요크샤 종모돈의 정액성상, 동결-융해 후 정자의 생존성 및 테스토스테론의 농도에 미치는 봄과 여름의 영향 (Effect of Spring and Summer on Semen Characteristics, Frozen-Thawed Sperm Viability and Testosterone Concentration in Yorkshire Boars)

  • 본 연구는 요크샤 종모돈의 정액성상, 동결-융해후의 정자생존성 및 혈청 중 테스토스테론의 농도에 미치는 영향을 봄 (3~5월)과 여름 (6~8월)으로 나누어 실시한 바, 그 얻어진 결과는 다음과 같았다. 1. 요크샤 종모돈의 정액량, pH, 희박정자 부분의 정자농도는 봄과 여름철에 큰 차이가 없었다. 그러나 봄철 농후정자 부분의 정자농도는 여름철 농후정자 부분의 정자농도보다 높았다 (P<0.05). 2. 원정액 정자에서는 운동성 및 정상첨체에서 봄과 여름철간에 차이가 없었다. 그러나 동결-융해정자의 운동성 및 정상첨체는 봄철의 종모돈에서 생산한 정자가 여름철의 종모돈에서 생산한 정자보다 우수하였다. 3. 요크샤 종모돈의 혈청 중 테스토스테론의 농도 변화는 봄과 여름에 각각 4.04 ng/$m\ell$과 2.85ng/$m\ell$l으로, 봄철 테스초스테론의 농도가 여름철의 농도보다 높았다(P<0.05). 4. 이상의 결과를 종합해 보면, 요크샤 종모돈에 있어 정자의 내동성은 혈청 중 테스토스테론의 농도가 높을수록 우수한 것으로 나타났다.

Antibacterial Activity and Synergism of Hydnocarpi Semen Extracts with Ampicillin or Oxacillin against Methicillin-resistant Staphylococcus aureus

  • Methicillin-resistant Staphylococcus aureus (MRSA) is a serious clinical and an urgent problem worldwide. Few new drugs are available against MRSA, because MRSA has the ability to acquire resistance to most antibiotics, which consequently increases the cost of medication. In the present study, the antibacterial activity of Hydnocarpi Semen was investigated. The most effective method is to develop antibiotics from the natural products without having any toxic or side effects. Therefore, there is a need to develop alternative antibacterial drugs for the treatment of infectious diseases. Five Clinical isolates (MRSA) were obtained from five different patients at Wonkwang University Hospital (Iksan, South Korea). The Other 2 strains were ATCC 33591 (Methicillin-resistant strain) and ATCC 25923 (Methicillin-susceptible strain). Antibacterial activity (Minimal Inhibitory Concentrations, MICs) was determined by broth dilution method, disk diffusion method, MTT test, and checkerboard dilution test. Antibacterial activity of n-hexane fraction was remarkable, and had a MICs ranging from $31.25-125{\mu}g/m{\ell}$. FICI values for HFH+AM and HFH+OX were 0.13-0.19 and 0.04-0.29, showing the increase of synergistic effect. When combined together, these antibacterial effects were dramatically increased.

A New Device for Intrauterine Artificial Insemination in the Dog

  • The intrauterine inseminator (IUI) was developed to provide the method of depositing dog semen into the uterine body instead of the vagina. The IUI consists of a vaginal endoscope, a balloon sheath, and injection catheter. When the endoscope is inserted into the vagina and the balloon expanded with air, the cervical os becomes visible so a injection catheter can be inserted through the cervix for deposition of the frozen-thawed semen. The efficacy of the IUI device was compared to intra-vaginal artificial insemination using semen that had been collected and frozen from pooled sperm-rich fraction of ejaculates collected from two Jindo dog donors. Aliquots of semen were extended with a Tris-egg yolk diluent, centrifuged, the seminal plasma removed, the pellet resuspended with the same diluent, and cooled to $5^{\circ}C$ over a 2 h period. A Tris-egg yolk-glycerol extender was added at $5^{\circ}C$; after 1 h, semen was loaded into 0.5 ml straws, and straws were frozen in LN vapor for 5 min, and immersed in LN for storage. The final sperm concentration for freezing was approximately $100{\times}10^{6}cells/ml$. The straws were thawed at $70^{\circ}C$ for precisely 6 sec, 1.5 ml Tris-egg yolk buffer at $38^{\circ}C$ added, and the 2 ml of thawed semen was used for a single insemination using the IUI device. Each bitch was inseminated at optimal insemination point, which was estimated by vaginal epithelial cells staining and progesterone concentration analysis. Use of the IUI device resulted in 21 of 26 females giving birth to 89 pups ($4.2{\pm}1.6$ pups per litter), while intra-vaginal AI resulted in 6 of 15 females whelping a total of 17 pups ($2.8{\pm}1.2$ pups per litter). We believe the IUI device is easier to use than previously described devices used for intrauterine insemination. In our experience the expansion of the balloon has a calming effect on the bitch that aids the inseminator. These results indicate that the IUI device was able to provide high fertility with 50 million frozen sperm per insemination and two inseminations.

자소자 항산화성분의 분리 (Isolation of Antioxidative Components of Perillae semen)

  • 자소자의 탈지시료로부터 FPA, SPA 및 IPA 형태의 phenol성 물질을 추출하여 이중 항산화활성이 가장 강한 FPA 추출물에 존재하는 항산화성분을 column chromatography 및 HPLC에 의하여 분리하였다. Chlorogenic acid를 표준물질로하여 측정한 탈지된 자소자에 함유되어 있는 phenol성 물질의 총함량은 0.38% 였고, 총 phenol성 물질중 FPA, SPA 및 IPA 추출물이 차지하는 비율은 71.1%, 15.8% 및 13.1%였다. 세가지 형태의 phenol성 물질의 항산화활성을 electron donating ability (EDA)와 linoleic acid를 기질로 하여 TBA값을 측정하여 비교하였을 경우 FPA 추출물이 가장 높은 항산화활성을 나타내었다. 항산화활성이 가장 높게 나타난 FPA 추출물을 silica gel column chromatography로 분획하여 항산화활성을 비교한 결과 acetone : methanol의 비가 8 : 2인 획분에서 가장 높은 항산화활성을 나타내었다. 항산화활성이 가장 높았던 획분을 HPLC에 의하여 분리한 결과 HPLC chromatogram 상에서 5가지 활성물질 획분이 분리되었으며, 이들 획분을 분취하여 항산화활성을 비교한 결과 F-I 획분이 가장 높은 항산화활성을 나타내었다.

돼지 난모세포의 체외성숙 및 체외수정시 배양액과 액상정액의 효과 (Effect of Matruation Media and Liquid Boar Semen on Maturation and Fertilization of Pig Oocytes In Vitro)

  • This present study was carried out to examine the effect of maturation media and liquid boar semen on in vitro maturation and feritilization of pig oocytes. The results obtained were as follows : When the oocytes were cultured for 36∼42 hours in mTCM-199, Waymouth MB 725/1 and mTLP-PVA medium, the maturation rates were 90%, 92% and 88%, respectively. The sperm penetration rates of pig oocyte matured in vitro were 87%(mTCM-199), 90%(Waymouth MB 725/1) and 86%(mTLP-PVA), respectively. The rates of nuclear maturation and fertilization of pig oocytes among three different media did not differ. However, the rate of male pronucleus formation of pig oocytes was significantly higher in pig oocytes matured in Waymouth MB 725/1(91%) than oocytes matured in mTCM-199(66%) and mTLP-PVA(62%) medium (P<0.05). When the collected sperm-rich fraction without diluent was used fro in vitro fertilization in mTCM-199 fertilization medium, the fertilization rate was 87.9%. However, when the liquid boar semen diluted with B tschwiler diluent was used at day 3 and 5 after dilution, the fertilization rate was 40.8% and 0.0%, respectively.

  • Rumex crispus (Polygonaceae) is a well known perennial plant. which is called So-Ri-Jaeng-Yi. growing in the field and on the roadside. It has been used as a Korean Folk medicine in treating of acute and chronic cutaneous disease. cathartics, fever and jaundice. Also, the seed of this plant has been used as only a folk medicine for the treatment of digestion problems, liver diseases and many sorts of tumor. So we examined analgesic activity, anti-inflammatory activities and hepatoprotective activity using MeOH extraction and BuOH fraction in this plant. (omitted)

  • 전통기호음료성분의 기능특성을 조사하기 위한 연구의 일환으로 결명자, 들깨, 대추, 모과, 오미자, 오갈피 및 생강 추출물을 여러가지 획분으로 분획하여 아질산염 소거작용을 조사한 결과, 본 실험에 사용한 시료는 모두 아질산염 소거작용이 있었으며, 수용성 획분을 에탄올 가용성 획분이 에탄올 침전획분으로 분획하였을 때, 결명자, 들깨, 오갈피, 생강의 경우에는 에탄올 가용성 획분이 에탄올 침전획분보다 아질사염 소거작용이 뛰어났으며 대추, 모과, 오미자의 경우에는 그 반대의 경향을 나타내었다. 시료 추출물 중에 함유되어 있는 환원성 물질의 관여도 클 것으로 생각되어 전자공여작용을 조사해 본 결과, 기호음료 원료에서 추출한 수용성 획분의 전자공여 작용은 오갈피>들깨>모과>생강>대추>오미자>결명자의 순으로 나타났으며, 에탄올 침전획분과 에탄올 가용성 획분의 전자공여효과도 아질산염 소거효과와 경향이 일치하였다.

  • These experiments were carried out to develop new techniques for In Vitro separatin of X-and Y-bearing spermatozoa. The bull semen was applied to the various Gel-Columns filled with swellen Sephadex G-50 Fine and then elutriated wtih Locke solution (elutriation rate; 1ml/3-4min., 1ml/1-2min.). Elutriated solution was fractionated into 1ml by automatic Fraction Collector and spermatozoa included in each fraction were subjected to the estimation of viability and recovery rate, and to B-body test. The results obtained in these experiments were summarized as follows: 1. When the column size and the elutriation rate were adjusted to 15$\times$1.6cm and 1ml/3-4min., respectively, the highest sperm concentration was obtained from the 8th to the 12th fraction. 2. As a trend, the viability of spermatozoa was improved by chromatography, and the degree of improvement ranged 5 to 10 percentage. 3. The average recovery rate of spermatozoa applied to column was 73.2 percentage and ranged 52.6 to 81.3 percentage. 4. The lowest rate of B-body bearing spermatozoa following chromatography was obtained when the column size and the elutriation rate were adjusted to 15$\times$0.8cm and 1ml/1-2min., respectively.

  • This experiment was conducted to investigate the effects of temperature and time of sedimentation and dilutor on the a, pp.arance of B-body in top and bottom fractions at separation of X and Y- chromosome bearing spermatozoa in boar semen. 1. The top fraction showed higher a, pp.arence rate of B-body than the bottom. 2. Sixty minutes at 5 and 15$^{\circ}C$ and 90 min, at 25$^{\circ}C$ showed highest difference of B-body a, pp.arence rate between top and bottom fractions. The highest difference was shown in the treatments of Sg at 5$^{\circ}C$, C at 15$^{\circ}C$ and P at 25$^{\circ}C$. 3. The highest difference was shown in the treatments of 25$^{\circ}C$ and Sg for 30 min, 15$^{\circ}C$ and P for 60 min. and 25$^{\circ}C$ and P for 90 min. 4. Sixty minutes in C, P, S and Sg dilutors showed the highest difference. 5. 25$^{\circ}C$ of the temperature levels, 60 min of the time levels and P of the dilutor levels showed the highest difference. 6. The difference was given due to the individual boar.
