Isopod Parasite Induced Secondary Microbial Infection in Marine Food Fishes

  • Isopods are parasitic crustaceans that pose serious threat to fisheries. Several studies have tried to explore the host-pathogen relationship between marine fishes and isopods. The present study aims to understanding the secondary infections in marine fishes pertaining to isopods. To assess the secondary infection in infected fishes, parasite infested and healthy tissues of fishes were collected. The samples were subjected to standard microbiological procedure to identify the presence of pathogenic bacteria and fungi. Our results showed the branchial region had the higher microbial load of non-sporulating cenocytic fungi in infected fishes. Moreover, fungal strains isolated from the parasitic lesion confirmed that the parasitation and body lesion facilitates the entry of several pathogenic microbes at the damaged host tissue. More over the immune regulation of fish fights back by producing minute cysts, trying to encapsulate the growing fungus. But this may eventually lead to systemic infestation and death of the fish.

  • Arbuscular mycorrhizal fungi (AMF) have mutualistic relationships with more than 80% of terrestrial plant species. This symbiotic relationship is ancient and would have had important roles in establishment of plants on land. Despite their abundance and wide range of relationship with plant species, AMF have shown low species diversity. However, molecular studies have suggested that diversity of these fungi may be much higher, and genetic variation of AMF is very high within a species and even within a single spore. Despite low diversity and lack of host specificity, various functions have been associated with plant growth responses to arbuscular mycorrhizal fungal colonization. In addition, different community composition of AMF affects plants differently, and plays a potential role in ecosystem variability and productivity. AMF have high functional diversity because different combinations of host plants and AMF have different effects on the various aspects of symbiosis. Consequently, recent studies have focused on the different functions of AMF according to their genetic resource and their roles in ecosystem functioning. This review summarizes taxonomic, genetic, and functional diversities of AMF and their roles in natural ecosystems.

한국산 균식성 미기록종, 갈색가시거저리의 분류학적 검토와 숙주버섯 (First Record of the Fungivorous Tenebrionid, Bolitotrogus kurosonis Miyatake (Coleoptera: Tenebrionidae), from Korea with a New Host Fungus)

  • 버섯에서 서식하는 거저리류 가운데 갈색가시거저리(Bolitotrogus kurosonis Miyatake)를 국내에서 처음 보고한다. 갈색가시거저리는 균식성으로 활엽수림에 나는 민주름버섯류, 특히 시루뻔버섯속(Inonotus)의 황갈색시루뻔버섯[Inonotus mikadoi (Lloyd) Imaz.]에서 서식하는 것으로 관찰되었다. 본 종의 성충에 대한 기재문을 기술하였고, 본 종의 상위 분류군인 한국산 가시거저리족(Bolitophagini)에 대한 검색표를 작성하였다. 성충의 사진, 진단 형질과 숙주버섯에 대한 생태적 정보를 제공하였다.

우포 습지에 분포하는 수지상균근균 포자의 다양성 (Spore Diversity of Arbuscular Mycorrhizal Fungi in Upo Wetland)

  • 우포 습지에 서식하는 식물의 근권 토양에서 수지상균근균(AMF) 포자를 추출하였다. 추출한 포자는 형태적 특성과 18S rDNA 염기서열의 분자적 분석을 이용해 동정하였다. 그 결과 6속 9종의 AMF 포자가 분리된 것을 확인하였으며, 종 다양성 지수를 계산하여 분석한 결과 습지에서 AMF 포자의 종 다양성은 식물에 의해 영향을 받는 것을 확인하였다. 연구 과정에서 국내 미기록종 1종을 확인하였으며, 확인된 종은 Diversispora epigaea이다. 미기록종 AMF 포자의 형태적 특성 및 분자적 분석의 결과에 대해 서술하였다.

Diversity and Plant Growth Promoting Capacity of Endophytic Fungi Associated with Halophytic Plants from the West Coast of Korea

  • Five halophytic plant species, Suaeda maritima, Limonium tetragonum, Suaeda australis, Phragmites australis, and Suaeda glauca Bunge, which are native to the Muan salt marsh of South Korea, were examined for fungal endophytes by sequencing the internal transcribed spacer (ITS) region containing ITS1, 5.8S rRNA, and ITS2. In total, 160 endophytic fungal strains were isolated and identified from the roots of the 5 plant species. Taxonomically, all 160 strains belonged to the phyla Ascomycota, Basidiomycota, and Zygomycota. The most dominant genus was Fusarium, followed by the genera Penicillium and Alternaria. Subsequently, using 5 statistical methods, the diversity indices of the endophytes were determined at genus level. Among these halophytic plants, P. australis was found to host the greatest diversity of endophytic fungi. Culture filtrates of endophytic fungi were treated to Waito-C rice seedlings for plant growth-promoting effects. The fungal strain Su-3-4-3 isolated from S. glauca Bunge provide the maximum plant length (20.1 cm) in comparison with wild-type Gibberella fujikuroi (19.6 cm). Consequently, chromatographic analysis of the culture filtrate of Su-3-4-3 showed the presence of physiologically active gibberellins, $GA_1$ (0.465 ng/mL), $GA_3$ (1.808 ng/mL) along with other physiologically inactive $GA_9$ (0.054 ng/mL) and $GA_{24}$ (0.044 ng/mL). The fungal isolate Su-3-4-3 was identified as Talaromyces pinophilus.

Re-identification of Colletotrichum acutatum Species Complex in Korea and Their Host Plants

  • Colletotrichum acutatum species complex is one of the most important groups in the genus Colletotrichum with a high species diversity and a wide range of host plants. C. acutatum and related species have been collected from different plants and locations in Korea and deposited into the Korean Agricultural Culture Collection (KACC), National Institute of Agricultural Sciences since the 1990s. These fungal isolates were previously identified based mainly on morphological characteristics, and a limitation of molecular data was provided. To confirm the identification of species, 64 C. acutatum species complex isolates in KACC were used in this study for DNA sequence analyses of six loci: nuclear ribosomal internal transcribed spacers (ITS), betatubulin 2 (TUB2), histone-3 (HIS3), glyceraldehyde3-phosphate dehydrogenase (GAPDH), chitin synthase 1 (CHS-1), and actin (ACT). The molecular analysis revealed that they were identified in six different species of C. fioriniae (24 isolates), C. nymphaeae (21 isolates), C. scovillei (12 isolates), C. chrysanthemi (three isolates), C. lupini (two isolates), and C. godetiae (one isolate), and a novel species candidate. We compared the hosts of KACC isolates with "The List of Plant Diseases in Korea", previous reports in Korea and global reports and found that 23 combinations between hosts and pathogens could be newly reported in Korea after pathogenicity tests, and 12 of these have not been recorded in the world.

사구에 서식하는 단자엽식물로부터 식물 생장 촉진 활성 내생 진균류의 분리 (Isolation of Endophytic Fungi Capable of Plant Growth Promotion from Monocots Inhabited in the Coastal Sand Dunes of Korea)

  • 내생성 진균류는 초본류의 식물체 내에 주로 서식하며, 식물의 병원균으로부터 숙주를 보호할 뿐만 아니라, 식물 생육에 유리하게 작용하는 다양한 대사 산물을 생산한다. 매우 흔히 접할 수 있는 사구식물의 뿌리로부터 49종의 균류를 분리하여 식물 생장 촉진 활성이 있는 균주를 선별하였다. 결과적으로 37균주(75.5%)는 awito-c 벼의 생육을 촉진하였으며, 11균주(22.5%)는 생육을 저해하였으며, 1균주(2%)는 생육에 아무런 영향을 미치지 않았다. Gibberella fujikuroi와 증류수 및 Czapek broth 배지를 control 로 사용하여 실험을 수행하였다. 결론적으로 사구식물 내생균류의 많은 부분은 숙주식물의 생육과 발달을 조장하는 대사 산물을 생산함을 알 수 있었다.

Rhizospheric fungi of Panax notoginseng: diversity and antagonism to host phytopathogens

  • Background: Rhizospheric fungi play an essential role in the plantesoil ecosystem, affecting plant growth and health. In this study, we evaluated the fungal diversity in the rhizosphere soil of 2-yr-old healthy Panax notoginseng cultivated in Wenshan, China. Methods: Culture-independent Illumina MiSeq and culture-dependent techniques, combining molecular and morphological characteristics, were used to analyze the rhizospheric fungal diversity. A diffusion test was used to challenge the phytopathogens of P. notoginseng. Results: A total of 16,130 paired-end reads of the nuclear ribosomal internal transcribed spacer 2 were generated and clustered into 860 operational taxonomic units at 97% sequence similarity. All the operational taxonomic units were assigned to five phyla and 79 genera. Zygomycota (46.2%) and Ascomycota (37.8%) were the dominant taxa; Mortierella and unclassified Mortierellales accounted for a large proportion (44.9%) at genus level. The relative abundance of Fusarium and Phoma sequenceswas high, accounting for 12.9% and 5.5%, respectively. In total,113 fungal isolates were isolated from rhizosphere soil. They were assigned to five classes, eight orders (except for an Incertae sedis), 26 genera, and 43 species based on morphological characteristics and phylogenetic analysis of the internal transcribed spacer. Fusarium was the most isolated genus with six species (24 isolates, 21.2%). The abundance of Phoma was also relatively high (8.0%). Thirteen isolates displayed antimicrobial activity against at least one test fungus. Conclusion: Our results suggest that diverse fungi including potential pathogenic ones exist in the rhizosphere soil of 2-yr-old P. notoginseng and that antagonistic isolates may be useful for biological control of pathogens.

Cutaneous Microflora from Geographically Isolated Groups of Bradysia agrestis, an Insect Vector of Diverse Plant Pathogens

  • Larvae of Bradysia agrestis, an insect vector that transports plant pathogens, were sampled from geographically isolated regions in Korea to identify their cutaneous fungal and bacterial flora. Sampled areas were chosen within the distribution range of B. agrestis; each site was more than 91 km apart to ensure geographical segregation. We isolated 76 microbial (fungi and bacteria) strains (site 1, 29; site 2, 29; site 3, 18 strains) that were identified on the basis of morphological differences. Species identification was molecularly confirmed by determination of universal fungal internal transcribed spacer and bacterial 16S rRNA gene sequences in comparison to sequences in the EzTaxon database and the NCBI GenBank database, and their phylogenetic relationships were determined. The fungal isolates belonged to 2 phyla, 5 classes, and 7 genera; bacterial species belonged to 23 genera and 32 species. Microbial diversity differed significantly among the geographical groups with respect to Margalef's richness (3.9, 3.6, and 4.5), Menhinick's index (2.65, 2.46, and 3.30), Simpson's index (0.06, 0.12, and 0.01), and Shannon's index (2.50, 2.17, and 2.58). Although the microbial genera distribution or diversity values clearly varied among geographical groups, common genera were identified in all groups, including the fungal genus Cladosporium, and the bacterial genera Bacillus and Rhodococcus. According to classic principles of co-evolutionary relationship, these genera might have a closer association with their host insect vector B. agrestis than other genera identified. Some cutaneous bacterial genera (e.g., Pseudomonas) displaying weak interdependency with insect vectors may be hazardous to agricultural environments via mechanical transmission via B. agrestis. This study provides comprehensive information regarding the cutaneous microflora of B. agrestis, which can help in the control of such pests for crop management.

Biological Control of Powdery Mildew by a Hyperparasite, Ampelomyces quisqualis 94103: From isolation to a commercial biofungicide product.

  • Total 291 isolates of Ampelomyces quisqualis were obtained from 32 species of powdery mildew fungi and a selected isolate, Ampelomyces quisqualis 94013 (AQ94013) effectively hyperparasitized 6 species of Sphaerotheca and one species of Erysiphe which cause serious damage on many important crops in Korea. Moreover, AQ94013 showed antagonistic effects against 12 major fungal plant pathogens as well. Results indicated that the present isolate is not a host specific hyperparasite and has a broad spectrum of biocontrol potential. Providentially, AQ94013 revealed resistance to a number of agrochemicals so as to be applied with the chemicals reciprocally.(중략)
