Enhanced Berberine Production in Sucrose Solution by Plant Cell Suspension Cultures of Thalictrum rugosum (자당 수용액을 이용한 Thalictrum rugosum 식물세포배양에서의 berberine 생산성 증진)

  • The effects of sucrose solution on cell growth and berberine production were studied in Thalictrum rugosum cell cultures. Application of sucrose solution without any other components enhanced berberine accumulation significantly in spite of slower cell growth. At high sucrose concentration, cells became very compact and fourfold increase in specific berberine content was achieved. Optimum concentration of sucrose in plain water to maximize the productivity was found to be 8%. Time course experiment in 8% sucrose solution showed that more than 5 days were required to utilize the advantage of this system efficiently. Addition of vitamins, growth regulators, and inorganic salts into the solution was not effective in increasing berberine productivity.

Antioxidant and Antibacterial Activities of Chitosan-Phloroglucinol Conjugate

  • In an effort to develop biopolymer-based antioxidant and antibacterial materials, a chitosan-phloroglucinol conjugate was prepared and cellular antioxidant activity and minimum inhibitory concentration against foodborne pathogens and methicillin-resistant Staphylococcus aureus (MRSA) evaluated. The chitosan-phloroglucinol conjugate showed higher antioxidant activities than the unmodified chitosan (P < 0.05). The chitosan-phloroglucinol conjugate showed 62.29% reactive oxygen species scavenging activity, 56.11% lipid peroxidation inhibition activity, and 2.21-fold increase of glutathione expression in mouse macrophage cells. Additionally, the chitosan-phloroglucinol conjugate exhibited higher antibacterial activities than the unmodified chitosan, and the chitosan-phloroglucinol conjugate showed fourfold higher antibacterial activities against MRSA and clinical isolates and twofold higher activities against foodborne pathogens compared to the unmodified chitosan.

  • A fast and sensitive protein staining method in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) using both an acidic dye, Coomassie Brilliant Blue R-250 (CBBR) and a basic dye, Neutral Red (NR) is described. It is based on a counter ion-dye staining technique that employs oppositely charged two dyes to form an ion-pair complex. The selective binding of the free dye molecules to proteins in an acidic solution enhances the staining effect of CBBR on protein bands, and also reduces gel background. It is a rapid staining procedure, involving fixing and staining steps with short destaining that are completed in about 1 h. As the result, it showed two to fourfold increase in sensitivity comparing with CBBR staining. The stained protein bands can be visualized at the same time of staining.

  • We investigate the structures and the formation energy of inversion domain boundaries (IDBs) using the Tersoff empirical potential. Four kinds of IDBs ( A and B types for IDB* and Holt ) are considered. The IDBs with A type are energetically favorable compared to B type with the structural instability. The IDB* is also more stable than the Holt type in spite of fourfold and eightfold rings of bonds. We calculate the atomic configurations of the Holt IDBs induced by the interactions of the IDB* with the stacking faults $I_1$ and $I_2$. The stacking fault $I_2$ interacted with $I_1$ on the IDB induces the structural transformation from IDB* to Holt type.

  • During the last decade the world had experienced oil crisis twice: the first one was the fourfold jump in oil prices in 1973 and 1974, the second one came in 1979 with 2.6 times price hike. The current crude oil prices stand around the level of $30 a barrel. The first reaction came quick, with reduced oil consumption and a general decline in economic activity. Furthermore, the second oil crisis has brought about tremendous and varied impacts upon the shipping industry, which is now undergoing an adaptation process to the changing environments. This article is divided into five chapters: chapter I is the introduction on the subject under examination; chapter II is devoted to the trends of the seaborne trade cargoes after the oil crisis; chapter III reveals the impact of the oil price hike on ships and their desion; chapter IV deals with the challenges shipping enterprises face in terms fo economic modus operandi and new international political environments; chapter V sets forth conclusion.

Calculation of the NMR Chemical Shift for a 3d$^2$ System in a Strong Crystal Field of Octahedral Symmetry

  • The NMR chemical shift arising from 3d electron spin dipolar nuclear spin angular momentum interactions for a 3d$^2$ system in a strong crystal field environment of octahedral symmetry has been investigated when the fourfold axis is chosen to be our axis of quantization. The NMR shift is separated into the contribution of 1/R$^5$ and 1/R$^7$ terms. A comparision of the multipolar terms with nonmultipolar results shows that the 1/R$^5$ term contributes dominantly to the NMR shift and there is in good agreement between the exact solution and the multipolar results when R ${\ge}$ 0.25. A temperature dependence analysis may lead to the results that the 1/T$^2$ term has the dominant contribution to the NMR shift for a paramagnetic 3d$^2$ system but the contribution of the 1/T term may not be negligible.

SPUTTERING PRESSURE EFFECTS ON MAGNETIC ANISOTROPY IN Co/Pt MULTILAYERS

  • We have investigated the effects of sputtering Ar gas pressure on magnetic anisotropy of Co/Pt multilayers, where sputtering Ar gas pressure was varied from 2 to 20 mTorr. The surface and volume anisotropies were found to be strongly dependent on sputtering Ar gas pressure. In particular, the surface anisotropy exhibited more than fourfold enhancement as Ar pressure was decreased from 20 to 5 mTorr. We have found that the surface anisotropy was closely correlated with the low-angle x-ray diffraction intensity. We believe that these results are mainly ascribed to the variation of microstructure in the Co/Pt multilayer thin films with sputtering Ar gas pressure.

  • A bionomic study of the coralligenous concretionary hard bottom in the northwestern Mediterranean was carried out at four stations : three stations(Arnette, Laurons, Auguette) under the influence of intense multisource pollution(gradient decreasing from north to south) in the Gulf of Fos, west of Marseille, France, and one control station(Moyade islet) in an unpolluted area near Riou island, east of Marseille. Along the increasing pollution gradient from the outer to the inner part of the Gulf of Fos, there is a qualitative and quantitative impoverishment of the fauna. On the whole, the species richness, the numerical abundance. and the species diversity index are all on the decrease. Accordingly, the innermost station in the Gulf of Fos(Auguette) is most heavily affected by the industrial, and to a lesser extent by the domestic wastes, from the nearby industrial complex and urban areas. The impact on the benthic coralligenous community of this serious alteration has been analysed in view of community composition, functional aspect, and ecological stocks. The faunal affinity between stations has been studied by means of the two coefficients : fourfold point correlation coefficient and Jaccard's community coefficient. The upper layer and inferior face communities of the coralligenous concretionary structures are also compared.

Effects of Hyperhomocysteinemia on the Immunohistochemical Reactivity for Vimentin in the Retinal Glial Cell (면역조직화학적 방법을 이용한 흰쥐의 호모시스테인 수준과 망막 손상의 관련성 연구)

  • It has been suggested that the elevated plasma homocysteine may lead to retinal dysfunction. We investigated the effects of plasma levels of homocysteine and folate on the retinal glial cells' injuries. Male Sprague-Dawley rats were raised either on a control diet or on an experimental diet containing 3.0 g/kg homocystine without folic acid for 10 weeks. Plasma homocysteine concentrations were measured by a HPLC-fluorescence detection method. Plasma folate and vitamin B/sub 12/ levels were analyzed by a radioimmunoassay. The response of Muller cells which are the principal glial cells of the retina was immunohistochemically examined using an antibody for vimentin, a cytoskeletal protein belonging to the family of intermediate filament. At 2 weeks, the homocystine diet induced a twofold increase in plasma homocysteine, and a concomitant increase in the expression of vimentin in the Muller cells' processes spanning from the inner to outer membranes of the retina indicating arterial degeneration. At 10 weeks, the homocystine diet induced a fourfold increase in plasma homocystine, but vimentin immunoreactivity in the retinas was similar in both groups. In conclusion, increased plasma homocysteine levels have influence on morphological and functional changes of Muller cells in the retina. (Korean J Nutrition 38(2): 96~103, 2005)
