Development of Cryopreservation Technique of Transgenic Bovine Embryos (형질전환 소 난자의 동결보존기술 개발)

  • The purpose of this study is to improve production efficiency of vitrified-thawed transgenic bovine embryos. Transgenic bovine embryos were produced by injection of FIV-GFP lentiviral vector into perivitelline space of in vitro matured MII stage oocytes, and then in vitro fertilization. EGFP-expressing transgenic bovine blastocysts were cultured in serum-containing and serum-free medium. These blsatocysts were vitrified by pull and cut (PNC) container made with 0.25 cm plastic straw. Results indicate that total developmental rates of normal IVF embryo cultured in serum-containing and-free medium into blastocyst were not significantly different (22.3 vs 21.5%) and those of GFP-expressing transgenic bovine embryo into blastocyst showed no significant difference between serum-containing (13.9%) and-free medium (13.1%). However, developmental rate of GFP transgenic embryo was significantly (P<0.05) lower than its of normal IVF embryos. In additional study, we vitrified GFP transgenic normal bovine blastocysts using PNC vitrification method. Survival rate of vitrified-thawed GFP transgenic blastocyst (23.1%) was significantly (P<0.05) lower than its of normal blastocysts (68.9%). Although, survival rate of vitrified-thawed GFP transgenic blastocyst was lower than its of normal blastocyst, our result may suggested that PNC vitrification method is feasible to cryopreserve transgenic embryos. Our next plan will be the production of GFP express transgenic bovine derived from vitrified-thawed embryos using PNC method.

Synthesis of Nanostructured Si Coatings by Hybrid Plasma-Particle Accelerating Impact Deposition (HP-PAID) and their Characterization (하이브리드 플라즈마 입자가속 충격퇴적(Hybrid Plasma - Particle Accelerating Impact Deposition, HP-PAID) 프로세스에 의한 Si 나노구조 코팅층의 제조 및 특성평가)

  • Using a recently developed Hybric Plasma-Particle Accelerating Impact Deposition (HP-PAID) process, synthesis of nanostructured silicon coatings has been investigated by injecting vapor-phase TEOS (tetraethosysilane, (C$_2$H$\_$5/O)$_4$Si) into an Ar hybrid plasma. The plasma jet with reactants was expanded through nozzle into a deposition chamber, with the pressure dropping from 700 to 10 torr. Ultrafine particles accelerated in the free jet downstream of the nozzle, deposited by an inertial impaction onto a temperature controlled substrate. By using this process, nanostructured amorphous silicon coatings with grain size smaller than 10 nm could be synthesized. These samples were annealed in an Ar and crystallized at 900$^{\circ}C$ for 30 min. TEM analysis showed that the annealed coatings were also composed of nanoparticles smaller than 10 nm, which showed a good consistency that the average grain size of 7 nm was also estimated from a peak shift of 2.39 cm$\^$-1/ and Full Width at Half Maximum (FWHM) 5.92 cm$\^$-1/ of Raman analysis. The noteworthy is that a strong PL peak at 398 nm was also obtained for this sample, which indicates that the deposited coatings also contained 3∼4 nm nanostructured grains.

  • The purpose of this research is to investigate the characteristics of heat transfer in the downward axisymmetric free water jet system impinged on a flat oblique plate which has the uniform heat flux. Experimental conditions considered were Reynolds number, distance between nozzle and Bat plate, inclination angle of heater surface and nozzle exit velocity. Local Nusselt number was subjected to the influence of Re number, Pr number, oblique angle of heating surface and local position of flat plate. In the wall region of downward surface, The secondary peak point of heat transfer appeared at the local point of X/D=-8 from the stagnation point. The stagnation heat transfer rate of this experimental study augments 2.4 times than that of laminar theorical solution. The stagnation nusselt number is function of Reynolds number, nozzle-plate spacing Prandtl number and oblique angle of impinging plate.

  • The characteristics of lifted laminar propane flames diluted with nitrogen have been investigated experimentally to elucidate self-excitation and the effects of flame curvature. Flame oscillation modes are classified as follows: oscillation induced by heat loss, a combination of oscillations induced by heat loss and buoyancy, and a combination of the oscillations induced by heat loss and diffusive thermal instability. It is shown that the oscillation induced only by heat loss is not relevant to the diffusive thermal instability and hydrodynamic instability caused by buoyancy; this oscillation is observed under all lift-off flame conditions irrespective of the fuel Lewis number. These experimental evidences are displayed through the analysis of the power spectrum for the temporal variation of lift-off height. The possible mechanism of the oscillation induced by heat loss is also discussed.

  • Atmospheric pressure microwave induced plasmas are used to excite and ionize chemical species for elemental analysis, for plasma reforming, and for plasma surface treatment. Microwave plasma differs significantly from other plasmas and has several interesting properties. For example, the electron density is higher in microwave plasma than in radio-frequency (RF) or direct current (DC) plasma. Several types of radical species with high density are generated under high electron density, so the reactivity of microwave plasma is expected to be very high [1]. Therefore, useful applications of atmospheric pressure microwave plasmas are expected. The surface characteristics of SUS304 stainless steel are investigated before and after surface modification by microwave plasma under atmospheric pressure conditions. The plasma device was operated by power sources with microwave frequency. We used a device based on a coaxial transmission line resonator (CTLR). The atmospheric pressure plasma jet (APPJ) in the case of microwave frequency (880 MHz) used Ar as plasma gas [2]. Typical microwave Pw was 3-10 W. To determine the optimal processing conditions, the surface treatment experiments were performed using various values of Pw (3-10 W), treatment time (5-120 s), and ratios of mixture gas (hydrogen peroxide). Torch-to-sample distance was fixed at the plasma edge point. Plasma treatment of a stainless steel plate significantly affected the wettability, contact angle (CA), and free energy (mJ/$m^2$) of the SUS304 surface. CA and ${\gamma}$ were analyzed. The optimal surface modification parameters to modify were a power of 10 W, a treatment time of 45 s, and a hydrogen peroxide content of 0.6 wt% [3]. Under these processing conditions, a CA of just $9.8^{\circ}$ was obtained. As CA decreased, wettability increased; i.e. the surface changed from hydrophobic to hydrophilic. From these results, 10 W power and 45 s treatment time are the best values to minimize CA and maximize ${\gamma}$.

  • The laser Doppler technique is well-established as a velocity measurement technique of high precision for flow velocity. Recently, the laser Doppler technique has also been used to measure acceleration of fluid particles. Acceleration is interesting from a fluid mechanics point of view, since the Navier Stokes equations, specifically the left-hand-side, are formulated in terms of fluid acceleration. Further, there are several avenues to estimating the dissipation rate using the acceleration. However such measurements place additional demands on the design of the optical system; in particular fringe non-uniformity must be held below about 0.0001 to avoid systematic errors. Relations expressing fringe divergence as a function of the optical parameters of the system have been given in the literature; however, direct use of these formulae to minimize fringe divergence lead either to very large measurement volumes or to extremely high intersection angles. This dilemma can be resolved by using an off-axis receiving arrangement, in which the measurement volume is truncated by a pinhole in front of the detection plane. In the present study an optical design study is performed for optimizing laser Doppler systems for fluid acceleration measurements. This is followed by laboratory validation using a round free jet and a stagnation flow, two flows in which either fluid acceleration has been previously measured or in which the acceleration is known analytically. A 90 degree off-axis receiving angle is used with a pinhole or a slit.

  • Air interlacing serves to protect the yarn against damage, strengthens inter-filament compactness or cohesion, and ensures fabric consistency. The air interlacing nozzle is used to introduce intermittent nips to a filament yarn so as to improve its performance in textile processing. This study investigates the effect of interlacing nozzle geometry on the interlacing process. The geometries of interlacing nozzles with multiple air inlets located across the width of a yarn channels are investigated. The basic interlacing nozzle is the yarn channel, with a perpendicular single air inlet in the middle. The yarn channel shapes are cross sections with semicircular or rectangular shapes. This paper presents three doubled sub air inlets with main air inlet and one of them is slightly inclined doubled sub air inlets with main air inlet. The compressed air coming out from the inlet hits the opposing wall of the yarn channel, divides into two branches, flows trough the top side of yarn channel, joins with the compressed air coming out from the sub air inlet and then creates two free jets at both ends of the yarn channel. The compressed air moves in the shape of two opposing directional vortices. The CFD-FASTRAN was used to perform steady simulations of impinging jet flow inside of the interlace nozzles. The vortical structure and the flow pattern such as pressure contour, particle traces, velocity vector plots inside of interlace nozzle geometry are discussed in this paper.

Dispersion Analysis of Surface Discharged Heat Water In Shallow Coastal Area (천해역에서의 표층온배수 확산해석)

  • Dispersion characteristics of surface discharge heat water in shallow region are investigated for coastal power plant with nearly constant depth of 20 meters by observing the seasonal depthwide temperature in several stations, which give or precise horizontal distribution and vertical structure of heat water. Surface discharged heat water in shallow coast in the Yellow Sea relies mainly on ambient tidal flow. so it behaves as free jet when the ambient now is strong and shows plumelike behavior during stagnant tide. According to observation the neat field region is estimated as 200-300 meters and shows distinct vertical profile and exponentially decreasing pattern from discharge point for this region. But there are no remarkable vertical distortion of temperature beyond 800 meters even though it is discharged from surface. Characteristic length scale model, CORMIX3, is applied and compared with the field date Overall tendency of CORMIX3 results resemble well with field data especially in near field and intermediate region.

  • The objective of this study was to examine the effect of in vitro maturation (IVM) medium, cytochalasin B (CB) treatment during intracytoplasmic sperm injection (ICSI), and electric activation on in vitro development ICSI-derived embryos in pigs.   • The characteristics of lifted butane flames diluted with nitrogen have been investigated experimentally in order to elucidate the mechanism of individual flame oscillation modes. Flame oscillations in laminar free-jet lift-off flames are classified into the following five regimes: a stabilized lift-off regime (I), a heat-loss-induced oscillation (II), a buoyancy-induced oscillation along with a heat-loss-induced oscillation (III), a combined form of an oscillation prior to blow-out and a heat-loss-induced oscillation (IV), and a combination of an oscillation prior to blow-out and a buoyancy-induced oscillation along with a heat-loss-induced oscillation (V). The characterization of the individual flame oscillations modes are presented and discussed using Strouhal numbers and their relevant parameters by the analysis of the power spectrum for temporal variation of the lift-off height.
