STUDIES ON THE EXTRACTION OF SEA WEED PROTEINS 2. Extraction of NaCl and Alcohol Soluble Proteins (해조단백질 추출에 관한 연구 2. 식염가용성 및 알콜가용성 단백질의 추출)

  • In present study, the effects of various factors including the solvent concentration, extraction time and temperature, the ratio of sample vs extraction solvent, (w/v) and pH upon the extractability of the NaCl and alcohol soluble proteins of marine algae were investigated. Eight species of fresh algae, the major ones in consumption as food, namely Porphyra suborbiculata, Undarie pinnatifida, Hizikia fusiforme, Sargassum, fulvellum, Enteromorpha linza, Sargassum kjellmanianum, Codium coarctatum, and Ulva pertusa were used for the extraction of NaCl soluble protein and dried materials of four species, Perphyra suborbiculata, Undaria pinnatifida, Enteromorpha linza and Sargassum fulvellum were used for the extraction of alcohol soluble protein. The frozen and mascerated samples were prepared by the same method described in previous paper (Ryu, 1977). And the dried materials were moistened with alcohol solution before freezing. The effect of solvent concentration on the extractability of NaCl soluble protein differed from species. The extractability of Undaria Pinnatifide, Hizikia fusiforme, Perphyra suborbiculata, Enteromorpha linza, and Ulva pertusa reached maxima at 0.25M NaCl solution while the 1.0M for Sargassum fulvellum, Saygassum kjellmanianum and Codium coarctatum. In case of alcohol soluble proteins, it was shown at $20\%$ ethanol solution for Porphyra suborbiculata, Undaria pinnatifida, Enteromorpha linza, and Sargassum fulvellum. Variation of the ratio of sample vs solvent gave slight effect upon the extractability, but the ratio of 1:30(w/v) seemed most efficient for the extraction of NaCl soluble proteins and 100 ml solvent added to 1 g dried sample was effective in case of alcohol soluble proteins. Extraction time has a minimal effect upon the extraction of alcohol soluble protein, and approximately 21 to $43\%$ of algal protein was extracted within 1 hour. But in case of NaCl soluble protein extraction, the effect of time revealed differently from species to species resulting in that the extraction for 1 hour gave a maximum extractability in Ulva pertusa and Enteromorpha linza, 2 hours in Porphyra suborbiculata, Codium coarctatum and 3 hours in Undaria pinnatifica, Hizikia susiforme, Sargassum fulvellum and Sargassum kjellmanianum. When the NaCl soluble protein of Undaria pinnatifida and Enteromopha linza was extracted at various temperature, the most effective extraction temperature was $40^{\circ}C$ while the temperature was $50^{\circ}C$ for Undaria pinnatifida and $60^{\circ}C$ for Hixikia fusiforme, Sargassum fulvellum, Sargassum kjellmanianum and Codium coarctatum. Bus in case of alcohol soluble extraction, the optimum temperature was $30^{\circ}C$ for Enteromorpha linza and $40^{\circ}C$ for Undaria pinnatifida, Sargassum fulvellum and Porphyra suborbiculata. In the effect of pH on extractability, the maximum extractability of NaCl soluble proteins was obtained at pH 7to 8 and pH 8 to 9 for alcohol soluble protein.

Residues of Azoxystrobin during Cultivation and Processing of Ginseng (인삼의 재배 및 가공단계 별 Azoxystrobin 잔류성)

  • The aim of this study was to determine the processing and reduction factors for ginseng and its commodities during ginseng processing to obtain information of pesticide residue in ginseng. For this study, azoxystrobin was used in two field containing 6 years old ginseng plants. Ginsengs were harvested and processed to obtain different commodities (Dried ginseng, red ginseng and ginseng water and alcohol extracts, red ginseng water and alcohol extracts) for pesticide analysis. The amount of residue levels from wonju and icheon for fresh ginseng were 0.05, $0.03\;mg\;kg^{-1}$ dried ginseng were 0.12, $0.14\;mg\;kg^{-1}$, red ginseng were both $0.05\;mg\;kg^{-1}$, ginseng alcohol extract were 0.28, $0.33\;mg\;kg^{-1}$, ginseng water extract were 0.22, $0.16\;mg\;kg^{-1}$, red ginseng alcohol extract were 0.31, $0.20\;mg\;kg^{-1}$ and red ginseng water extract were 0.09, $0.11\;mg\;kg^{-1}$ respectively. These data were under MRLs notified by KFDA. The processing factors for ginseng products were 3.25, 1.34, 7.84, 4.63, 6.15 and 2.56 respectively. The reduction factors for ginseng products were 1.19, 0.51, 3.41, 1.91, 2.74 and 1.00 respectively. These data showed increment during processing which could be due to concentration but considering water contents, residue levels were similar or decreased than the initial residue level during processing.

Effects or $H_2O$ Fraction or Dioscorea japonica Thunb with selenium on plasma Glucose and Lipid Metabolism in Streptozotocin Induced Diabetic Rats (당뇨 흰쥐에서의 참마 $H_2O$ 분획물과 Selenium 보충시 혈당 및 지질대사에 미치는 영향)

  • The purpose of this study was to investigate the effect of H$_2$O fraction of Dioscorea japonica Thunb(DJT) with selenium(Se) treatment on blood glucose and lipid metabolism in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats(180-220g) weighing were divided into five groups, that is one normal group and four diabetic experimental groups : the STZ-control group, the DJT group, the DJT-Se group and the Se group. Diabetes mellitus was induced in the male rats by injection of STZ into the tail vein at a dose of 45mg/kg B.W. The H,0 fraction of DJT(500mg/kg) given orally were administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na$_2$SeO$_3$(2mg/kg diet), which was prepared fresh daily. The body weight and food intake was monitored daily and plasma levels of glucose, insulin, hematocrit and protein were determined. The plasma concentrations of cholesterol, HDL-cholesterol, triglyceride and fire fatty acid were measured. The activities of aminotrans ferase were analysed. The body weight gain was shown to be significantly higher in the normal group than all diabetic groups. The blood glucose levels of the DIT-Se group was significantly lower compared to those of the experimental groups. The administration of H$_2$O fraction of DJT and selenium showed an increase in plasma protein concentrations. The plasma cholesterol levels of all STZ-groups were not significantly different and HDL-cholesterol levels were increased in the diabetic experimental groups fed on H$_2$O fraction of DJT or Se supplementation. Plasma triglyceride levels were lower in the DJT-Se and Se group than in the STZ-control group. free fatty acid levels were not significantly differ among STZ-control groups. STZ treatment increased aminotransferase activity and that of DJT group was highest. In conclusion, the data from the present experiments indicate that the treatment of the H$_2$O fraction of DJT with selenium showed a synergistic effect and the two can have an influence on hyperg1ycemia and lipid metabolites when administered together. Effect of LED as Light Quality on the Germination, Growth and Physiological Activities of Broccoli Sprouts (LED 광질이 브로콜리 새싹의 발아, 생장 및 생리활성에 미치는 영향)

  • This study was carried out to investigate into the effect of light-emitting diode (LED) for the light quality as a light source on the broccoli seed germination and the physiological activity of vegetable sprouts. We have also germinated seeds of the broccoli and applied LED as a light quality such as blue, green, red, white, yellow and red + blue color lights to their sprouts for 14 hours and kept dark for 10 hours at the temperature of $25^{\circ}C$ (day)/$18^{\circ}C$ (night). Broccoli sprouts were extracted by methanol and their physiological activities were examined. All broccoli seeds were germinated at 3 days after seeding regardless of the light color. Total sprout fresh weight were mostly became highest by 0.389g (10 plants) at 8 days after seeding when their sprouts were grown under blue color light. Total phenol compound contents in broccoli sprouts were extremely increased by $83.0\;mg{\cdot}L^{-1}$ under the white light, and total flavonoid contents were most much more by $72.6\;mg{\cdot}L^{-1}$ under the blue light. DPPH radical scavenging activity at $2,000\;mg{\cdot}L^{-1}$ were most highest by 93.5% in broccoli sprouts grown under the white light. Nitrite radical scavenging activity at the concentration of $500\;mg{\cdot}L^{-1}$ in sprout extracts were the most increased by 66.9% under the yellow light, and tyrosinase inhibition activity at $2,000\;mg{\cdot}L^{-1}$ in sprout extracts were by 14.5% under red light.

  • The transmissible spongiform encephalopathies (TSEs) disease group are fatal neurodegenerative disorders affecting a wide range of hosts. The group includes kuru and Creutzfeldt-Jakob disease (CJD) in humans, scrapie in sheep and goats and Bovine spongiform encephalopathy (BSE) in cattle. The exact nature of the infectious agent involved in the transmission of these diseases remains controversial. However, a central event in their pathogenesis is the accumulation in infected tissues of an abnormal form of a host-encoded protein, the prion protein (PrP). Whereas the normal cellular protein is fully sensitive to protease ($PrP^{sen}$), the disease-associated prion protein ($PrP^d$) is only partly degraded ($PrP^{res}$), its amino-terminal end being removed. BSE was first reported in the mid-80s in the UK. Ten years later, a new form of human prion disease, variant CJD (vCJD) developed in the wake of the BSE epidemic, and there is now strong scientific evidence that vCJD was initiated by the exposure of humans to BSE-infected tissues, thus indicating a zoonotic disease. However, the ban on the feeding of animal-derived proteins to ruminants, and the apparent lack of vertical transmission of BSE, have led to a decline in the incidence of the disease within cattle herd and therefore, an assumed decreased risk for human contacting vCJD. The origin of the original case(s) of BSE still remains an enigma even though three hypotheses have been raised. Hypotheses are i) sheep- or goat-derived scrapie-infected tissues included in meat and bone meal fed to cattle, ii) a previously undetected sporadic or genetic bovine TSE contaminating cattle feed or iii) originating from a human TSE through animal feed contaminated with human remains. A host cellular membrane protein ($PrP^C$), which is abundant in central nervous system tissue, appear to be conformationally altered in the diseased host into a prion protein ($PrP^{Sc}$). This $PrP^{Sc}$ is detergent insoluble and partially protease-resistant ($PrP^{res}$). The term $PrP^{res}$ is normally used to describe the protein detected after protease treatment, in techniques such as Western immunoblotting, and enzyme-linked immunosorbant assay using fresh/frozen tissue. Immunohistochemistry may performed with formalin-fixed tissues. Also, clinical signs of the BSE are one of the major diagnostic indicators. Recently, atypical forms (known as H- and L-type) of BSE have appeared in several European countries, Japan, Canada and the United States. An unusual case was also reported in a miniature zebu. The atypical BSE fall into two groups based on the relative molecular mass (Mm) of the unglycosylated $PrP^{res}$ band relative to that of classical BSE, one of the higher Mm (H-type) and the other lower (L-type). Both types have been detected worldwide as rare cases in older animals, at a low prevalence consistent with the possibility of sporadic forms of prion diseases in cattle. This raises the unwelcome possibility that vCJD could increase in the human population. Now, active surveillance program against BSE is going on in Korea. In regional veterinary service lab, ELISA is applied to screen the BSE in slaughter and confirmatory tests by Western immunoblotting and immunohistochemisty are carried out if there are positive or suspect in the screening test. Also, the ruminant feed ban is rigorously enforced. Removal of specified risk materials such as brain and spinal cord from cattle is mandatory process at slaughter to prevent the infected material from entering the human food chain.

Effect of Temperature on the Quality and Storability of Cherry Tomato during Commercial Handling Condition (유통중 온도관리가 방울토마토의 품질과 저장성에 미치는 영향)

  • This study was carried out in order to investigate the effect of temperature of treatment and storage on the longevity of 'Unicorn' tomatoes of light red maturity stage during commercial handling conditions encountered while exporting over long distances. Tomato stored at $5^{\circ}C$ and $11^{\circ}C$ temperature with 85% relative humidity by pre-treating handling temperature that was using from field to before shipment as a winter temperature $5^{\circ}C$, spring temperature $11^{\circ}C$ and summer temperature $25^{\circ}C$ for 3 days. On the final storage day, $25^{\circ}C/11^{\circ}C$ (treated/stored) tomatoes showed the highest respiration and ethylene production rate; whereas the lowest respiration and ethylene production rate was found for $5^{\circ}C/5^{\circ}C$ treated and stored tomatoes. Tomatoes treated and stored at $5^{\circ}C/5^{\circ}C$ showed higher marketability, without evidence of fungal rot, decay or spots for 23 days. The fresh weight loss under all treatment conditions increased gradually during $5^{\circ}C$ and $11^{\circ}C$ storage temperatures. The higher firmness and soluble solids were determined from $5^{\circ}C/5^{\circ}C$ and $5^{\circ}C/11^{\circ}C$ treated and stored tomatoes repectively, than from others tomatoes on the final day of storage. In addition, $5^{\circ}C/5^{\circ}C$ tomatoes showed higher vitamin C contents than tomatoes stored at other temperatures, on the final day of storage. As the ripening and storage period progressed, the titratable acidity increased, but declined (P < 0.05) thereafter, due to over ripe tomatoes under all treatment conditions. These results show that $5^{\circ}C/5^{\circ}C$ treated and stored light red maturity stages of 'Unicorn' tomatoes are optimum to export because they show the highest storability and marketability. Moreover, the marketability of light red maturity stage of 'Unicorn' tomato maintained for 2 weeks in $25^{\circ}C/11^{\circ}C$ treated and stored temperature that might be the export temperature from Korea to Japan in summer season. This research result could be useful in helping tomato growers and exporters to get optimum market value by satisfying the buyer and consumer with a fresher product.

Effect of Submergence and Air Exposure of the Shoot on Growth, Nutrient Uptake and Photosynthesis in Monochoria vaginalis Presl. (물달개비 경엽(莖葉)의 침수여부(沈水與否)에 따른 생장(生長), 양분흡수(養分吸收) 및 광합성(光合成) 비교(比較))

  • Growth, nutrient uptake and photosynthesis as affected by submersion of shoot in pickerel weed (Monochoria vaginalis Presl.) were determined. The shoots of pickerel weeds in hydroponic culture were subjected to the submerged or emerged condition at 3- or 5-leaf stage for 8 or 10 days. Under submerged condition, growth in plant height was enhanced, but leaf number, leaf area, fresh and dry weight were reduced compared to those under the emerged condition. Similar responses in growth to submergence were obtained with the pickerel weeds rooted in the soil. Under submergence, chlorophyll content increased during the first 2 days, but thereafter remarkably decreased at 3-leaf stage and after the first 4 days at 5-leaf stage. Compared to the emerged condition, uptakes of $NH_4\;^+$-N, $NO_3\;^-$-N, $P_2O_5$ and $K^+$ were reduced, but uptakes of $Ca^{++}$ and $Mg^{++}$ increased under the submerged condition. Photosynthetic rate of shoot under water, measured by $CO_2$electrode, showed the maximum by 210 ${\mu}$moles $HCO_3\;^-$/g F.W. at the 8th day after submergence(DAS) at 3-leaf stage and 320 ${\mu}$moles $HCO_3\;^-$/g F. W. at 6 DAS at 5-leaf stage. These results indicate that pickerel weeds grow much better when the shoot is air-exposed and are less tolerable to submergence at 3 leaf-stage than at 5-leaf stage.

Growth Characteristics, Seed and Forage Productivity of New Naked Oat (Avena nuda L.) Cultivar "Jungmo2005" (귀리 품종 '중모2005'의 생육특성과 종실 및 조사료 생산성)

  • A new naked oat cultivar, 'Jungmo2005'(Avena nuda L.), was developed for food and forage use by National Institute of Crop Science, RDA in 2010. It was derived from a cross between $F_1$['Early80'/'Gwiri33'] and $F_1$['Early80'/'Gwiri23']. 'Early80', a covered oats, has early heading and high yielding, while 'Gwiri23' of covered type and 'Gwiri33' of naked type has early heading with large-size grain. 'Jungmo2005' has the characteristics of narrow and long leaves of pale green color, middle diameter culm of yellow color and medium grain of whitish yellow color. Cultivar 'Jungmo2005' had 2 days earlier heading date (May 9) than the check cultivar 'Seonyang' (May 11) in field condition. The 'Jungmo2005' showed better winter hardiness than that of the check cultivar, and similar to the check cultivar in respect to lodging resistance. The 'Jungmo2005' had 105 cm of culm length, 21.5 cm of spike length, 658 spikes per $m^2$, 82 grains per spike, 23.3 g of 1,000-grain weight, and 622 g of test weight. The grain yield of 'Jungmo2005' was averaged $3.38MT\;ha^{-1}$, which was 6% higher than that of the check. Average forage fresh and dry matter yield of 'Jungmo2005' harvested at milk-ripe stage were 44.8 and $12.6tone\;ha^{-1}$, respectively, compared with 47.5 and $12.5tone\;ha^{-1}$ of the check. The protein content of the 'Jungmo2005' was similar to the check (7.6% and 7.5%, respectively), while ADF (28.6%) and NDF (51.5%) were lower than the check (31.4% and 57.0%, respectively). TDN content and RFV were higher than those of the check (66.3%, 120.3 and 64.1, 105.2, respectively). 'Jungmo2005' is recommended for fall sowing cropping only in the south area where daily minimum mean temperatures are averaged higher than $-4^{\circ}C$ in January, and it should not be cultivated in mountain areas, where frost damages is likely to occur. The areas would do better only to sow in spring season.

Characterization of a new commercial strain 'Goni' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 백색느타리 품종 '고니'의 육성 및 그 특성)

  • To develop new white variety of Oyster mushroom, all white varieties which have been collected and kept in the lab were revived and screened their cultural characteristics. 84 intra-specific Oyster mushroom hybrids between the white-colored mutants Suhan and Wonhyeong were developed using hyphal anastomosis technique in 2007. The Po2007-63 ($2842-7{\times}0205-7$) was shown the best cultural characteristics, selected to be a new variety and named as 'Goni'. The new commercial strain, 'Goni' has white pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Goni' are of an excellent quality in that not only the stipe is thick and long but also the pileus is small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25-30^{\circ}C$ and $10-16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body is about 3 to 5 days depending on the temperatures. The shape of fruiting body is thin funnel shape. Fruiting body production per bottle was about $91{\pm}13$ g which is almost 97% quantity compared to that of other variety 'Miso'. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Goni' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the growth of mushroom were much faster in mushroom complete medium at $20-25^{\circ}C$, but not at $25^{\circ}C$. Similar results were observed with other variety 'Miso'. Analysis of the genetic characteristics of the new commercial strain 'Goni' showed a major DNA profile as that of the parental Suhan when primer URP 1 was used, but different to 'Miso' that was used as a control. When screens were performed with primer URP 2, DNA patterns were similar both to that of the parents and 'Miso'. This new variety of the white Oyster mushroom has a clean and fresh image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.

  • Aquaculture of olive flounder started in the middle of 1980's and now farming has been taken place in many places along the coastal line in Korea. The taste of olive flounder has a good chewy texture because of high collagen content, low fat content, so it is popular for sliced raw fish. Olive flounder is popular among Koreans but the consumption pattern is uniformly so as to be used as sliced raw fish but not other ways. So, now there needs to develop high valued-processed food using olive flounder. This study was set to investigate the processing of terrine by using olive flounder, in which terrine is French style meat loaf that is well favored around the world. In this study, terrine was prepared by chopping olive flounder meat with 39 g egg white and 10 mL fresh cream (per 50 g fillet) and then seasoned with 5 mL lemon juice, 5 mL brandy, 0.05 g salt and 0.05 g pepper. The 25 g of dough was placed on a vinyl wrap, put with 2 g cheese, and layered an another 25 g dough, and then rolled up and wrapped by aluminium foil. Two different cooking methods were used for terrine processing in this study. Terrine-1 was cooked by vacuum sealed in polyethylene film ($20{\times}30{\times}0.05mm$) after boiling for 5 min and stored at $-20^{\circ}C$ for 7 days. Terrine-2 was prepared by vacuum sealed in polyethylene film ($20{\times}30{\times}0.05mm$) and stored at $-20^{\circ}C$ for 7 days. After 7 days, Terrine-1 was thawed and then heated up in microwave for 2 min (Sample-1), while Terrine-2 was thawed and then boiled in water for 5 min (Sample-2). Viable bacterial count, chemical composition, pH, salinity, hardness, TBA, free amino acid content, and sensory evaluation were measured for both Sample-1 and Sample-2. Especially, the scores of sensory evaluation of Sample-2 is slightly higher than that of Sample-1. On the other hand, there were no significant differences on color, odor, taste, texture, and overall acceptance between Sample-1 and Sample-2.
