The aim of the present study was to investigate whether self-fermented pine extract for 2 years (SFPE2) and ethyl acetate (EtOAc) fraction from self-fermented pine needle extract may affect the contractility of the isolated aortic strips and blood pressure of normotensive rats. SFPE2 ($360-1440\;{\mu}g/mL$) significantly depressed both phenylephrine ($10\;{\mu}M$)- and high potassium (56 mM)-induced contractile responses of the isolated rat aortic strips in dose-dependent fashion. The EtOAc-fraction ($400\;{\mu}g/mL$) also inhibited both phenylephrine ($10\;{\mu}M$)- and high potassium (56 mM)-induced contractile responses. Also, in anesthetized normotensive rats, intravenous injection of the EtOAc fraction (1.0~10.0 mg/kg) dose-dependently elicited hypotensive responses. The EtOAc fractions (1.0 and 3.0 mg/kg/30 min) inhibited norepinehrine-induced pressor responses. Intravenous infusion of SFPE2 fraction (3.0 and 10.0 mg/kg/30 min) also inhibited norepinehrine-induced pressor responses in both anesthetized spontaneously hypertensive rats (SHRs) and normotensive rats. In conclusion, these results suggest that both SFPE2 and the EtOAc fraction cause vascular relaxation in the aortic strips isolated from normotensive rats and SHRs as well as vasodepressor responses. Based on these experimental data, it seems that SFPE2 or the EtOAc fraction possesses active antihypertensive components, which are available to prevent or treat hypertension in future.
In preliminary tests, we examined the effect of several fractions isolated from fermented pine needle extract on pacemaker potentials in cultured interstitial cells of Cajal (ICCs) from the mouse colon using a whole cell patch clamp technique. Among these fractions, Fraction 3 (F3) elicited the most powerful depolarization of membrane. Therefore, the aim of the present study was to investigate the effect of F3 obtained from fermented extract of Pinus densiflora needle on pacemaker potentials in ICCs and to establish its mechanism of action. Colonic ICCs generated spontaneous periodic pacemaker potentials in the current-clamp mode. F3 depolarized the membrane and decreased the frequency and amplitude of pacemaker potentials in a dose-dependent fashion. The F3-induced effects on pacemaker potentials were blocked by methoctramine, a muscarinic $M_2$ receptor antagonist, and by glycopyrrolate, a muscarinic $M_3$ receptor antagonist. The F3-induced effects on pacemaker potentials were blocked by external $Na^+$-free solution and by flufenamic acid, a non-selective cation channel blocker, as well as by the removal of external $Ca^{2+}$ and in the presence of thapsigargin, a $Ca^{2+}$-ATPase inhibitor in the endoplasmic reticulum. Taken together, these results suggest that F3 of pine needle extract modulates the pacemaker activity of colonic ICCs by the activation of non-selective cation channels via muscarinic $M_2$ and $M_3$ receptors. And external $Ca^{2+}$ influx and intracellular $Ca^{2+}$ release are involved in F3 actions on ICCs.
솔잎과 쑥의 기능성 검토를 위한 연구의 일환으로 열수 및 70% acetone으로 처리하여 얻은 추출물의 항변이원성, angiotensin I converting enzyme(ACE) 저해작용, 전자 공여작용, polyphenol oxidase(PPO)에 대한 저해작용, 아질산염 소거작용, 항산화 작용을 조사하였다. 솔잎의 열수 및 70% acetone 추출물 모두 총 polyphenol 성분 중 flavanol형 tannin의 함량이 60% 이상이었고, 간접변이원인 Trp-P-1을 이용한 항변이원성 실험결과 70% acetone 추출물은 처리구는 82.2%로 열수 추출물보다 높은 억제 활성을 나타내었으나 ACE 저해작용은 열수 추출물이 61%로 70% acetone 추출물의 50%보다 저해능이 높았다. 전자공여작용은 두가지 추출물 모두 80% 이상의 높은 효과를 나타내었다. 미나리에서 분리한 PPO 조효소에 대한 70% acetone 추출물의 저해효과는43%로 나타났고 또한 아질산염 소거작용은 산성 부근(pH 3.0)에서 각 추출물 모두 $80{\sim}90%$의 높은 효과를 보였다. 한편 쑥추출물의 경우 항변이원성은 70% acetone 추출물이 84.1%로 솔잎과 유사하였으나 ACE 저해작용은 열수 추출물이 75.1%로 acetone 추출물의 57.6%보다 높았고, 전자공여 작용은 두 처리구 모두 45% 정도였으며, 아질산염 소거작용은 솔잎에 비해 $20{\sim}30%$ 분해능이 떨어지는 것으로 판명되었다 또한 솔잎 열수 추출물을 chloroform, ethyl acetate, butanol을 이용하여 얻은 분획물을 대두유에 첨가하여 저장기간 중 항산화성을 조사한 결과 ethyl acetate 분획물이 가장 효과가 있었다.
The purpose of this study was to investigate the phenolic acid, proanthocyanidin (PAs), and flavonol glycoside contents, as well as the antioxidant activities of pine needle extracts from six species of young pine trees. The extracts were prepared from Section Pinus (Diploxylon): P. densiflora, P. sylvestris, P. pinaster and P. pinea, and Section Strobus (Haploxylon): P. koraiensis and P. strobus. Phenolics were extracted from pine needles with 80% acetone to obtain the soluble free fraction, and insoluble residues were digested with 4 M NaOH to obtain bound ethyl acetate and bound water fractions. Phenolics were analyzed by HPLC, and the hydrophilic antioxidant activity was measured using oxygen radical absorbance capacity (ORAC). Total phenolic and flavonoid contents of the soluble free fraction were higher than those of the bound ethyl acetate and bound water fractions. The main phenolics were monomers and polymers of PAs in the soluble free fraction, and phenolic acids and flavonol glycosides in bound ethyl acetate fraction. Flavonol glycosides found in different species of pine needles were qualitatively similar within fractions, but composition varied among Pinus sections. High levels of kaempferol arabinoside and an unknown compound were present in all Strobus species. The soluble free fraction had the highest antioxidant activity, followed by bound ethyl acetate and bound water fractions.

Most cervical cancers are associated with high-risk human papillomavirus (HPV) infection. Currently, cervical cancer treatment entails surgical removal of the lesion, but treatment of infection and preventing tissue damage are issues that still remain to be addressed. Herbal medicine and biological studies have focused on developing antiviral drugs from natural sources. In this study, we analyzed the potential antiviral effects of Pinus densiflora Sieb. et Zucc. leaf extracts against HPV. The pine needle extracts from each organic solvent were analyzed for antiviral activity. The methylene chloride fraction (PN-MC) showed the highest activity against HPV pseudovirus (PV). The PN-MC extract was more effective before, rather than after treatment, and therefore represents a prophylactic intervention. Mice were pre-treated with PN-MC via genital application or oral administration, followed by a genital or subcutaneous challenge with HPV PV, respectively. The HPV challenge results showed that mice treated via genital application exhibited complete protection against HPV. In conclusion, PN-MC represents a potential topical virucide for HPV infection.
The aim of the present study was to investigate the effects of several fractions obtained from methylene chloride ($CH_2Cl_2$) extract of self-fermented pine needle (SFPNE) on the acetylcholine (ACh)-evoked CA release from the isolated perfused model of the rat adrenal medulla and to establish the mechanism of the most active fraction (Fr.)-induced inhibitory action on the CA release. We obtained 6 fractions from $CH_2Cl_2$ extract of self-fermented pine needle. For the ACh (5.32 mM)-evoked CA release, the following rank order of inhibitory potency was obtained: Fr.4-5 > Fr.8-11 ${\gg}$ Fr.3 > Fr.6 = Fr.7 > Fr.1-2. Fr. 4 - 5 (60 ${\mu}g/mL$) perfused into an adrenal vein for 90 min produced relatively time-dependent inhibition of the CA secretory responses to ACh (5.32 mM), DMPP (100 ${\mu}M$), McN-A-343 (100 ${\mu}M$) and high $K^+$ (56 mM). Fr. 4 - 5 itself did not affect basal CA secretion. Also, in the presence of Fr. 4 - 5 (60 ${\mu}g/mL$), the CA secretory responses to angiotensin II (AngII, 0.1 ${\mu}M$), veratridine (50 ${\mu}M$), Bay-K-8644 (10 ${\mu}M$), and cyclopiazonic acid (10 ${\mu}M$) were significantly reduced, respectively. In the simultaneous presence of Fr. 4 - 5 (60 ${\mu}g/mL$) and L-NAME (30 ${\mu}M$), the inhibitory responses of Fr. 4 - 5 on the CA secretion evoked by ACh, DMPP, high $K^+$, AngII, Bay-K-8644 and veratridine were considerably recovered to the extent of the corresponding control secretion compared with that of Fr. 4 - 5-treatment alone. The level of NO released from adrenal medulla after the treatment of Fr. 4 - 5 (60 ${\mu}g/mL$) was greatly elevated compared with the basal level. Taken together, these results demonstrate that Fr. 4 - 5 inhibits the CA secretion from the isolated perfused rat adrenal medulla evoked by stimulation of cholinergic receptors as well as by direct membrane-depolarization. It seems that this inhibitory effect of Fr. 4 - 5 is mediated by blocking the influx of $Ca^{2+}$ and $Na^+$ into the adrenomedullary chromaffin cells as well as by inhibition of $Ca^{2+}$ release from the cytoplasmic calcium store, which is evoked at least partly through the increased NO production due to the activation of NO synthase. Based on these results, it is also thought that Fr. 4 - 5 isolated from $CH_2Cl_2$ extract of pine needle may contain beneficial antihypertensive components to prevent or treat hypertension.
솔잎 EtOAc획분을 하루 25, 50, 100 mg/kg체중으로서 SD계 랫트에 45일동안 투여하여 간장획분의 세포막 유동성(membrane fluidity MF), 기초 및 유도활성산소(BOR 및 IOR), 산화적 스트레스로서 과산화지질(lipid peroxide : LPO) 및 산화단백질(oxidized protein : OP), 그리고 리포푸신(lipofuscin : LF)의 축적에 미치는 영향을 평가하여 보았다. 솔잎 EtOAc획분의 투여에 의하여 간장의 mitochondria 및 microsome획분에서 어느 정도 MF가 증가하였지만, 유의성은 EtOAc-100 투여그룹의 mitochondria획분에서만 인정되었다. 기초 및 유도활성산소로서 BOR 및 IOR의 생성 억제효과는 간장의 두 획분의 EtOAc-50 및 EtOAc-100투여그룹에서 유의적인 활성산소의 생성 억제 효과가 인정되었다. 산화적 스트레스에 미치는 영향에서는 mitochondria획 분에서 는 EtOAc-100투여그룹에서, 그리고 microsome획분에서는 EtOAc-50 및 EtOAc-100투여그룹에서 유의적인 LPO의 생성 억제효과가 인정되었고, mitochondria 획분에서는 EtOAc-50 및 EtOAc-100투여그룹에서 유의적인 OP의 생성 억제효과가 인정되었지만, microsome 획분에서는 아무런 유의성도 인정할 수 없었다. 그렇지만, 간장의 클로로포름층에서 측정한 LF의 축적은 EtOAc-25, EtOAc-50 및 EtOAc-100 투여그룹에서 다같이 유의적인 LF의 축적 억제효과가 인정되었다. 이상의 결과에서 평가하여 볼 때 솔잎 EtOAc획분의 투여는 간장의 유동성을 촉진하고 산화적 스트레스를 효과적으로 억제할 수 있을 것으로 기대된다.
솔잎 추출물의 BuOH획분을 하루 25, 50, 100 mg/kg BW로써 SD계 흰쥐에 45일동안 투여하여 간장 세포막의 유동성(MF), 기초 및 유도산소라디칼(BOR 및 IOR), 산화적 스트레스로서 과산화지질(LPO) 및 산화단백질(OP), 리포푸신(LF)의 축적에 미치는 영향을 평가하였다. MF은 간장의 mitochondria에서 BuOH-50 및 BuOH-100 투여군에서 16∼22%의 유의적인MF의 증가효과가 인정되었지만(p<0.01∼0.001), 간장 microsomes에서는 BuOH획분의 투여에 의한 MF의 유의적인 증가효과를 기대할 수 없었다. BOR 및 IOR의 생성은 간장의 mitochondria에서 BuOH-25, BuOH-50, BUOH-100 투여군은 각각 11∼27% 및 11∼28%의 BOR 및 IOR의 생성 억제효과가 인정되었고(p<0.05∼0.001), microsomes에서 BuOH-50, BuOH-100 투여군 및 BuOH-25, BuOH-50, BuOH-100 투여군은 각각 l1∼24% 및 l5∼24%의 유의적인 IOR의 생성 억제효과가 인정되었다(p<0.05∼0.001, p<0.01∼0.001). LPO의 생성은 간장의 mitochondria에서 BuOH-100투여군과 microsomes에서는 BuOH-50, BuOH-100투여군에서 각각9%, 9∼l3%의 유의적인 LPO의 생성 억제효과가 인정되었고(P<0.05∼0.01), OP의 생성은 간장의 mitochondria에 서 BuOH-50 및 BuOH-100투여군에 서 10∼12%의 유의적인 OP의 생성 억제효과가 인정되었지만(p<0.05), 간장의 microsomes에서는 유의성을 인정할 수 없었다. 그렇지만, 간장의 chloroform층에서 LF의 축적은 BuOH-50, BuOH-100투여군에서 8∼9%의 유의적인 LF의 축적 억제효과가 인정되었다(p<0.05). 따라서 솔잎 추출물의 BuOH 획분의 투여는 간장의 유동성을 촉진하고 산화적 스트레스를 효과적으로 억제하여 생리적 노화를 효과적으로 억제할 수 있을 것으로 기대된다.
한국에 널리 분포하고 있는 곰솔, 리기다, 잣나무, 적송의 에탄올추출물과 각 용매 분획물을 이용하여, in vitro assay로서 spore rec-assay와 Salmonella typhimurium reversion assay법으로 시료자체 변이원성 유무와 4가지 양성변이원 물질(MNNG, 4NQO, Trp-P-1 및 B(a)p)에 대한 돌연변이 억제효과를 고찰하였다. 곰솔, 리기다, 잣나무, 적송의 에탄올 추출물은 spore rec-assay와 S. typhimurium를 이용한 mutagenicity assay에서 변이원성을 나타내지 않았다. 돌연변이 억제 시험에서는 4가지 시료 모두 농도 증가에 따른 억제율을 나타내었고 또 각각의 시료들의 분획물 중 잣나무와 소나무의 에테르 분획물들이 간접변이원인 Trp-P-1의 변이원성에 대하여 다른 분획물에 비하여 높은 억제율을 보여주었다.
SD계 랫트를 사용하여 솔잎 추출물의 BuOH획분을 하루에 각각 25, 50 및 100mg/kg B.W. 이 섭취될 수 있도록 BuOH 획분을 사료에 각각 0.025%, 0.05% 및 0.1% 첨가하여 조제된 사료로싸 45일 동안 투여하였다. 솔잎 BuOH획분의 투여는 체중변화 및 사료 섭취랑에는 유의적인 영향을 주지 못했다. 그렇지만, BuOH-25, BuOH-50 및 BuOH-100 투여군은 대조군 대비 각각 12.8%, 19.1% 및 21.6%의 현저한 항콜레스테롤효과가 인정되었고, 또한 10.2%, 15.6% 및 23.7%의 유의적인 LDL-콜레스테롤의 증가효과가 인정되었다. BuOH-25, BuOH-50 및 BuOH-100 투여군은 대조군 대비 각각 24.8%, 30.4%, 36.2%의 매우 유의적인 동맥경화지수 (AI)의 감소효과가 인정되었다. BuOH-25, BuOH-50 및 BuOH-100 투여군은 대조군 대비 각각 13.3%, 13.3% 및 16.7%의 매우 유의적인 LPO의 억제효과가 인정되는데, 이러한 사실은 9.8%, 19.7% 및 21.2%의 ·OH 라디칼의 생성 억제효과 때문으로 기대된다. 또한 활성산소의 제거효소로서 BuOH-50 및 BuOH-100 투여군에서 SOD의 활성은 대조군 대비 12.1% 및 23.3%의 유의적인 증가효과가 인정되었다. 이상의 결과를 종합하여 볼 때 솔잎의 BuOH획분의 투여는 성인병을 효과적으로 억제할 수 있을뿐만 아니라 노화도 매우 효과적으로 억제할 수 있을 것으로 기대된다.

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
