클로탈라리아 시들음병을 일으키는 Fusarium udum의 특성 (Characterization of Fusarium udum Causing Fusarium Wilt of Sunn Hemp in Korea)

  • 클로탈라리아는 국내에서 토양개량을 위한 질소고정, 토양침식 감소, 잡초 및 선충 억제를 위해 풋거름작물로 이용하고 있다. 2014년, 클로탈라리아를 풋거름작물로 재배하는 완주 지역의 포장에서 시들음 증상이 관찰되었다. 감염된 식물체의 잎은 아래잎부터 황화 되면서 시들기 시작하였고, 위쪽 잎도 황화 되었으며, 결국 식물체는 완전히 고사하였다. 감염된 식물체 줄기에서 어두운 색의 자낭각이 다수 관찰되었고, 이 자낭각으로부터 자낭포자를 단포자 분리하여 6개 균주를 분리하였다. 균학적 특성에 의해 분리균은 Fusarium udum(완전세대: Gibberella indica)로 동정되었다. 대형포자의 정단세포는 대부분 갈고리 모양으로 굽어 있고, 소형포자는 단경자에서 false head 상으로 형성되었다. 후벽포자는 균사에서 단일 혹은 무리지어 풍부하게 형성되었다. 이와 같은 동정 결과는 translation elongation factor 1 alpha(TEF), calmodulin (CAL), histone 3 (HIS3) 유전자 염기서열 분석으로 재확인되었다. 그 결과, 분리 균주는 NCBI GanBank에 등록된 F. udum과 TEF 유전자는 94.4~96.2%, CAL 유전자는 99.7%, HIS3 유전자는 99.6~99.8%의 상동성을 보였다. 클로탈라리아와 두 품종의 콩을 대상으로 포자현탁액을 토양에 관주 접종하여 병원성 검정을 수행한 결과, 접종 14~21일 이내에 접종한 클로탈라리아와 태광콩에서 병징이 관찰되었다. 따라서 이 병을 F. udum에 의한 클로탈라리아 시들음병으로 명명하고자 제안하며, 국내에서 처음으로 보고한다.

  • Biological control of Fusarium udum causing wilt disease of pigeonpea was studied in vitro, as well as, in vivo. Aspergilluspavus, Anergillus niger, Bacilius licheniformis (strain-2042), Gliocladium virens, Peniciliium citrimum, and Trichoderma harzianum, which were found to be the most potent ones in inhibiting the radial colony growth of the test pathogen, were used as biological control by amending their inocula at diffeyent concentrations in pots and in pathogen-infested soil in the fields. Maximum reduction of the wilt disease was observed with G. vireos both in pots and in the fields. The population of E. udum was found to be markedly reduced when the antagonists were applied in the soil. The study establishes that G. virens can be exploited for the biological control of wilt disease at field level.

  • The fungicidal effects of four commercial fungicides, two herbicides and two insecticides have been examined on Fusarium udum, causing wilt disease of pigeonpea in vitro and in vivo. The fungicides Bavistin and MeMc inhibited the growth of the test pathogen completely at 8 and 30 ppm. The herbicide Butachlore inhibited the growth of the test pathogen up to 80.4%. The insecticides, Ekalux and Thiodane partially inhibited the radial growth at 1000 ppm. In unsterilized and sterilized soil MeMc was most effective in controlling the disease in comparison to Bavistin and Ekalux. Maximum rhizosphere fungal population was recorded in MeMc amended soil and minimum in case of Bavistin.

Antifungal Activity of Methanolic of Centella asiatica and Andrographis panicuiata

  • The antifungal activity of methanolic extracts of Centella asiatica and Andrographis paniculata leaves was observed against fourteen fungi, viz., Alternaria alternata, A. brassicae, A. brassicicola, A. solani, A. tenuissima, Cercospora blumae, Curvularia lunata, C. penniseti, and Drechslera monoceras, D. oryzae, D. turitica, Fusarium albizziae and F. udum. Different concentrations of the methanolic extract (1000, 2000, 3000, 5000, 7000, 10000ppm) were used. The effect of mixed leaf extract (1500 ppm of C. asiatica + 1500 ppm of A. paniculata) and its 1:2 ad 1:4 dilutions were also studied. The individual extracts of both the plants showed significant inhibitory effect on spore germination of all the fungi tested. F. udum, F. albizzae, D. oryzae, D. turtica, and D. monoceras were particularly sensitive to these extracts. In general, the extract of C. asiatica showed a higher inhibitory effect in all concentrations against all the fungi as compared to A. paniculata, except for A. brassicae A. solani, D. oryzae, D. penniseti and Curvularia sp. The inhibitory effect of extracts increased when they were used in combination with or without dilutions against A. brassicicola, A. solani A. brassicae, A. alternata, A. tenussima, C. blumae, C. lunata, C. penniseti and Curvularia species. Higher efficacy of active ingredient of these extracts under field condition is envisaged against plant pathogens.

  • Narceine methyl ester and narceine are potent alkaloids which were isolated from Corydalis longipes were found effective in vitro at very low concentration, i.e., $100{\sim}500\;ppm$ against spore germination of some test plant pathogenic fungi (Alternaria solani, A. tagetica, Cercospora abelmoschi, Curvularia maculans, Erysiphe cichoracearum, E. pisi, Fusarium udum, Helminthosporium oryzae, H. penniseti, Ustilago cynodontis). Among the test, phytopathogens the spores of F. udum, C. maculans and H. penniseti were highly sensitive at 200 ppm. However, spores of E. pisi, A. solani and A. tagetica were less sensitive at low concentration followed by other test fungi. Most of the fungi showed zero or nearly zero percent spore germination at 400 and 500 ppm.

  • Inheritance of resistance to Fusarium wilt (FW) disease caused by Fusarium udum was investigated in pigeonpea using four different long duration FW resistant genotypes viz., BDN-2004-1, BDN-2001-9, BWR-133 and IPA-234. Based on the $F_2$ segregation pattern, FW resistance has been reported to be governed by one dominant gene in BDN-2004-1 and BDN-2001-9, two duplicate dominant genes in BWR-133 and two dominant complimentary genes in resistance source IPA-234. Further, the efficacy of six simple sequence repeat (SSR) markers namely, ASSR-1, ASSR-23, ASSR-148, ASSR-229, ASSR-363 and ASSR-366 reported to be associated with FW resistance were also tested and concluded that markers ASSR-1, ASSR-23, ASSR-148 will be used for screening of parental genotypes in pigeonpea FW resistance breeding programs. The information on genetics of FW resistance generated from this study would be used, to introgress FW resistance into susceptible but highly adopted cultivars through marker-assisted backcross breeding and in conventional breeding programs.

  • The inhibitory activity of ent-norsecurinine alkaloid was evaluated against spore germination of some plant pathogenic fungi(Curvularia maculans, Curvularia species, C. palliscens, Colletotrichum gloeosporioides, Colletotrichum species, Afternaria solani, A. brassicae, Fusarium udum, Helminthosporium echinoclova and H. penniseti). It inhibited spore germination of all the test fungi. C. maculans, C. species, and C. palliscens were the most sensitive as complete inhibition of spore germination was observed at 1000 ppm. A. solani was not inhibited by this chemical.

  • Phytoterpenoid isolated from Artabotrytis odoratissimus inhibited spore germination of some plant pathogenic as well as saprophytic fungi e.g. Alternaria alternata, A. solani, Cercospora sp., Curvularia maculans, C. pennisetti, Fusarium udum, Helminthosporium echinochlova, H. frumentacie, H. penniseti and Ustilago cynodontis. In Curvularia maculans and H. frumentacie, spore germination was completely inhibited at 2000 ppm. However, Curvularia maculans and C. pennisetti showed considerable sensitivity to this chemical even at 500 ppm.

It inhibited mild spore germination of all the fungi tested. Curvularia lunata, Curvularia sp., Collectotrichum sp., C. musae and Heterosporium sp. were most sensitive as complete inhibition of spore germination was observed at very low concentrations.

Effect of 1-corydalmine, an Alkaloid Isolated from Corydalis chaerophylla Roots on Spore Germination of Some Fungi

  • 1-Corydalmine, an alkaloid isolated from roots of Corydalis ehaerophylla inhibited spore germination of some plant pathogenic as well as saprophytic fungi e.g. Alternaria brassicae, A. brassicieola, A. solani, Curvularia lunata, C. maculans, C. sp., C. pallseens, Erysiphe pisi, Fusarium udum, Helminthosporium species, H. penniseti and a Heterosporium species. 1-Corydalmine significantly inhibited spore germination of all the fungi at 100 to 1500 ppm. It was effective against all the fungi at 1500 ppm. C. lunata was highly sensitive to this chemical even at 250 ppm.
