Influence of hydrogel encapsulation during cryopreservation of ovarian tissues and impact of post-thawing in vitro culture systems in a research animal model

  • Objective: Using domestic cats as a biomedical research model for fertility preservation, the present study aimed to characterize the influences of ovarian tissue encapsulation in biodegradable hydrogel matrix (fibrinogen/thrombin) on resilience to cryopreservation, and static versus non-static culture systems following ovarian tissue encapsulation and cryopreservation on follicle quality. Methods: In experiment I, ovarian tissues (n=21 animals; 567 ovarian fragments) were assigned to controls or hydrogel encapsulation with 5 or 10 mg/mL fibrinogen (5 or 10 FG). Following cryopreservation (slow freezing or vitrification), follicle viability, morphology, density, and key protein phosphorylation were assessed. In experiment II (based on the findings from experiment I), ovarian tissues (n=10 animals; 270 ovarian fragments) were encapsulated with 10 FG, cryopreserved, and in vitro cultured under static or non-static systems for 7 days followed by similar follicle quality assessments. Results: In experiment I, the combination of 10 FG encapsulation/slow freezing led to greater post-thawed follicle quality than in the control group, as shown by follicle viability (66.9%±2.2% vs. 61.5%±3.1%), normal follicle morphology (62.2% ±2.1% vs. 55.2%±3.5%), and the relative band intensity of vascular endothelial growth factor protein phosphorylation (0.58±0.06 vs. 0.42±0.09). Experiment II demonstrated that hydrogel encapsulation promoted follicle survival and maintenance of follicle development regardless of the culture system when compared to fresh controls. Conclusion: These results provide a better understanding of the role of hydrogel encapsulation and culture systems in ovarian tissue cryopreservation and follicle quality outcomes using an animal model, paving the way for optimized approaches to human fertility preservation.

  • The novel coronavirus 2019 is called COVID-19 has outspread swiftly worldwide. An early diagnosis is more important to control its quick spread. Medical imaging mechanics, chest calculated tomography or chest X-ray, are playing a vital character in the identification and testing of COVID-19 in this present epidemic. Chest X-ray is cost effective method for Covid-19 detection however the manual process of x-ray analysis is time consuming given that the number of infected individuals keep growing rapidly. For this reason, it is very important to develop an automated COVID-19 detection process to control this pandemic. In this study, we address the task of automatic detection of Covid-19 by using a popular deep learning model namely the VGG19 model. We used 1300 healthy and 1300 confirmed COVID-19 chest X-ray images in this experiment. We performed three experiments by freezing different blocks and layers of VGG19 and finally, we used a machine learning classifier SVM for detecting COVID-19. In every experiment, we used a five-fold cross-validation method to train and validated the model and finally achieved 98.1% overall classification accuracy. Experimental results show that our proposed method using the deep learning-based VGG19 model can be used as a tool to aid radiologists and play a crucial role in the timely diagnosis of Covid-19.

Design and Implementation of a Network-Adaptive Mechanism for HTTP Video Streaming

  • This paper proposes a network-adaptive mechanism for HTTP-based video streaming over wireless/mobile networks. To provide adaptive video streaming over wireless/mobile networks, the proposed mechanism consists of a throughput estimation scheme in the time-variant wireless network environment and a video rate selection algorithm used to increase the streaming quality. The adaptive video streaming system with proposed modules is implemented using an open source multimedia framework and is validated over emulated wireless/mobile networks. The emulator helps to model and emulate network conditions based on data collected from actual experiments. The experiment results show that the proposed mechanism provides higher video quality than the existing system provides and a rate of video streaming almost void of freezing.

  • Pulsatile swelling behaviors and their application to drug delivery system were studied by using interpenetrating polymer networks(IPN) hydrogels constructed with poly(vinyl alcohol) and poly(acrylic acid). The PVA/PAAc IPNs hydrogels were symthesized by UV irradiation tallowed by repetitive freezing and thawing method. These hydrogels showed pH and temperature sensitive swelling behaviors. From the release experiment, the release amount of model drug incorporated into these hydrogels showed pulsatile patterns. Permeability coefficients obtained by various solutes differed in response to changes of permeation conditions.

An experimental study on occurrence of intermediate peaks in ice load signals

  • The purpose of this study is to analyze the relationship between the occurrence of intermediate peak and time duration, and to conduct a review for the causes of the intermediate peak. In this test, ice impact tests were conducted using a bow side shell frame and ice specimen. A total of 70 samples were manufactured. Two types of ice specimen with relatively different surface conditions were used. The criterion for dividing the two types of ice specimen was the different exposure times to room temperature after freezing. This experiment was conducted for each parameter in order to reproduce the actual icebreaking situation. As a result of the analysis, the intermediate peak in the ice load signal have been found to be caused by mechanisms by which the inner surface of broken ice contact with hull immediately after the initial hitting point of ice has been broken.

An Experimental Study for the Mechanical Properties of Model Ice Grown in a Cold Room (Cold Room을 이용한 모형빙의 재료특성에 관한 실험적 연구)

  • A full-scale field experiment is an important part in the design of ships and offshore structures. Full-scale tests in the ice-covered sea, however, are usually very expensive and difficult tasks. Model tests in a refrigerated ice tank may substitute this difficulty of full-scale field tests. One of the major tasks to perform proper model tests in an ice towing tank is to select a realistic material for model ice which shows correct similitude with natural sea ice. This study focuses on the testing material properties and the selection of model ice material which will be used in an ice model basin. The first Korean ice model basin will be constructed at the Maritime & Ocean Engineering Research Institute (MOERI) in 2009. With an application to the MOERI ice model basin, in this study the material properties of EG/AD/S model ice of IOT (Institute for Ocean Technology) Canada, were tested. Through comprehensive bending tests, the elastic modulus and the flexural strength of EG/AD/S model ice were evaluated and the results were compared with published test results from Canada. Instead of using an ice model basin, a cold room facility was used for making a model ice specimen. Since the cold room adopts a different freezing procedure to make model ice, the strength of the model ice specimen differs from the published test results. The reason for this difference is discussed and the future development for a making model ice is recommended.

Sensing Model for Reducing Power Consumption for Indoor/Outdoor Context Transition (실내/실외 컨텍스트 전이를 고려한 저전력 센싱 모델)

  • With the spread of smartphones containing multiple on-board sensors, the market for context aware applications have grown. However, due to the limited power capacity of a smartphone, users feel discontented QoS. Additionally, context aware applications require the utilization of many forms of context and sensing information. If context transition has occurred, types of needed sensors must be changed and each sensor modules need to turn on/off. In addition, excessive sensing has been found when the context decision is ambiguous. In this paper, we focus on power consumption associated with the context transition that occurs during indoor/outdoor detection, modeling the activities of the sensor associated with these contexts. And we suggest a freezing algorithm that reduces power consumption in context transition. We experiment with a commercial application that service is indoor/outdoor location tracking, measure power consumption in context transition with and without the utilization of the proposed method. We find that proposed method reduces power consumption about 20% during context transition.

Prediction Model of Remaining Service Life of Concrete for Irrigation Structures by Measuring Carbonation (중성화 측정을 통한 콘크리트의 잔존수명 예측 모델)

  • Recently, the researches on the durability design of concrete structures have been studied. As the examples, models to evaluate the service life prediction of the structure have been developed. The purpose of this article is to develop the model for predicting remaining service life. The final aim is to provide the user time for repairing the concrete structures. In addition, it makes possible to maintain the concrete structure economically. 70 reservoirs out of the inland concrete structures were selected and concrete structures of their components were surveyed. Two methods were used for measuring carbonation; TG/DTA method and Phenolphtalein indicator and, the value of pH was measured by the pH meter, After deriving correlations of calcium carbonate and used year, duration from completion year to 2002, pH value, and concrete cover depth the model was developed for predicting remaining service life by measuring data as small as possible. The conventional models had been developed on the basis of experiment data obtained from the restricted lab environment like as carbon gas exposure. On the other hand this model was developed on the basis of measuring data obtained from the real field that the complex deterioration actions are occurred such as freezing and thawing, carbonation, steel corrosion, and so on. The reliability of the developed model will be evaluated high in this point and this model can help to maintain concrete structures economically by providing the manager time to repair the deteriorated concrete structures in site of facility management.

Numerical Investigation of the Urea Melting and Heat Transfer Characteristics with Three Different Types of Coolant Heaters (냉각수 순환 방식 가열원 형상에 따른 요소수 해동 특성에 관한 수치적 연구)

  • Urea-SCR system, which converts nitrogen oxides to nitrogen and water in the presence of a reducing agent, usually AdBlue urea solution, is known as one of the powerful NOx reduction systems for mobile as well as stationary applications. For its consistent and reliable operation in mobile applications, such various problems as transient injection, ammonia slip, and freezing in cold weather have to be resolved. In this work, therefore, numerical study on three-dimensional unsteady heating problems were analyzed to understand the melting and heat transfer characteristics such as urea liquid volume fraction, temperature profiles and generated natural convection behavior in urea solution by using the commercial software Fluent 6.3. After validating by comparing numerical and experimental data with pure gallium melting phenomena, numerical experiment for urea melting is conducted with three different coolant heating models named CH1, 2, and 3, respectively. Finally, it can be found that the CH3 model, in which more coolant is concentrated on the lower part of the urea tank, has relatively better melting capability than others in terms of urea quantity of $1{\ell}$ for start-up schedule.

Characteristics of Liquid-Liquid Direct Contact Heat Exchanger for a Solar System (태양열 이용을 위한 직접접촉식 액-액 열교환기 특성)

  • In most direct contact liquid-liquid heat exchangers, oil or hydrocarbon with a density less than water is normally used as dispersed working fluid. The main difficulty that arises with this arrangement lies in the control of the interface at the top of the column. When it is connected with a solar collector which uses water as its working fluid, the main difficulties arise from the fact that the water can be frozen during winter time. In order to solve these problems and to demonstrate the technical feasibility of a direct contact liquid-liquid heat exchanger, liquids heavier than water with low freezing temperature has been utilized as dispersed phase liquids in a small laboratory scale model made of pyrex glass. In the present investigation, dimethyl phthalate(C/sub 6/H/sub 4/)COOCH/sub 3/)/sub 2/) and diethyl phthalate (C/sub 6/H/sub 4/(CO/sub 2/C/sub 2/H/sub 5/)/sub 2/) are utilized as heavy dispersed phase working fluids. The results of the present investigation the technical in the utilization of heavier dispersed working liquid in the spray-column liquid-liquid heat exchanger for a solar system. The overall average temperature difference along the column is found to be almost half of the initial temperature difference between the dispersed and the continuous phase. Despite the fact that the two phthalates tested in the experiment differ significantly in some of their physical properties, the volumetric heat transfer coefficients in terms of dispersed fluid superficial velocities were found to be similar for both phthalates tested.
