Relationship between Planthoppers (Nilaparvata lugens and Sogatella furcifera) and Rice Diseases (멸구류(類)(벼멸구 및 흰등멸구)와 수도병해(水稻病害)의 복합발생피해(複合發生被害)에 관(關)한 연구(硏究))

  • The locational preference of the brown planthopper (BPH) Nilaparvata lugens ($St{\aa}l$) and the whitebacked plant hopper (WBPH) Sogatella furcifera(Horvath) was studied on rice cultivars IR22 and IR36 as an integral part of subsequent research on insect-fungal pathogen relationships. The BPH was observed to stay consistently on the basal portion while the WBPH showed a general preference for the upper portion regardless of varieties, rice growth stages and insect population density levels. The habitat preference of both species (BPH and WBPH) was found not to be affected by the presence of the other species when both species are present on the same host plant Five rice cultivars with different reactions to BPH biotype 2 were used in the study on BPH-Rhizoctonia solani relationship: IR22 and TN1 (susceptible); Triveni and ASD7 (moderately resistant); and IR42 (resistant). Test plants were inoculated with R. solani (Kuhn) $3{\sim}4$days after insect infestation. Sheath blight disease severity/incidence was significantly higher in the treatment where BPH+R. solani were together than in the treatment with only the pathogen. Symptom expression of the disease in the BPH-pathogen combination was faster and mycelial growth was more profuse inducing the formation of more infection structures. Regardless of varietal reaction to BPH biotype 2, the degree of hopperburn was significantly higher in the combination of the two pests as compared with that of BPH alone. There could be a synergistic relationship between the insect pest and the pathogen indicated by a positive interaction between the two species.

Growth Response of Pinus rigida × P. taeda to Mycorrhizal Inoculation and Efficiency of Pisolithus tinctorius at Different Soil Texture and Fertility with Organic Amendment (리기테다 소나무의 균근(菌根) 접종(接種) 반응(反應)과 토양비옥도(土壤肥沃度)에 따른 모래밭 버섯의 효과(効果) 및 그 생태학적(生態學的) 의미(意味))

  • Potted, germinating Pinus rigida ${\times}$ P. taeda seedlings were inoculated with Pisolithus tinctorius (Pt) ectomycorrhizal fungus to test the effectiveness of Pt in relation to organic amendment and changes in soil fertility and soil texture. Pt was cultured as mycelia in vermiculite-peat moss mixture with nutrients and added to sterilized pot soils with or without organic amendment (fully fermented compost) at three soil texture levels (sand, loamy sand, and sandy loam) in a factorial design. Plants were grown in a greenhouse for 4 months and harvested to compare their growth with non-mycorrhizal plants and plants infected by natural fungi. Regardless of sod texture, soil fertility, or organic amendment, seedlings inoculated with Pt were better in dry weight and height than non-mycorrhizal plants or those infected by natural fungi. An exception was observed in the most fertile soil (0.075% N and 1.32% organic matter content in sandy loam with organic amendment), where non-mycorrhizal plants were slightly bigger (8%) and heavier (18%) than Pt-inoculated plants. In over-all average, Pt-inoculated seedlings were 30% taller and 107% heavier than those infected by natural fungi and 31 % taller and 60% heavier than non-mycorrhizal plants. Growth stimulation of seedlings by Pt was more pronounced in less fertile sand soil when organic was not amended. Mycorrhizal frequency of Pt (% of mycorrhizal root tips) was reduced to about half (from 84 to 33% in sandy loam and from 77 to 40% in loamy sand) by organic amendment, while that of natural fungi was not significantly affected. Severe nitrogen deficiency was observed in the needles of non-mycorrhizal plants (1.38% N), while both Pt-inoculated plants (1.68% N) and those infected by natural fungi (1.89% N) did not develop symptom, suggesting an active role of mycorrhizae in absorption of soil nitrogen. Top to root ratio increased with organic amendment to non-mycorrhizal plants, but was not significantly affected by fungal treatment. It was concluded from this study that relative effectiveness of Pt was determined by soil fertility. Organic amendment to less fertile sand soil increased effectiveness of Pt, while the same amendment to more fertile loamy sand and sandy loam decreased effectiveness of Pt. Benefits of Pt mycorrhizae would be expected most either when organic was not added to the soil, or when soil nutrients were not abundant.

Brief Introduction of Research Progresses in Control and Biocontrol of Clubroot Disease in China

  • Clubroot disease of crucifers has occurred since 1957. It has spread to the whole China, especially in the southwest and nourtheast where it causes 30-80% loss in some fields. The disease has being expanded in the recent years as seeds are imported and the floating seedling system practices. For its effective control, the Ministry of Agriculture of China set up a program in 2010 and a research team led by Dr. Yueqiu HE, Yunnan Agricultural University. The team includes 20 main reseachers of 11 universities and 5 institutions. After 5 years, the team has made a lot of progresses in disease occurrence regulation, resources collection, resistance identification and breeding, biological agent exploration, formulation, chemicals evaluation, and control strategy. About 1200 collections of local and commercial crucifers were identified in the field and by artificiall inoculation in the laboratories, 10 resistant cultivars were breeded including 7 Chinese cabbages and 3 cabbages. More than 800 antagostic strains were isolated including bacteria, stretomyces and fungi. Around 100 chemicals were evaluated in the field and greenhouse based on its control effect, among them, 6 showed high control effect, especially fluazinam and cyazofamid could control about 80% the disease. However, fluzinam has negative effect on soil microbes. Clubroot disease could not be controlled by bioagents and chemicals once when the pathogen Plasmodiophora brassicae infected its hosts and set up the parasitic relationship. We found the earlier the pathogent infected its host, the severer the disease was. Therefore, early control was the most effective. For Chinese cabbage, all controlling measures should be taken in the early 30 days because the new infection could not cause severe symptom after 30 days of seeding. For example, a biocontrol agent, Bacillus subtilis Strain XF-1 could control the disease 70%-85% averagely when it mixed with seedling substrate and was drenching 3 times after transplanting, i.e. immediately, 7 days, 14 days. XF-1 has been deeply researched in control mechanisms, its genome, and development and application of biocontrol formulate. It could produce antagonistic protein, enzyme, antibiotics and IAA, which promoted rhizogenesis and growth. Its The genome was sequenced by Illumina/Solexa Genome Analyzer to assembled into 20 scaffolds then the gaps between scaffolds were filled by long fragment PCR amplification to obtain complet genmone with 4,061,186 bp in size. The whole genome was found to have 43.8% GC, 108 tandem repeats with an average of 2.65 copies and 84 transposons. The CDSs were predicted as 3,853 in which 112 CDSs were predicted to secondary metabolite biosynthesis, transport and catabolism. Among those, five NRPS/PKS giant gene clusters being responsible for the biosynthesis of polyketide (pksABCDEFHJLMNRS in size 72.9 kb), surfactin(srfABCD, 26.148 kb, bacilysin(bacABCDE 5.903 kb), bacillibactin(dhbABCEF, 11.774 kb) and fengycin(ppsABCDE, 37.799 kb) have high homolgous to fuction confirmed biosynthesis gene in other strain. Moreover, there are many of key regulatory genes for secondary metabolites from XF-1, such as comABPQKX Z, degQ, sfp, yczE, degU, ycxABCD and ywfG. were also predicted. Therefore, XF-1 has potential of biosynthesis for secondary metabolites surfactin, fengycin, bacillibactin, bacilysin and Bacillaene. Thirty two compounds were detected from cell extracts of XF-1 by MALDI-TOF-MS, including one Macrolactin (m/z 441.06), two fusaricidin (m/z 850.493 and 968.515), one circulocin (m/z 852.509), nine surfactin (m/z 1044.656~1102.652), five iturin (m/z 1096.631~1150.57) and forty fengycin (m/z 1449.79~1543.805). The top three compositions types (contening 56.67% of total extract) are surfactin, iturin and fengycin, in which the most abundant is the surfactin type composition 30.37% of total extract and in second place is the fengycin with 23.28% content with rich diversity of chemical structure, and the smallest one is the iturin with 3.02% content. Moreover, the same main compositions were detected in Bacillus sp.355 which is also a good effects biocontol bacterial for controlling the clubroot of crucifer. Wherefore those compounds surfactin, iturin and fengycin maybe the main active compositions of XF-1 against P. brassicae. Twenty one fengycin type compounds were evaluate by LC-ESI-MS/MS with antifungal activities, including fengycin A $C_{16{\sim}C19}$, fengycin B $C_{14{\sim}C17}$, fengycin C $C_{15{\sim}C18}$, fengycin D $C_{15{\sim}C18}$ and fengycin S $C_{15{\sim}C18}$. Furthermore, one novel compound was identified as Dehydroxyfengycin $C_{17}$ according its MS, 1D and 2D NMR spectral data, which molecular weight is 1488.8480 Da and formula $C_{75}H_{116}N_{12}O_{19}$. The fengycin type compounds (FTCPs $250{\mu}g/mL$) were used to treat the resting spores of P. brassicae ($10^7/mL$) by detecting leakage of the cytoplasm components and cell destruction. After 12 h treatment, the absorbencies at 260 nm (A260) and at 280 nm (A280) increased gradually to approaching the maximum of absorbance, accompanying the collapse of P. brassicae resting spores, and nearly no complete cells were observed at 24 h treatment. The results suggested that the cells could be lyzed by the FTCPs of XF-1, and the diversity of FTCPs was mainly attributed to a mechanism of clubroot disease biocontrol. In the five selected medium MOLP, PSA, LB, Landy and LD, the most suitable for growth of strain medium is MOLP, and the least for strains longevity is the Landy sucrose medium. However, the lipopeptide highest yield is in Landy sucrose medium. The lipopeptides in five medium were analyzed with HPLC, and the results showed that lipopeptides component were same, while their contents from B. subtilis XF-1 fermented in five medium were different. We found that it is the lipopeptides content but ingredients of XF-1 could be impacted by medium and lacking of nutrition seems promoting lipopeptides secretion from XF-1. The volatile components with inhibition fungal Cylindrocarpon spp. activity which were collect in sealed vesel were detected with metheds of HS-SPME-GC-MS in eight biocontrol Bacillus species and four positive mutant strains of XF-1 mutagenized with chemical mutagens, respectively. They have same main volatile components including pyrazine, aldehydes, oxazolidinone and sulfide which are composed of 91.62% in XF-1, in which, the most abundant is the pyrazine type composition with 47.03%, and in second place is the aldehydes with 23.84%, and the third place is oxazolidinone with 15.68%, and the smallest ones is the sulfide with 5.07%.

Fish Safety and Antimicrobial Activity of Natural Sulfur Solution on Aquatic Microorganisms (Saprolegnia parasitica) Isolated from Misgurnus mizolepis (미꾸라지(Misgurnus mizolepis)에서 분리된 수생균 (Saprolegnia parasitica)에 대한 천연유황수의 항균 활성 및 처리에 대한 어류 안전성)

  • Basic dyes such as malachite green and methylene blue have been used as disinfectants to control water fungal infections since the 1930s. However, after succeeding reports of carcinogenicity and bioaccumulation of the dye, their use was forbidden in lieu of public health. This study undertook to evaluate the therapeutic effect of sulfur solution processed by effective microorganisms (EM-PSS) against Saprolegnia parasitica infection, and its safety in fish. In vitro antifungal evaluation of EM-PSS inhibited the growth of S. parasitica mycelia at concentrations of 50 ppm or higher. The acute toxicity test of EM-PSS to the mud fish (Misgurnus mizolepis) measured a no effect concentration (NOEC) at 100 ppm, the lowest effect concentration (LOEC) at 125 ppm, and the half-lethal concentration ($LC_{50}$) at 125 ppm in juvenile and 250 ppm in the immature stage. In addition, the ecotoxicity test of EM-PSS using Daphnia magna inhibited swimming of D. magna at concentrations of 100 ppm or less. Lastly, the EM-PSS prevented infection of S. parasitica to mud fish, at concentrations of 50 ppm. Furthermore, at 100 ppm concentration, the EM-PSS showed no acute toxicity on mud fish, nor any eco-toxic effects on D. magnano. Therefore, we conclude that carcinogenic disinfectants such as malachite green and methylene blue could be replaced by EM-PSS to remove S. parasitica in mud fish farming, and might be a potential eco-friendly disinfectant in aquaculture.

Characteristics of Everbearing Strawberry Cultivars and the Effect of Precooling Treatment to Maintain Quality of 'Charlotte' Cultivar Grown on Highland in Summer Season (고랭지 사계성 딸기 품종 특성 비교 및 'Charlotte' 품종의 예냉 처리 효과)

  • This study was conducted to investigate the quality characteristics of eight everbearing strawberry cultivars and the effect of precooling treatment to maintain the quality during storage and shelf life of 'Charlotte'. Several quality parameters including firmness, soluble solids content (SSC), color, soluble sugars, and organic acids were evaluated. For Successful marketing everbearing strawberries required more than 1 N of firmness and $6^{\circ}Brix$ of SSC, respectively. 'Albion', 'Charlotte', and 'Goha' cultivars were higher in fruit SSC and 'Charlotte' cultivar was higher in fruit firmness among eight cultivars examined in this study. Fruit had more bright red color in 'Charlotte', 'Flamingo', 'GW-4', and 'San Andreas', which may reflect the consumer acceptance. Physicochemical characteristics of 'Charlotte' strawberry after with/without room precooling treatment were evaluated during storage at $4^{\circ}C$, $10^{\circ}C$, and $25^{\circ}C$ and additional 3 days at $25^{\circ}C$ for shelf-life. Weight loss increased about 8% in the fruits stored at $25^{\circ}C$ for 3 days, while in low temperature storage was about 2% for 7 days showing precooling was more effect on during shelf life periods rather than storage periods. Decay was not found during storage at $4^{\circ}C$ regardless of precooling treatment, but fungal growth was found in all the treatments after transferred to room temperature. Precooling treatment did not affect fruit color in shelf life period. For advanced marketing, everbearing strawberry should maintain in low temperature less than $4^{\circ}C$ to sustain quality, but the room precooling showed less significant effect to maintain quality on 'Charlotte' cultivar.

Improvement of Shelf-life and Quality in Fresh-Cut Tomato Slices:

  • Quality of fresh-cut tomato slices was compared during cold storage under various modified atmosphere packaging conditions. Chilling injury of slices in containers sealed with Film A was higher than with Film B; these films had oxygen transmission rates of 87.4 and 60.0 ml $h^{-1}\;m^{-2}\;atm^{-1}$ at $5^{\circ}C\;and\;99\%$ RH, respectively. While slices in containers with an initial atmospheric composition of air, $4\%\;CO_2+1\;or\;20\%\;O_2,\;8\%\;CO_2+1\;or\;20\%\;O_2,\;or\;12\%\;CO_2+20\%\;O_2$ showed fungal growth, slices in containers with $12\%\;CO_2+1\%\;O_2$ did not. Low ethylene in containers enhanced chilling injury. Modified atmosphere packaging provided good quality tomato slices with a shelf-life of 2 weeks or more at $5^{\circ}C$. Experiments were conducted to compare changes in quality of slices of red tomato (Lycopersicon esculentum Mill. 'Sunbeam') fruit from plants grown using black polyethylene or hairy vetch mulches under various foliar disease management systems including: no fungicide applications (NF), a disease forecasting model (Tom-Cast), and weekly fungicide applications (WF), during storage at $5^{\circ}C$ under a modified atmosphere. Slices were analyzed for firmness, soluble solids content (SSC), titratable acidity (TA), pH, electrolyte leakage, fungi, yeasts, and chilling injury. With both NF and Tom-Cast fungicide treatments, slices from tomato fruit grown with hairy vetch (Vicia villosa Roth) mulch were firmer than those from tomato fruit grown with black polyethylene mulch after 12 days storage. Ethylene production of slices from fruit grown using hairy vetch mulch under Tom-Cast was about 1.5- and 5-fold higher than that of slices from WF and NF fungicide treatments after 12 days, respectively. The percentage of water-soaked areas (chilling injury) for slices from tomato fruit grown using black polyethylene mulch under NF was over 7-fold that of slices from tomato fruit grown using hairy vetch under Tom-Cast. When stored at $20^{\circ}C$, slices from light-red tomato fruit grown with black polyethylene or hairy vetch mulches both showed a rapid increase in electrolyte leakage beginning 6 hours after slicing. However, slices from tomato fruit grown using the hairy vetch mulch tended to have lower electrolyte leakage than those grown with black polyethylene mulch. These results suggest that tomato fruit from plants grown using hairy vetch mulch may be more suitable for fresh-cut slices than those grown using black polyethylene mulch. Also, use of the disease forecasting model Tom-Cast, which can result in lower fungicide application than is currently used commercially, resulted in high quality fruit for fresh-cut processing. Experiments were conducted to determine if ethylene influences chilling injury, as measured by percentage of slices exhibiting water-soaked areas in fresh-cut tomato slices of 'Mountain Pride' and 'Sunbeam' tomato (Lycopersicon esculentum Mill.). Ethylene concentration in containers without ventilation significantly increased during storage at $5^{\circ}C$, whereas little or no accumulation of ethylene occurred in containers with one or six perforations. Chilling injury was greatest for slices in containers with six perforations, compared to slices in containers with one perforation, and was over 13-fold greater than that of slices in control containers with no perforations. An experiment was also performed to investigate the effectiveness of including an ethylene absorbent pad in containers on subsequent ethylene accumulation and chilling injury. While ethylene in the no-pad controls increased continually during storage of both 'Mountain Pride' and 'Sunbeam' tomatoes at $5^{\circ}C$ under modified atmosphere conditions, no increase in accumulation of ethylene was observed in containers containing ethylene absorbent pads throughout storage. The ethylene absorbent pad treatment resulted in a significantly higher percentage of chilling injury compared with the no-pad control. In studies aimed at inhibiting ethylene production using AVG during storage of slices, the concentration of ethylene in control containers (no AVG) remained at elevated levels throughout storage, compared to containers with slices treated with AVG. Chilling injury in slices treated with AVG was 5-fold greater than that of controls. Further, we tested the effect of ethylene pretreatment of slices on subsequent slice shelf-life and quality. In slices treated with ethylene (0, 0.1, 1, or $10\;{mu}L\;L^{-1}$) immediately after slicing, ethylene production in non-treated controls was greater than that of all other ethylene pre-treatments. However, pretreatment of slices 3 days after slicing resulted in a different pattern of ethylene production during storage. Ihe rate of ethylene production by slices treated with 1 L $L^{-1}$ ethylene 3 days after slicing was greater during storage than any of the other ethylene treatments. With slices pre-treated with ethylene, both immediately and 3 days after slicing, the rate of ethylene production tended to show an negative correlation with chilling injury.   • Quercus acutissima ectomycorrhizae were classified as apical type, linear type, clavate type, diffuse type, pyramidal type, coralloid type, and nodular type. The surface texture of the fungal mantle at the initial stage of mycorrhizal formation was velvety. The surface texture of Pisolithus tinctorius(Pt) mycorrhizae at 30 September was well-developed felty mantle, Yellowish white. Except Pt all mycorrhizae formed by ectomycorrhizal fungi used to experiment were white with felty mantle. Mycorrhizae at the initial stage of mycorrhizae formation were creamy or creamy brown and swelled with thin mantle. Transverse and longitudinal sections showed radially-elongated cortical cell layers and epidermal cell with Hartig net. The transversal wideth of radially-elongated cortical and epidermal cells in the mycorrhizae with thick mantle on 30 September did not different with the mycorrhizae with thin mantle on the initial stage. Pt #250 formed coralloid mycorrizae but Pt KJ-1 did not although they are same species. On the mean length of linear type ectomycorrhizae of Pt KJ-1(2.21mm) was 1.5 times longer than that of Pt #250(1.32mm). The total dry weight of seedlings inoculated with Pt KJ-1, Pt #250, Lycoperdon pedicellatum, Scleroderma verrucosum were significantly heavier than those of suillus granulatus, Laccaria laccata.

Food Waste Composting by Using an Inoculum-Mixture Containing New Facultative Anaerobic Bacteria (신규 통성혐기성 세균으로 제조한 발효흙에 의한 음식물 쓰레기의 퇴비화)

  • Four newly isolated bacteria from soil were used to manufacture microbial inoculum to compost food waste. The bacteria, GM103, V25, V31, and V35, were identified as Bacillus licheniformis, B. subtilis, B. stearothermophilius, and B, subtilis, respectively. The bacterial strains were efficient to degrade protein and starch and also able to inhibit the growth of plant pathogenic fungus Rhizopus stronifer. The GM103 showed distinct capability in degrading starch, but grow only aerobically. The other three bacterial strains. V25, V31, and V35, could grow both aerobically as well as anaerobically, in 10%(w/v) salt, at $50^{\circ}C$, and had good viability and survival rate in soil. These characteristics of the bacterial strains are very adquate in Korean food composting containing high concentration of salt, especially at home. By mixing the 4 bacterial culture broth with molasses, beet pulp, zeolite, The bacterial inoculum for food waste composting-BIOTOP-CLEAN-was made. The performance of food waste composting by the BIOTOP-CLEAN was compared with that by control(not treated) and HS(other demestic company's inoculum product for food waste composting). The maximum temperature of the food waste during the composting with the BIOTOP-CLEAN was $50^{\circ}C$, while those of the control and HS were $30^{\circ}C$ and $35^{\circ}C$, respectively. The BIOTOP-CLEAN gave the good smell and showed dark brown color, while the control gave bad smell and HS gave less bad smell. These indicates that the food waste composting by the BIOTOP-CLEAN had been well accomplished. The culture broth of V25, V31, V35 were sparyed to the plants of tomato, chinese cabbage, raddish, red pepper every month and the spraying the culture broth to these plant significantly improved the production yield of the crops, due to the control effect of the bacterial strains against the plant pathogens.

  • A bacterial strain YC4963 with antifungal activity against Colletotrichum orbiculare, a causal organism of cucumber anthracnose was isolated from the rhizosphere soil of Siegesbeckia pubescens Makino in Korea. Based on physiological and biochemical characteristics and 16S ribosomal DNA sequence analysis, the bac­terial strain was identified as Pseudomonas aurantiaca. The bacteria also inhibited mycelial growth of several plant fungal pathogens such as Botrytis cinerea, Fusarium oxysporum and Rhizoctonia solani on PDA and 0.1 TSA media. The antifungal activity was found from the culture filtrate of this isolate and the active compound was quantitatively bound to XAD adsorption resin. The antibiotic compound was purified and identified as phenazine-l-carboxylic acid on the basis of combined spectral and chemical analyses data. This is the first report on the production of phenazine-l-carboxylic acid by Pseudomonas aurantiaca.

Resistance of Cucurbits to Podosphaera xanthii Race 1 (Podosphaera xanthii Race 1에 대한 박과 작물의 저항성)

  • This study was conducted to establish an efficient screening method to identify cucurbits resistant to powdery mildew. Powdery mildew fungus was obtained from a single lesion of infected cucumber leaf in 2010 at Daejeon. The fungus was identified as Podosphaera xanthii race 1 based on morphological characteristics and resistance responses of four melon differentials. Development of powdery mildew caused by the fungal isolate on 34 commercial cultivars of cucumber was investigated at three plant growth stages in a greenhouse. The degree of resistance of cotyledons of each cultivar to the fungus was not correlated with that of whole plant, but powdery mildew occurrence in the first true leaf was highly correlated with resistance at the level of the whole plant. Based on these results, the first true leaf of cucurbit cultivars can be used for screening of resistance to powdery mildew. In addition, variation of resistance of commercial 12 cucumber and 26 melon cultivars to the powdery mildew fungus due to different growing seasons was tested. In the case of cucumber, the resistance response in some cultivars was influenced by growing season. The resistant cultivars showed higher resistance in the warm season than in the cool season. By contrast, the resistant melon cultivars demonstrated strong resistance in all the tested growing seasons. Interestingly, the tested powdery mildew pathogen, a member of P. xanthii race 1, was not pathogenic on seven cultivars of watermelon (Citrullus lanatus). To follow up on this, diverse race 1 isolates of P. xanthii should be collected and tested.
