Functional Characteristics of C-terminal Lysine to Cysteine Mutant Form of CTLA-4Ig

  • CTLA-4Ig is regarded as an inhibitory agent of the T cell proliferation via blocking the costimulatory signal which is essential for full T cell activation. To improve applicability, we developed the CTLA-4Ig-CTKC in which the c-terminal lysine had been replaced by cysteine through single amino acid change. The single amino acid mutation of c-terminus of CTLA-4Ig was performed by PCR and was checked by in vitro transcription and translation. DNA construct of mutant form was transfected to Chinese hamster ovary (CHO) cells by electroporation. The purified proteins were confirmed by Western blot and B7-1 binding assay for their binding ability. The suppressive capacity of CTLA-4Ig-CTKC was evaluated by the mixed lymphocyte reaction (MLR) and in the allogeneic pancreatic islet transplantation model. CTLA-4Ig-CTKC maintained binding ability to B7-1 molecule and effectively inhibits T cell proliferation in MLR. In the murine allogeneic pancreatic islet transplantation, short-term treatment of CTLA-4Ig-CTKC prolonged the graft survival over 100 days. CTLA-4Ig-CTKC effectively inhibits immune response both in MLR and in allogeneic islet transplantation model, indicating that single amino acid mutation does not affect the inhibitory function of CTLA-4Ig. CTLA-4Ig-CTKC can be used in vehicle-mediated drug delivery system such as liposome conjugation.

AMT용 새로운 변속자동 메커니즘 설계 및 해석 (Design and Analysis of a New Shift Automation Mechanism for Automated Manual Transmission)

  • This article proposes a novel shift automation mechanism for an automated manual transmission (AMT). The development of an automated manual transmission is currently being paid considerable attention by vehicle manufacturers, with the prospects of combining the comfort of an automatic transmission and the high efficiency of a manual transmission. In order to automate the shift mechanism of a manual transmission, the proposed shift automation mechanism consists of two electric motors, cross shaped pinion gears, rack type shift rails, and a ball splined hollow shaft. First we describe the shift mechanism and operating principles of a manual transmission to investigate important design criteria for the shift automation device. And a new shift automation mechanism is described with its structure, elements, and operating principles in detail. Using a conventional manual transmission, we develop a full three-dimensional CAD model of an AMT which includes main components of the manual transmission and the designed shift automation mechanism. Finally we investigate the operating performances and feasibility of the designed AMT by a dynamic analysis.

다축 로드 시뮬레이터의 노면 프로파일 재현 소프트웨어 개발 (Realization Software Development of Road Profile for Multi-axial Road Simulator)

  • Full scale durability test in the laboratory is an essential of any fatigue life evaluation of components or structures of the automotive vehicle. Component testing is particularly important in today's highly competitive industries where the design to reduce weight and production costs must be balanced with the necessity to avoid expensive service failure. Generally, hydraulic road simulator is used to carry out the fatigue test and the vibration test. In this paper, the algorithm and software to realize the real road profile are developed. The operation software for simultaneously controlled multi-axial road simulator is developed and the input and output data are displayed window based PC controller in the real time. Futhermore, the software to generate the real road profile are developed. The validity of the software are verified by applying the belgian road, the city road, the highway, and the gravel road. The results of the above experiment show that the real road profiles are realized well after 10th iteration.

  • This study proposes a 7kW low-voltage DC-DC converter (LDC) using a coupled inductor (CI) for heavy hydrogen electric transport vehicles. The LDC uses a phase-shift manner for soft switching. SiC-MOSFET is used to reduce the loss of reverse recovery current through the use of a high switching frequency. LDC is require large transformer and inductor because of large output current. The size of transformer and inductor can be reduced by deviding the transformer and inductor into two pieces each. This work presents the experimental results of the proposed circuit.

  • 본 논문에서는 무인항공기인 방송용 멀티콥터를 이용한 Full-HD급 이상 화질의 이미지를 효율적으로 전송하기 위해 이미지 압축 센싱 기법을 적용하고, Sparse 신호의 효율적 복원을 위해 Turbo 알고리즘과 Markov chain Monte Carlo (MCMC) 알고리즘의 복원 성능을 모의실험을 통해 비교 분석하였다. 제안된 복원 기법은 압축 센싱에 기반하여 데이터 용량을 줄이고 빠르고 오류 없는 원신호 복원에 중점을 두었다. 다수의 이미지 파일로 모의실험을 진행한 결과 Loopy belief propagation(BP) 기반의 Turbo 복원 알고리즘이 Gibbs sampling기반 알고리즘을 수행하는 MCMC 알고리즘 보다 평균 복원 연산 시간, NMSE 값에서 우수하여 보다 효율적인 복원 방법으로 생각된다.

A Dynamic Test Facility for Mobile Air Conditioning Systems

  • Mobile air conditioning systems work under widely changing operating conditions. To understand the system behavior under such dynamic conditions, a test facility that can impose transient loads as well as conducting dynamic measurements is needed. To test mobile air conditioning systems including their dynamic performance under various drive cycle patterns without using full scale vehicles in a wind tunnel, a new test facility, called "dynamic simulator," is described. It can replicate real vehicle operating conditions by interacting with the system being tested based on the measured system performance and subsequently adjusting the air properties returning to the test system based on the results of a numerical cabin model. A new dynamic simulator has been designed, constructed, and verified for performing dynamic tests. It was successful in controlling the temperature and relative humidity of the air returning to the test unit within ${\pm}0.7^{\circ}C$ and ${\pm}4%$ of their respective intended values. The verification test under the New European Driving Cycle demonstrated that detailed transient behavior of the mobile air conditioning system could be measured by using this dynamic simulator.

Design of Small-Scaled Derailment Simulator for Investigating Bogie Dynamics

  • The dynamic stability of railway vehicle has long been one of the important issues in railway safety. The dynamic simulator has been used as a tool for investigating the dynamic stability of railway vehicles and wheel/rail interfaces. In particular, small scale simulators have been widely used in laboratory studies instead of full scale roller rigs which can be quite costly and rather inconvenient for testing out the effect of diverse design parameters. But techniques for design of a small scale simulator for the fundamental study about the dynamic characteristics of the wheel-rail systems and the bogie systems have not been well developed in Korea. Therefore, a research on the development of a small scale simulator for investigating bogie dynamics needs to be undertaken. The present paper investigates design of a small-scaled derailment simulator and the design of a small scale bogie. The simulator developed can be used to investigate the effect of diverse parameters such as attack angle, wheelbase and cant on dynamic behavior of the bogie and key dynamic performance parameters such as derailment coefficient and critical speed.

철도차량 부유상구조의 Floor support 재질이 차량 실내소음에 미치는 영향에 관한 연구 (A Study for Interior Noise Contribution of Support Material used in Railway Vehicle Floor)

  • To reduce interior noise of running vehicles, a floating floor construction has been widely used in recent railway industry. Among the key factors of the floating floor design, dynamic stiffness is of most important in acoustical point of view. Sometimes hard rubber type supports have often been selected due to the other design constraints such as heavy load condition, durability of rubber element and its cost etc., even though it seems like the softer support, the better isolation of noise and vibration. In this paper two representative floor supports have been considered to evaluate their effectiveness in interior noise contribution: one is a soft rubber and another is a relatively hard one. From the measured dynamic stiffness of the specimens, equivalent stiffness of actual floating floor has been derived to use in the analytical models. Calculated air-borne and structure-borne noise insulation properties of the floating floors have been compared with experiments in prototype car. In full car model interior noise levels of running vehicles have been predicted to quantify the effectiveness of the two different floating support materials and verified through the measured inside noise levels of actual train as well. By comparison with difference of running noise levels two materials for floor support can be investigated quantitatively so that it could be applied in floating floor design.

  • Vehicular Ad Hoc Network (VANET) is considered to be a subclass of Mobile Ad Hoc Networks (MANET). It has some challenges and issues of privacy which require to be solved before practical implementation of the system i.e., location preservation privacy. Many schemes have been proposed. The most prominent is pseudonym change based location preservation scheme. Safety message can be compromised when it sends via a wireless medium, consequently, an adversary can eavesdrop the communication to analyze and track targeted vehicle. The issue can be counter by use of pseudo identity instead of real and their change while communication proves to be a sufficient solution for such problems. In this context, a large amount of literature on pseudonym change strategies has been proposed to solve such problems in VANET. In this paper, we have given details on strategies proposed last two decades on pseudonym change based location preservation along with issues that they focus to resolve and try to give full understanding to readers.

  • 철도차량의 동적 거동 및 안정성 등을 예측하고 물리적인 특성을 이해하기 위하여 철도선진국인 영국, 프랑스, 일본 등에서는 축소모델 제작 및 시험하는 방법과 컴퓨터 시뮬레이션을 통하여 해석적으로 접근하는 방법이 사용되고 있다. 실물 규모의 동특성 시험은 대규모의 시험설비 구축에 따른 많은 비용과 시간의 소요, 그리고 시험 환경 설정의 어려움 등의 문제점이 발생하게 된다. 이러한 단점들을 극복하고자 상사법칙을 적용한 축소대차 모델을 이용하여 동적 시험 및 컴퓨터 시뮬레이션을 통한 해석적 연구를 수행하고 있다. 본 연구에서는 소형탈선시뮬레이터를 이용한 1/5 축소대차의 주행안정성 시험에서 신뢰성이 검증된 해석모델을 이용하여 주행안정성에 미치는 매개변수 중 주행속도, 차체 하중의 2가지 변수에 따른 1/5 축소대차의 주행안정성 해석을 수행하고, 그에 따른 동적특성을 검토하였다.
