도두(刀豆)의 조혈증진 작용에 관한 연구 (Enhancing effect of Canavalia gladiata DC semen on the hematopoietic expansion and function of stem cells)

  • Objective : Canavalia gladiata DC semen (CGS) have been used to improve hematopoietic activity. In the current study, we investigated whether CGS regulate hemato-potentiating function using hematopoietic stem cells (HSCs) as a testing system. Methods : HSCs isolated from femur in mice with leukopenia and thrombocytopenia induced induced by CTX. Then, Real-time PCR was performed to measure the mRNA expression and hematopoietic related gene (EPO, IL-3, SCF, c-kit, GM-CSF), the phoaphorylation of GATA-1 and STAT-5a/b were observed by ELISA method, and the number of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E), semisolid clonogenic assay was performed. Result : When HSCs were treated with CGS, the expression of hematopoietic related genes (EPO, IL-3, SCF, c-kit, and GM-CSF) were significantly increased at the levels of mRNA as well as production in HSCs. Additionally, CGS enhanced phosphorylation of STAT-1 and signal transducer and activator of transcription-5a/b (STAT-5a/b) in HSCs. Furthermore, CGS significantly enhanced the growth rate of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E) in vitro. Conclusion : These result suggest that CGS has hematopoietic enhancement via hematopoietic cytokine-mediated GATA-1/STAT-5a/b pathway.

Real-time PCR과 Colony forming unit법을 이용한 타액 내 2종의 구강미생물 총량분석 (Analysis of total oral microorganisms in saliva using real-time PCR and colony forming unit)

  • Objectives: The purpose of this study was to compare colony forming unit (CFU) method and multiplex real-time polymerase chain reaction (MRT-PCR) method for accurate quantitative analysis of bacteria. Methods: We compared the CFU method and the MRT-PCR method, which are still used in Korea, for Prevotella intermedius (P. intermedius), a periodontal disease pathogen selected by MRT-PCR, and Streptococcus mutans (S. mutans), a dental caries causative organism. The subjects of this study were 30 patients who visited the C dental hospital. Results: Total microorganisms in MRT-PCR method were significantly higher in both types of bacteria (p<0.05), since DNA of dead bacteria was also analyzed. This was because the periodontal dise(-) anaerobes, and even dead bacteria contain large amounts of toxic substances called LPS in the extracellular membrane, and fimbriae and pili, which are motility structures, still remain as a strong toxic substance in periodontal tissue. Conclusions: Therefore, in terms of the total amount of bacteria found, the MRT-PCR method will be a useful technique for searching all the bacteria in the oral cavity including live bacteria, as well as sterilization.

Development of software for computing forming information using a component based approach

  • In shipbuilding industry, the manufacturing technology has advanced at an unprecedented pace for the last decade. As a result, many automatic systems for cutting, welding, etc. have been developed and employed in the manufacturing process and accordingly the productivity has been increased drastically. Despite such improvement in the manufacturing technology, however, development of an automatic system for fabricating a curved hull plate remains at the beginning stage since hardware and software for the automation of the curved hull fabrication process should be developed differently depending on the dimensions of plates, forming methods and manufacturing processes of each shipyard. To deal with this problem, it is necessary to create a "plug-in" framework, which can adopt various kinds of hardware and software to construct a full automatic fabrication system. In this paper, a framework for automatic fabrication of curved hull plates is proposed, which consists of four components and related software. In particular the software module for computing fabrication information is developed by using the ooCBD development methodology, which can interface with other hardware and software with minimum effort. Examples of the proposed framework applied to medium and large shipyards are presented.

중성빔 입사장치에서 빔형성 구조의 입자모사 모형 (Particle Simulation Modelling of a Beam Forming Structure in Negative-Ion-Based Neutral Beam Injector)

  • 중성입자입사 장치의 효율적인 빔형성 구조를 목적으로 정전기장 내에서 하전 입자의 움직임을 시간의 흐름에 따라 계산해 볼 수 있는 프로그램을 만들어 입자 모사 모형을 찾았다. 가속관 내의 입자의 움직임은 일정 시간 간격으로 계산하였고 전위는 유한차분법에 의해 Poisson 방정식에서 구하였다. 행렬식은 반복해법인 successive overrelaxation법을 사용하였고 전하밀도와 임자에 미치는 전기장의 힘을 구할 때는 cloud-in-cell모델을 사용하였다. 이 전자계산 코드를 사용하여 가속관 내 전극의 여러 조건들을 변화시켜가면서 빔형성 구조의 최적 설계를 수행하였다. 중성자 입사 장치의 가속관에서 가속 감속-전극간의 간격변화, 감속전극의 두께 변화, 가속 전극의 형태변화 등을 통하여 이들이 빔의 모양에 끼치는 영향을 조사하여 몇 가지 경우에 있어서 일정한 시간 간격으로 나타나는 입자들의 움직임을 예시하였다. 이 입자 모사모형을 통하여 가속전극의 형태가 빔 퍼짐에 가장 주요한 역할을 하는 것을 알았다.

Preparation and Characterization of Chemically Stable PVDF-HFP Asymmetric Microfiltration (MF) Membranes

  • Chemically stable Polyvinylidene fluoride-hexa-fluoropropane (PVDF-HFP) copolymer asymmetric membranes were prepared by the conventional phase inversion process, using Dimethyacetamide (DMAc) as a solvent and water as a non-solvent. To control the pore size and porosity of the PVDF-HFP membranes, tetra-ethoxysilane (TEOS) was used as a pore-forming agent. The prepared membranes were characterized, using several analytical methods such as Fourier Transform Infrared spectroscopy (FTIR), Thermo-gravimetric analyzer (TGA), Field Emission Scanning Electronic Microscopy (FESEM). TEOS turned out to increase porosity and make homogeneous pores on the membranes. Depending on the composition of the dope solutions, the pore size was ranged from 0.1 to 1.0 ${\mu}m$. The flux of the PVDF-HFP membranes prepared by using TEOS as a pore forming agent was increased substantially without much decrease in the rejection. When 15 wt% PVDF-HFP solution was blended with 13 wt% TEOS solution at composition ratio of 70/30 in wt%, the water flux at 2 bars was about 2 $m^3/m^2day$.

No-Bridge Blank의 공정 해석 및 성형 공정 개선에 관한 연구 (A Study on the Analysis and Improvement of Forming Process of a No-Bridge Blank)

  • Deep drawing process, one of sheet metal forming methods, is used widely. Circular or square shape blanks are currently studied mainly. Especially, circular blank for coating case of chip condenser remains bridges when it is made out of aluminum coil. The bridge reduces Material-withdrawal-rate of aluminum coil to $60\%$. This paper proposes a no-bridge blank instead of circular blank. To get the different values of two cases, comparison circular blank with no-bridge blank is accomplished in the point of thickness strain in the vicinity of flange. In order to find optimal condition in new proposed blank, several process variables - those are blank holder shape, die shape radii, punch shape radii and blank holding force - are changed.

알루미늄 원통컵 스피닝 작업을 위한 롤러이송경로 설계 (Design of roller path for spinning of cylindrical cups of aluminum sheet metal)

  • Spinning is a chipless forming method for producing axially symmetric parts by using axial-radial motions of a spinning roller. This process has still some advantages in such a view point that a variety of complex shapes which can not be formed in a press can be easily spun at a low cost although it is one of the oldest forming methods for spinning mainly cookware parts for a long time. This study is to investigate the optimum roller path in order to obtain the maximum spinnability in producing cylindrical cups of Aluminum(A1050-H16) sheet metal. Working conditions applicable to any size of blank were predetermined through preliminary spinning tests. 9 types of roller path were proposed and experiments were carried out. The modified involute curve was shown to give the maximum drawing ration and more uniform quality of spun cups as compared with other results of this study. in addition thickness distribution and dimensional accuracy of spun cups were examined and discussed.

  • The explicit and implicit time integration methods are applied effectively to analyze sheet metal stamping processes, which include the forming stage and the springback stage consecutively. The explicit time integration method has better merits in the forming stage including highly complicated three-dimensional contact conditions. By contrary, the implicit time integration method is better for analyzing springback since the complicated contact conditions are removed and the computing time to get the final static state is short. In this work, brief descriptions of the formulation and the factor study for springack simulations are presented. Further, the simulated results for the S-rail and the roof panel stamping processes are shown and discussed.

고무패드 변형의 3차원 유한요소해석 (Three-dimensional Finite Element Analysis of Rubber Pad Deformation)

  • This paper applies the FE analysis procedure, developed in the Part I of the companion article, to the three-dimensional rubber pad deformation during rubber-pad forming process. Effects of different algorithms corresponding to incompressibility constraint and time integration methods on numerical solution responses are investigated. Laboratory scale experiments support the validity of the developed FE procedure an demonstrate the accuracy of the numerical models. Full scale model responses are also predicted using the reasonable method and parameters obtained in laboratory modeling.

  • In the analysis of metal forming processes by the finite element method, there are many numerical instabilities such as element locking, hourglass mode, shear locking. These instabilities may have a bad effect upon accuracy and convergence. The present work is concerned with improvement of stability and efficiency in two dimensional rigid-plastic finite element method using various type of elements and numerical integration schemes. AS metal forming examples, upsetting and backward extrusion are taken for comparison among the methods : various element types and numerical integration schemes. comparison is made in terms of stability and efficiency. As a result, it has been shown that the finite element computation is stabilized from the viewpoint of computational time, convergency, and numerical instability.
