잿빛곰팡이균(Botrytis cinerea)에 대한 N-phenylbenzenesulfonamide 유도체들의 살균활성에 관한 CoMFA 분석 (CoMFA Analyses on the Fungicidal Activity with N-phenylbenzensulfonamide Analogues against Gray Mold (Botrytis cinerea))

  • 잿빛곰팡이균(Botrytis cinerea)에 대한 N-phenylbenzenesulfonamides 유도체(1-45)들의 살균활성에 관한 비교 분자장 분석(CoMFA)을 정량적으로 검토하였다. 통계적으로 CoMFA 모델의 예측성과 상관성이 비교분자 유사성 지수분석(CoMSIA) 모델보다 월등히 좋았다. 최적화 된 CoMFA I 모델의 통계값은 예측성이 $r^2_{cv.}(or\;q^2)=0.457$ 그리고 상관성이 $r^2_{ncv.}=0.959$이었고 살균활성은 기질 분자들의 입체장(51.9%)과 정전기장(35.6%)에 의존적이었다. 또한 progressive scrambling 분석으로 얻어진 섭동에 대한 감도($d_q^{2'}/dr^2_{yy'}=0.898$)와 예측성($q^2=0.346$ 및 SDEP=0.614)에 의하여 최적의 CoMFA I 모델은 우연 상관성에 의존적이지 않음을 알았다. 그러므로 CoMFA I 모델의 등고도 분석 결과로부터, N-phenyl 고리상 $R_3$$R_4$-치환기는 입체적으로 크고 $R_1$-치환기로서 S-phenyl 고리상 para-치환기는 입체적으로 작은 치환체가 살균활성에 기여 할 것으로 기대되었으며 최적화 된 CoMFA I 모델은 잿빛곰팡이균에 대한 살균활성을 예측하는데 유용하게 활용될 수 있을 것이다.

천연물 Polygodial과 Imidazole계 화합물의 병용에 의한 항진균 활성의 증진 (Synergistic Effect of Polygodial with Imidazole Drugs on the Antifungal Activity)

  • The fungistatic and fungicidal activities of amphotericin B, fluconazole, miconazole, econazole, and 5-fluorocytosine against Saccharomyces cerevisiae were estimated in the presence of 1/2 minimum inhibitory concentration (MIC) and 1/2 minimum fungicidal concentration (MFC) of polygodial, respectively. Among them, the antifungal activities of miconazoles by polygodial was still shown against several yeast-like fungi including Candida albicans, Candida utilis, Cryptococcus neoformans, except for Candida krusei. The combination of polygodial with imidazole drugs against Saccharomyces cerevisiae was further examined using the macrobroth dilution checkerboard method. The fractional inhibitory concentration (FIC) and the fractional fungicidal concentration (FFC) index between polygodial and miconazole were 0.16 and 0.19, while the combination of polygodial with econazole exhibited the FIC index of 0.19 and the FFC of 0.25, respectively. These results suggest that polygodial and the imidazoles on the fungistatic and fungicidal action are highly synergistic.

상이한 정렬에 따른 N-phenylthionocarbamate 유도체들의 살균활성에 관한 비교 분자장 분석 (Comparative Molecular Field Analyses on the Fungicidal Activities of N-phenylthionocarbamate Derivatives based on Different Alignment Approaches)

  • 새로운 N-phenylthionocarbamate 유도체들의 N-phenyl-치환기(X) 변화에 따른 벼잎집무늬마름병균(Rhizoctonia solani) 및 고추역병균(Phytophthora capsici)에 대한 살균활성과의 3D-QSARs 관계를 상이한 정렬에 따른 비교 분자장 분석(CoMFA) 방법으로 연구하였다. 그 결과, field fit 정렬 조건에서 유도된 CoMFA 모델들이 atom based fit 정렬 조건의 모델보다 양호하였다. 따라서 두 균주에 대한 살균활성을 가장 잘 설명하는 CoMFA 모델들은 field fit 정렬과 CoMFA장들의 조합 조건에서 유도된 CoMFA 모델(RS: RF2 및 PC: PF2)들이었다. 두 CoMFA 모델들은 통계적으로 높은 예측성($r^2_{cv.}$=RS: 0.557 및 PC: 0.676)과 상관성($r^2_{ncv.}$=RS: 0.954 및 PC:0.968)을 보였다. 그리고 두 균주에 사이의 선택적인 살균활성은 기질분자의 정전기장에 의존적이었으며 CoMFA 등고도는 두 균주에 대한 살균활성이 기질분자의 치환기(X) 변화로 개전 될 수 있을 것임을 나타내었다.

Efficacy of the Rhinacanthus nasutus Nees Leaf Extract on Dermatophytes with Special Reference to Trichophyton mentagrophytes var. mentagrophytes and Microsporum canis

  • The effect of Rhinacanthus nasutus leaf extract on the growth of dermatophytes had been investigated. In vitro the extract exhibited high activity against various species of dermatophytes (Trichophyton mentagrophytes var. mentagrophytes, T. mentagrophytes var. interdigitale, T. rubrum, Microsporum canis and M. gypseum). The minimal inhibitory concentration (MIC) values of the extract revealed that all the dermatophytes tested had MIC values of 13.6 mg/ml. The extract exhibited fungistatic activity at lower concentrations $({\leq}13.6\;mg/ml\;or\;below\;the\;MIC\;value)$ and fungicidal activity at higher concentrations $({\geq}13.6\;mg/ml$\;or\;above\;the\;MIC\;value)$. The results suggested that the extract acted on the cell wall of the dermatophytes which subsequently leading to the formation of cytopathological and membrane structural degeneration and finally leading to cell lysis and death.

Candicidal Action of Resveratrol Isolated from Grapes on Human Pathogenic Yeast C. albicans

  • Resveratrol (3,5,4'-trihydroxystilbene) is a naturally occurring, multi-biofunctional chemical existing in grapes and various other plants as a polyphenol type, and it is one of the best known natural anticancer and antiatherosclerosis reagents. In this study, we investigated the antifungal action by resveratrol in Candida albicans, which is a human infectious fungi as an agent of candidiasis. Resveratrol displayed potent fungicidal activity in an energy-dependent manner, without any hemolytic effects against human erythrocytes. It was found that the serum-induced mycelial forms, which playa crucial role in the pathogenesis of C. albicans during host tissue invasion, were disrupted by resveratrol. To understand the correlation between lethal effects and resveratrol action, we examined the physiological changes of C. albicans. A significant accumulation of intracellular trehalose was induced by stress responses to resveratrol action, and a remarkable arrest of cell-cycle processes at the S-phase in C. albicans occured. Therefore, the fungicidal effects of resveratrol demonstrate that this compound is a potential candidate as an antifungal agent in treating infectious diseases by candidal infections.

  • Rice blast is the most serious disease of rice due to its harmfulness and its world wide distribution. $Magnaporthe$ $grisea$ is the cause of rice blast disease and destroys rice enough to feed several tens of millions of people each year. Fungicides are commonly used to control rice blast. But $M.$ $grisea$ acquires resistance to chemical treatments by genetic mutations. 2-Phenylimino-1,3-thiazolines were proposed as a novel class of fungicides against $M.$ $grisea$ in the previous study. To develop compounds with a higher biological activity, a new series of 2-phenylimino-1,3-thiazolines was synthesized and its fungicidal activity was determined against $M.$ $grisea$. The QSAR analysis was carried out on a series of 2-phenylimino-1,3-thiazolines. The QSAR results showed the dependence of fungicidal activity on the structural and physicochemical features of 2-phenylimino-1,3-thiazolines. Our results could be used as guidelines for the study of the mode of action and further design of optimal fungicides.

Fungicidal Effect of Resveratrol on Human Infectious Fungi

  • Resveratrol, a phenolic antioxidant found in grapes, has been known to mediate various biological activities on the human body. In the present study, we tested the antifungal a ctivity of resveratrol against human pathogenic fungi before carrying out further studies to elucidate the antifungal mechanism(s) of resveratrol. Resveratrol displayed potent antifungal activity against human pathogenic fungi at concentration levels of 10-20 ${\mu}g$/mL. Furthermore, time-kill curve exhibited fungicidal effect of resveratrol on C. albicans, but the compound had no hemolytic activity against human erythrocytes. The destruction of C. albicans cells by resveratrol was confirmed by scanning electron microscopy. These results suggest that resveratrol could be employed as a therapeutic agent to treat fungal infections of humans.

In vitro Evaluation of the Antifungal Activity of Propolis Extract on Cryptococcus neoformans and Candida albicans

  • The antifungal activities of propolis on Cryptococcus neoformans and Candida albicans were evaluated. In microbroth culture assay, the MIC(minimum inhibitory concentration) of propolis for C. neoformans and C. albicans were 2 and 16 mg/ml, respectively. In propolis-included solid medium assay, the MIC of propolis for C. neoformans and C. albicans were 4 and 16 mg/ml, respectively. Propolis showed fungicidal activity against C. neoformans, whereas propolis possesed fungistatic activity against C. albicans. The MFC(minimum fungicidal concentration) for C. neoformans was 8 mg/ml. Cell morphology of C. neoformans was affected by treatment of propolis. In scanning electron microscope, the appearance of cell rupture was observed.

벼잎집무늬마름병균 및 고추역병균에 대한 N-Phenyl-O-phenyl-thionocarbamate 유도체들의 살균활성에 관한 비교분자 유사성 분석 (Comparative Molecular Similar Indice Analysis on Fungicidal Activity of N-phenyl-O-phenylthionocarbamate Derivatives against Rice Sheath Blight and Phytophthora Blight)

  • 벼잎집무늬마름병균(RS: Rhizoctonia solani) 및 고추역병균 (PC: Phytophtora capsici)에 대한 N-phenyl-O-phenylthionocarbamate 유도체 중, N-phenyl 치환체(X)들의 살균활성에 관한 비교분자 유사성 지수분석(CoMSIA) 모델을 유도하고 기질분자와 ${\beta}-tubulin$ 사이의 수소결합성을 정량적으로 검토하였다. CoMSIA 모델들의 기여도로부터 두 균주에 대한 살균활성은 공통적으로 수소결합 받게장이 가장 크게 기여하였다. 두 균주 사이의 살균활성에 있어서 선택성은 N-phenyl 고리상 para- 및 meta-치환기로서, RS에서는 수소결합 주게가 아닌 작용기를 그리고 PC에서는 수소결합 받게가 아닌 작용기들의 역할에 기인하는 것으로 예측된다. 또한, RS에서는 meta-위치에 입체적으로 작고 음하전을 선호하는 치환기(X)가 그리고 PC에서는 para-위치에 수소결합 받게가 도입된다면 두 균주에 대한 살균 활성이 증가 할 것이다.

  • 새로운 10종의 N-[1-(benzotriazol-1-yl)alkyl]aniline유도체(4)를 합성하여 그중 4h의 결정을 확인한 바, 단사정계이고 S형의 절대배열을 나타내었으며 분자간에는 1개의 수소결합($N_{11}-H_{11}{\cdots}N3$)이 $2.300(11){\AA}$길이로 존재함을 알았다. 또한, 4 유도체들의 구조와 잿빛 곰팡이균(Botriyis cineria, BC), 고추 역병균(Phytophthora casici, PC), 및 마늘 흑색썩음균핵병(Sclerotium cepinorum, SC)등 곰팡이균에 대한 항균 활성값($pI_{50}$)과의 관계(SAR)를 검토한 바, SC>BC>PC의 순서로 SC에 대하여 가장 큰 항균활성을 보였다. X-치환체보다는 주로 Z의 소수성(${\pi}$)이 클수록 큰 항균활성을 나타내는 경향이었으며 bromo 치환체인 4f가 3종의 균에 대하여 가장 큰 항균활성을 나타내었다. 그리고 항균활성에 미치는 치환기들의 자유에너지 관계(LFER)를 검토하였다.
