In vitro Anti-fungal Activity of Various Hydroxylated Fatty Acids Bioconverted by Pseudomonas aeruginosa PR3

  • The in vitro anti-fungal activity of hydroxylated fatty acids obtained from microbial conversion by Psuedomonas aeruginosa PR3 using ricinoleic acid(RA), eicosadienoic acid(EDA) and conjugated linoleic acid(CLA) as substrates, was investigated. Bioconverted hydroxylated fatty acids showed different anti-fungal activities potentials against the range of phytopathogenic fungi such as Botrytis cinerea, Rhizoctonia solani, Fusarium oxysporum, Sclerotonia sclerotiorum, Colletotricum capsici, Fusarium solani and Phytophthora capsici. RA and EDA showed up to 50% fungal mycelial inhibition at the concentration of $5{\mu}l\;ml^{-1}$. RA, EDA and CLA also exhibited anti-fungal activities with minimum inhibitory concentration(MIC), ranging from 500 to $1000{\mu}g\;ml^{-1}$. Screening was also carried out using varied concentrations of bioconverted RA and EDA for determining the anti-fungal effect on the spore germination of different fungi. Bioconverted RA and EDA showed a considerable degree of spore germination inhibition.

당근의 모상근 배양에서 Fungal Elicitor에 의한 Anthocyanin 생산의 향상 (Improved Anthocyanin Production in Hairy Root Culture of Daucus carota by Fungal Elicitor)

  • In order to improve anthocyanin production, effects of fungal elicitation in hairy root culture were investigated. fungal elicitor prepared from Fusarium moniliforme was the best in enhancement of anthocyanin production among the eight fungal elicitors tested. The optimum treat-ment time and concentration of treated elicitor for anthocyanin production were 12 hours and 3.28 mg carbohydrates per liter medium. Also, fungal elicitor was treated to hairy root culture in flat-bottomed fluidized-bed bioreactor. The anthocyanin production of elicited culture was enhanced 227% than non-treated.

  • Chronic invasive fungal sinusitis (CIFS) is a relatively rare disease which occurs in immunocompetent or immunocompromised patients. We report a case of CIFS in ethmoid sinus with concurrent fungal ball in maxillary sinus. After surgical debridement and antifungal therapy, CIFS recurred with submucosal invasion in nasopharynx and osteomyelitis of clivus. Although CIFS encompasses a slowly progressing invasive fungal disease but potentially aggressive. It should be noted that CIFS could progress and extend through submucosal route without evidence of mucosal lesion.

Effect of Vibration on Dispersal of Cladosporium cladosporioides Bioaerosols

  • The vibration of fungal cultures was evaluated to determine its potential effect on the dispersal of airborne fungal microorganisms suspected of being pathogens. An artificial vibration system, which simulates the actual environmental vibration of fungal structures, was designed and constructed for this purpose. Experiments featured the use of low-frequency vibrations similar to those induced by earthquakes. Within the range of conditions tested, the vibration of fungal cultures was found to affect the airflow-driven generation of bioaerosols.

  • NADPH oxidases (Noxes), transmembrane proteins found in most eukaryotic species, generate reactive oxygen species and are thereby involved in essential biological processes. However, the fact that genes encoding ferric reductases and ferric-chelate reductases share high sequence similarities and domains with Nox genes represents a challenge for bioinformatic approaches used to identify Nox-encoding genes. Further, most studies on fungal Nox genes have focused mainly on functionality, rather than sequence properties, and consequently clear differentiation among the various Nox isoforms has not been achieved. We conducted an extensive sequence analysis to identify putative Nox genes among 34 eukaryotes, including 28 fungal genomes and one Oomycota genome. Analyses were performed with respect to phylogeny, transmembrane helices, di-histidine distance and glycosylation. Our analyses indicate that the sequence properties of fungal Nox genes are different from those of human and plant Nox genes, thus providing novel insight that will enable more accurate identification and characterization of fungal Nox genes.

숙시닐화가 Aspergillus fumiagtus 균체단백질의 기능적 특성에 미치는 영향 (Effect of Succinylation on Functional Properties of Aspergillus fumiagtus Cell Protein)

  • 숙시닐화가 균체 단백질(Aspergillus fumigatus)의 여러가지 기능적 특성에 미치는 영향을 검토하였다. 균체 단백질은 succinic anhydride 2.5와 10% 첨가에 20.7 과 85.3%가 숙시닐화되었다. 숙시닐화된 균체 단백질은 흡광도, 핵산 및 탄수화물의 양은 감소하였으나 단백질소, 단백질 추출율은 증가되었다. 숙시닐화된 균체 단백질의 질소용해도는 milk casein과 soy flour 보다 32와 51% 증가되었다. 유화활성도와 유화 안정성은 균체 단백질의 숙시닐화 비율에 따라서 증가 되었으며, 80% 숙시닐화된 균체 단백질은 약 8.4배 증가율을 나타내어 숙시닐화를 증가시킴에 따라 유화활성도와 유화안정성이 증가 되었다. 숙시닐화 시킨 균체 단백질은 숙시닐화 시키지 않는 균체 단백질, milk casein 및 soy flour에 비하여 향상된 기능적 특성을 나타내었다.

Roles of Forkhead-box Transcription Factors in Controlling Development, Pathogenicity, and Stress Response in Magnaporthe oryzae

  • Although multiple transcription factors (TFs) have been characterized via mutagenesis to understand their roles in controlling pathogenicity and infection-related development in Magnaporthe oryzae, the causal agent of rice blast, if and how forkhead-box (FOX) TFs contribute to these processes remain to be characterized. Four putative FOX TF genes were identified in the genome of M. oryzae, and phylogenetic analysis suggested that two of them (MoFKH1 and MoHCM1) correspond to Ascomycota-specific members of the FOX TF family while the others (MoFOX1 and MoFOX2) are Pezizomycotina-specific members. Deletion of MoFKH1 (${\Delta}Mofkh1$) resulted in reduced mycelial growth and conidial germination, abnormal septation and stress response, and reduced virulence. Similarly, ${\Delta}Mohcm1$ exhibited reduced mycelial growth and conidial germination. Conidia of ${\Delta}Mofkh1$ and ${\Delta}Mohcm1$ were more sensitive to one or both of the cell cycle inhibitors hydroxyurea and benomyl, suggesting their role in cell cycle control. On the other hand, loss of MoFOX1 (${\Delta}Mofox1$) did not show any noticeable changes in development, pathogenicity, and stress response. Deletion of MoFOX2 was not successful even after repeated attempts. Taken together, these results suggested that MoFKH1 and MoHCM1 are important in fungal development and that MoFKH1 is further implicated in pathogenicity and stress response in M. oryzae.

Mycoflora and Enzymatic Characterization of Fungal Isolates in Commercial Meju, Starter for a Korean Traditional Fermented Soybean Product

  • Mycoflora was assessed in the commercial meju from four well-separated geographic origins. A total of 112 fungal isolates were identified by phenotypic characteristics and molecular taxonomy using sequencing the internal transcribed spacer of the rDNA and revealed 19 species from 13 genera. Enzymatic characteristics of protease and amylase, and mycotoxin production were analyzed.

Eukaryotic DNAJ/K Database: A Comprehensive Phylogenomic Analysis Platform for the DNAJ/K Family

  • Proteins in DNAJ/K families are ubiquitous, from prokaryotes to eukaryotes, and function as molecular chaperones. For systematic phylogenomics of the DNAJ/K families, we developed the Eukaryotic DNAJ/K Database (EDD). A total of 12,908 DNAJs and 4,886 DNAKs were identified from 339 eukaryotic genomes in the EDD. Kingdom-wide comparison of DNAJ/K families provides new insights on the evolutionary relationship within these families. Empowered by 'class', 'cluster', and 'taxonomy' browsers and the 'favorite' function, the EDD provides a versatile platform for comparative genomic analyses of DNAJ/K families.

Hybrid Fungal Genome Annotation Pipeline Combining ab initio, Evidence-, and Homology-based gene model evaluation

  • Fungal genome sequencing and assembly have been trivial in these days. Genome analysis relies on high quality of gene prediction and annotation. Automatic fungal genome annotation pipeline is essential for handling genomic sequence data accumulated exponentially. However, building an automatic annotation procedure for fungal genomes is not an easy task. FunGAP (Fungal Genome Annotation Pipeline) is developed for precise and accurate prediction of gene models from any fungal genome assembly. To make high-quality gene models, this pipeline employs multiple gene prediction programs encompassing ab initio, evidence-, and homology-based evaluation. FunGAP aims to evaluate all predicted genes by filtering gene models. To make a successful filtering guide for removal of false-positive genes, we used a scoring function that seeks for a consensus by estimating each gene model based on homology to the known proteins or domains. FunGAP is freely available for non-commercial users at the GitHub site (https://github.com/CompSynBioLab-KoreaUniv/FunGAP).
