A report of 29 unrecorded bacterial species in Korea, belonging to the Alphaproteobacteria

  • As a subset study to discover indigenous prokaryotic species in Korea, a total of 29 bacterial strains assigned to the classes Alphaproteobacteria were isolated from various environmental samples collected from plant root, ginseng soil, forest soil, marsh, mud flat, freshwater and seawater. From the high 16S rRNA gene sequence similarity (>99.1%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 29 species included in Alphaproteobacteria is have been described in Korea; therefore 14 species of 9 genera in the order Rhizobiales, 7 species of 6 genera in the order Sphingomonadales and 4 species of 2 genera in the order Caulobacterales and 3 species in the order Rhodobacterales and 1 species in the order Rhodospirillales found in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are also described in the species description section.

  • In order to investigate indigenous prokaryotic species diversity in Korea, various environmental samples from diverse ecosystems were examined. Isolated bacterial strains were identified based on 16S rRNA gene sequences, and those exhibiting at least 98.7% sequence similarity with known bacterial species, but not reported in Korea, were selected as unrecorded species. 28 unrecorded bacterial species belonging to the phylum Bacteroidetes were discovered from various habitats including wastewater, freshwater, freshwater sediment, wet land, reclaimed land, plant root, bird feces, seawater, sea sand, tidal flat sediment, a scallop, marine algae, and seaweed. The unrecorded species were assigned to 18 different genera in five families: Flavobacterium, Epilithonimonas, Dokdonia, Gillisia, Flavicella, Chryseobacterium, Algibacter, Aquimarina, Lacinutrix, Gaetbulibacter, Cellulophaga, Tenacibaculum, and Maribacter of Flavobacteriaceae, Dyadobacter of Cytophagaceae, Draconibacterium of Draconibacterium_f, Sunxiuqinia of Prolixibacteraceae, and Fulvivirga of Fulvivirga_f. The selected isolates were subjected to further taxonomic characterization including analysis of Gram reaction, cellular and colonial morphology, biochemical activities, and phylogenetic trees. Descriptive information of the 28 unrecorded species is provided.

  • In 2016, as a part of the research program 'Survey of Korean Indigenous Species', diverse environmental samples were collected from various sources of freshwater, seawater, soil, wetland, reclaimed land, sand, pine forest, plant root, ginseng field, solar saltern, and caves. Thousands of bacterial strains were isolated from the diverse samples and identified based on 16S rRNA gene sequence analyses. The present study, as a phylogenetic subset of the primary research program, reports 24 unrecorded bacterial species in Korea that belong to the orders Rhizobiales and Sphingomonadales in the class Alphaproteobacteria. Based on the high 16S rRNA gene sequence similarities (>98.8%) and formation of a robust phylogenetic clade with the closest type species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 24 bacterial species have been described in Korea; therefore, 10 species of nine genera in the order Rhizobiales and 14 species of seven genera in the order Sphingomonadales are described for unreported alphaproteobacterial species in Korea. Gram reaction, colony and cell morphology, biochemical properties, and isolation sources are also provided in the species description section.

Evaluation of Meals Students Consumed in College Foodservice (일부 서울지역 대학식당의 메뉴분석)

  • A survey was conducted in 591 college students using 8 different cafeterias serving 31 meals. The information was obtained about the name and amount of dishes they consumed from the menu offered by college foodservice. Mean nutrient contents per meal in offered menu were higher than 1/3 of RDA for their age, sex in offered menu. The mean energy content was 466kcal from rice, 113kcal from soup, 141kcal from side dish and 21kcal from kimchi. When side dishes were classified by cooking methods, side dishes using grilling, frying, roasting methods had high energy content and fat percent above 200kcal, 40% per dish. By main ingredients, side dishes with animal food were higher energy than with plant food. Students consumed 94% of the energy provided in offered menu. When compared to proportion of foods consumed by sex, there was 43% of female and 22% of male consumed less than half in soup and 31% of female and 12% of male in kimchi. The most prevalent menu patterns of subjects included rice, soup, 2 side dishes, kimchi and were the same in both sexes. Mean energy intakes per meal were 989kcal for males and 842kcal for females which were enough to meet 1/3 of RDA for their age and sex. Most nutrient intake except fat and vitamin B1 were higher in set menu than in cafeteria. Nutrient adequacy ratio(NAR) were above 0.9 except calcium and vitamin A. The mean energy intake was 542kcal from rice, 70kcal from soup, 164kcal from side dish and 20kcal from kimchi. In conclusion, intake of most of the nutrients of students obtained from males in college foodservice were sufficient but calcium intake was insufficient and fat content was above 20% of energy. High fat consumption was due to side dishes from frying, grilling, roasting with animal food. To provide desirable meals in college foodservice, use of fat in cooking must be decreased.

  • A pilot plant scale freeze dryer was designed and manufactured and its performance was tested. Freezing capacity of water vapor in the developed freeze dryer was 8㎏/batch and heating and defrosting methods were conduction and hot gas types, respectively. And exhausting time of vacuum pump was 7-8 minutes and temperature of cold trap was kept below $-50^{\circ}C$. In the freeze drying of fruits and vegetables, its optimum and maximum drying capacities were 2 and 4㎏/batch, respectively. Performance of the new freeze dryer was compared quite well with a foreign made freeze dryer.

A case study on the Occurrence Category of aircraft accidents and serious incidents in Korea in the 2000's (2000년대 국내 항공기 사고·준사고 발생유형 사례연구)

  • Since year 2001 to the present time, the aircraft accidents and serious incidents in our country have surpassed 150 occurrences. The Boeing has published the statistical summary of commercial jet airplane accidents annually for the past 10 years on the basis of the occurrence categories defined by the CICTT(CAST/ICAO Common Taxonomy Team), and the number of occurrences is in order of loss of control(LOC-I), controlled flight into terrain(CFIT) and runway excursion (RE). Like the NTSB and the EASA, when fatal and non-fatal accidents are aggregated, though fatality rate is low, abnormal runway contact(ARC), system/component failure(SCF-PP/NP), ground handling(RAMP) rank high in the CICTT occurrence categories. With the less occurrence frequency, it is difficult to statistically analyze the aircraft accidents in our country, thus customarily the accidents and the serious incidents on aggregate are consolidated, and the statistical analysis is performed. This study categorizes the accidents and serious incidents to the domestic transportation aircraft in the past 10 years according to the CICTT occurrence categories, that is compared with foreign practices, and the implications have been discussed. From years 2001 through 2010, the accidents to the domestic transportation aircraft occurred in order of system failure(SCF-NP), ARC and power plant failure(SCF-PP), and when the accidents and the serious incidents are consolidated and analyzed, it is verified that a distribution appears similar to the European accident occurrence categories defined from 300 accident occurrence data.

  • The sensitive technique of activation analysis is well suited for this study since the elements such as As, Br, and Se in tobaccoes are expected to be high concentration. As, Br, and Hg were determined by Bethge destruction method and subsequent neutron activation analysis. $^{77m}Se$ was also by non-destruction activation analysis. The quantities of the element determined in Korean tobaccoes are given as follows in ppm: As, 0.65 ppm. Hg, 0.74 ppm. Se, 1.18 ppm. Br, 7.1 ppm. From the date given it seems that Korean tobaccoes and foreign tobaccoes contained considerably high concentration of selenium and mercury.

  • As a subset work for the collection of indigenous prokaryotic species in Korea, 38 actinobacterial strains were isolated from various environmental samples obtained from plant root, ginseng cultivating soil, mud flat, freshwater and seawater. Each strain showed higher 16S rRNA gene sequence similarity (>99.1%) and formed a robust phylogenetic clade with closest actinobacterial species which were defined and validated with nomenclature, already. There is no official description on these 38 actinobacterial species in Korea. Consequently, unrecorded 37 species of 24 genera in the 12 families belonging to the order Actinomycetales of the phylum Actinobacteria were found in Korea. Morphological properties, basic biochemical characteristics, isolation source and strain IDs are described in the species descriptions.

  • In order to prevent the corrosion of component contacted primary water designed alloy 600 material in the nuclear power plant. But the primary water stress corrosion cracking(PWSCC) of alloy 600 and weld area occurs continuously due to the residual stress. The leakage accident resulted from PWSCC in the drain nozzle of the steam generator of domestic power plants. Heater sleeves of the pressurizer are welded with alloy 600 weld material and therefore exposed to the primary water environment. PWSCC occurred in heater sleeve material and weld area of many foreign power plants. The current issue of domestic nuclear power plants are consequently concentrated to PWSCC of similar material. In order to improve the detection and the sizing of the PWSCC in the welding sleeve of the pressurizer, the automatic UT system and multi-directions probe sets have been developed. The experimental studies have been performed using the mock-up block containing artificial reflectors(ID connected EDM notch) and semi-artificial cracks made from thermal fatigue. The automatic UT System is applied in the detection and the length sizing of the ID/OD on the tube and the J-groove weld area of the artificial reflectors and results of the detection and the sizing are compared respectively. Also, the developed automatic UT system is successfully accomplished to inspect the heater sleeve and the J-groove weld area on the pressurizer for the detection of PWSCC.
