In vitro fertilization have been performed to know whether the frozen semen has fertilizing ability and can be used clinically. The results of cultured and developed embryos obtained are as follows: 1. The semen was frozen in three media for the good viability. The viability was more than 50% and the motility was also moderate (grade III), 2. As the 33 oocytes were collected from 45 follicles, the oocyte recovery rate was 73.3%. Among them, mature and immature ova were 5% each, and premature ova were 69.7%, When the first polar body was appeared, above ova were inseminated after adequate incubation with activated sperms. 3. The main components of three freezing medium containing egg yolk, glycerol and pyruvate respectively were the best for sperm viability, and Ham's F-10 medium was used for the fertilization and culture of eggs. 4. The results of in vitro fertilization of 33 ova, showed the second polar body developed in 12%, polyspermia in 24%, 1-cell embryo in 21% and 2-cell embryo in 9%. One mature ova developed to blastocyst via 16-cell to 32-cell embryo. The fertilization rate was 66%. 5. Above mentioned results represent that the frozen semen has fertilizing ability and can be used practically in the clinic.
This study was carried out to investigate the effects of season influencing semen characteristics, frozen-thawed sperm viability and testosterone concentration in Duroc boars. There were no significant differences in the semen volume and sperm concentration of Duroc boars among spring, summer, autumn and winter. However, the pH of sperm-rich and sperm-poor fractions in autumn and winter season was higher than in spring and summer season in Duroc boars. Sperm motility and normal acrosome of raw semen in Duroc boars did not differ significantly among spring, summer, autumn and winter. However, motility and normal acrosome of frozen-thawed sperm were higher in spring season than in summer, autumn and winter. Serum testosterone concentrations in Duroc were higher in spring than summer, autumn and winter. In conclusion, when serum testosterone concentrations were higher in seasons, frozen-thawed sperm viability in Duroc boars were higher.
Heparin binding proteins (HBPs) are produced by accessory glands. These are secreted into the seminal fluid, bind to the spermatozoa at the time of ejaculation, favour capacitation, acrosome reaction, and alter the immune system response toward the sperm. The present study was conducted with an objective to assess the effect of purified seminal plasma-HBPs (SP-HBPs) on cross bred cattle bull sperm attributes during two phases of cryopreservation: Pre freezing and freezing-thawing. SP-HBPs were purified from pooled seminal plasma by heparin affinity chromatography. Three doses of SP-HBPs i.e. 10, 20, $40{\mu}g/mLs$ semen were standardized to find out the optimum dose and $20{\mu}g/mLs$ was found to be an optimum dose. Semen as such and treated with SP-HBPs was diluted with sodium citrate-egg yolk diluter and cryopreserved as per the standard protocol. Sperm parameters i.e. motility, viability, Hypo-osmotic swelling test (HOST), acrosome damage, in vitro capacitation and lipid peroxidation were evaluated in SP-HBP treated and untreated (control) semen at both phases of cryopreservation. A considerable variation in percent sperm motility, viability, membrane integrity (HOST), acrosome damage, acrosome reaction and lipid peroxidation was observed at both phases among the bulls irrespective of the treatment. Incubation of neat semen with $20{\mu}g/mL$ SP-HBP before processing for cryopreservation enhanced the average motility, viability, membrane integrity by 7.2%, 1.5%, 7.9%, and 5.6%, 6.6%, 7.4% in pre-frozen and frozen-thawed semen in comparison to control. There was also an average increase of 4.1%/3.9% in in vitro capacitation and acrosome reaction in SP-HBPs-treated frozen-thawed semen as compared to control. However, binding of SP-HBPs to the sperm declined acrosome damage and lipid peroxidation by 1.3%/4.1% and 22.1/$32.7{\mu}M$/$10^9$ spermatozoa in SP-HBP treated pre-frozen/frozen-thawed semen as compared to control, respectively. Significant (p<0.05) effects were observed only in motility, HOST and in vitro acrosome reaction. It can be concluded that treatment of neat semen with SP-HBPs before cryopreservation minimized the cryoinjury by decreasing the generation of reactive oxygen species.
This study was undertaken to determine whether the presence of fertility-associated antigen (FAA) in semen would influence semen characteristics and conception rate of artificial insemination in Hanwoo. The response to FAA of 36 heads of proven bull, 7 heads of young bull, and 27 heads of performance-tested bull was that one proven bull was FAA-negative and the others were FAA-positive, therefore FAA-negative bull was 1.4%. FAA-negative bull was lower in first and second semen concentrations than those of FAA-positive bull in 5,301 semen of 21 heads of proven bull, then FAA-negative bull was fewer as 11.5% in total sperm counts. The estrus of 22 heads was 70d-nonreturned in 36 cows first inseminated with frozen semen of FAA-negative bull, but that of 249 heads in 378 cows first inseminated with frozen semen of FAA-positive bull. Each conception rate was 61.1% and 65.9%, respectively. The difference of conception rates was 4.8%. These results indicate that the response of FAA to semen were influenced semen characteristics and conception rate of artificial insemination, but further investigations are needed to confirm the results.
This cepseiment was carried out to cerify the effect of thawing methods and preservative temperature on the sperm motility and fertility after thawing semen with plastic straws in fresh and warm water. Sperm motility in vitro stored at room temperature after thawing were conducted by the various storage hours. A field trial after thawing semen with warmed water in straws from Cheju native cows involving 4 technicians and 800 cows first (or second) services gave the following results. The thawing methods of warmed water for one minute in semen motility were considerably higher than that in iced water during 12 hours after thawing semen, however, the sperm survival index of ice-water shwed a better results according as the time passed away, but not significant differences. Preservative temperaure at 5$^{\circ}C$ (iced water) after thawing gave significantly better results than that of thawed at 3$0^{\circ}C$ (warmed water). The N R rate to 175 inseminations with semen thawed at 15-2$0^{\circ}C$ (fresh water) was 82.8%, 80.9% for 610 inseminations thawed in warm water. Conception rate ofthe semen thawed in warm water for 10-60 secs gave no significant difference among storage hours, because the semen used to be inseminated within one hour almost, but in decreased when semen thawed at the period of one minute over.
본 연구는 진도개 동결정액 제조기술을 정립하기 위하여 진도개 정액성상과 동결 전후 정액의 활력과 생존율 및 CASAs를 이용한 운동성 등에 대하여 조사하였고, 백구와 황구간, 개체간의 정액성상과 내동성을 비교, 조사하였다. 이상의 연구결과는 다음과 같다. 1. 총 63회 정액채취 후 신선정액의 평균 정액량 3.8 $m\ell$, 농도 145.6$\times$$10^{6}$$m\ell$, 총정자수 396.2$\times$08/$m\ell$, 전진운동율 79.7% 및 생존율 89.5% 였다. 황구와 백구간에는 황구가, 개체간에는 황구 2호가 정자농도, 총정자수, 전진운동정자율 및 생존율 등의 정액성상에서 유의적으로 우수하였다(P<0.05). 2. 동결전.후 정자의 전진운동율과 생존율을 46회 조사한 결과 동결전 73.5%와 82.3%를, 동결후 51.1%와 64.9%를 나타내 동결과정이 전진운동율과 생존율에 영향이 있었으며, 역시 황구와 백구간에는 황구가, 개체간에는 황구 2호가 동결전후 전진운동율과 생존율에서 유의적인 차이가 인정되었다 (P<0.05). 3. 동결.융해정자의 보다 객관적인 평가를 위하여 CASA system을 이용한 총 44회 평가한 결과, 생존율 65.6%, 전진운동율 54.8%, VAP 75.3 $\mu\textrm{m}$/sec, VCL 90.0 $\mu\textrm{m}$/sec, VSL 69.4 $\mu\textrm{m}$/sec 및 ALH 4.4 $\mu\textrm{m}$로 동결 융해 정액의 운동성은 양호하였고, 황구와 백구간의 운동성에는 유의적인 차이가 없었으나, 개체간에는 역시 황구 2호가 운동성이 우수하여 유의적인 차이가 인정되었다 (P<0.05). 4. 채취된 정액중 백구는 46%(13/28), 황구는 94%(33/35)가 동결정액 제조가 가능해 황구가 내동성이 좋았으며, 전체적으로 73%(46/63) 동결정액 제조가 가능하였다. 결론적으로 진도개의 동결정액을 제조하기 위하여 정액성상 및 동결 전후 운동성을 조사한 결과 동결정액 제조와 생산체계의 구축이 가능하였으며, 황구와 백구간, 개체간의 정액성상과 동결전후의 운동성에 차이가 인정되므로 정액성상과 내동성을 고려한 종견선발 체계가 필요하다고 사려되었다.
This study was conducted to establish a freezing method of miniature pig spermatozoa. The semen 더aculated from PWG M-type miniature pig was collected by gloved-hand method. The semen was diluted with same volume extender (m-Modena B). The frozen solution used frozen solution of four different (LEY, TCG, BF-5 and m-Modena+egg yolk) for find optimal frozen solution in miniature pig sperm. The diluted semen for frozen rate assay was added to LEY solution (solution I: 11% lactose+egg yolk; solution II: solution I+glycerol+OEP), and frozen depending on freezing rate by the three different freezing methods (A: until $5^{\circ}C$ for 1 hrs, holding at $-102^{\circ}C$ for 10 min; B: until $5^{\circ}C$ for 2 hrs, holding at $-102^{\circ}C$ for 10 min; C: until $5^{\circ}C$ for 3 hrs, holding at -80 and $-102^{\circ}C$ for 10 min). Semen cooled until $5^{\circ}C$ was added with glycerol 1, 3 and 5%, and take a equilibrium time for 0, 10 and 30min. Frozen-thawed sperm were evaluated for viability, acrosomal status and morphological abnormality. The results of frozen-thawed sperm ability by frozen solution, viability was higher in LEY solution compared to other three different frozen solution. AR pattern of LEY solution were lower than other three different frozen solution. The results of freezing rate, viability was higher in B method compared to other methods (p<0.05). Acrosomal statute was intacted in A and B methods than C method. The experiment for glycerol condition was showed that sperm viability was higher in extender with 1% and 3% glycerol and equilibrium time of 0 min. The acrosome damage was lower in extender with 1% glycerol and equilibrium time of 10 min than other conditions. In conclusion, the optimal conditions for cryopreservation of miniature pig spermatozoa obtained in LEY frozen solution, cooling rate of 1~2 hrs, 1~3% glycerol concentrations and glycerol equilibrium time of 0~10 min.
This study was designed to analyze boar sperm to compare motility, acrosome morphology, viability and ATP by various preservation methods between Duroc boar A with cold shock resistance sperm and Duroc boar B with cold shock sensitivity sperm. Semen volume, sperm concentration, motility and normal acrosome between Duroc boar A and B did not show any differences within 2 h after collection. There were no differences in sperm motility and normal acrosome between boar A and B at 1 day of preservation at $17^{\circ}C$ and $4^{\circ}C$, respectively. However, sperm motility and normal acrosome from 2 day of preservation at $17^{\circ}C$ and $4^{\circ}C$, respectively, were higher for boar A than boar B. The frozen-thawed sperm motility and normal acrosome were higher for boar A than boar B. The sperm viability and ATP concentration according to storage period of liquid semen at $17^{\circ}C$ and $4^{\circ}C$ were higher for boar A than boar B. Also, the sperm viability and ATP concentration of frozen-thawed semen were higher for boar A than boar B. In conclusion, we found out that the original quality of boar semen with cold shock resistance sperm played an important role.
본 연구는 Duroc 종모돈의 정액성상, 동결-융해후의 정자생존성 및 혈청 중 테스토스테론의 농도에 미치는 영향을 봄 (3~5월)과 여름 (6~8월)으로 나누어 실시한 바, 그 얻어진 결과는 다음과 같았다. 1. Duroc 종모돈의 정액량, pH, 정자농도는 봄과 여름철에 큰 차이가 없었다. 2. 봄철 및 여름철의 전체 정액량, 정자농후부분 그리고 정자희박부분에서 봄과 여름간의 차이는 나타나지 않았다. 3. 원정액 정자에서는 운동성 및 정상첨체에서 봄과 여름철간에 차이가 없었다. 그러나 동결-융해정자의 운동성 및 정상첨체는 봄철의 종모돈에서 생산한 정자가 여름철의 종모돈에서 생산한 정자보다 우수하였다. 4. 듀록 종모돈의 혈청 중 테스토스테론의 농도 변화는 봄과 여름에 각각 2.15 ng/$m\ell$과 0.65 ng/$m\ell$으로, 봄철 테스토스테론의 농도가 여름철의 농도 보다 높았다(P<0.05). 5. 이상의 결과를 종합해 보면, 듀록 종모돈에 있어 정자의 내동성은 혈청 중 테스토스테른의 농도가 높을수록 우수한 것으로 나타났다.
