Anti-inflammatory Activity of Peel Extracts in Color-fleshed Potatoes (컬러감자외피 추출물의 항염활성)

  • Potatoes were first introduced outside the Andes region four centuries ago, and have become an integral part of much of the world's food. Potatoes were first introduced into Europe in the 16th century and Korea in the early 19th century. In the nutritional aspects, potatoes contain abundant vitamins and minerals, as well as an assortment of phytochemicals such as carotenoids and natural phenols. Chlorogenic acid constitutes up to 90% of potato natural phenols. Due to the high content of potato functional compounds, it has known that potatoes are effective in the prevention of various human diseases. Recently, color-fleshed potatoes 'Hongyoung' and 'Jayoung' were developed by RDA, and it has reported that they have high content of anthocyanin. Additionally they show higher radical scavenging activity compared to white or yellow fleshed potatoes. So it will be expected that the consumption of color-fleshed potatoes grandually increase by pre-peeled potatoes and color potato chips. This study was conducted to enhance the peel of color-fleshed potatoes utilization and to determine the biological activity of peel of color-fleshed potatoes extract. The anti-inflammatory effects on ethanol extract and its solvent fraction were also evaluated. The anti-inflammatory activities of $CHCl_3$ fraction was evaluated for inhibitory activities against lipopolysacchride(LPS) induced nitric oxide(NO) and prostaglandin $E_2(PGE_2)$ production as well as inducible nitric oxide synthase(iNOS) and cyclo oxygenase-2(COX-2) protein expressions in RAW264.7 cell lines. The fraction inhibitory activity for both tests with $IC_{50}$ values showed in the ranges of $25{\sim}50{\mu}g/ml$. This result revealed that $CHCl_3$ fraction of Jayoung's peel is expected to be good candidate for development into source of anti-inflammatory agent.

Comparison of Antioxidant Effects of Different Korean Pear Species (종에 따른 배나무 열매의 항산화 활성 비교)

  • In this studies, contents and activities of antioxidative substances in the fruits of a pear cultivar and five Korean wild pear lines were investigated. The content of total phenolic compounds was highest with Pyrus pyrifolia var. culta "Shingo" (7.68 $mg{\cdot}g^{-1}$, D.W.) while total flavonoids was highest with P. ussuriensis var. acidula (5.66 $mg{\cdot}g^{-1}$, D.W.). The contents of total polyphenols and flavonoids varied according to plant species, but in general pyrifolia lines had higher total polyphenols than total flavonoids whereas in ussuriensis lines total flavonoid contents were higher than total polyphenols. The scavenging effects of DDPH radicals was highest with highest with P. ussuriensis var. seoulensis ($RC_{50}$=1.45 $mg{\cdot}mL^{-1}$) and that of ABTS radicals was highest with P. ussuriensis var. acidula ($RC_{50}$=0.69 $mg{\cdot}mL^{-1}$). A pear cultivar "Shingo" showed highest total polyphenol content, but scavenging activity of DDPH and ABTS radicals was lowest. The fruits of P. ussuriensis var. seoulensis contained low level of phenolic compounds, but their scavenging effects on two radicals was superior. Ferrous ion chelating effect was high in P. pyrifolia var. culta (98.93%), and in P. ussuriensis var. sinensis (68.71%), but two species had low scavenging activity. The content of phenolic compounds in wild pears was not always directly proportional to the radical scavenging activity. Neither radical scavenging activity was to ferrous ion chelating activity. In conclusion the fruits of P. ussuriensis var. acidula, P. ussuriensis var. seoulensis, and P. ussuriensis var. sinensis had great potential for the development of new antioxidative functional food because their antioxidative activity is higher or at least similar to P. pyrifolia var. culta. However, due to their different levels of antioxidative effects, the development of methods of using them jointly, not singly, to obtain maximum level of antioxidants is recommended.

Inhibitory Effects of Extracts from Arabis glabra on Lipopolysaccharide Induced Nitric Oxide and Prostaglandin E2 Production in RAW264.7 Macrophages (RAW264.7 대식세포에서 장대나물 추출물의 Nitric oxide 및 Prostaglandin E2생성 저해효과)

  • Arabis glabra is a localized common rhizomatous flowering plant, This plant is often used in Korean traditional systems of medicine as a remedy for blood cleaning, detoxification, abscess, gastrospasm, arthritis, contraction and diarrhea. Generally drugs that are used for arthritis have antinociceptive and anti-inflammatory properties. However, validity of the anti-inflammatory activity has not been scientifically investigated so far. Therefore, the aim of this study was to investigate the anti-inflammatory potential of A. glabra using the ethanolic extract and its sub-fractions. To evaluate the anti-inflammatory effects, we examined the inflammatory mediators such as nitric oxide (NO) and prostaglandin E2 (PGE2) on RAW 264.7macrophages. Our results indicated that hexane and chloroform fraction significantly inhibited the LPS-induced NO and PGE2 production in the cells. The hexane fraction inhibitory activity for NO tests with IC50 values showed in 21.0 ㎍/㎖. The chloroform fraction inhibitory activity for PGE2 tests with IC50 values showed in 18.0 ㎍/㎖. These efficacy are expected to be able to present the potential for the development of health functional food for the prevention inflammatory diseases because it has sufficient preventive medical possibilities. Further, it is determined that it is necessary to further study the mechanism of cytokine and protein expression associated with inflammation.

Screening of Effective Extraction Conditions for Increasing Antioxidant Activities of Licorice Extracts from Various Countries of Origin (원산지별 감초추출물의 항산화활성 증가를 위한 효율적인 추출조건 탐색)

  • In this work, licorice extracts were prepared using various extraction conditions such as extraction solvent, temperature, and time from Glycyrrhiza uralensis (G. uralensis) produced in Korea and China and Glycyrrhiza glabra (G. glabra) in Uzbekistan. The optimum extraction condition was selected from the extraction yields and antioxidative activities of extracts. Korea licorice extracts showed the highest free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity (46.05%) under the extraction condition of 85% ethanol at $60^{\circ}C$ for 6 hours. The prominent ROS (reactive oxygen species) scavenging activity using luminol-dependent chemiluminescence assay and the cellular protective effect against $^1O_2$ induced cellular membrane damage were also shown from the extracts obtained from the same condition. Especially, Korea G. uralensis extracts exhibited the higher prominent protective effect (${\tau}_{50}$ = 116.4 min) than (+)-(+)-${\alpha}$-tocopherol (${\tau}_{50}$ = 28.5 min) and the extraction yield of Korea licorice extract was 18.75%, which is 1.2 times and 2.5 times higher than that of Uzbekistan and China, respectively. These results indicate that the extraction condition of 85% ethanol at $60^{\circ}C$ for 6 hours is optimal to prepare licorice extracts, which can be applicable as anti-oxidative cosmetic materials.

Establishment of Optimal Rearing Conditions for the Production of Tenebrio molitor Larvae (갈색거저리 유충 생산을 위한 효율적인 사육조건 확립)

  • Tenebrio molitor larvae contain large amounts of proteins, lipids and other functional materials, enabling this insect to be used as an edible food source in animal feeds and for industrialization. Although many efforts have been made to set up mass rearing systems, few studies have been conducted to establish optimal rearing conditions for the production of high quality T. molitor larvae. Herein we investigated 1) the effects of additional diets on the survival and fecundity of the insect, 2) the relationship between oviposition period and the uniformity of larval size, 3) the effects of rearing density and temperature on insect development, and 4) the storage stability of eggs and pupae at low temperatures given possible temporary production discontinuation. The addition of carrot and zucchini to the traditional wheat bran diet significantly increased the survival and fecundity rate of adult T. molitor. Of the three different oviposition sampling periods (3, 7, and 14 days) used to investigate the uniformity of the hatched larvae in each treatment, the period of 3 and 7 days provided higher uniformity than the 14 days oviposition period. Larval development was faster at $30^{\circ}C$ than at 20, 25, and $35^{\circ}C$. Interestingly, oviposition rates were highest at $20^{\circ}C$ but showed much slower larval development and lower uniformity at $30^{\circ}C$. Regarding the effect of larval rearing densities (1, 10, 20, 30, 40, and 50 larvae per 90 mm diam. dish), larval weight was significantly reduced at higher rearing densities, but larval longevity and length were not influenced by rearing density. The 30 larvae/dish is suggested to be a reasonable density to be applied to mass production systems. When kept at $4^{\circ}C$, T. molitor eggs showed a significant reduction in hatching rate; however, when stored under the same conditions, pupae emergence rates remained high until 10 weeks, suggesting that storage at low temperatures is more suitable for the pupal stage than the egg stage. Our findings suggest that an increase in T. molitor adult survival and fecundity rates and a uniformity of hatched larval development can be achieved with the following recommendations: a combination diet (including wheat bran), a 7-day oviposition period; a larvae-rearing temperature of $30^{\circ}C$, a rearing density of 30 larvae/dish, and the storage of pupal stages at low temperatures in the case of rearing discontinuation. This study serves as a strong foundation for the successful mass production of high quality T. molitor larvae.

Quality Properties of Madeleine added with Black Bean Chungkukjang Flour (검은콩 청국장 가루를 첨가한 마들렌의 품질 특성)

  • The chromaticity, mechanical, and sensory properties of madeleine were investigated during its manufacturing process based on additions of black bean chungkukjang flour containing functional health ingredients and a high nutritient value. 1. The moisture content decreased with increasing content of the chungkukjang flour$(16.1{\sim}13.7%)$. 2. The brightness of the Madeleine decreased significantly from 75.24 to 53.61 by increasing the content of the black bean chungkukjang flour. The degree of red color increased significantly from -0.31 to 4.93 by increasing the content of the black bean chung kukjang flour. 3. For the mechanical properties, the hardness increased significantly with increasing amounts of the black bean chungkukjang flour (p<0.05). The degree of fracturability, adhesiveness, and gumminess also increased with the increasing the content, but the fracturability showed no difference by either addition. The springiness and cohesiveness decreased significantly by increasing the content of the black bean chungkukjang flour, showing an opposite result compared to the hardness. 4. The results of the sensory evaluation indicated significant differences in color among the samples(p<0.05). The color of the madeleine samples with added black bean chungkukjang flour was more preferred than that of the sample without any additive. The degree of preference increased in the order of the quantity of the additive. The moistness and softness became significantly drier and less soft, respectively, with an increasing quantity of the black bean chungkukjang flour (p<0.05). For the overall acceptance, the BC10 sample added with 10% of black bean chungkukjang flour added had the highest acceptance. This may be because it did not have a strong chungkukjang odor and had the fewest differences from the control Madeleine without the additive, in terms of its moistness and softness. Based upon these results, adding10 % of black bean chungkukjang flour in the manufacture of Madeleine is an appropriate quantity with regard to its structural and sensory characteristics.

Trends of mushroom science and mushroom industry (버섯과학과 버섯산업의 동향)

  • World production of mushrooms has been increasing 10-20% every year. Recently, Pleurotus eryngii and P. nebrodensis are very popular as new mushroom species for cultivation. Two kinds of mushrooms, Gumji (Ganoderma) and Soji, were described in old book of Samguksagi (History of the three kingdoms; 1145) in Koryo-dynasty. Many kinds of mushrooms were also described in more than 16 kinds of old books during Chosun-dynasty in Korea. One hundred and sixty commercial strains of 25 species in mushrooms were distributed to cultivators. By the way, only 8 varieties of them have registered variety protection. Mushroom industry as important export products developed from 1960 to 1980. Production of mushrooms as food was 181,828 metric tons valued at 800 billion Korean won in 2003. Isolated and identified substances from mushrooms are promising antifungal, antiinflammatory, antitumor, antiviral (anti-HIV), antibacterial & antiparasitic, antidiabetic, immunomodulating, kidney tonic, hepatoprotective, nerve tonic, and sexual potentiator. These substances can also be used for blood pressure regulation and effective against cardiovascular disorders, hypocholesterolemia & hyperlipidemia, and chronicbronchitis. Mushroom products including pharmaceuticals, tonics, healthy beverages, functional biotransformants, and processed foods have also became available on the markets. Compost and feed can likewise be made from mushroom substrates after harvest. The mushroom industry is already one of the fastest growing investment sectors in Korea. By the way, there is a need to strain improvement for variety protection, advanced cultivation technology at low cost for growers, and control of demand and supply for marketing in order to more upgrade development of mushroom industry in the future.

Collection, Identification and Hepatic Effect of Native Cordyceps militaris (새로운 번데기 동충하초의 수집, 동정 및 간기능에 미치는 효과)

  • Entomopathogenic fungus Cordyceps militaris is famous for its medicinal efficacies. It has been reported to have various pharmacological activities such as anti-tumour, insecticidal, antibacterial, immunomodulatory and antioxidant. In this study, we investigated the effect of the extract of C. militaris (MPUN8501), which was identified by the analysis of the nucleotide sequences of 5.8S ribosomal RNA, on the function of liver. C. militaris powder was extracted using hot water extracts method as time, volume and temperature and using method as differential polarity of organic solvent. Each fraction was tested for the improvement of hepatic enzyme alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity. The BuOH extracts (CME) had highest activity which was used for the test of toxicity and efficacy of C. militaris. The enhancing effect of CME on the activity of ADH and ALDH was much more than medicine, drink, natural tea etc. Thus CME promoted the resolution of alcohol and acetaldehyde in rats, inducing recovery to normal condition rapidly. Furthermore, oral administration of CME effectively protected the carbon tetrachloride-induced acute hepatic injury as revealed by the hematological parameters (levels of sGOT and sGPT) and histological observation. CME was ascertained to be safe by regulatory toxicity studies of single dose toxicity and genotoxicity. These results suggest that CME would be useful for the maintaining normal hepatic activity as a functional health food.

Comparative Analysis of Anti-oxidative, Anti-inflammatory, Anti-allergy, and Whitening Effects of Different Solvent Extracts from Zizania latifolia (고장초 추출 용매의 에탄올 함량에 따른 항산화, 항염증, 항알러지, 미백 활성 비교 분석)

  • This study was performed to evaluate the anti-oxidative, anti-inflammatory, anti-allergy, and whitening effects of Zizania latifolia ethanol extracts prepared from 5 different ethanol concentrations (10, 30, 50, 70, and 90%). As the ethanol concentration in the extraction solvent was increased, the radical scavenging activities also increased. The inhibitory activity of Z. latifolia ethanol extracts on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells tended to increase as the content of ethanol increased. The highest inhibitory activity was obtained with 70% ethanol extract. The antiallergy effects of Z. latifolia ethanol extracts were tested by measuring the release of ${\beta}-hexosaminidase$ in IgE-sensitized RBL-2H3 cells. The suppressive effect of Z. latifolia ethanol extracts increased in a dose-dependent manner as the proportion of ethanol increased, except for the 10% ethanol extract. Furthermore, the inhibitory effects of Z. latifolia ethanol extracts against melanin production in ${\alpha}-melanocyte$ stimulated hormone (MSH)-stimulated B16F0 cells increased as the ethanol ratio increased, and 70 and 90% ethanol extracts showed similar inhibitory activities to arbutin, a positive control, at $250{\mu}m$. The present study confirmed the efficacy of Z. latifolia ethanol extracts in various areas, demonstrating antioxidative, anti-inflammation, antiallergy, skin protective, and skin whitening effects, with no cytotoxicity. It could be used as a raw material in functional foods, as well as in cosmetics.

Production Medium Optimization for Monascus Biomass Containing High Content of Monacolin-K by Using Soybean Flour Substrates (기능성 원료를 기질로 이용하는 Monacolin-K 고함유 모나스커스 균주의 생산배지 최적화)

  • During the last decade, monacolin-K biosynthesized by fermentation of red yeast rice (Monascus strains) was proved to have an efficient cholesterol lowering capability, leading to rapid increase in the market demand for the functional red yeast rice. In this study, the production medium composition and components were optimized on a shake flask scale for monacolin-K production by Monascus pilosus (KCCM 60160). The effect of three different soybean flours on the monacolin-K production were studied in order to replace the nitrogen sources of basic production medium (yeast extract, malt extract and beef extract). Among the several experiments, the production medium with dietary soybean flour to replace a half of yeast extract was very good for monacolin-K production. Plackett-Burman experimental design was used to determine the key factors which are critical to produce the biological products in the fermentation. According to the result of Plackett-Burman experimental design, a second order response surface design was applied using yeast extract, beef extract and $(NH_4)_2SO_4$ as factors. Applying this model, the optimum concentration of the three variables was obtained. The maximum monacolin-K production (369.6 mg/L) predicted by model agrees well with the experimental value (418 mg/L) obtained from the experimental verification at the optimal medium. The yield of monacolin-K was increased by 67% as compared to that obtained with basic production medium in shake flasks.
