Generation of Transgenic Rice without Antibiotic Selection Marker through Agrobacterium-mediated Co-transformation System (아그로박테리움 동시 형질전환 시스템을 통한 항생제 선발 마커가 없는 형질전환벼의 생산)

  • Development of transgenic plant increasing crop yield or disease resistance is good way to solve the world food shortage. However, the persistence of marker genes in crops leads to serious public concerns about the safety of transgenic crops. In the present paper, we developed marker-free transgenic rice inserted high molecular-weight glutenin subunit (HMW-GS) gene ($D{\times}5$) from the Korean wheat cultivar 'Jokyeong' using Agrobacterium-mediated co-transformation method. Two expression cassettes comprised of separate DNA fragments containing only the $D{\times}5$ and hygromycin resistance (HPTII) genes were introduced separately into Agrobacterium tumefaciens EHA105 strain for co-infection. Each EHA105 strain harboring $D{\times}5$ or HPTII was infected into rice calli at a 3: 1 ratio of EHA105 with $D{\times}5$ gene and EHA105 with HPTII gene expressing cassette. Then, among 66 hygromycin-resistant transformants, we obtained two transgenic lines inserted with both the $D{\times}5$ and HPTII genes into the rice genome. We reconfirmed integration of the $D{\times}5$ and HPTII genes into the rice genome by Southern blot analysis. Wheat $D{\times}5$ transcripts in $T_1$ rice seeds were examined with semi-quantitative RT-PCR. Finally, the marker-free plants containing only the $D{\times}5$ gene were successfully screened at the $T_1$ generation. These results show that a co-infection system with two expression cassettes could be an efficient strategy to generate marker-free transgenic rice plants.

  • Relative entropy is a divergence measure between two probability density functions of a random variable. Assuming that the random variable has only two alphabets, the relative entropy becomes a cross-entropy error function that can accelerate training convergence of multi-layer perceptron neural networks. Also, the n-th order extension of cross-entropy (nCE) error function exhibits an improved performance in viewpoints of learning convergence and generalization capability. In this paper, we derive a new divergence measure between two probability density functions from the nCE error function. And the new divergence measure is compared with the relative entropy through the use of three-dimensional plots.

  • The entropy and heat capacity are calculated for the Morse oscillator model in order to test the quality of the partition function recently deduced by two of us. It is found that these analytical expressions are more reliable than the usually accepted one and give better results in the calculation of thermodynamic properties.

  • For a stationary field $\{X_{\b{j}},\b{j}{\;}\in{\;}{\mathbb{Z}}^d_{+}\}$ of associated random variables with distribution function $F(x)\;=\;P(X_{\b{1}}\;{\leq}\;x)$ we study strong consistency and asymptotic normality of the empirical distribution function, which is proposed as an estimator for F(x). We also consider strong consistency and asymptotic normality of the empirical survival function by applying these results.

Factors Associated with Lung Function Recovery at the First Year after Lung Transplantation

  • Purpose: Post-operative pulmonary function is an important prognostic factor for lung transplantation. The purpose of this study was to identify factors affecting recovery of forced expiratory volume in 1 second (FEV1) at the first year after lung transplantation. Materials and Methods: We retrospectively reviewed the medical records of lung transplantation patients between October 2012 and June 2016. Patients who survived for longer than one year and who underwent pulmonary function test at the first year of lung transplantation were enrolled. Patients were divided into two groups according to whether they recovered to a normal range of FEV1 (FEV1 ${\geq}80%$ of predicted value vs. <80%). We compared the two groups and analyzed factors associated with lung function recovery. Results: Fifty-eight patients were enrolled in this study: 28 patients (48%) recovered to a FEV1 ${\geq}80%$ of the predicted value, whereas 30 patients (52%) did not. Younger recipients [odds ratio (OR), 0.92; 95% confidence interval (CI), 0.87-0.98; p=0.010], longer duration of mechanical ventilator use after surgery (OR, 1.14; 95% CI, 1.03-1.26; p=0.015), and high-grade primary graft dysfunction (OR, 8.08; 95% CI, 1.67-39.18; p=0.009) were identified as independent risk factors associated with a lack of full recovery of lung function at 1 year after lung transplantation. Conclusion: Immediate postoperative status may be associated with recovery of lung function after lung transplantation.

Prediction of Postoperative Lung Function in Lung Cancer Patients Using Machine Learning Models

  • Background: Surgical resection is the standard treatment for early-stage lung cancer. Since postoperative lung function is related to mortality, predicted postoperative lung function is used to determine the treatment modality. The aim of this study was to evaluate the predictive performance of linear regression and machine learning models. Methods: We extracted data from the Clinical Data Warehouse and developed three sets: set I, the linear regression model; set II, machine learning models omitting the missing data: and set III, machine learning models imputing the missing data. Six machine learning models, the least absolute shrinkage and selection operator (LASSO), Ridge regression, ElasticNet, Random Forest, eXtreme gradient boosting (XGBoost), and the light gradient boosting machine (LightGBM) were implemented. The forced expiratory volume in 1 second measured 6 months after surgery was defined as the outcome. Five-fold cross-validation was performed for hyperparameter tuning of the machine learning models. The dataset was split into training and test datasets at a 70:30 ratio. Implementation was done after dataset splitting in set III. Predictive performance was evaluated by R2 and mean squared error (MSE) in the three sets. Results: A total of 1,487 patients were included in sets I and III and 896 patients were included in set II. In set I, the R2 value was 0.27 and in set II, LightGBM was the best model with the highest R2 value of 0.5 and the lowest MSE of 154.95. In set III, LightGBM was the best model with the highest R2 value of 0.56 and the lowest MSE of 174.07. Conclusion: The LightGBM model showed the best performance in predicting postoperative lung function.

Antioxidative and Protective Effects of Ulmus davidiana var. japonica Extracts on Glutamate-Induced Cytotoxicity in PC 12 Cells (느릅나무 추출물의 항산화 효과 및 L-glutamate 유래 PC12 세포독성 보호효과)

  • Antioxidative and protective effects of Ulmus davidiana var. japonica against oxidative damages induced by glutamate in PC 12 cells were investigated. Inhibitory activity against $FeSO_{4}-H_{2}O_{2}$-induced oxidative stress and DPPH radical-scavenging activity were detected in ethyl acetate and butanol fractions of ethanol extracts from stems and roots. Ethyl acetate and butanol fractions of ethanol extracts from roots significantly inhibited glutamate-induced cytotoxicity and reactive oxygen species in PC 12 cells. These results demonstrate ethyl acetate and butanol fractions of ethanol extracts of U. davidiana var. japonica have potent protective effect against glutamate-induced oxidative stress.

Antioxidant Activity and Contents of Bioactive Components in Polar Microalgae (극지미세조류의 유용성분 함량 및 항산화 활성)

  • In this study, bioactive components such as polyohenols, flavonoids and tocopherols were determined in cultured polar microalgae (Fragilariopsis pseudonana, Chaetoceros neogracile, Stellarima microtrias, Porosiara pseudodenticular). Antioxidant activity of methanol extracts of polar microalgae was also investigated. ${\alpha}-Tocopherol$ contents in Fragilariopsis pseudonana were almost two times higher than those of Chaetoceros neogracile in. The antioxidant activity of methanol extracts of Fragilariopsis pseudonana methanol extracts determined by ABTS assay was higher than other algae. Total polyphenol contents of methanol extracts also showed a similar trend as antioxidant activity. The protective activity against oxidative damages induced by glutamate in PC 12 cells was shown in only Chaetoceros neogracile.

Effect of degree of milling on the chemical composition of rice oil

  • The degree of milling is an important factor in terms of the nutritional value and the economic return of the milled rice. This study was to investigate the changes in compositions of the oils obtained from rice with different degree of milling. The content of total tocols in rice significantly (p<0.05) decreased, as degree of milling increased. The relative % of -tocopherol in brown rice had lower than those in milled rices whereas the relative % of -tocotrienol, and -tocotrienol in brown rice higher than those in milled rices. -Oryzanol content significantly (P<0.05) decreased from 198.5 mg/kg rice for brown rice (0% milling) to 65.5 125.0 mg/kg rice for rice samples with 5.6, 8.0, and 9.6 % milling. -Sitosterol was most abundant sterol representing 50 % - 56% of the total sterol content in all analyzed samples. The content of total sterol as well as each sterol isomers in rice significantly (P<0.05) decreased, as degree of milling increased. A similar trend was observed in changes of octacosanol and squalene.

  • This paper propose ZVS-HB type resonant DC/DC converter have earth different output characteristics using division ratio, not only a source division function but a resonant function and soft switching technique(ZVS, ZCS) instead of conventional source division capacitor. Circuit analysis generally described using normalized parameters most of characteristics with division ratio of source division capacitor. Also, this paper citified a rightfulness of characteristic analysis in comparison with a theoretical values and a experimental values obtain from experiment using Power-MOSFET.
