Development of a multiplex qRT-PCR assay for detection of African swine fever virus, classical swine fever virus and porcine reproductive and respiratory syndrome virus

  • Background: African swine fever virus (ASFV), classical swine fever virus (CSFV), and porcine reproductive and respiratory syndrome virus (PRRSV) are still prevalent in many regions of China. Co-infections make it difficult to distinguish their clinical symptoms and pathological changes. Therefore, a rapid and specific method is needed for the differential detection of these pathogens. Objectives: The aim of this study was to develop a multiplex real-time quantitative reverse transcription polymerase chain reaction (multiplex qRT-PCR) for the simultaneous differential detection of ASFV, CSFV, and PRRSV. Methods: Three pairs of primers and TaqMan probes targeting the ASFV p72 gene, CSFV 5' untranslated region, and PRRSV ORF7 gene were designed. After optimizing the reaction conditions, including the annealing temperature, primer concentration, and probe concentration, multiplex qRT-PCR for simultaneous and differential detection of ASFV, CSFV, and PRRSV was developed. Subsequently, 1,143 clinical samples were detected to verify the practicality of the assay. Results: The multiplex qRT-PCR assay could specifically and simultaneously detect the ASFV, CSFV, and PRRSV with a detection limit of 1.78 × 100 copies for the ASFV, CSFV, and PRRSV, but could not amplify the other major porcine viruses, such as pseudorabies virus, porcine circovirus type 1 (PCV1), PCV2, PCV3, foot-and-mouth disease virus, porcine parvovirus, atypical porcine pestivirus, and Senecavirus A. The assay had good repeatability with coefficients of variation of intra- and inter-assay of less than 1.2%. Finally, the assay was used to detect 1,143 clinical samples to evaluate its practicality in the field. The positive rates of ASFV, CSFV, and PRRSV were 25.63%, 9.36%, and 17.50%, respectively. The co-infection rates of ASFV+CSFV, ASFV+PRRSV, CSFV+PRRSV, and ASFV+CSFV+PRRSV were 2.45%, 2.36%, 1.57%, and 0.17%, respectively. Conclusions: The multiplex qRT-PCR developed in this study could provide a rapid, sensitive, specific diagnostic tool for the simultaneous and differential detection of ASFV, CSFV, and PRRSV.

Pork Production in China, Japan and South Korea

  • Global pork markets are becoming more competitive, riding the wind of the bilateral free trade agreement. China is the world's largest pork producer with nearly 50% of the world's total production. China's fast growing economy has provided its people with higher purchasing power, resulting in a rapid expansion of the Chinese swine industry over the past decades. Worldwide, China consumes the greatest amount of pork and it is believed that this trend will continue. Japan is the world's largest pork importing country, even though it also produces a lot of pork. The Japanese swine industry encounters weighty obstacles in production costs and environmental limitations which result in reduced domestic supply and creates the situation in which Japan has to import a significant amount of pork for their consumption. South Korea is also a large buyer of pork, with a status greatly influenced by the struggle that the country has faced with Foot and Mouth Disease (FMD) which occurred in 2000, 2002 and 2010. High production costs, low production efficiency, and forced culling following the FMD outbreak resulted in the loss of many hog farming households in the country, reducing supply of domestic pork in the face of continued demand. Overall, pork production in these economically important countries can greatly impact the industry globally. The goal of this review paper is to describe pork production in China, Japan, and South Korea and discuss these countries' role in global pork export markets.

Effect of far-infrared radiating products on cold hypersensitivity of lower limbs using Cold Stress Test (CST) : A pilot study (냉부하검사(CST)로 살펴 본 원적외선 기능성 제품이 족부냉증에 미치는 영향 : A pilot study)

  • Purpose: We investigated the effect of far-infrared radiating products on cold hypersensitivity of lower limbs using CST. Methods: 7 patients with cold hypersensitivity of lower limbs were investigated in this study. Exclusive criteria was skin diseases, spinal nervous disease and external wounds. They were asked to answer the VAS of cold hypersensitivity at baseline and after 3 weeks. We measured temperature of lower limbs with Spectrum 9000 MB (Dorex Inc, USA). We performed cold stress test (CST) by 3 thermographic observation using DITI : 1st was taken after 15 minutes resting at $25^{\circ}C$, the 2nd was immediately taken after 1 minute soak in $20^{\circ}C$ water, the 3rd was taken at 15 minutes after soak. We performed 3 times of CST : 1st CST was perfomed at baseline, 2nd CST was perfomred after 1 week and just observation, 3rd CST was performed after 1 week using far-infrared radiating products (lasner, UMT, Korea). Results: After using products, temperature of foot incresed more than thigh area, but there was no significance. There was no statistical difference of VAS, change of temperature and CST between before and after using far-infrared radiating products. Conclusion: There was no statistical effect of far-infrared radiating products on change of temperature of lower limbs.

  • Currently, each farm bears both the outbreak of foot-and-mouth disease and the damage caused by AI. In addition, complaints about odors in the livestock industry are constantly being recovered and are expected to occur in the future. The purpose of this study is to improve the indoor air quality of enclosed facilities such as barns, houses, pigsty, and etc. This paper develops low-temperature plasma waste heat ventilation system to be installed in ventilation unit location and standardizes heat exchange element, low-temperature plasma lamp, and ballast for enhanced air cleaning function. In addition, this study intends to develop a new control system so that the farmers can connect with existing weather systems, flow fans, and other facility equipment by incorporating ICT.

  • Facioscapulohumeral muscular dystrophy (FSHD), the third most common inherited muscular dystrophy, is an autosomal dominant disease characterized by progressive weakness and wasting of the facial, shoulder-girdle, upper arm, foot extensor, and pelvic girdle muscles. FSHD is caused by contraction of the polymorphic D4Z4 repeat in the subtelomere of chromosome 4q. However, there has been no report of genetically confirmed FSHD in Korea. We report a patient with FSHD who was found to have a deletion of D4Z4 repeat on chromosome 4q35.

  • 구제역은 소, 돼지 등 발굽이 두 개로 갈라진 가축들에게 감염을 유발하는 전염성이 매우 높은 바이러스성 질병이다. 구제역 감염 시 입 주변, 구강 내, 코, 발굽사이 등에 수포가 생기며 고열과 식욕이 저하되어 심하게 앓거나 죽게 되는데, 강한 감염 전파력을 가졌음에도 치료제가 없고, 감염확인 즉시 확산 방지를 위한 살 처분만이 이루어지고 있다. 따라서 무엇보다도 빠른 감염여부 진단이 중요하다. 현재까지 구제역을 진단하는 방법으로는 감염 된 가축의 혈액에서 구제역 항원 단백질에 대한 항체형성 여부를 확인하는 항체진단법과 수포액과 같은 체액을 채취하여 세포배양을 통한 구제역 바이러스 분리방법이 있지만 두 가지 모두 짧은 잠복기를 갖는 구제역 바이러스를 빠른 시간 내 진단하기는 어렵다. 따라서 본 연구에서는 보다 빠른 구제역 진단 키트개발을 위해 NCBI Pubmed를 이용하여 구제역바이러스가 가지는 6개의 주요 단백질을 확인하였고, NCBI BLAST를 이용하여 6개의 단백질 중 구제역 바이러스에 특이적인 항원 단백질 peptidase C28을 선정하였다. 선정된 단백질의 아미노산 서열을 이용하여 IEDB analysis resource를 통해 peptidase C28의 epitope 부위를 예측하였다. 예측 된 부위의 아미노산 서열을 NCBI BLAST에서 정상 동물과 비교하여 구제역바이러스 특이 항원 단백질 epitope peptide를 최종 선정하였다. 이를 이용한 구제역 바이러스 진단키트 제작은 보다 빠른 진단을 통해 감염 확산을 조기에 차단하고 경제적 손실과 피해를 최소화 할 수 있다.

A survey on the incidence rate of foot diseases in dairy cattle

  • This study was carried out to investigate the status of hoof diseases and to develope treatment method for cattle with hoof diseases during the period of January to December in 2000. Out of 435 heads, 34 heads(7.8%) had hoof diseases. In respect to season, incidence rate was higher in August to September than that of other seasons. The incidence rates of hoof rot, pododermatitis verrucosae, laminitis, other cases and trauma were 14 cases(41%), 10 cases (29%), 7 cases(21%), 2 cases(6%) and 1 case(3%), respectively. In respect to age and milk production, Incidence rate was higher in the cattle with high milk production. Incidence rate of hoof diseases on the hind-limbs was higher than that of the fore-limbs. E. coli(8 strains), Clostridium spp(8 strains), Staphylococcus spp(6 strains), Fusobacterium spp(6 strains), and Bacteroides spp(6 strains) were isolated from the hoof lesions. All isolates were sensitive to composite preparation made of mainly formalin with picric acid and phenol. (Treatment against each strains isolated was sensitive to composite preparation made out of the main constitutions of formalin with picric acid and phenol).

Ergonomic Problems and Their Improving Measures in Office Environment of General Hospitals

  • Objective: The purposes of this study are to investigate ergonomic problems and to propose their improving measures in office environment of big general hospitals. Background: Office tasks have varying risk factors of work-related musculoskeletal disorders (WMSDs). The first symptom resulted from the office work was recognized as an occupational disease in Korea in 1986. Although the symptoms have increased since its first recognition, there has been few study on the effect of office work environment settings. Method: First, the author took pictures of working scenes performed in three big university hospitals. Next, the pictures were analyzed in view point of ergonomics. Based on the analysis, their improving measures were proposed for reducing work stress. Results: The results showed that most physical office environment settings such as dimensions of tables/desks and chairs, leg room, thigh, knee and foot clearances, and chairs used did not satisfy the ergonomically recommended design guidelines. In addition, some clerks placed personal belongings under their desks, put monitors in high position and did not lean against the backrest of chairs in seated tasks, which resulted in poor working postures of leg, back, neck etc. It is recommended that the hospital management should provide their clerks with ergonomically designed office furniture and continuously perform ergonomics training program for raising clerks' recognition for office ergonomics. Conclusion: Most office environment settings investigated in this study were not in good condition in view point of ergonomic design for the settings. Application: It would be useful as basic data for establishing ergonomically good office environment in hospitals.

Development of the Tailored Health Promotion Program for Rural Elderly: Based on the PRECEDE Model (농촌형 노인 건강증진프로그램 개발 연구: PRECEDE 모형을 중심으로)

  • Purpose: The purpose of this study was to develop the health promotion program for rural elderly through PRECEDE process. Method: The health promotion program was developed based on the preliminary diagnosis. The data collection was performed from March 10th to April 9th, 2003. The subjects were selected at Mari Myun, Geochang Gun, in Korea. The preliminary diagnosis was examined with the 115 elders. Data was analyzed by descriptive statistics and Cronbach's n test with SPSS/Win 10.0 program. Results: The health promotion program was developed based on diagnostic result using PRECEDE model. The developed program was corrected and revised with the advices from 6 experts. The final program included health responsibility(cancer prevention and early detection, hygienic, false teeth management no smoking and moderation in drink, and chronic disease prevention and management), physical activity(6 kinds of exercise), nutrition(low sodium diet calcium intake, and right eating habit), spiritual growth(spiritual demand and death preparation teaching), interpersonal relations(relationship with couple, children, grandchildren, neighborhood), and stress management(sports dance, foot massage, positive thought, and song class). Conclusion: I propose that it is necessary to identity the effect of health promotion program for rural elderly. And strategy development that can spread the health promotion program elderly is needed.

A Possible Role of Trehalose as a Regulatory Molecule in Plant Drought Resistance

  • In many organisms, trehalose has been Down as an energy source and a protectant against various environmental stresses such as desiccation, freezing, heat and osmotic pressure. Previously, we have isolated and characterized the genes encoding trehalose-6-phosphate synthase (ZrTPS1) and trehalose-6-phosphate phosphatase (ZrTPS2) from one of the most osmotolerant yeasts, Zygosaccharomyces rouxii. We have also generated transgenic plants by co-introduction of ZrTPS2 and ZrTPS2 into potato plant (ZrTPS2-2A-ZrTPS1 plant) in an attempt to metabolically engineer trehalose in the transgenic plant using the foot-and-mouth disease virus(FMDV) 2A system and to generate drought resistant crop plants. In this research, we assayed previously generated the ZrTPS2-2A-ZrTPS1 plant biofunctionally by drought treatment, and measured the amount of trehalose in the ZrTPS2-2A-ZrTPS1 transgenic plants. The ZrTPS2-2A-ZrTPS1 transgenic plant showed strong drought resistance in spite of little or no accumulation of transgenic in he transgenic plant compared with control plant.
