Two-Stage Fed-Batch Culture of Candida magnoliae for the Production of Erythritol using an Industrial Medium (산업용 배지를 이용한 Candida magnoliae의 2단계 유가식 배양에서 에리스리톨의 생산)

  • Experiments were carried out to select an industrial nitrogen source and optimize erythritol production by Candida magnoliae in fed-batch culture. Among the industrial nitrogen sources tested, light steep water (LSW) was found to be the best nitrogen source for producing erythritol, based on erythritol yield and raw material price. The maximum erythritol concentration obtained a 131.6 g/L, with a 52.6% yield and 0.52 g/L-hr productivity from a 250 g/L glucose and 43.3 mL/L LSW in batch culture. Two-stage fed-batch culture was chosen to improve the volumetric productivity and the yield of erythritol. High cell density culture in cell growth stage was achieved by batch type culture containing 100 g/L glucose and 500 mL/L LSW. The cell concentration was 71.0 g/L after 23 hours of culture. Erythritol productivity was decreased by increasing glucose concentration in the production stage. But 37.3% of the maximum erythritol yield was obtained with 185.5 g/L of erythritol and 1.66 g/L-hr of productivity when 820 g of glucose powder was directly added to a concentration of 450 g/L glucose in production stage.

High Production of L-Threonine using Controlled Feeding of L-Methionine and Phosphate by Escherichia coli Mutant (L-Methionine과 Phosphate의 제한 공급에 의한 Escherichia coli MT201로부터의 고농도 L-Threonine 생산)

  • L-Threonine fermentation process was constructed on batch and fed-batch culture by using Escherichia coli MT201. The production type of L-threonine was observed as growth-associated production in batch culture. In fed-batch culture studying optimal concentration of yeast extract in feeding media, when 600 g/l of glucose and 60 g/l of yeast extract were added in feeding media, 87 g/$\ell$ of L-threonine was produced. To improve cell growth and L-threonine production, the culture of high cell density was performed in fed-batch culture with oxygen enriched air and feeding media containing L-methionine and phosphate. Under the conditions, we could achieve the highest L-threonine production of98 g/$\ell$ at 60 h. The highest productivity of L-threonine was about 3.85 g/$\ell$/h.

Optimization of Xylitol Production by Candida tropicalis in Two-stage Fed-batch Culture (Candida tropicalis의 2단계 유가식 배양에 의한 Xylitol 생산의 최적화)

  • Two-stage fed-batch culture of Candide tropicalis that was designated primarily to cultivate the cell in the glucose medium (1st stage) and then produced the xylitol from xylose medium (2nd stage) was developed to improve a xylitol yield and productivity. In the growth stage, glucose was automatically supplied to the fermentor by pH-stat mode when the pH was up 5.7, When a feeding medium was added in order to reach the glucose and yeast extract concentrations up to 100 and 40 g/L, respectively, a high cell concentration and a relatively low ethanol concentration were obtained in 18.5 h culture. In the production stage, initial xylose concentration of 150 g/L was the most favorable for obtaining the final xylitol concentration and productivity. The addition of mineral salts was also enhanced a xylitol production. But the aeration rate was not significantly affected a xylitol production. When the addition of 16 g yeast extract and 232.5 g xylose powder at the production stage was used, xylitol yield and productivity were significantly increased. With these conditions, xylitol concentration, yield and productivity of 108.9 g/L, 74%) and 3.3 g/L·h, respectively, were obtained in a final volume of 1.58 L. The further addition of 16 g yeast extract and 232.5 g xylose powder increased the working volume partly (1.67 L) and resulted in a relatively high xylitol concentration, yield and productivity of 193 g/L, 70% and 3.6 g/L·h, respectively.

Composition of Vitamin A, E, $B_l$ and $B_2$ Contents in Korean Cow's Raw Milk in Korea (국내산 원유 중 비타민 A, E, $B_l$$B_2$ 함량에 관한 연구)

  • This study was performed to investigate the changes of vitamin A, vitamin E, vitamin $B_l$ and vitamin $B_2$ contents in cow's raw milk collected from dairy farms in Chungcheong-do and Jeolla-do for a year. The contents of fat soluble vitamin A and E were changed as seasonal effect, but water soluble vitamin $B_l$ and $B_2$ contents were not changed as seasonal effect. Vitamin A content in cow's raw milk was as follows [minimum ${\sim}$maximum (mean), ug/100 mL]; $35.1{\sim}59.0$ (44.4) in spring, $36.7{\sim}65.6$ (50.0) in summer, $28.7{\sim}61.2$ (46.8) in autumn and $29.9{\sim}57.8$ (43.1) in winter. In case of vitamin E was as follows [minimum${\sim}$maximum (mean), ug/100 mL]; $28.3{\sim}59.2$ (45.8) in spring, $39.6{\sim}69.9$ (58.8) in summer, $35.0{\sim}62.8$ (46.2) in autumn and $26.0{\sim}55.4$ (41.5) in winter. In case of vitamin $B_l$ was as follow [minimum${\sim}$maximum (mean), ug/100 mL]; $27.7{\sim}57.9$ (42.84) in spring, $32.4{\sim}66.1$ (49.39) in summer, $34.1{\sim}63.7$ (46.69) in autumn and $20.6{\sim}61.4$ (43.20 in winter. The amounts of vitamin $B_2$ in cow's raw milk was as follows [minimum${\sim}$maximum (mean), ug/100 mL]; $150{\sim}182$ (160 in spring, $145{\sim}185$ (163) in summer, $149{\sim}180$ (166) in autumn and $148{\sim}190$ (167) in winter.

Proguction of $_{L}$-Iysine by Continuous Culture of Corynebacterium glutamicum (Corynebacterium glutamicum의 연속배양에 의한 $_{L}$-Iysine 생산)

  • Fed-batch culture, single stage and two stage continuous cultures of Corynebacterium glutamicum SH 35 for the production of $_{L}$-Iysine were performed and compared. In the case of fed batch culture, $_{L}$-Iysine concentration, $_{L}$-Iysine yield and $_{L}$-Iysine productivity was 129.2 g/L, 47.0% and 3.08 g/L/h, respectively. In a single-stage continuous culture, optimum dilution rate and pH was 0.1 h$^{-1}$ and 6.9, respectively, and optimum concentration of sugar and ammonium sulfate in a medium reservoir was 108 g/L and 25 g/L, respectively. Under the optimized conditions, 67 of cell concentration($OD_{610}$), 44.2 g/L of lysine concentration, 41% of $_{L}$-Iysine yield and 4.39 g$L^{-1}$ of $_{L}$-Iysine productivity were obtained. In a two-stage continuous culture, optimum dilution rate was 0.075 $h^{-1}$. Under the conditions, 103 of cell concentration($OD_{610}$) 84.0g/L of $_{L}$-Iysine concentration and 46% of $_{L}$-Iysine yield were obtained.

Direct Alcohol Fermentation of Starch by Schwanniomyces castellii (Schwanniomyces castellii에 의한 전분의 직접 알콜발효)

  • Alcohol fermentations were carried out to confirm the capacity of ethanol production from glucose, starch and soluble starch(dextrin) by Schwanniomyces castellii NRRL Y-2477. Schw. castellii NRRL Y-2477 was able to produce the 63.9g/l ethanol using 94% subtrate from 150g/l-glucose medium. The direct alcohol fermentation of starch having the maximum solubility of 20g/1 at $30^{\circ}C$ yielded 9.1g/l ethanol upon complete depletion of starch, whereas 34.5g/1 ethanol was produced by utilizing 82% of 100g/1 soluble starch medium. The fermentation of 150g/1 soluble starch produced 52.1g/l ethanol using about 79% of substrate. Thus, it was found that the limiting step of direct alcohol fermentation of starch by Schwanniomyces castellii NRRL Y-2477 was a hydrolysis of starch.

  • An injective coloring of a graph G is an assignment of colors to the vertices of G so that any two vertices with a common neighbor receive distinct colors. A graph G is said to be injectively $k$-choosable if any list $L(v)$ of size at least $k$ for every vertex $v$ allows an injective coloring ${\phi}(v)$ such that ${\phi}(v){\in}L(v)$ for every $v{\in}V(G)$. The least $k$ for which G is injectively $k$-choosable is the injective choosability number of G, denoted by ${\chi}^l_i(G)$. In this paper, we obtain new sufficient conditions to be ${\chi}^l_i(G)={\Delta}(G)$. Maximum average degree, mad(G), is defined by mad(G) = max{2e(H)/n(H) : H is a subgraph of G}. We prove that if mad(G) < $\frac{8k-3}{3k}$, then ${\chi}^l_i(G)={\Delta}(G)$ where $k={\Delta}(G)$ and ${\Delta}(G){\geq}6$. In addition, when ${\Delta}(G)=5$ we prove that ${\chi}^l_i(G)={\Delta}(G)$ if mad(G) < $\frac{17}{7}$, and when ${\Delta}(G)=4$ we prove that ${\chi}^l_i(G)={\Delta}(G)$ if mad(G) < $\frac{7}{3}$. These results generalize some of previous results in [1, 4].

The Effect of Concentration of Glucose and Salts on both the Growth and the production of Lipid and DHA of Thraustochytrium aureum ATCC 34304 (당농도 및 염농도가 해양미생물 Thraustochytrium aureum ATCC 34304의 성장 및 지질과 DHA 생성에 미치는 영향 규명)

  • The polyunsaturated fatty acids (PUFAs) are mostly synthesized by the marine original microorganisms. In this study, the effect of concentrations of glucose, NaCl, and $MgSO_4{\cdot}7H_2O$ on both the growth and the production of lipid and Docosahexaenoic acid (DHA) was investigated using Thraustochytrium aureum ATCC 34304. $Y_{Lipid/X}\;and\;Y_{DHA/Lipid}$ increased according to the increase of initial glucose concentration until 15 g/L of glucose. The maximum values of $Y_{Lipid/X}\;and\;Y_{DHA/Lipid}$ were 0.18 g/g and 0.41 g/g respectively at 15 g/L of glucose. The biomass yield, $Y_{x/s}$, however, was constant as 0.378 g/g regardless of initial glucose concentrations in the range of 5 g/L to 25 g/L. The specific growth rate of T. aureum increased also with initial glucose concentration from 5 g/L and 15 g/L. The maximum value was $0.79\;day^{-1}$ at 15 g/L of glucose, and the growth rate decreased to $0.57\;day^{-1}$ and remained constant according to initial sugar concentration in the range of higher than 15 g/L of glucose. The concentration of NaCl was changed from 0 g/L to 48 g/L, in order to study the characteristics of the growth and the production of lipid and DHA according to NaCl concentration. The growth and the production of lipid as well as DHA stopped all at 0 g/L of NaCl and the maximum values of all the three variables occurred at 24 g/L. The effect of $MgSO_4{\cdot}7H_2O$ concentration was also investigated not to find the growth and the production of lipid and DHA at 0 g/L. However, the growth and the Production with $MgSO_4{\cdot}7H_2O$ concentration increased to reach the maximum values at 18 g/L of $MgSO_4{\cdot}7H_2O$.

L-Arabinose Production from Diluted Sulfuric Acid Hydrolysis of Corn-fiber (Corn-fiber의 희석된 황산 가수분해에 의한 L-arabinose의 생산)

  • The demand of L-arabinose has been increased recently because of its advantages including clinical effect. L-arabinose can be produced from dilute acid hydrolysis of agricultural wastes. In this study, optimum conditions of L-arabinose production using dilute acid hydrolysis of agricultural wastes and nutshells were determined. Among the tested various agricultural wastes and nutshells, corn fiber was selected as the best raw material for the production of arabinose. The highest arabinose production was achieved an acid hydrolysis of corn fiber for 1 h at 130$^{\circ}C$ with 0.4% sulfuric acid. Above optimal conditions, it was obtained 20.1 g/L glucose, 10.1 g/L xylose, 7.8 g/L arabinose and 1.8 g/L galactose from 90 g/L of corn fiber. For the purification of arabinose, it was carried out to remove all of sugars except arabinose by the Candida tropicalis cultivation of acid hydrolyzate and an organic contaminants such as pigments by the active carbon treatment of fermentation broth. Moreover, experiments were carried out to eliminate an ions by exchange chromatography. Finally, we obtained 3.1 g of partially purified L-arabinose powder with about 40% yield by evaporation and vacuum drying.

A Effect on Physiological Metabolism of Microorganism Which Irradiated by Non-ionization Radiation (비 전리 방사선을 조사한 미생물에서 생리적 대사에 미치는 영향)

  • A Effect on physiological metabolism of microorganism which irradiated by visible light of non-ionization radiation(12,000 Lux) was investigated. The microorganism used in this experiment was a Rhodospirillum rubrum KS-301 of chemosynthetic microorganism. Batch fermentation of glucose were implement, and based on the data resulted from the fermentation. First, physiological characteristic of microorganism which was not irradiated was investigated. As a result, the decrease of the residual quantity of the substance(5.03 g/L - 2.17 g/L) was increased with the quantity of the bacteria(1.08 g/L - 3.14 g/L)and the quantity of the hydrogenous production(0.186 g - 0.3 g) respectively. Second, physiological characteristic of microorganism which was irradiated was investigated. As a result, the decrease of the residual quantity of the substance(13.17 g/L - 5.2 g/L) was increased with the quantity of the bacteria(4.7 g/L - 10.57 g/L)and the quantity of the hydrogenous production(0.186 g - 0.3 g) respectively. As the physiological characteristic of microorganism which was irradiated by visible light of non-ionization radiation(12,000 Lux) was active with its life, but the cell damages irradiated by with gamma-ray, X-ray, electron-ray in ionization radiation were appeared at cell.
