Effects of Dioscoreae Rhizoma on Excercise Practice Ability and Blood Fatigue Variable Factor (산약의 운동수행능력 증진 및 항피로 효과)

  • Objectives In this study, we analyzed the effect of Dioscoreae Rhizoma on exercise practice ability and blood fatigue variable factor in the forced swimming test. Methods Male mice were divided into four group. Mice were administrated with saline, Discorae Rhizoma extracted by water (DRW), Discorae Rhizoma extracted by 30% ethanol (DRE), red ginseng (RG), orally once per day, for 1 week. Exercise performance was evaluated by exhaustive swimming time and weights. Glycogen contents of liver and soleus muscle were measured at the end of the experiment. In addition, the biochemical parameters such as asparate aminotransferase (AST), alanine aminotransferase (ALT), blood urea nitrogen, creatine, lactate dehydrogenase (LDH), ammonia, triglycerides (TG), total cholesterol (TC) and free fatty acid (FFA) were analyzed. Results Both DRW and DRE showed an improvement of swimming endurance compared with control group. Levels of ALT, ammonia, TG, TC and FFA were significantly decreased in both DRW and DRE compared with control group. Levels of AST, Creatine were significantly reduced in DRE group compared with control group. Moreover LDH showed only a tendency to increase in the DRW and DRE groups. Conclusions In summary, the present study showed that Dioscoreae Rhizoma improved in physical fatigue and exercise practice ability and DRE improved the superior than those of DRW.

Hotwater Extract of Rhus Verniciflua Stokes Improves Exercise Performance in Mice (옻나무 열수추출물에 의한 운동수행능력향상에 미치는 효과 및 기전 연구)

  • The aim of this was to evaluate the effects of Rhus Verniciflua Stoke (RVS) extract powder on the endurance capacity for the forced swimming mice. Thirty mice were divided into 3 groups including negative control, 250 (RVS-250) and 500 (RVS-500) mg/kg RVS extract powder groups for 4-times swimming exercises. The swimming times to exhaustion in RVS-250 and RVS-500 mice were prolonged 1.6- and 2.0-fold at 4 weeks compared with negative control mice, respectively. Blood biochemical parameters for AST, ALT, T-CHO and TG were not significantly different between RVS fed or negative control mice. However, blood concentration of creatinine was significantly increased in RVS-250 mice, but not in RVS-500 mice. During the longer swimming exercise in RVS group, blood glucose and lactate levels were significantly decreased, but free fatty acid not changed. And also LDH levels were significantly decreased in RVS groups compared to negative control mice. Hepatic lipid peroxidation was not affected by RVS, but SOD and catalase activity were significantly increased in RVS groups. Interestingly, the levels of testosterone and free testosterone were significantly increased in RVS groups before swimming exercise, but they were not significantly changed among groups after swimming exercise. The present results suggest that RVS extract powders may enhance swimming exercise performance by recovering the exercise-fatigue via reduction of blood LDH activity and by burning blood glucose as an energy source. These results imply that RVS-produced testosterone may act as an energy buster to enhance physical activity.

Effects of the Soyosan and Chyengansoyosan on the Regional Brain Monoamines Contents of Cold Swimming Stressed Mice (소요산(逍遙散)과 청간소요산(淸肝逍遙散)이 스트레스 생쥐의 뇌부위별(腦部位別) Monoamines 함량(含量)에 미치는 영향(影響))

  • This study aimed to evaluate the anti-stress effects of soyosan and chyengansoyosan on the contents cold swimming stress.The experimental animals were forced cold swimming stress for 3 minutes, and administered 4㎎/10g of soyosan and chyengansoyosan extract for 7 days before stress. The monoamines contents were measured by HPLC method in various part of mouse brain. The following results were obtained :1. In frontal cortex, the contenrs of norepinephrine ans dopamine increased significantly in the group administered chyrngansoyosan compared with in the group administered non chyengansoyosan and soyosan(control group) of mice stressed by cold swiming.2. In hypothalamus, the content of norepinephrine increased with statistical significance in the group administered soyosan compared with control group of mice stressed by cold swimming. 3. The content of dopamine increased with statistical significance on the group administered chyenfansoyosan and compared with control group of mice stressed by cold swimming in hypothlaamus and hippocampus.4. The content of serotonin increased with statistical significance in the group administered soyosan and compared with control group of mice stressed by cold swimming in hippocampus and corpus striatum.5. In corpus striatum and hippocampus, the content of norepineohrine decreased with statistical significance in the group administered soyosan and chyengandoyosan compared with control group of mice stressed by cold swimming.Judgding from the above results, this sthdy reaches a conclusion that soyosan or chyengansoyosas has significant effects in reducing stress.

Synthesis and Studies on Anticonvulsant and Antidepressant Activities of 5-Alkoxy-tetrazolo[1,5-a]quinolines

  • A series of 5-alkoxy-tetrazolo[1,5-a]quinolines were synthesized to evaluate their anticonvulsant and antidepressant effects. Anticonvulsant effects and neurotoxicity of the compounds when injected intraperitoneally to mice were determined by a maximal electroshock (MES) test and a rotarod test, respectively. Only three of the synthesized compounds (4a, 4b, 4c) displayed anticonvulsant activity at a dose of 300 mg/kg. Most of the compounds significantly reduced immobility times during the forced swimming test (FST) at a dose of 100 mg/kg, indicative of antidepressant activity. Among the compounds, 5-(2-fluorobenzyloxy)tetrazolo[1,5-a]quinoline (4k) reduced immobility time by 66.85% at 30 mg/kg compared with the same dose of Fluoxetine, which reduced immobility time by 52.30%. According to the results of the 5-Hydroxytryptophan induced head-twitch test and yohimbine toxicity potentiation test, the noradrenergic system seems not to be involved in the antidepressant-like effect of compound 4k while the serotonergic system seems a little to be involved.

Experimental Study on the Antidepressant Effect of Sam-Jeong-Hwan (삼정환(三精九)의 항우울 효과에 대한 실험적 연구)

  • Objective: The purpose of this study was to investigate the protective effects of Sam-Jeong-Hwan(SJH) on the animal model of depression induced immobilization stress. Method: The subject were divided into 4 groups(l. normal 2. saline solution administered during immobilization stress treatment 3. SJH of 100mg/kg administered 4. BKJ of 400mg/kg administered). Immobilization stress was treated for 1 hours on day. During 2 days of immobilization stress treatment, they were executed forced swimming test, passive avoidance test, elevated plus maze test. Corticosterone and ACTH in blood were measured. Results: In forced swimming test, SJH of 400mg/kg group showed decreased immobilization. In passive avoidance test, SJH of 400mg/kg group showed increased learning execution. In EPM test, SJH of 400mg/kg group showed decreased anxiety. In locomotor activity test, SJH groups showed significantly increased locomotor activity. Stress group showed significantly increase in serum level of corticosterone, SJH of 400mg/kg group showed decreased serum level of corticosterone. Stress group showed significantly increase in serum level of ACTH, SJH of 400mg/kg group showed decreased serum level of ACTH. Conclusion: These results suggest that Sam-Jeong-Hwan(SJH) is effective in the treatment of depression.

Anti-Depressive Effects of OnDam-Tang with Addition of Linderae Radix (ODT-L) after Chronic Immobilization Stress in C57BL/6 Mice (우울증 유발 생쥐에서 온담탕가오약(溫膽湯加烏藥)의 항우울 효과)

  • The purpose of this study was to examine the anti-depressive effects of OnDam-Tang with addition of Linderae Radix (ODT-L) on the animal model of depression induced by chronic immobilization stress. Depression model was made by chronic immobilization stress for 2 hours for 21 days. And we performed forced swimming test, analysis of the neurotransmitter and immunohistochemical staining, measured expression levels of serotonin in the brain. ODT-L has decreased immobilization time in forced swimming test. ODT-L has increased amount of melatonin in the brain. ODT-L has increased expression levels of serotonin in the brain. ODT-L prevented damage in the hippocampal region. ODT-L has reduced the expression level of CRF receptors in in hippocampus region. These results suggest that ODT-L may have anti-depressive effects on depression.

Effects of Forsythiae Fructus on CRF, c-Fos and TH in the Forced Swimming Test (연교(連翹)가 강제수영부하시험에서 CRF, c-Fos와 TH에 미치는 영향)

  • Objectives : In order to investigate the antidepressant effects of Forsythiae Fructus(FF), we performed the Forced Swimming Test(FST). Also the expressions of corticotropin-releasing factor (CRF), c-Fos and tyrosine hydroxylase(TH) were measured by immunohistochemical method at paraventricular nucleus(PVN), ventral tegmental area(VTA) and locus coeruleus(LC). Methods : Spraque-Dawley rats were administered FF extract(100 mg/kg, 400 mg/kg) intragastrically three times prior to the FST. Results : The duration of immobility in the FST was significantly decreased in the FF 100 mg/kg, 400 mg/kg groups(p<0.05). The expression of CRF was significantly reduced in the FF 100 mg/kg and 400 mg/kg groups(p<0.001). c-Fos expression was significantly decreased at PVN in the FF 100 mg/kg group(p=0.069). TH expression at VTA was significantly increased in the FF 100 mg/kg and 400 mg/kg groups(p<0.05). TH expression at LC was not significantly changed(p=0.346). Conclusion : According to the results, it can be suggested that Forsythiae Fructus has antidepressant effect via the decreased immobility through the reduction of CRF and c-Fos expression at PVN.

Anti-inflammatory effects of porcine placenta in forced swimming tested fatigue mice and RAW264.7 cells

  • Inflammation has been linked to various diseases. Especially, fatigue is a frequent symptom in several inflammatory disorders. Therefore, blocking inflammatory process is effective in fatigue. We investigated whether Denmark porcine placenta (DPP) alleviates fatigue by inhibiting inflammatory reaction using forced swimming test (FST) animal model and RAW264.7 cells. In FST-induced fatigue animal model, the mice which received the DPP for 21 days showed decreases of interleukin $(IL)-1{\beta}$ and IL-6 serum levels. Furthermore, our data revealed that lipopolysaccharide (LPS)-induced $IL-1{\beta}$, IL-6, and tumor necrosis $factor-{\alpha}$ secretion were markedly inhibited by DPP in RAW264.7 cells without inducing cytotoxicity. LPS-enhanced nitric oxide secretion and inducible nitric oxide synthase expression were inhibited by DPP. The present study also figured out that these effects of DPP were mediated by blockade of caspase-1 and nuclear $factor-{\kappa}B$ activation. Taken together, our results indicated that DPP could be alleviating fatigue as candidate of anti-inflammatory agent.

Effects of Chengwhabosimtang on depression, anxiety, TH and c-Fos of the brain in the CMS treated rats (청화보심탕(淸火補心湯)이 우울증(憂鬱症) 막형동물(模型動物)의 절망행동(絶望行動), 불안(不安) 및 뇌(腦)의 TH 와 c-Fos 발현(發展)에 미치는 효과(效果))

  • Objective : This study was designed to assess the protective effects of Chengwhabosimtang on the animal model of depression, chronic mild stress(CMS). Method : Male Sprague-Dawley rats Were used for this experiment. The subjects Were divided into 3 groups (1. CMS-drug: Chengwhabosimtang administered during CMS treatment, 2. CMS-vehicle: water administered, 3. normal ). After 4 weeks of CMS treatment they were executed Forced swimming test(FST) and Elevated plus maze. Tyrosine hydroxylase(TH) in ventral tegmental area(VTA) and c-Fos in paraventricular nucleus(PVN) were measured. Result : 1. In FST, CMS-drug group showed significantly decreased immobility behavior. 2. CMS-drug group showed no significantly lower TH level in VTA than CMS-vehicle group. 3. CMS-drug group showed significantly less c-Fos expressed cell bodies in PVN than CMS-vehicle group. 4. In Elevated plus maze, CMS-drug group showed no significantly anxiety. Conclusion : These results suggest that Chengwhabosimtang may have protective antidepressant effects in CMS model rats. And these effects could be explained by the elevated stress-copying behaviors which are related with PVN of hypothalamus and dopaminergic neurons in VTA.
