located and highly expressed. We have applied the AgNOR staining to identify the distribution of NORs in the chromosomes of Korean Cattle. We have also studied the NORs pattern on the cells originated from different breeds, tissues and sex. Peripheral blood from forty-four Korean Cattle and Holstein was cultured for chromosme preparation. The fibroblast culture from biopsied ear skins was also conducted for chromosome analysis. The distribution of NORs was analyzed by sequential Ag staining and G-banding on metaphases of the cells. In Korean Cattle, the NORs are localized on the telomeres of the five chromosome pairs number 2, 3, 4, 11 and 28. The number of NORs per metaphase ranged from 2 to 10 giving a mean value of 5.6. The number of NORs per cell varied among individuals and cells within same individual. The size of NORs also differed in NO-chromosomes. The number of NORs was significantly different between Korean Cattle and Holstein, fibroblasts and lymphocytes, and male and female. However, the distribution and frequency of NORs were similar among the cells regardless of breeds, tissues, and sex.

Background: Cytogenetic abnormalities have important implications in diagnosis and prognosis of acute leukemia and are now considered an important part of the diagnostic workup at presentation. Karyotype, if known at the time of diagnosis, guides physicians to plan appropriate management strategies for their patients. Aim and Objectives: To determine the cytogenetic profile of acute lymphoblastic leukemia (ALL) in Pakistani children in order to have insights regarding behavior of the disease. Materials and Methods: A retrospective analysis of all the cases of ALL (<15years old) diagnosed at Aga Khan University from January 2006 to June 2011 was performed. Cytogenetic analysis was made for all cases using the trypsin-Giemsa banding technique. Karyotypes were interpreted using the International System for Human Cytogenetic Nomenclature (ISCN) criteria. Results: A total of 153 patients were diagnosed as ALL during the study period, of which 127 samples successfully yielded metaphase chromosomes. The male to female ratio was 1.8:1. A normal karyotype was present in 51.2% (n=65) of the cases whereas 48.8% (n=62) had an abnormal karyotype. Most of the abnormal cases showed hyperdiploidy(13.4%) followed by t(9;22)(q34;q11.2) (7.08%). Conclusions: This study revealed a relative lack of good prognostic cytogenetic aberrations in Pakistani children with ALL.
