Structural Design and Characterization of a Channel-forming Peptide

  • A 16-residue polypeptide model with the sequence acetyl-YALSLAATLLKEAASL-OH was derived by rational de novo peptide design. The designed sequence consists of amino acid residues with high propensity to adopt an alpha helical conformation, and sequential order was arranged to produce an amphipathic surface. The designed sequence was chemically synthesized using a solid-phase method and the polypeptide was purified by reverse-phase liquid chromatography. Molecular mass analysis by electro-spray ionization mass spectroscopy confirmed the correct designed sequence. Structural characterization by circular dichroism spectroscopy demonstrated that the peptide adopts the expected alpha helical conformation in 50% acetonitrile solution. Liposome binding assay using Small Unilamellar Vesicle (SUV) showed a marked release of entrapped glucose by interaction between the lipid membrane and the tested peptide. The channel-forming activity of the peptide was revealed by a planar lipid bilayer experiment. An analysis of the conducting current at various applied potentials suggested that the peptide forms a cationic ion channel with an intrinsic conductance of 188 pS. These results demonstrate that a simple rational de novo design can be successfully employed to create short peptides with desired structures and functions.

The Response to Impulse Signal on Three Phase Transformer using Vector Network Analyzer (벡터 회로망 분석기 측정을 기반으로 한 3상 변압기의 시간영역 펄스 신호에 대한 응답 분석)

  • Transformer is widely used element on power system and industrial area. Especially the transformers installed at power system are exposed to an environment of arbitrary changed. Thus the prediction of degradation and the analysis of response to impulse are important. To conduct those works, the electrical characteristics of system should be analyzed, effectively. But the analysis of electrical characteristic in electric machine level such as pole and pad-mounted transformer is almost no, thus commercial VNA (Vector Network Analyzer) is used to getting the response in wide frequency range. However, the output power of VNA is usually under 10mW, so verification for effectiveness of measuring electrically large component should be conducted, firstly. Next, after getting total S-parameter of transformer, predicting impulse response can be performed in time-domain with circuit simulator. In this paper, it is introduced that verification effectiveness of VNA using transfer function from SFRA (Sweep Frequency Response Analyzer), firstly. Next, total S-parameter, six by six matix form, was built using measured 2 port S-parameter from vector network analyzer. To get the response to impulse which is defined by IEC 60060-1, time-domain simulation is conducted to ADS (Advenced Design System) circuit simulator.

  • This paper deals with the distribution characteristics of the current density and axial magnetic flux density on the vacuum interrupter with axial magnetic field type using 3 dimension finite element analysis. An axial magnetic field parallel to the current flow in the arc column can improve the current breaking capacity of vacuum interrupter by affecting the arc mode. The axial magnetic flux density on the contact electrode surface is analyzed by inputting external current as a function of the transient time for sine half wave. And it also is analyzed within the gap distance of the contact electrode. The peak value of current but is decreased with the descending current on the contact electrode surface and within the gap distance of the contact electrode. The residual magnetic field is generated on the contact electrode surface and within the gap distance in the instant of zero current, which is due to the influence of eddy currents. The phase shift due to eddy currents, defined as time difference between the maximum value of current and axial magnetic field, is about 1ms in the center point of gap distance.

  • In this paper, various modulation techniques, including the legacy Global Navigation Satellite System (GNSS) signal modulation techniques, are introduced and the spectral characteristics and correlation characteristics of signals with various modulation techniques are analyzed based on numerical simulation. With the development of various GNSS services, the limited frequency band has become increasingly saturated, and issues of interoperability and compatibility have emerged in the new GNSS design. Since the efficient allocation of frequency resources is closely related to spectrum design, modulation techniques are one of the important signal design parameters of new signal design. Signal modulation techniques are closely related to various figure of merits (FoMs) as well as spectrum characteristic, and in some cases there is a complicated trade-off between FoMs. Thus, the FoMs associated with modulation technology should be analyzed and the best signal candidates should be chosen carefully via the trade-off analysis for FoMs. In this paper, we define the modulation technique based on Phase Shift Keying (PSK), Binary Offset Carrier (BOC) and Continuous Phase Modulation (CPM) for the design of KPS signals, and the FoMs of signals in terms of spectrum and correlation function are evaluated. Signals with various modulation techniques are implemented through a numerical simulation, and the relevant FoMs are analyzed.

  • Onboard ATC performs the train speed ristriction function by receiving the speed limit on sections of track from wayside ATC. Onboard ATC can protect passenger from collision and derailment, and so on. So, the high level of safety and reliability for Onboard ATC is required, and by this reason, it is also required to improve the safety of Onboard ATC by applying the result of safety analysis on the Onboard ATC into its design and developing from the concept definition to the detailed desing phase. The paper introduces, when developing Onboard ATC, the safety improvement of Onboard ATC can be accomplished by applying system requirements from Risk Analysis into software and hardware.

  • This paper presents a design and analysis solutions for the general class and control parameter estimation of slotted iron-cored Permanent Magnet Linear Synchronous Motor with Halbach magnetization stator and 3-phase winding mover. In our design and analysis, stator consisting of permanent magnets and slotted iron-cored coil mover are treated in a uniform way via magnetic vector potential. For one such motor structure, we give analytical formulas for its magnetic field, back electromotive force and trust force. Moreover, the detent force are calculated by the relative permeance function. In good agreement with the estimated values from the analyses, the experimental results and Finite Element Method (FEM) confirm the validity of the design and analysis framework.

  • This paper presents the d-q axis inductances calculations method of Interior Permanent Magnet Synchronous Motor(IPMSM) by using Finite Element(FE) analysis. The inductances of d-q axis in IPMSM, which is the function of current magnitude and phase angle are computed by energy dual method. The appropriateness of the proposed FE analysis has been verified by comparing with experimental results.

  • This paper analyzes the operation characteristics of a self excited induction generator with Steinmetz connection. For this analysis, the symmetrical components analysis is used to obtain the related expressions and the excitation capacitance and the magnetizing reactance are determined in turn by the condition of self excitation which includes the input impedance of the generator as viewed across load terminals. Two simultaneous equations of the condition of self excitation itself are solved by using the real and imaginary function in an application software. This method is applied to simulate the operation characteristics when the generator is driven at rated speed and the specified excitation capacitor is connected across the lagging phase. The results show that better operation characteristics except generated frequency are obtained by using relatively large excitation capacitance and resistive load.

  • Design VE (Value Engineering) has been basic design and implementation design separately, but due to the recent change in guidelines, it is changing to implement only once in the implementation design stage. However, in the implementation design stage, construction costs, plans, and specifications based on the volume details are almost completed. Therefore, it is effective to perform the function analysis step after identifying information analysis and key issues through CostModeling based on functional center for the selected target in the process of implementing the VE performance procedure.

  • In this paper, we present a noise analysis and measurement results of a bio-radar system that can detect human heartbeat and respiration signals. The noise analysis including various phase noise effects is very important in designing the bio-radar system, since the frequency difference between the received signal and local oscillator is very small and the received power is very low. All of the noise components in a bio-radar system are considered from the point of view of SNR. From this analysis, it can be concluded that the phase noise due to antenna leakage is a dominant factor and is a function of range correlation. Therefore, the phase noise component with range correlation effect, which is the most important noise contribution, is measured using the measurement setup and compared with the calculated results. From the measurement results, our measurement setup can measure a closed-in phase noise of a free-running oscillator. Based on these results, it is possible to design a 2.4 GHz bio-radar system quantitatively which has a detection range of 50 cm and low power of 1 mW without additional PLL circuits.
