Isolation and Phylogenetic Analysis of Orchid Rotting Fungus, Fusarium sp. KS-01 (난 썩음병균 Fusarium sp. KS-01의 분리 및 계통학적 분석)

  • A orchid rotting fungus was isolated and identified. The isolate was consistent with the genus Fusarium in morphological and cultural characteristics. The partial 18S rRNA sequence of the isolate showed high similarity with anamorph or telemorph of Fusarium and other Fusarium species. In phylogenetic analysis, the isolates was poorly related to other Fusarium species. The isolate closely related to Fusarium sp. LP-A2/3.

Germination of Some Fungal Spores under Different Soil Conditions (토양조건(土壤條件)에 따른 수종(數種) 진균포자(眞菌胞子)의 발아(發芽))

  • To investigate germination characteristics of soils on Fusarium species under different soil conditions, this study was carried out to test spore germination of 4 Fusarium species. Among 102 soil samples, spore germinations of Fusarium solani f. sp. pisi, F. oxysporum f. sp. cucumerium, F. oxysporum f. sp. raphani and F. coccophilum were under 50% in 98, 85, 82 and 83 soil samples, respectively. The highest spore germination of F. solani f. sp. pisi, F. oxysporum f. sp. cucumerium and F. coccophilum was obtained in 21 soil samples of pH 5.1-6.0, whereas F. oxysporum f. sp. raphani was in 33 soil samples of pH 4.1-5.0. The highest spore germination of F. solani f. sp. pisi, F. coccophilum and F. oxysporum f. sp. cucumerium was obtained in 4 soil samples holding soil moisture content of 41-60%, while F. oxysporum f. sp. raphani was in 22 soil samples of 21-30%. F. oxysporum f. sp. raphani only showed a direct correlation(r=0.29) between spore germination and soil moisture content. On the basis of each spore germination in cultivated and non-cultivated soils, there was significant difference(p=0.01) in 4 Fusarium species. F. solani f. sp. pisi only indicated significant difference(p=0.0l) between two contrasting-soils, cultivated and non-cultivated soils. The numbers of microbial population were higher in soils suppressive to spore germination than soils conducive to spore germination.

Taxonomy and Identification of Fungi Isolated from Round Bale Silage (원형 곤포사일리지에 발생한 곰팡이의 분류 동정)

  • To identification of fungi that occurs round bale silages, 253 fungal contaminated samples were collected from 2009 to 2011. Total 253 silage samples from Italian ryegrass, sudan grass, rye, corn, barley and oat were analysed. Total 270 strains were purely isolated from contaminated round bale silages. The fungi were identified with morphological characteristics and rDNA sequence analysis. Nineteen species of fungi(Rhizopus sp., Fusarium spp., Coprinus sp., Blastomyces sp., Aureobasidium sp., Polypaecilum sp., Botryoderma sp., Mucor sp., Scytalidium sp., Sphaeropsis sp., Aspergillus spp., Trichocladium sp., Humicola sp., Staphylotrichum sp., Periconia sp., Verticillium sp., Diplococcium sp., Penicillium spp. and Trichoderma spp.) were identified by morphological characteristics. On the other hand, fungi isolated from silage were identified to Acremonium strictum, Aspergillus tubingensis, Bionectria ochroleuca, Dipodascaceae sp., Fusarium proliferatum, Fusarium oxysporum, Fusrium solani, Gelasinospora reticulata, Gibberella moniliformis, Gibberella zeae, Nectria mauritiicola, Penicillium paneum, Pseudallecheria boydii, Schizophyllum commune, Scopulariopsis brevicaulis and Simplicillium lamellicola by rDNA sequence analysis. Penicillium sp. and Trichoderma sp., were isolated 74 and 64 strains, respectively. Humicola sp., Aspergillus sp., Coprinus sp., and Fusarium spp. were identified 10 to 30 strains. Most fungi were isolated together with more than one species in a sample looked like one species with the naked eyes.

  • The vegetative nuclear divisions in hyphae and chromosome numbers were studied with the aid of Giemsa-HCl techniques from 10 strains of Fusarium oxysporum. The entire nuclear division process occurred within an intact nuclear envelope like other fungus. The results confirmed that 2 strains(F. oxysporum S Hongchun D2, F. oxysporum S Jinyang 4) were n=4; 3 strains(F. oxysporum f. sp. lini KFCC 32585, F. oxysporum f. sp. melongenae KFCC 34743 and F. oxysporum f. sp. raphani) n=5; 2 strains(F. oxysporum f. sp. vasinfectum, and F. oxysporum f. sp. mori KFCC 34742) n=6; 3 strains(F. oxysporum f. sp. cucumerium, F. oxysporum f. sp.niveum, and F. oxysporum f. sp. pisi) n=7.

Biological Control of Fusarium Wilt of Carnation Plants by Antagonistic Microorganism in Greenhouse (카네이션의 시설재배에서 길항성 세균을 이용한 Fusarium Wilt 의 생물학적 방제)

  • This study was carried out to screen and select the effects of antifungal bacterial strains which inhibit the growth of plant pathogen, Fusarium oxysporum(fusarium wilt) occurred in carnation plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it's identification. Twenty bacterial strains which strongly inhibited Fusarium oxysporum were isolated from roots of carnation plants and the soil in greenhouse, and the best antifungal bacteria designated as C121, was finally selected. Antagonistic bacterial strain, C121 was identified to be the genus Bacillus sp. based on the morphological, biochemical and cultural characterizations. The Bacillus sp. C121 showed 58.1% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the cultural broth and the heat bacterialization culture filtrate of it, Bacillus sp. C121 was shown 92.1% and 21.0% of antifungal activity, respectively.

  • This study was carried out to investigate the germination of four fungi under non-leaching and leaching incubation. The range of germination rates of Fusarium solani f. sp. pisi, Fusarium oxysporum f. sp. cucumerium, Mortierella, and Helminthosporium sativa incubated on sand saturated with various solutions without leaching were 90-99%, 31-45%, 38-62% and 86-98% respectively. The range of germination rates of Fusarium solani f. sp. pisi, Fusarium oxysporum f. sp. cucumerium, Mortierella n. sp and Helminthosporum sativa incubated on sand undergoing leaching were 51-63%, 13-25%, 15-37% and 47-80%, respectively. The germination rates of the four fungi tested were suppressed under leaching incubation.

  • This study was carried out to clarify the effects of antagonistic microorganism, Bacillus sp. JC181 isolated from the greenhouse soil grown cucumber plants on the growth inhibition of plant pathogen, fusarium wilt (Fusarium oxysporum) occurred in cucumber plants in greenhouse. Antagonistic bacterial strains were isolated and were investigated into the antifungal activity of the antagonistic microorganism against fusarium wilt. Screened fourteen bacterial strains which strongly inhibited F. oxysporum were isolated from thc greenhouse soil grown cucumber plants, and the best antagonistic bacterial strain designated as JC181, was finally selected. Antagonistic bacterial strain JC181 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. Bacillus sp. JC181 showed 58.2% of antifungal activity against the plant pathogen growth of F. oxysporum. By the bacterialization of culture broth and heated filtrates of culture broth, Bacterial strain, Bacillus sp. JC181. showed 91.2% and 260% of antifungal activity against F. oxysporum, respectivrly.

Occurrence of Fusarium Wilt on Lisianthus (Eustoma grandiflorum) Caused by Fusarium oxysporum f. sp. eustomae (Fusarium oxysporum f. sp. eustomae에 의한 꽃도라지 시들음병(가칭) 발생)

  • Fusarium wilt of lisianthus occurred severely throughout all cultivation areas, especially in alpoine areas during summer season and the disease incidence was 5 to 30 percent in Korea. The major symptoms of the disease were wilt with chlorosis and water deficiency, stunted plants and scorched leaves. Severe symptoms appeared just after high temperature period around late August in Daekwallyong area. Whit and pale red colored mycelia were developed on stems of infected plants near the soil surface. The causal organism of Fusarium wilt of lisianthus was isolated and identified as Fusarium oxysporum f. sp. eustomae on the basis of pathogenic and cultural characteristics. The causal organism was reisolated from all infected and inoculated stems of plants. This is the first report referring to F. oxysporum f. sp. eustomae of lisianthus in Korea.

RAPD Analysis for the Evaluation of Genetic Diversity Among the Fusarium Species from Various Sources (각종 작물로부터 분리한 Fusarium속 균의 RAPD 기법을 이용한 유전분석)

  • In this study, we evaluated the use of RAPD method to discriminate among strains of Fusarium species including F. oxysporum and f. sp. of F. oxysporum. As a result of the amplication, fifteen primers showed total 180 bands ranging from 0.2 to 3 Kb. Among those 180 bands, 126 polymorphic bands were used for bionominal matrix code (0, 1), and UPGMA dendrogram analysis. Fusarium oxysporum isolate 355 showed high similarity with F. oxysporum isolate 358 at 0.9603. Fusarium roseum isolate 87 and F. oxysporum isolate 358, F. o. f. sp. lycopersici isolate 69 and F. o. f. sp. melongena 68 showed low similarity of 0.3809. Fusarium oxysporum isolate 361 and F. o. f. sp. raphani isolate 218 showed similarity of 0.8730, F. oxysoprum isolate 354 and unidentified Fusarium sp. isolate 228 showed similarity matrix of 0.7936, and F. roseum isolate 87 and F. o. f. sp. raphani isolate 57 showed similarity matrix of 0.5873.
