Gene-Diet Interaction on Cancer Risk in Epidemiological Studies

  • Genetic factors clearly play a role in carcinogenesis, but migrant studies provide unequivocal evidence that environmental factors are critical in defining cancer risk. Therefore, one may expect that the lower availability of substrate for biochemical reactions leads to more genetic changes in enzyme function; for example, most studies have indicated the variant MTHFR genotype 677TT is related to biomarkers, such as homocysteine concentrations or global DNA methylation particularly in a low folate diet. The modification of a phenotype related to a genotype, particularly by dietary habits, could support the notion that some of inconsistencies in findings from molecular epidemiologic studies could be due to differences in the populations studied and unaccounted underlying characteristics mediating the relationship between genetic polymorphisms and the actual phenotypes. Given the evidence that diet can modify cancer risk, gene-diet interactions in cancer etiology would be anticipated. However, much of the evidence in this area comes from observational epidemiology, which limits the causal inference. Thus, the investigation of these interactions is essential to gain a full understanding of the impact of genetic variation on health outcomes. This report reviews current approaches to gene-diet interactions in epidemiological studies. Characteristics of gene and dietary factors are divided into four categories: one carbon metabolism-related gene polymorphisms and dietary factors including folate, vitamin B group and methionines; oxidative stress-related gene polymorphisms and antioxidant nutrients including vegetable and fruit intake; carcinogen-metabolizing gene polymorphisms and meat intake including heterocyclic amins and polycyclic aromatic hydrocarbon; and other gene-diet interactive effect on cancer.

  • Age specific characteristics in Korean diet and their changes were analyzed with the demand model including the number of family members by age group as independent vasiables Urban household budget data, 773 household data in 1977 and 947 household data in 1983, was applied to the demand model. A series of F-test was carried out to set up the age groups by food item. The main results can be summarized as follows; 1. Food consumption patterns are remarkably different between 0-13 years old age generation and over 40 years old generation. The 0-13 years old generation reveals higher preference for milk, wheat product, fruit and milk and lower preference for rice and vegetable than any other age generation. This characteristics of the childhood diet has been formed In the generation born after 1970's and thus will be reserved as their generational characteristics. On the other hand, over 40 years old age group prefers rice and fish to fruit, milk and wheat product. Therefore we can conclude that over 40 years old age group maintains the traditional Korean diet pattern. 2. The preference for fish is low. in 20-30 years old age group than in any other age group and wheat product is Preferred highest in 6-13 years old age group of all age groups. Meanwhile the most remarkable change In food consumption pattern is shown in meats and vegetables. Namely in 30 40 years old age group the preference for meat has increased substantially and that vegetable has decreased to a great extent. 3. The declining per capita consumption of rice spreads like a wave to higher age. groups, but over 40 years old age group bas played an important role in reducing the decreasing rate of rice consumption. In order to restrain the declining rice consumption a special integrated food policy is required to induce rice consumption in less 30 years old generation.

Evaluation of the effect of a school garden as an educational didactic tool in vegetable and fruit consumption in teenagers

  • BACKGROUND/OBJECTIVES: Increasing the consumption of vegetables and fruits in Mexico remains a challenge. Promoting sustainable food production systems through schools may be an effective way to educate young people about food and nutrition issues. A study of nutritional education in adolescents, based on the school garden, is necessary in order to evaluate its effects on the consumption of fruits and vegetables among middle- and upper-income segments of the population. The objective of this study was to evaluate the effect of an educational intervention, accompanied by a school garden as an educational teaching tool, to improve vegetable and fruit consumption by Mexican teenagers attending a private middle/high school. SUBJECTS/METHODS: Teenagers between 12 and 18 years of age (n = 126) attending a private middle/high school in Queretaro, Mexico participated in a 3-arm, controlled, comparative impact study using a vegetable and fruit consumption frequency questionnaire, food consumption diaries, a psychosocial factor assessment questionnaire of vegetable and fruit consumption, and structured interviews. The participants were randomized into 3 experimental groups: 1) food education + school garden (FE + SG), 2) FE only, and 3) control group (CG). RESULTS: The FE + SG and FE groups significantly increased the frequency and daily intake of vegetables and fruits compared to the CG. The FE + SG group showed greater understanding of, reflection upon, and analysis of the information they received about vegetable and fruit consumption, as well as a greater willingness to include these in their daily diet. CONCLUSIONS: FE accompanied by a SG as a teaching tool is more effective at promoting vegetable and fruit consumption than either education alone or control in teenagers in middle-upper income segments of the population.

Dietary Patterns and Risk of Colorectal Cancer: A Systematic Review of Cohort Studies (2000-2011)

  • Objectives: This systematic review of cohort studies aimed to identify any association between specific dietary patterns and risk of colorectal cancer (CRC). Dietary patterns involve complex interactions of food and nutrients summarizing the total diet or key aspects of the diet for a population under study. Methods and materials: This review involves 6 cohort studies of dietary patterns and their association with colorectal cancer. An exploratory or a posteriori approach and a hypothesis-oriented or a priori approach were employed to identify dietary patterns. Results: The dietary pattern identified to be protective against CRC was healthy, prudent, fruits and vegetables, fat reduced/diet foods, vegetables/fish/poultry, fruit/wholegrain/dairy, healthy eating index 2005, alternate healthy eating index, Mediterranean score and recommended food score. An elevated risk of CRC was associated with Western diet, pork processed meat, potatoes, traditional meat eating, and refined grain pattern. Conclusion: The Western dietary pattern which mainly consists of red and processed meat and refined grains is associated with an elevated risk of development of CRC. Protective factors against CRC include a healthy or prudent diet, consisting of vegetables, fruits, fish and poultry.

비파잎 추출물이 고지방식이로 유도한 비만쥐에 미치는 항비만 효과 (Anti-Obesity Effect of Eriobotrya japonica Leaves Extract on Obese Mice Induced by High-Fat Diet)

  • 본 연구는 비파잎 추출물이 고지방식이로 유도한 비만쥐에 미치는 항비만 효과를 조사하였다. 8주 동안 고지방식이에 100 mg/kg b.w., 500 mg/kg b.w.의 비파잎 추출물을 보충한 그룹과 고지방식이군만을 제공한 그룹을 비교하였다. 비파잎 추출물은 식이섭취량에는 영향을 주지 않으면서도 고지방식이군과 비교하여 유의적으로 체중증가를 줄이고 내장지방의 축적을 억제하였다. 또한, 비파잎 추출물을 급여한 군은 고지방식이군과 비교하여 혈중 지질, 간지질의 축적, 혈중 렙틴 농도를 감소시켰다. 이상의 실험 결과로 고지방식 이와 함께 급여한 비파잎 추출물은 체중, 간 및 지방조직의 무게 감소와 더불어 혈장 지질 농도를 개선하고 혈장 렙틴농도를 감소시키므로 항비만 효과를 가진 천연 기능성 식품의 소재로써 활용 가능성이 있을 것으로 생각된다.

로페로마이드로 유도한 변비랫드 모델에서 프락토올리고당 및 과채복합 추출물의 변비개선 효과 (Effects of Fructooligosaccharide and Fruit and Vegetable Complex Extracts on Loperamide-induced Constipation in Rats)

  • Loperamide에 의한 변비는 위 배출, 소장 및 대장 운동성을 감소시켰으며, 과채복합추출물(Fruit and vegetable complex extracts, FVCE)은 이러한 효과를 예방한다. 우리는 다음과 같은 실험에서 로페로마이드(3mg/kg, sc, 14일)로 유발된 변비 수컷 Sprague-Dawley 쥐에 대한 프락토올리고당 및 과채복합추출물의 효과를 조사하였다. 쥐를 무작위로 정상대조군 쥐(일반식이), 변비 쥐(일반식이 + 로페로마이드), 200mg FVCE(200mg/kg/day FVCE + 로페로마이드가 보충된 일반 식이)로 처리한 변비 쥐, 400mg FVCE로 처리한 변비 쥐(200mg/kg/day FVCE와 로페로마이드가 보충된 일반 식단)으로 시험군을 구성하였다. 대조군 쥐에 비해 로페로마이드로 처리된 쥐에서 대변 배설량이 적고 대변 수분 함량이 더 낮았다. FVCE을 경구투여 하면 분변배설량의 감소를 억제하고 로페로마이드로 처리된 쥐의 대변 수분 함량. 로페로마이드를 처리한 쥐에서는 대장선와세포의 점액 생성과 대변 및 점막표면의 점액 함량이 감소하였다. 그러나 FVCE 처리군에서는 대장선와세포의 뮤신 함량이 증가하였고, Aclain blue로 염색된 점액층은 로페로마이드 처리군에 비해 FVCE 처리군에서 유의적으로 두꺼워졌다. 쥐 회장에서 로페로마이드는 회장 운동성을 억제한다. 이러한 결과는 FVCE가 로페로마이드에 의한 대장 연동 운동 억제 완화에 효과적이며 FVCE추출물이 변비 예방에 효과적 일 수 있다는 것을 나타낸다.

Effects of Fruit By-product Extracts Supplementation on Growth Performance and Nutrient Digestibility in Growing Pigs

  • A total of 96 pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc] with an initial BW of $27.94{\pm}0.92kg$ were used in a 6-week experiment to determine the effects of dietary supplementation with fruit by-product extracts on growth performance and nutrient digestibility in growing pigs. Pigs were randomly allotted to four treatments : 1) CON (basal diet), 2) PRO (CON + 0.5% procyanidin), 3) HES (CON + 0.5% hesperidin), 4) TAN (CON + 0.5% tannin). There were six replications per treatment with four pigs per pen. Supplementation had no effect (p>0.05) on average daily gain, average daily feed intake, and G/F. The apparent total tract digestibility (ATTD) of dry matter and nitrogen was increased (p<0.05) in the HES treatment relative to the PRO treatment. Pigs fed the HES and TAN diets had greater (p<0.05) ATTD of energy than pigs fed PRO diet. The ATTD of ash was greatest (p<0.05) in HES treatments. In addition, the ATTD of calcium was greater (p<0.05) in HES treatments than in CON and PRO treatments. Overall, the results of this study indicated that dietary supplementation with 0.5% fruit by-products did not affect growth performance, but inclusion of 0.5% hesperidin increased nutrient digestibility in growing pigs.

  • 호박과실파리의 유충은 전북 완주군 진안의 호박 피해과에서 채집하였다. 실내 사육실 $24{\pm}2^{\circ}C$, RH $65{\pm}5%$, 14L:10D에서 용화를 유도하였으며 번데기에서 우화한 성충은 전지분유를 기본으로 한 diet를 제공하였다. 호박과실파리 짝짓기 관련 행동의 관찰은 우화 후 40일의 암, 수 성충을 대상으로 야외 케이지를 이용하여 2008년 7월과 10월 초 오후 자연광 아래에서 실시하였다. 성충 수컷 날개 떨림의 광 조건은 200 lux-20 lux였고, 30 lux에서 활발하였다. 날개 떨림 후 성충 암, 수의 올라타기는 150 lux-30 lux 사이에서 관찰 되었으며, 올라타기도 30 lux에서 빈도가 높았다. 교미의 경우 올라타기 광 조건과 유사한 150 lux-20 lux에서 관찰되었다. 날개 떨림 중의 수컷은 암컷과 교미 성공률이 64% 수준으로 날개 떨림 행동을 통해 이끌려 날아온 암컷이 수컷의 앞쪽에 있을 때 대부분 교미가 이루어졌다. 날개 떨림을 하지 않은 경우 앞쪽, 옆쪽, 그리고 어느 쪽에 암컷이 있어도 교미가 이루어지지 않았다.

저열량 생즙 식이가 성인의 혈청 페리틴 농도에 미치는 영향 (Effects of a Low-Calorie Raw Juice Diet on the Level of Serum Ferritin in Korean Adults)

  • This study examines the effects of a low-calorie raw juice diet on the level of serum ferritin in adults and analyzes nutrient intake from the diet. There were significant differences between juices; the highest calorie was provided by pear juice, highest crude protein, vitamin A, and vitamin B2 levels were from green Juice 1; and highest vitamin C and vitamin B1 levels were from fruit juices. The ratio of estimated energy requirements (EER) for the participants was 56.2% from the raw juice diet. The percentages of recommended intake (RI) from the raw juice diet of protein (57.9%), dietary fiber (19.1%), niacin (6.2%), calcium (0.1%), and magnesium (0.2%) were lower than 75%. However, those of RI of vitamin A, vitamin B1, vitamin B2, vitamin B6, and vitamin C were 1796.5%, 7481.7%, 1915.5%, 30858.7%, and 7500%, respectively, exceeding the tolerable upper intake level (UL) for vitamin A, vitamin B6, and vitamin C. There were significant decreases in weight, the body mass index (BMI), body fat mass, and skeletal muscle mass in males and females. After the diet program, serum iron and SOD (superoxide dismutase) showed significant decreases, whereas RBC, hemoglobin, hematocrit, and serum ferritin showed significant increases. There were negative correlations between serum ferritin and weight and between serum ferritin and skeletal muscle mass for all participants. There were negative correlations between serum ferritin and skeletal muscle mass for males and between serum ferritin and body fat mass for females. These results suggest that a raw juice diet can supplement a regular diet to prevent excess or deficient nutrient intake.

Organic acid and sugar contents in different domestic fruit juices

  • Organic acid and sugar contents in ten kinds of juices (two orange juices, two grape juices, two aloe juices, two citron juices, one bokbunja, and one grapefruit juice) were analyzed by high-performance liquid chromatography. Citric acid was detected in all the fruit juices analyzed. Grape juices contained the highest tartaric acid content (67.85 - 99.37 mg/100 mL) while orange and grape juices contained a high content of malic acid (151.67 - 211.18 mg/100 mL). Lactic acid was detected in all the aloe juices (35.12 - 65.27 mg/100 mL) as well as in one orange (203.8 mg/100 mL) and one grape juice (112.28 mg/100 mL). Citrus fruit juices (A, A', F) showed the highest content of total organic acids as 902.81 - 1,103.7 mg/100 mL. With regard to sugar contained in the juices, lactose and maltose were not detected in any of the fruit juices. Even when comparing the same kind of fruit juice, sugar (fructose, glucose, and sucrose) content showed a significant difference (p < 0.05) depending on the manufacturer. In the grape juices, contents of fructose (6.86 - 7.51) and glucose (6.23 - 7.3 g/100 mL) were higher than in other juices. One serving size of the juices (180 mL) analyzed in this study can provide approximately 3.3 - 4.9% of total daily energy requirements when consumed as part of the 2,200 kcal/day diet required for an adult.
