The present experiments aimed to investigate the metabolism of calcium during oocyte maturation in rat. The concentration of free calcium and calmodulin in oocytes was measured respectively by using of fluo-3/AM and FITC with microscope fluorescence spectrometer. The ultrastructural localization of calcium precipitates in oocytes was observed with the transmission electron microscope. Cumulus-free immature oocytes(GV-oocyte) were cultured in vitro through 15 hours. The free calcium concentration in GV oocyte was $55.9{\pm}3.5nM$. In calcium-containing medium, the free calcium concentration was increased in germinal vesicle breakdown(GVBD) oocyte($64.2{\pm}7.3nM$). In normal medium after calcium chelator treatment ($10{\mu}M$ BAPTA/AM), the free calcium contents were slightly lower than those in control group. In calcium-free medium, the free calcium content was drastically increased in GVBD($72.7{\pm}3.4nM$) and metaphase I - anaphase I ($88.0{\pm}3.4nM$) oocyte. In maturation rate of oocytes, GVBD rate was high in control group($82.9{\pm}6.55%$) and calcium chelator treatment group($91.2{\pm}4.4%$), but in calcium-free medium group, it was low and then the oocyte was degenerated without polar body formation. Relative content of calmodulin in oocyte was significantly(P<0.001) increased in metaphase I - anaphase I than in GV and GVBD oocyte. The calcium precipitates were observed in mitochondria and cytoplasm of GV oocyte but that were not observed in mitochondria of GVBD and metaphase I - anaphase I oocyte. And then the calcium precipitates reappeared in mitochondria of metaphase II oocyte. The above results indicate that changes in free calcium and calmodulin concentration of oocyte occur according to the maturational stages and the extracellular calcium is required during oocyte maturation. Also change of calcium localization in oocyte occurs according to the maturational stages.
The effect of calcium-free reperfusion for 5, 10, and 15 minutes, respectively, followed by continuous reperfusion with normal Tyrode solution containing 1.0mM calcium chloride, after global ischemia in the isolated perfused guinea pig heart by Langendorff techniques was examined with transmission electron microscope. Compared to the nomal Tyrode solution-perfused control hearts, the 5 minute calcium-free-reperfused hearts showed loss or thickening of Z lines, focal sarcolemmal disruption, mitochondrial swelling, clumping of chroma-tin, intracellular fluid accumulation, and some separation of cell junctions, especially the fasciae adherentes. These changes became more severe in the hearts of 10 minute calcium-free reperfusion. Subsarcolemmal larger bleb and near complete separation of cell junctions were noticed. In the 15 minute calcium-free-reperfused hearts, irreversible ultrastructural changes including contraction bands, biazrre mitochondria, and sarcolemmal destruction were widely distributed. The severity of myocardial changes were in accordance with the duration of calcium-free reperfusion. These changes indicate that calcium-free reperfusion regardless of its duration could not salvage the post-ischemic myocardium probably due to development of calcium paradox.
Vegetables contain more or less calcium. By the traditional method the total calcium in the vegetable was analyzed. A part or calcium in the vegetable is fixed as calcium oxalate and the calcium does not utilize for nutrition. In this investigation the free calcium was determined by extraction in boiling water, the combined calcium with oxalic acid was determined by extraction in 0.1 M hydrochloric aci and the other calcium combined with protein, amino acid etc. was estimated. The amount of calcium obtained from the difference between the total calcium and calcium exmtaining in calcium oxalate was estimated to be nutritious.
The determination of fluoride and the nutritious calcium in infusion of teas are explained. Tea leaves were pulverized and were immersed in boiling water. The solution was filtered and fluoride, calcium and oxalic acid were determined by the ion chromatography. The quantities of fluoride, calcium and oxalate ions extracted from 100 g of tea leaves were calculated. Tea leaves were also immersed in 0.5 M hydrochloric acid and extracted oxalate and calcium ions were analyzed. The free oxalic acid and calcium were extracted in boiling water and the total ones were extracted in hydrochloric acid. The quantity of calcium oxalate was calculated from the total and the free oxalic acids. The free calcium was estimated to be nutritious.
This study was to compare the bioavailability of calcium from anchovy and tofu to those of calcium from calcium carbonate(CaCO3)as the control diet and non-fat dry milk(NFDM). Rats weighing 50-60g were placed on experimental diets and deionized water at free access for 4 weeks. Diets contained 0.2% calcium from calcium-carbonate, NFDM, anchovy, tofu or 0.5% calcium as obtained were as follows : 1) No significant differences in the apparent absorption of calcium(62.5%-71.0%) were observed in the rats fed four different calcium sources at the level of 0.2% while 0.5% calcium diet group apparently absorbed calcium less efficiently(52.2%). 2) Bone length of tibia and femur was not significantly different among the groups, though 0.5% calcium of control group showed slightly longer length. 3) Tibia fat-free dry weights of 0.2% calcium of NFDM and anchovy diet groups were not significantly different from that of 0.5% calcium of control group. For femurs NFDM, anchovy and tofu groups were similar in their fat-free dry weight to that of 0.5% calcium group. 4) For calcium contents tibia contents tibia from anchovy treated group showed higher value than calcium-carbonate and tofu groups and the value was not significantly different from that of 0.5% calcium group. In femur NFDM, tofu and calcium-carbonate groups were not significantly different in their calcium content but 0.5% calcium group had higher level of calcium than 0.2% calcium groups. 5) The normalized values(NV) show that there was no significant differences in NV among 0.2% CaCO3 anchovy and tofu groups, while NV of NFDM group was significantly lower than that of calcium-carbonate group. NV of 0.5% calcium group was a little more than 50% of those in 0.2% calcium groups. Though the values obtained for the calcium bioavailability were somewhat variable among experimental products, it was demonstrated that anchovy and tofu are as good as NFDM for the dietary calcium provider when calcium intake is at marginal level.
In order to examine the effect of $Ca^{2+}$ -chelation on in vitro fertilization of rat zona-free oocyte, the formation of cortical granule envelope (CGE) and the rate of fertilization related to monospermy and polyspermy were determined. The ultrastructural characteristics of oocytes were observed with the scanning electron microscope and BAPTA/AM was used for calcium-chelation. The CGE formed by cortical reaction was observed in zona-free oocyte inseminated in vitro and it was also observed in the calcium chelator (1, 5, 10$\mu$M BAPTA/AM) treated zona-free oocytes inseminated in vitro. The CGE developed according to incubation time. The fertilization rate was decreased in the calcium chelator-treated group (59.8, 38.1, 37.0%) compared to the control group (60.6%) but monospermy rate was increased in the calcium chelator-treated group (45.0, 47.3, 50.9%) compared to control group (37.5%). The above results demonstrate that the CGE is formed during fertilization in rat and the extracellular calcium is used in cortical reaction. Also the results suggest that proper concentration of free calcium in oocyte acts as important factor in fertilization.n.
This study was conducted to determine the interaction of dietary wheat bran and dietary calcium levels n)n calcium utilization in post-breeding female rats. It was designed to compare the effects of four different levels (2.5, 5, 10 and 20%) of wheat bran and two different levels (0.5 and 1%) of calcium on bone and calcium balance in post-breeding female rats over a ten-week period. The effects of diet on animal weight gain, serum calcium, femur weight, femur calcium concentration, bone mass and calcium balance were determined and statistically analyzed. The addition of 20% wheat bran significantly (p$\leq$0.05) decreased the weight gain of rats. Serum calcium and bone calcium contents were more affected by dietary calcium level than by dietary wheat bran level. There was no significant difference in fat-free solid, ash, percentage of ash to fat-free solid and percentage of calcium to ash among groups. Groups fed the 1% calcium diet had a higher percentage of calcium to fat-free solid. All rats were in positive calcium balance during the three-4ay experimental period. The average calcium balance of the rats fed 1% calcium diet ranged from 25.34 to 53.90mg and the average calcium balance of the rats fed the 0.5% calcium diet ranged from 26.71 to 32.90mg. In rats receiving 2.5% wheat bran, the difference in calcium balance between the group fed the 1% calcium diet and the group fed the 0.5% calcium diet was only 1.37mg, which was not significantly (p$\leq$0.05) different. In rats receiving 20% wheat bran, the difference in calcium balance between the group fed the 1% calcium diet and the group fed the 0.5% calcium diet was 19.S7mg, which was significantly (p$\leq$0.05) different. The addition of wheat bran caused an increase in the calcium balance of the rats adminslesed the 1% calcium diet. On the other hand, the addition of wheat bran had no effect on the calcium balance of the rats adminislesed the 0.5% calcium diet. In conclusion, calcium utilization of rats wire more positively affected by the interaction of both dietary wheat bran levels and dietary calcium levels than either dietary wheat bran levels or dietary calcium levels alone. Moderate wheat bran consumption did not interfere with the calcium metabolism of rats when calcium intake was high enough.
This study was designed to investigate the role of calcium in the function of an isolated perfused rabbit kidney and its effect on the diuretic action of furosemide. The administrations of hydralazine and verapamil produced remarkable diuretic actions mainly by decreasing renal resistance. The administration of furosemide in combination with hydralazine or verapamil produced remarkable diuretic action and there was no difference between the two groups. The administration of quinidine produced a diuretic action in spite of vasoconstriction and potentiated the diuretic action of furosemide. In the calcium-free perfusion medium, the administration of calcium produced a marked diuretic action in spite of vasoconstriction and potentiated significantly the diuretic action of furosemide. The administration of quinidine did not alter renal function and the diuretic action of furosemide, but the combined administration of quinidine and calcium showed antidiuretic effect due to excessive vasoconstriction in the calcium-free perfusion medium. Although the administration of verapamil produced a slight diuretic action in the calcium-free perfusion medium, verapamil did not alter the diuretic action of calcium as well as the diuretic actions of furosemide alone and in combination with calcium. The results of this experiment show that calcium, verapamil and quinidine produced diuretic actions and calcium potentiates the diuretic action of furosemide.
To clarify the effect of extracellular ATP in cultured C6 glioma cells, ATP-induced cytosolic free calcium ($[Ca^{2+}]_i$) mobilization and cell proliferation were investigated. ATP-induced $[Ca^{2+}]_i$ increased in a dose-dependent manner $(10^{-7}\;M{\sim}10^{-3}\;M)$. ATP-induced $[Ca^{2+}]_i$ increases were slightly slowed in extracellular calcium-free conditions especially in sustained phase. ATP-induced $[Ca^{2+}]_i$ increment was also inhibited by the pretreatment of U73122, a phospholipase C (PLC) inhibitor, in a time-dependent manner. Suramin, a putative $P_{2Y}$ receptor antagonist, dose-dependently weakened ATP-induced $[Ca^{2+}]_i$ mobilization. Significant increases in cell proliferation were observed at 2, 3, and 4 days after ATP was added. Stimulated cell proliferation was also observed with adenosine at days 2 and 3. This cell proliferation was significantly inhibited by the treatment with suramin. Ionomycin also stimulated cell proliferation in a concentration-dependent manner. In conclusion, we suggest that extracellular ATP stimulates C6 glioma cell proliferation via intracellular free calcium mobilization mediated by purinoceptor.

The present study was carried out to investigate the effects of dietary protein and magnesium levels on calcium and magnesium metabolism in male Sprague-Dawley rats. Forty-eight male rats(average weight, 210g) divided into six diet groups; protein 8% and Mg-free diet, protein 8% and Mg 400mg/kg diet, protein 8% and Mg 900mg/kg diet, protein 20% and Mg-free diet, protein 20% nd Mg 400mg/kg diet, and protein 20% and Mg 800mg/kg diet group. After the rats were fed with experimental diets concentration were examined. Kidney weight was significantly higher in protein 8% and Mg-free group compared with other groups. Serum calcium concentration of protein8% group tended to decrease with the increase of the level of magnesium. Serum magnesium concentration in protein 8% and 20% group was lower in Mg-free group than that in other groups(p<0.05) and it also increased as the dietary magnesium level was increased. Urinary calcium and magnesium concentration in Mg-free group were significantly lower than those of other groups.
