Studies on the Constituents of Higher Fungi of Korea (XX)

  • To investigate constituents of Strobilomyces floccopus (Fr.) Karst. and Coprinus comatus (Fr.) S. F. Gray, free and total amino acids of the two mushrooms were quantitatively analyzed by G. L. C. and an amino acid analyzer. Free amino acids were extracted from both mushrooms with ethanol. Fourtenn free amino acids were detected from the ethanol extract of S. floccopus and fifteen free amino acids from C. comatus by G. L. C. And the dry carphopores of both mushrooms were hydrolyzed with hydrochloric acid and then the total protein amino acids were analyzed by A. A. A. Seventeen total amino acids were detected from each acid-hydrolysate of S. floccopus and C. comatus. Lipids were extracted from the carpophores of S. floccopus and saponified with alcoholic potassium hydroxide. The isolated sterols were subjected to G. L. C. and two sterols were detected. The isolated free fatty acids were methylated with diazomethane and subjected to column chromatography and G. L. C. Eleven saturated and nine unsaturated free fatty acids were detected from the carpophores of S. floccopus. The presence of these nutrient components shows that the two mushrooms can be utilized as edible ones.

Chemical composition of different parts of ramie (Boehmeria nivea)

  • Ramie (Boehmeria nivea) is a perennial herbaceous plant belonging to the family Urticaceae. It was used in folk remedies for diuretic or anti-pyretic purposes and as an hepatoprotective, anti-oxidant, and anti-inflammation agent. In this study, we investigated the composition of free sugars, amino acids, fatty acids, organic acids, and total polyphenol contents in parts of ramie (root, stem, and leaf) and different harvest areas (10 areas). Overall, free sugars were found as sucrose, glucose, fructose, and galactose at concentrations ranging from 24.5 to 1173.8 mg/g. Amino acids lysine, threonine, tyrosine, phenylalanine, and histidine were detected at concentrations of 33.8 to 3735.3 mg/g. Major fatty acids were linoleic acid, palmitic acid, and linolenic acid at concentrations of 7.3 to 364.4 mg/g. In organic acids, the concentration of malic acid was highest of all with 672.2 mg/g. The content of free sugars, amino acids, fatty acids, organic acids, and total polyphenol was higher in leaves than in roots and stems. In contrast, the total polyphenol content was higher in roots with 33.7 - 219.4 mg/g than in leaves or in stems. As seen in collecting region, the contents of free sugars, amino acids, fatty acids, organic acids, and total polyphenol of Bn-33, 39, 55, 65, and 90 were higher than other samples. Chemical compositions of different parts of ramie are important factors to consider in manufacturing functional foods. The results of this study provide fundamental information on the chemical compositions of ramie parts and would help develop new functional foods from ramie leaves or the whole plant.

Changes of Lipid and Free Fatty Acid in Mung-bean Sprouts during Growth (녹두나물 성장과정중의 지질 및 지방산변화에 관한 연구)

  • The changes of various lipid content and fatty acids composition of mung-bean sprout during 6 days. The results are summarized as follows 1) The total lipid contents of mung-bean sprouts are generally decreased during growing. 2) On T.L.C. gram, triglyceride contents are decreased and free fatty acids and sterol are increased with time. 3) The ratio of unsaturated fatty acids to saturated fatty acids U/S are increased by six times after 4 days of mung-bean germination. 4) The contents of palmitic acid as main fatty acid in the mangbean but oleid, linoleic acid were increased after 2 days, 4 days, 6 days. 5) Total lipids from mung-bean sprouts contained fatty acid in order of linoleic acid, oleic acid. 6) The content of essential fatty acid are rich in 4 days.

  • The free fatty acids were prepared from the ethereal fraction of Panax ginseng. The prepared acids were methylated with diazomethzne. The methyl esters of saturated and unsaturated fatty acids were separated by the means of mercuric acetate method and column chromatography. The separated methyl esters were gaschromatographed and analyzed. The obtained conclusions were as follows. 1. The root of six-year old Korean Panax ginseng contains 0.28% of free fatty acids. 2. It was found that 24 kinds of free fatty acids existed in Panax ginseng. Among them, 22 kinds of free fatty acids were indentified by the gas chromatogram and the graphical method but the rest, 2 kinds of them were not identified by the only gas chromatographical data. The amount of each free fatty acid which was not identified was predominant and they were supposed to be unusual free fatty acids which would not commonly exist in nature. These results were shown in Table III. 3. $L_{EE}$ and $L_{EE}$ reported that n18:3 existed in Panax ginseng. However, in this experiment, n18:3 did not exist in Panax ginseng, and instead, peak XVI appeared between n18:2 and n18:3 as shown in Fig. 9.

  • Changes in taste components of Backilju, an traditional alcoholic beverage of Korea, were investigated. Ethanol(17%) was the most abundant, and then isoamylalcohol(23mg%) and methanol(8mg%) were also detected in a small amount in Backilju. Major non-volatile organic acids were lactic and malic acid, followed by citric, fumaric and succinic acid. Arginine, phenylalanine and glutamic acid were major free amino acids and essential amino acid content was 230∼560 mg%, which was 45∼48% of total free amino acids. The major fatty acid of total lipid was palmitic acid(37∼43%). It has been found that the Backilju contained more free amino acids and alcohols than other Korean Yokjus.

  • Till the 21st century, fatty acids were considered as merely building blocks for triglycerides, phospholipids, or cholesteryl esters. However, the discovery of G protein-coupled receptors (GPCRs) for free fatty acids at the beginning of the 21st century challenged that idea and paved way for a new field of research, merged into the field of receptor pharmacology for intercellular lipid mediators. Among the GPCRs for free fatty acids, free fatty acid receptor 4 (FFA4, also known as GPR120) recognizes long-chain polyunsaturated fatty acids such as DHA and EPA. It is significant in drug discovery because it regulates obesity-induced metaflammation and GLP-1 secretion. Our study reviews information on newly developed FFA4 agonists and their application in pathophysiologic studies and drug discovery. It also offers a potency comparison of the FFA4 agonists in an AP-TGF-α shedding assay.

The Effect of Alisma orientale Extract on Free Fatty Acid-induced Lipoapoptosis in HepG2 Cells (택사(澤瀉)가 유리지방산으로 유발된 HepG2 cell의 lipoapoptosis에 미치는 영향)

  • Objectives : This study was designed to investigate the effect on lipoapoptosis of Alisma orientale extract against free fatty acid-induced cellular injury. Methods : HepG2 cells were used in an vitro model. HepG2 cells were treated with free fatty acids to generate a cellular model of nonalcoholic fatty liver disease (NAFLD). Using this cellular model, the anti-apoptotic effect and reducing steatosis of Alisma orientale extract against free fatty acid-induced cellular injury was evaluated by measuring steatosis and apoptosis. Results : Alisma orientale extract significantly attenuated free fatty acid-induced intracellular steatosis. Alisma orientale extract inhibited free fatty acid-mediated activation of pJNK, PUMA, BAX, caspase-3, and -9, and apoptotic kinases that are correlated with NAFLD. Alisma orientale extract also promoted Bcl-2, a anti-apoptotic protein. Conclusions : From the above, the Alisma orientale extract decreased the hepatocyte steatosis and showed the hepatocelluar protective effect by the regulation of apoptosis-related protein. It proposes the possibility of Alisma orientale extract to the treatment of nonalcoholic fatty liver disease in clinics.

Effect of Controlled Atmosphere and Modified Atmosphere Storage on the Fatty Acid of Fresh and Red Ginseng (수삼의 CA 및 MA 저장이 홍삼의 지방산에 미치는 영향)

  • Fifteen free fatty acids including myristic acid were isolated and identified from red ginseng processed from CA and MA stored fresh ginseng. Linoleic acid (80%) and palmitic acid (10.5%) content were the major components accounting for more than 90% of the total free fatty acid. The contents of free and identified fatty acids were not greatly changed in all the treatments. Especially when preservative was treated, the change of free fatty acid was quite stable over the control. Each fraction of neutral, glyco- and phospholipid was constant in terms of quantities before and after the treatment.   • The concentration of free fatty acids and fatty acid composition as well as cholesterol supersaturation in bile may be an important factor in the gallstone formation. Therefore, we simultaneously determinded 23 fatty acids in bile by selected ion monitoring (SIM) method of gas chromatography-mass spectrometry (GC-MS). Biliary fatty acids were extracted by aminopropyl column and the extracts with (phospholipid fraction) or without (free fatty acid fraction) alkaline hydrolysis of phospholipid were derivatized with MSTFA/TMCS (N-methyl-N-trimethylsilyl-trifluoroacetamide/trimethylsilylchloride) mixture in order to be detected on the GC-MS. The recovery range of this method was 61.1-99.0% and the RSD value of within-a-day and day-to-day test were 3.1-25.6% and 3.8-27.0%, respectively. Using this method, biliary profile was investigated in the bile of normal controls and patients with gallstones. The amounts and their distribution of free and phospholipid fatty acids showed different pattern between normal subjects and patients.

Manganese-induced Oxidative Stress in Nucleus Accumbens of Rat Brain (흰쥐 뇌의 측중격핵에서 망간에 의한 산화적 손상에 관한 연구)

  • This study was carried out to identify the effect of oxidative stress on the pathogenesis of manganese intoxication. Five rats in experimental group were given with $MnCl_2$intraperitoneally for 4 weeks(4 mg/kg once daily 5 days per week) and another five rats for control group were given with normal saline. In experimental group, manganese concentrations increased significantly in nucleus accumbens by 142% (p<0.05), SOD activities increased significantly by 124%(p<0.01), and MDA concentrations increased significantly 148%(p<0.05) compared with control group. Among fatty acids, total n-6 polyunsaturated fatty acids(PU) increased significantly by 231%(p<0.05) compared with control group. Arachidonic acids(AA) increased by 224%(p<0.05), and these increase were composed mostly of n-6 polyunsaturated fatty acids(PUFA). Among n-3 PUFAs except linolenic acids, eicosapentanolc acid(EPA) decreased significantly by 38%(p 0.01) and docosahexanoic acids(DHA) decreased by 30% p<0.05) compared with control group. Our results suggest that the oxygen free radicals produced by manganese may cause compositional changes of fBtty acids in nucleus accumbens of the rat. Characteristics of fatty acids compositional changes by manganese were the decrease of EPAs and DHAs(n-3 PUFAs), and increase of AAs(n-6 PUFAs). These changes with the increase of MDA, suggest that manganese neurotokxcity is caused by lipid perokidation mediated with oxygen free radicals, especially superoxide radicals.
