A Study on the Method of Manufacturing Lactic Acid from Seaweed Biomass (해조류 바이오매스로부터 Lactic acid를 제조하는 방법에 관한 연구)

  • With the spread of COVID-19 worldwide, non-face-to-face services have grown rapidly, but at the same time, the problem of plastic waste is getting worse. Accordingly, eco-friendly policies such as carbon neutrality and sustainable circular economy are being promoted worldwide. Due to the high demand for eco-friendly products, the packaging industry is trying to develop eco-friendly packaging materials using PLA and PBAT and create new business models. On the other hand, Ulva australis occurs in large quantities in the southern seas of Korea and off the coast of Jeju Island, causing marine environmental problems. In this study, lactic acid was produced through dilute acid pretreatment, enzymatic saccharification, and fermentation processes to utilize Ulva australis as a new alternative energy raw material. In general, seaweeds vary in carbohydrate content and sugar composition depending on the species, harvest location, and time. Seaweed is mainly composed of polysaccharides such as cellulose, alginate, mannan, and xylan, but does not contain lignin. It is difficult to expect high extraction yield of the complex polysaccharide constituting Ulva australis with only one process. However, the fusion process of dilute acid and enzymatic saccharification presented in this study can extract most of the sugars contained in Ulva australis. Therefore, the fusion process is considered to be able to expect high lactic acid production yield when a commercial-scale production process is established.

A Cyclophilin from Griffithsia japonica Has Thermoprotective Activity and Is Affected by CsA

  • Members of the multifunctional Cyp family have been isolated from a wide range of organisms. However, few functional studies have been performed on the role of these proteins as chaperones in red alga. For studying the function of cDNA GjCyp-1 isolated from the red alga (Griffithsia japonica), we expressed and purified a recombinant GjCyp-1 containing a hexahistidine tag at the amino-terminus in Escherichia coli. An expressed fusion protein, $H_6GjCyp-1$ maintained the stability of E. coli proteins up to $50^{\circ}C$. For a functional bioassay for recombinant $H_6GjCyp-1$, the viability of E. coli cells overexpressing $H_6GjCyp-1$ was compared with that of cells not expressing $H_6GjCyp-1$ at $50^{\circ}C$. After high temperature treatment for 1 h, E. coli overexpressing $H_6GjCyp-1$ survived about three times longer than E. coli lacking $H_6GjCyp-1$. Measurement of the light scattering of luciferase (luc) showed that GjCyp-1 prevents the aggregation of luc during mild heat stress and that the thermoprotective activity of GjCyp-1 is blocked by cyclosporin A (CsA), an inhibitor of Cyps. Furthermore, the Cyp-CsA complex inhibited the growth of E. coli under normal conditions. The results of the GjCyp-1 bioassays as well as in vitro studies strongly suggest that Cyp confers thermotolerance to E. coli.

A Comparative Study on the Aesthetic Characteristics of Contemporary Nomad Fashion and Korean Traditional Costume -Focused on Deleuze's Nomadism Aesthetics- (현대 노마드 패션과 한국 전통복식의 미적 특성 비교연구 -들뢰즈의 노마디즘(유목주의) 미학을 중심으로-)

  • This study focused on similarities between modern philosophical Nomadism and the thoughts of Korean people as well as analyzed the correlation between modern Nomad fashion and Korean traditional costumes. This study aims are as follows. First, to establish aesthetic characteristics by examining modern Nomadism and Korean thoughts. Second, draw the principles of design expressions of modern Nomad fashion and Korean traditional costumes. Third, analyze correlations between them. The method of this study was a literature review and an analysis of related photos. The study results are as follows. First, Nomadic aesthetics are characterized by the formation of complex diversity with the variability of fusion and separation through liquidity in an ambiguous boundary; in addition, various possibilities and harmony are the characteristics of Korean aesthetics, held by a holistic thought based on the vision of the universe of Qi (氣). Second, Nomad fashion appeared as variable designs through repetition and the overlapping of forms, and irregular designs were noticed through repetition and the overlapping of circles, quadrangles, and angles in Korean traditional costumes. Third, similarities in the creation of liquidity, variability, and de-territoriality based on modules can be found; in addition, common principles can also be drawn from the appearing design expressions.

Suggestion for Science Education through the Analysis of Archimedes' Creative Problem Solving Process (Archimedes의 창의적 문제해결과정 분석을 통한 과학교육에의 함의 고찰)

  • In this study, we developed a model for analyzing scientists' creative thinking processes, and analyzed Archimedes' thinking process in solving the golden crown problem. As results show, scientists' complex problem solving processes could be represented as a repeating circular model, and the fusion of processes of diverse thinking required for scientists' creativity could be analyzed from the case. Also in this study, we represented the role of experiments in scientists' creative discovery, and investigated the reasons for the difference between the viewpoints of textbooks and historic facts. We found the importance of abductive reasoning and advance knowledge in creative thinking. Archimedes solved the golden crown problem creatively by crossing the scientific thought of dynamics and the daily thought of baths. In this process, abductive reasoning and advance knowledge played an important role. Besides Archimedes' case, if we would reconstruct the creative discovery processes of diverse scientists' in textbooks, students could raise their creative thinking ability by experiencing these processes as educational steps.

  • Selective laser melting (SLM) is an additive manufacturing process by melting metallic powders and stacking into layers, and can product complex shapes or near-net-shape (NNS) that are difficult to product by conventional processes. Also, SLM process is able to raise the efficiency of production by creating a streamlined manufacturing process. For manufacturing in SLM process using Ti-6Al-4V powder, analysis of microstructural evolution and evaluation of mechanical properties are essential because of rapid melting and solidification process of powders according to high laser power and rapid scan speed. In addition, it requires a post-processing because the soundness and mechanical properties are degraded by defects such as pore, un-melted powder, lack-of-fusion, etc. In this study, hot isostatic press (HIP) was conducted as a post-processing on SLM-printed Ti-6Al-4V alloy. Microstructure of post-processed Ti-6Al-4V alloy was compared to as-built Ti-6Al-4V, and the evolution of quasi-static (Vickers hardness, room temperature tensile characteristic) and dynamic (high-cycle fatigue characteristic) mechanical properties were analyzed.

A Study on the Trace Analysis of Germanium in Inorganic Matrices by Differential Pulse Polarography (Differential Pulse Polarography에 의한 무기물 시료 중 Germanium의 미량 분석에 관한 연구)

  • Analytical method for the determination of trace germanium in inorganic matrices by differential pulse polarography(DPP) was studied. The reduction peak of germanium(IV) in perchloric acid solution containing 1, 2, 3-trihydroxy benzene appeared at -0.45V(vs. Ag/AgCl) and the peak current for germanium complex varied linearly with concentration variation. Factors affecting sensitivity and precision for germanium quantification were studied and detection limit under the investigated parameters was 1ng/ml. Inorganic samples were decomposed by fusion with potassium pyrosulfate. Serious interferences of Se(IV), Pb(II), As(III) for the determination of germanium were discussed. Interferences of these elements could be avoided by extraction of germanium from decomposed matrices by $CCl_4$ in 10M HCl solution. The germanium contents of inorganic samples(Pb bf. dust, Cu bf. dust, gneiss, Cu anode slime) were determined by the above method.

Characterization of the Gene for the Light-Harvesting Peridinin-Chlorophyll-Protein of Alexandrium tamarense

  • Photosynthetic dinoflagellates contain a water-soluble, light-harvesting antenna called the peridinin-chlorophyll-protein (PCP) complex, which has an apoprotein with no sequence similarity to other known proteins. There are two forms of PCP apoproteins; the 15-kDa short form and the 32- to 35­kDa long form. The present study describes the PCP protein and its cDNA from Alexandrium tamarense. A cDNA library was constructed from mRNA isolated from A. tamarense. The complete PCP cDNA was generated by reverse-transcription coupled to polymerase chain reaction (RT-PCR), together with rapid-amplification of cDNA ends (RACE). The A. tamarense PCP cDNA encoded a 55-amino acid signal peptide and a 313-amino acid mature protein with a calculated mass of 32 kDa, which corresponded to that of the long form of PCP. Phylogenetic analysis indicated that the sequence of A. tamarense PCP did not cluster with the short-form PCPs, to which it was only about $55\%$ identical, but which were $79-83\%$ identical to other long-form PCPs. The deduced amino acid sequence of A. tamarense PCP contains an internal duplication, which suggests the possibility that long-form PCPs arose by gene duplication or by the fusion of genes encoding the short form. The abundance of PCP mRNA changed substantially in response to different light conditions, indicating the possible existence of a photo-acclimation response in A. tamarense.

  • The fourth industrial revolution and so on, the manufacturing environment is based on the latest IT technology, and ICT-based convergence that links with various industrial fields is actively proceeding. For this reason, convergence security based on changing environment in the field of security is emerging as a new issue, and various demands for convergence security are increasing. Such changes in the industrial environment not only provide advantages of convergence environment, but also cause various fusion and complex security problems. In particular, the government is actively promoting policies to build 10,000 smart plants by 2020, and has implemented projects to build 2,800 smart plants from 2014 to 2016. Therefore, we will examine the security threats and countermeasures against smart factories that are being built on the basis of ICT rather than the existing closed type.

  • To solve the energy problem and finding new growth engines, interest for the smart grid is increasing and related technologies are making great efforts to secure in the world. AMI (Advanced Metering Infrastructure) Among them is the first to be constructed and getting attention as a key component of smart grid. A fusion of various technologies in technology development and demonstration is underway on Jeju Island Smart Grid Demonstration Complex in Korea, and focusing on broadband power line communication technology infrastructure is actively underway in Korea Electric Power Corporation. AMI system using power line communication technology without building a separate communication lines are available for power supply lines, but communication is impossible in occurs because admission to the power company or the ideal infrastructure for communication is not considered. In this paper, we analyze the requirements to build AMI system using Binary CDMA and powerline communications technology, and design the basic communication protocol based on Binary CDMA, implement network management and relay feature. By doing so, ways to apply Ad-hoc Binary CDMA indigenous technology to the AMI system were derived, and could build a system to make use of Wired (PLC) and wireless (Binary CDMA) simultaneously.

Quality Assessment Model for Practical Wearable Computers (실용적 웨어러블 컴퓨터 품질평가모델)

  • Recently, the progress of smart phone market has retarded by oversupply therefore wearable computer has been the focus of new growth engine. Wearable computing system is a complex fusion of a variety of technologies such as wireless network, embedded, sensor and new material. Because these technologies involves utilization and mobility in addition to quality characteristic in existing software, application of ISO/IEC 9126 is not perfect when assessing quality of wearable computer. In this study, author suggested new quality assessment model for wearable computer by sorting quality attribute in ISO/IEC 9126 and adding new quality attribute. For this, author investigated features and functional requirements related to wearable computer. and then author suggested quality standard and metrics by identifying quality characteristic. Author confirmed practicality of quality assessment model by using suggested model in scenario and comparing quality assessment of three goods such as company S, L, G. This quality assessment model is expected to use guidelines for assessing quality of wearable computer.
