  • 동치미에서 분리한 유산균인 Leu. mesenteroides subsp.cremoris DLAB19 균주의 aflatoxin Bl에 대한 항 돌연변이원성 물질 생산을 위한 최적 조건을 조사한 결과, 탄소원으로 glucose를 첨가시 가장 높은 항 돌연변이 효과를 나타내었으며, 질소원으로서는 yeast extract 첨가시 항 돌연변이 효과가 우수하였다. 탄소원으로 glucose의 농도을 2$\%$ 첨가시aflatoxin Bl에 대한 항 돌연변이 효과가 가장 우수하였으며,질소원으로서 yeast extract 1$\%$ 첨가시 가장 우수한 항 돌연변이 효과를 나타내었다. 항 돌연변이원성 물질 생산을 위한 최적 배양 조건은 초기 pH, 배양 온도, 진탕 속도가 각각 7.0, 30$^{\circ}C$ 및 150 rpm이었다. 상기의 최적 조건에서 36시간 배양시 가장 높은 항 돌연변이 효과를 나타내었는데 S. enterica serovar Typhimurim TA100과 TA98을 이용한 경우항 돌연변이 효과가 87.11$\%$와 75.04$\%$이었다.

  • 한국인의 분변으로부터 분리된 Bifidobacterium sp. Int-57은 다른 장내세균에 비해 ${\beta}-xylosidase$의 높은 역가를 보였으며, xylooligomer의 bifidogenic factor의 유용성을 고려하였을 때 효소생산에 초점을 둔 결과 최적 탄소원으로는 xylose였는데, 일반적으로 최종생산물은 feedback inhibition, feedback repression 등으로 효소 생산을 억제하는 일반 이론과는 상이한 결과를 보였다. 또한 xylose는 1.1%일 때 가장 높은 효소역가를 보였다. 질소원으로서는 yeast extract였고, 그 농도가 0.04%일 때, 무기염류 면에서는 $CoCl_2$였고, 농도가 0.0003%일 때 ${\beta}-xylosidase$의 최대 생산을 보였다.

하계 한반도 해역(동해, 서해, 남해 및 동중국해)의 하위영양단계 먹이망 구조 : 탄소 및 질소 안정동위원소 활용 (Food-Web Structures in the Lower Trophic Levels of the Korean Seas (East Sea, West Sea, South Sea, and East China Sea) during the Summer Season: Using Carbon and Nitrogen Stable Isotopes)

  • 본 연구는 2019년 8월 한반도 주변해역(동해, 서해, 남해, 동중국해)에서 탄소 및 질소 안정동위원소 기법을 활용하여 하위영양 단계에서의 먹이망 구조를 파악하였다. 입자성 유기물(POM)의 δ13C 범위는 -26.18 ~ 20.61 ‰, δ15N 범위는 5.36 ~ 15.20 ‰의 넓은 범위를 보였다. POM과 각 생물별 개체군 사이의 δ13C 분별작용의 결과는 대부분 micro-POM을 섭식하는 것으로 확인하였으나 해역 간 차이를 보였다. 각 생물별 영양단계는 chaetognaths (3.40±0.61)가 가장 높은 영양단계에 있음을 확인하였다. 동위원소 혼합모델을 적용한 결과에서 chaetognaths의 먹이원으로 copepods (13 ~ 48 %)와 euphausiids (20 ~ 51 %)가 가장 높은 기여도를 나타냈다. 본 연구결과 각 해역별 먹이원의 제한적 공급 및 다양성의 차이가 먹이망 구조 및 각 생물별 동위원소 비에 영향을 미친 것으로 판단된다.

Effect of Temperature and Carbon Source on the Expression of $\beta$-Galactosidase Gene of Lactococcus lactis ssp. lactis ATCC 7962

  • The effects of growth temperature and a carbon source on the expression of $\beta$-galactosidase gene of Lactococcus lactis ssp. lactis ATCC 7962 (L. lactis 7962) were investigated. At $25^{\circ}C$, L. lactis 7962 had a higher $\beta$-galactosidase activity than cells grown at $30^{\circ}C$ or $37^{\circ}C$, although cells grew most quickly at $37^{\circ}C$ The highest $\beta$-galactosidase activity was observed in cells grown in M17 with lactose (l %) followed by cells grown in a galactose (1 %) medium. L. lactis 7962 exhibited the minimum $\beta$-galactosidase activity in glucose media, indicating catabolite repression. When the cellular levels of $\beta$-galactosidase mRNA were examined using slot blot hybridization, no significant differences were observed between cells grown at $25^{\circ}C$ and cells at $30^{\circ}C$ or $37^{\circ}C$ in the same media. This suggests that the quantity of $\beta$-galactosidase mRNA may not be the reason for the higher $\beta$-galactosidase activities of L. lactis 7962 at $25^{\circ}C$ The level of ccpA (Catabolite Control Protein) transcript remained almost constant during the exponential growth phase irrespective of a carbon sourse.

사상균 FM04에 의한 Amylase 생산 및 음식물 쓰레기의 효소학적 가수분해 (Production of Amylase by a Filamentous Fungus, Strain FM04, and Enzymatic Hydrolysis of Food Waste)

  • 본 연구에서는 썩은 고구마 및 감자로부터 amylase 생산균주인 사상균 FM04를 분리하였으며, 분리균주의 섬유소분해 효소 생산에 관한 배양의 환경변수 및 기질특성을 조사하였다. 최적의 효소생산을 위한 온도, pH, 교반조건은 각각 28∼3$0^{\circ}C$, 5.0∼6.0, 100rpm이였다. FM04는 Amylase 생산을 위해 탄소원으로서 starch을 잘 이용하였다. 경제적인 효소생산을 위해 전분이 많이 함유된 음식물쓰레기를 이용하였고, 1% (w/v)의 기질농도에서 5.2 U/ml의 amylase 효소활성을 얻을 수 있었다. Amylase 활성의 최적온도 및 PH는 각각 6$0^{\circ}C$와 4.5이었으며, 열안정성은 5$0^{\circ}C$에서 2일 동안 90%의 활성이 유지되었다. 음식물쓰레기의 효소학적 가수분해에서는 2.5 U/ml의 amylase 배양상등액에 음식물쓰레기 20% (w/v)의 첨가한 반응물을 5$0^{\circ}C$, 48시간의 반응한 결과 72.6 g/L의 환원 당을 얻을 수 있었다.

Food web structure in a $Salix$ $subfragilis$ dominated wetland in Hangang estuary using stable isotopes and fatty acid biomarkers

  • We investigated food webs of a $Salix$ $subfragilis$-dominated wetland in the Janghang wetland in the Hangang estuary, which is very close to the Demilitarized Zone, along the west coast of Korea. Our study focused on understanding sesarmine crab ($Sesarma$ $dehaani$)-related food webs in a $S.$ $subfragilis$ forest. For our study, we used carbon and nitrogen stable isotopes and fatty acid biomarkers. We collected samples of plants, animals, and detrital sediment from four quadrats ($5{\times}5m^2$) set in the $S.$ $subfragilis$ community. Samples were collected from September 2006 to June 2009, except during the winter hibernation period of $S.$ $dehaani$. In the wet season, the sediment showed relatively high ${\delta}^{13}C$ and low ${\delta}^{15}N$ signatures compared with relatively low ${\delta}13C$ and high ${\delta}15N$ signatures in the dry season. Mature $S.$ $dehaani$ appeared to feed on fresh leaves and other carbon sources, such as immature individuals or fish, in addition to detrital sediment, which appeared to be the main carbon source for immature crabs. Principal component analysis of fatty acid biomarkers of $S.$ $dehaani$ showed a clear difference between immature individuals (10-30 mm) and mature ones (larger than 30 mm), indicating that the main food source for immature crabs was detrital sediment, whereas mature crabs foraged plants in addition to consuming detrital sediment. On the basis of our results from stable isotope and fatty acid analyses, mature $S.$ $dehaani$ appeared to feed on detrital sediment and fresh leaves of $S.$ $subfragilis$ in summer in addition to engaging in cannibalism of immature individuals.

음폐수의 열가수분해 최적조건 도출과 생물학적 탈질공정에서 열가용화액의 적용 가능성에 관한 연구 (A Study on Optimum Conditions Derivation on Thermal Hydrolysis of Food Wastewater and the Applicability of the Thermal Solubilization in Biological Denitrification Process)

  • The aim of this research is to derive an optimum operating condition for the thermal solubilization equipment that is employed to increase concentration of soluble organic materials and to assess whether it would be possible to use the waste sludge generated by thermal solubilization reaction as an external carbon source in biological denitrification process. For the purpose, we have constituted a laboratory-size thermal solubilization equipment and have assessed thermal hydrolysis efficiency based on various reaction temperature and reaction time. We have also derived SDNR using the waste sludge generated by thermal solubilization reaction through a batch experiment. As a result of research, the highest thermal hydrolysis efficiency of about 42.8% was achieved at $190^{\circ}C$ of reaction temperature and at 90 minutes of reaction time. And when SDNR was derived using the waste sludge, the value obtained was $0.080{\sim}0.094\;g\;NO_3{^-}-N/g\;MLVSS{\cdot}day$, showing SDNR that is higher than that obtained by the results of existing researches that used common wastewater as an external carbon source. Accordingly, in view of the fact that food wastes vary quite a bit in characteristics based on the area they are generated from and seasonal change, it seems that a flexible operation of thermal solubilization equipment is required through on-going monitoring of food wastes that are imported to food wastes recycling facilities.

된장으로부터 Angiotensin 전환효소 저해제 생산 세균의 분리 및 특성 (Isolation and Characterization of a Bacterium from Korean Soy Paste Doenjang Producing Inhibition of Angiotensin Converting Enzyme)

  • 우리 나라 전통발효식품인 된장으로부터 angiotensin 전환효소(angiotensin converting enzyme, ACE) 저해활성이 우수한 균주 SYG3를 분리하여 형태학적, 배양학적, 생리학적인 특성을 조사한 결과 B. pumilus 또는 그 유연균으로 동정되었다. 분리 균주의 ACE 저해물질 생산조건을 알아보기 위해 각종 배양조건에 따른 저해활성을 조사한 결과, 탄소원으로서는 glucose, 질소원으로서는 soybean powder에서 가장 높은 저해활성을 나타내었으며, skim milk와 casein에서도 비교적 높은 저해활성을 나타내었다. Soybean powder 농도에 따른 저해활성은 5%(w/v) 첨가시 가장 높은 저해활성을 나타내었으며. 그 이상의 농도에서는 저해활성의 변화가 거의 없었다. NaCl 농도는 3%(w/v) 첨가시 저해활성이 가장 높은 것으로 나타났으며 배양온도는 $32^{\circ}C$, 초기 pH 7.0에서 가장 높은 저해활성을 나타내었다. 최적배지조건에서의 배양시간에 따른 ACE 저해활성은 배양 36시간에 약 98%로서 최고에 도달하였다.

자연치즈가 함유된 칼슘강화 구워먹는 치즈두부 품질 특성 (Quality Characteristics of Grilled Cheese Tofu Containing Natural Cheese and High Calcium)

  • This study was conducted to develop a novel cheese tofu hybrid product by increasing the calcium content of cheese, an animal protein source, and tofu, a vegetable protein source. Both pH and moisture were lower in the cheese tofu hybrid than those values in the control group (6.03% and 72%, respectively). Protein and fat were slightly higher than in the control (15.43% and 9.91%, respectively). Total bacteria count increased at the end of the product's shelf life, but did not affect its texture. The cheese tofu hybrid displayed stronger a) red, and b) yellow coloration than the control, and its lightness (L) was lower than that of the control. The cheese tofu hybrid possessed high hardness, and displayed high values for gumminess and brittleness. Sensory evaluation by a specialized agency examined consumer preferences, purchase intentions, strengths, and weaknesses of the developed product line. A panel of 30 female volunteers in their 20s and 40s recorded an overall preference for cheese tofu of 5.40 points, fairly good. Cheese tofu was found to be better than the control tofu, with appealing differences in appearance, color, nutty aroma, chewiness, and cheese flavor.

Effects of Nitrite and Phosphate Replacements for Clean-Label Ground Pork Products

  • We investigated the effects of different phosphate replacements on the quality of ground pork products cured with sodium nitrite or radish powder to determine their potential for achieving clean-label pork products. The experimental design was a 2×5 factorial design. For this purpose, the ground meat mixture was assigned into two groups, depending on nitrite source. Each group was mixed with 0.01% sodium nitrite or 0.4% radish powder together with 0.04% starter culture, and then processed depending on phosphate replacement [with or without 0.5% sodium tripolyphosphate; STPP (+), STPP (-), 0.5% oyster shell calcium (OSC), 0.5% citrus fiber (CF), or 0.5% dried plum powder (DPP)]. All samples were cooked, cooled, and stored until analysis within two days. The nitrite source had no effect on all dependent variables of ground pork products. However, in phosphate replacement treatments, the STPP (+) and OSC treatments had a higher cooking yield than the STPP (-), CF, or DPP treatments. OSC treatment was more effective for lowering total fluid separation compared to STPP (-), CF, or DPP treatments, but had a higher percentage than STPP (+). The STPP (+) treatment did not differ from the OSC or CF treatments for CIE L* and CIE a*. Moreover, no differences were observed in nitrosyl hemochrome content, lipid oxidation, hardness, gumminess, and chewiness between the OSC and STPP (+) treatments. In conclusion, among the phosphate replacements, OSC addition was the most suitable to provide clean-label pork products cured with radish powder as a synthetic nitrite replacer.
