EMS-induced Mutagenesis for C18 Unsaturated Fatty Acids in Rapeseed (Brassica napus L.)

  • Rapeseed (Brassica napus L.) oil with high oleic acid content is of great interest for both food and non-food uses. The 'Tamla' variety, characterized by oleic acid content of approximately 69%, was treated with 1% ethyl methane sulfonate (EMS) to induce mutations in the fatty acid biosynthesis pathway. $M_1$ plants were selfed and subsequent generations ($M_2$, $M_3$, and $M_4$ mutants) were analyzed to identify mutants having increased levels of oleic acid. $M_2$ mutants showed oleic acid content ranging from 13.5% to 76.9% with some mutants (TR-458 and TR-544) having up to 74.7% and 76.9% oleic acid, which was an increase of nearly 5% and 7%, respectively, compared to untreated cv 'Tamla'. We selected two $M_3$ mutants with >75% oleic acid content. One mutant (TR-458-2) had increased oleic acid (75.9%) and decreased linoleic acid (12.5%) and linolenic acid (4.4%) contents. The other (TR-544-1) showed increased oleic acid content (75.7%) and decreased linoleic acid (13.5%) and linolenic acid (3.3%) contents. The accumulation or reduction of oleic acid content in the selected $M_4$ mutants was also accompanied by a simultaneous decrease or increase in linoleic and linolenic acid contents. The high-oleic lines could be utilized further in breeding programs for improvement of rapeseed oil quality.

다시마와 미역의 섭취가 발암물질에 의한 DNA 손상과 칼슘 및 철 흡수에 미치는 영향 (Effect of Seatangle and Seamustard Intakes on Carcinogen Induced DNA Adduct Formation and the Absorption of Calcium and Iron)

  • A number of epidemiological studies has indicated lifestyles including dietary habits are closely related to the development of certain forms of cancer. These findings have led several investigators to identify the ways in which these factors mdulate the risk of cancer. Seaweeds are rich sources of non-digestible polysaccharides which possibly posses physiological functions. In vitro studies showed several components in seaweeds inhibit tumor cell growth and mutagenicity of known food mutagens. On the other hand non-digestible polysaccharides of different food sources negatively affect mineral nutrition by decreasing mineral absorption. The objectives of this study was to investigate the effect of major seaweed intake on azoxymethane(AOM) - induced DNA damage a known cancer initiation step and on apparent absorption of calcium and iron. To accomplish these objectives twenty five ICR mice were divided into five groups and fed one of the following diets for 10 days : control diet d, diet containing 10% water-soluble fraction of seamustard or seatangle diet containing 10% water-insoluble fraction of seamustard or seatangle. AOM was injected 6 hours before sacrifice and N7-methylated guanines from the colonic DNA were quantified using a gas chromatography -mass spectroscopy. Fecal samples were collected on days 4 and 8. Caclium and iron contents of the diets and feces were analyzed using an atomic absorption spectrophotometry to determine the apparent absorption of these minerals. Results are as follows. AOM-induced guanine methylation of colon was decreased in animals fed diets containing water-soluble fractions of seamustard or seatangle compared to those in animals fed control diet although only the seatnagle fed group showed statistically significant effect. Apparent calcium absorption was significantly reduced in animals fed diets containing water-insoluble fractions of seaweeds. Iron absorption was significantly decreased and negatively balanced in animals fed diets containing water-insoluble fractions of both seaweeds, and water-soluble fraction of seatangle. In conclusion, seamustard and seatangle intakes may effectively prevent colon tumorigenesis by reducing a carcinogen-induced DNA damages, and more mechanistic studies on possible role of seaweeds on carcinogenesis are required. Also, adverse effects of seaweed diets cintaming a large amount of polysaccharides on mineral nutrition should be carefully monitored.

DMBA로 유발한 Mouse 피부암에 대한 Astaxanthin이 함유된 난황의 항암효과 (Inhibition of DMBA-Induced Mouse Epidermal Carcinogenesis by Astaxanthin-Containing Egg Yolks)

  • Anticarcinogenic activity of astaxanthin-containing egg yolks (designate AEY) was investigated for 7,12-dimethylbenz[a]anthracene (DMBA)-induced two stage mouse epidermal carcinogenesis. Female ICR mouse (6-7 weeks of age) were house in a humidity-and-temperature-controlled facility and subjected to feed and water ad libitum. AEY (10 mg/0.2 ml acetone) was painted on the back of mice 7 days, 3 days and 5 min before DMBA treatment (50 nmole/0.2 ml acetone). One week later after DMBA treatment, 6 ${\mu}g$ tetradecanoyl 12-phorbol 13-O-acetate (TPA) dissolved in 0.2 ml acetone was applied on the mouse twice weekly over a period of 22 weeks. No sample was given to control mice. Control egg yolk (CEY) and astaxanthin-containing oil (designate AO) from Phaffia rhodozyma were used as positive controls. Mouse treated with AEY exhibited 10 tumors per mouse whereas control mouse exhibited 15 tumors per mouse, the fact that 33% reduction of tumor per mouse by AEY treatment. Tumor incidence was also reduced to 15% by AEY treatment when compared to that of control group. Such effects were also seen in CEY and AO treatment groups, but leaser extent. AO gave reduction of food intake and body weights relative to those of AEY and CEY, indicating toxicity of AO. These results suggest that AEY exhibits anticarcinogenic activity for DMBA-induced mouse epidermal carcinogenesis.

황 흡착 알긴산의 Lactobacillus acidophillus 성장에 미치는 영향 및 항 돌연변이 작용 (The Antimutagenic Activity of Sulfated Alginate and Its Effect on the Growth of Lactobacillus acidophillus)

  • 톳으로부터 산ㆍ알칼리 (AASA) 방법으로 추출한 알긴산에 인위적으로 sulfate를 흡착시킨 후 ICP-AES를 이용해서 확인한 결과 3% alginate용액에서 1843 ppm 농도의 sulfate가 검출되었다. 배지 자체적으로 황을 함유하고 있는 MRS broth와 황이 없는 peptone water배지 에 S-alginate와 alginate를 0.25, 1, 2% 농도별로 첨가하여 Lactobacillus acidophilus를 접종한 후 배양하면서 유산균의 성장에 대한 영향을 살펴 본 결과 모든 농도에서 이들 배지에 S-alginate를 첨가한 것이 유산균의 성장을 증가시키는 것을 알 수 있었다. S-alginate와 alginate를 0.25% 첨가한 경우 유산균의 성장은 비슷한 값을 나타내었으나 농도가 증가할수록 S-alginate를 첨가한 군이 alginate만 첨가한 군보다는 유산균의 성장을 늘리는 것으로 나타났다. Ames test를 통해서 S-alginate는 돌연변이성을 전혀 나타내지 않았으며 항 돌연변이성는 직접변이원에 대하여 TA98균주는 14∼19%, TA100 균주에 대해서는 12∼17%까지의 억제율을 보였으며 2%의 S-alginate가 가장 높은 억제율을 나타내었다.

Effects of Six Different Starter Cultures on Mutagenicity and Biogenic Amine Concentrations in Fermented Sausages Treated with Vitamins C and E

  • This study was performed to determine changes in mutagenicity and biogenic amine concentrations in sausages fermented with six different starter cultures treated with vitamins C and E. Six different types of fermented sausages with different combination of starter cultures were manufactured. T1, Pediococcus acidilactici; T2, P. pentosaceus and Staphylococcus carnosus; T3, S. carnosus, S. xylosus, Debaryomyces hansenii, Lactobacillus curvatus, and P. pentosaceus; T4, S. carnosus and L. sakei; T5, S. xylosus and L. plantarum; and T6, Penicillium nalgiovensis. After treatment with vitamins C and E in fermented sausages, changes in mutagenicity and biogenic amine concentrations were measured. The sausages fermented with Staphylococcus xylosus and Lactobacillus plantarum starter cultures showed the most effective antimutagenic activity (p<0.05). The mutagenicity was further decreased in the sausages treated with vitamins C and E (p<0.05), regardless of the starter cultures. The use of Pediococcus acidilactici, S. xylosus, L. plantarum, and Penicillium nalgiovensis as starter cultures was effective in decreasing biogenic amine concentrations (p<0.05). In addition, vitamin E was more effective in decreasing the biogenic amine concentrations than vitamin C. In conclusion, we recommend the use of S. xylosus and L. plantarum as starter cultures, in addition to the use of vitamins C and E, to reduce the potential risk of meat mutagens in fermented sausages.

머루 과피와 종자 에탄올 추출물의 항산화 활성 및 항돌연변이 활성 분석 (Analysis of Antioxidative and Antimutagenic Activities of Ethanol Extracts from Pericarp and Seeds of Wild Grape (Vitis coignetiea))

  • The antioxidative activity and antimutagenic activity of the ethanol extracts from pericarp and seeds of wild grape (Vitis coignetiea) were analyzed in this study. The antioxidative activity of the extracts from wild grape was determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay. The antimutagenic activity of the extracts was evaluated on Salmonella typhimurium TA98 and TA100 by Ames test using 4-nitroquinoline 1-oxide (4-NQO) and sodium azide as mutagens. In the antioxidative activity determination, $IC_{50}$ values of the DPPH radical scavenging activity of the extracts from pericarp and seeds were 27.16 ppm and 7.61 ppm, respectively. Additionally, ABTS radical scavenging activities of pericarp and seed extract were 99.75% and 98.87% at 200 ppm, respectively. In the antimutagenic activity determination, pericarp extract at 5 mg/plate inhibited 72.6% and 74.3% of mutagenicity of S. typhimurium TA98 induced by 4-NQO and sodium azaid, respectively. Also, the mutagenicity inhibition rates of seed extract at 5 mg/plate were 77.8% and 74.5% in S. typhimurium TA100 induced by 4-NQO and sodium azaid, respectively. These results demonstrate that the ethanol extract from wild grape has remarkable antioxidant activity and antimutagenicity.

  • Rice(Oryza stiva L.) is a major cereal food providing nourishment to over half of the world's populations and was considered only as a source of energy. However, The recent many researches have been made to suggest that rice may relate to prevention chronic disease and health-promoting properties such as postprandial glucose response, hypocholesterolemic effect and blood pressure-lowering effect. There has been numerous observation supporting that rice has hypocholesterolemic effect. Rice, rice bran, rice bran oil and unsaponifiable matter of rice bran oil reduced plasma cholesterol in rat, hamster as well as human. Components of rice showing hypocholesterolemic effect include dietary fiber(hemicellulose, phytic acid). protein, ${\gamma}$-oryzanol, $\beta$-sitosterol, and tocotrienols. Crapo et al has been studied that the effect of various of starchy foods on the postprandial blood glucose and insulin responses in healthy and diabetic humans. The results showed that rice had lower blood glucose and insulin responses compared to potato, bread and dextrose. The different physical forms in the same starch also produce the different postprandial glucose and insulin responses. In recent years, several studies have shown that some components of rice have potent antioxidant activity against Fe$^{2+}$ -ascorbate induced lipid peroxidation in rat liver microsomal membranes. Cell culture and animal studies have shown that some components of rice have inhibitory effect on the growth and proliferation of several types of human cancer cell. It was also reported that the methanol extract of brown rice has antimutagenic activity against various mutagens. In addition, the pepsine hydrolysate from rice protein is reported to inhibit angiotensin converting enzyme activity. GABA (${\gamma}$ - aminobutyric acid) and GABA enriched rice germ is also effective for lowering blood pressure and triglyceride levels.s.

  • (l) 핵산관련물질 생산균주의 분리를 위한 변이 유기제와 최적 처리조건은 UV; 30 cm, 1 min, NTG; 200 $\mu\textrm{g}$/ml, 20min, DES; 0.32 %, 90 min, EMS; 5%, 60 min 이었으며 이때 변이솔는 각각 0.083 %, 0.67%, 1.1 %, 및 0.45%로 DES가 가장 양호하였다. (2) Purine 영양요구변이주 239 strain 중 5'-IMP 생산균주는 모두 6 strain이었으며 이중 adenine-guanine 동시요구주 D-2150이 4.8mg/ml로 가장 우수하였다. (3) D-21530 strain의 배양액에서 얻은 결정을 paper chromatogram의 Rf치, 자외선 및 적외선 흡수 spectum 비교, 가수분해에 의한 hypoxanthine 생성량 및 orcinol 반응에 의한 D-ribose의 생성량을 검토한 결과 5'-IMP로 동정되었다. 이 연구을 할때 협력하여 주신 제일제당주식회사 김홍집 상무, 그리고 핵산의 정량분석을 하여준 문화식 씨에게 감사드립니다.

Chitosan 가수분해물의 in vitro 항돌연변이 활성 (Antimutagenic Activity in vitro of Chitosan Hydrolysates)

  • Chitosan이 항돌연변이 활성을 나타내는 분자량 범위를 알아보기 위해, chitosan 가수분해물의 3-amino-1methyl-5H-pyrido[4,3-b]indole(Trp-P-2), aflatoxin $B_1$, 2-nitrofluorene 및 4-nitroquinoline oxide에 대한 억제활성을 Salmonella typhimurium reversion assay와 SOS chromotes로 조하였다. Chitosan을 산가수분해한 후 한외여과하여 6가지 fraction를(분가량 1,000 이하, $1,000{\sim}3,000,\;3,000{\sim}10,000,\;10,000{\sim}30,000,\;30,000{\sim}100,000$, 100,000 이상)의 chitosan 가수분해물을 제조하였다. 제조된 각 fraction의 탈아세틸화도는 큰 차이가 없었으며, 시료 자체의 pH가 복귀 돌연변이수와 SOS 유도 반응에 미치는 영향은 없는 것으로 나타났다. 얻어진 fraction별로 Salmonella typhimurium reversion assay를 실시한 결과, Trp-P-2에 대한 항돌연변이 활성은 fraction 6의 5% 농도에서 78%, aflatoxin $B_1$에 대해서는 fraction 5의 10% 농도에서 92%, 그리고 2-nitrofluorene에 대해서는 fraction 6의 5% 농도에서 51%의 최고 활성을 나타냈다. 한편 SOS chromotest에서는 Trp-P-2에 대하여 $0{\sim}54%$, 4-nitroquinoline oxide에 대하여 $0{\sim}77%$의 억제 활성을 나타냈다.

Chitosan의 in vitro 돌연변이 억제효과 및 세포내 작용 특성 (In vitro Antimutagenic Activity of Chitosan and Its Bio-antimutagenic Characteristics)

  • 키토산의 in vitro 돌연변이 억제활성을 Salmonella typhimurium reversion assay와 SOS chromotest 이용하여 살펴보았다. S. typhimurium에 의한 시험된 간접변이원 Trp-P-2에 대해서 0.1-1.0 mg/plate의 chitosan농도로 시험하였을 때, 24-65%의 돌연변이 억제활성을 나타내었다(p<0.01). Chitosan농도 0.1-0.5 mg/plate 범위에서는 용량-반응(dose-responese)관계를 나타내면서 저해효과를 나타내었으며, 0.5 mg/plate이상의 농도에서는 오히려 저해효과가 감소하는 경향이었다. 반면, 직접변이원인 SA 및 2-NF로 유도된 돌연변이에 대해서는 시험한 어느 농도에서는 chitosan에 의한 돌연변이 억제효과가 나타나지 않았다. Chitosan은 직접변이원인 4-NQO에 의한 SOS 유도에 대해 chitosan농도가 0.15 mg/assay 및 0.20 mg/assay일 때 4-NQO에 의해 유도된 유도지수(induction factor) 8.290을 4.226및 4.516으로 낮추어 46-49%의 저해활성을 나타내었다. 간접변이원인 Trp-P-2에 의한 SOS 유도에 대한 chitiosan의 억제효과는 시험한 chitosan의 농도범위에서 약 9-39%의 저해활성을 나타내었으며, chitosan농도가 0.1 mg/assay이 경우를 제외하고는 chitosan농도 증가에 따라 비례적으로 SOS유도 저해활성이 나타났다. Trp-P-2d 의해 DNA 손상을 유도한 다음 chitosan의 세포내 역제(bio-antimutagenicity) 활성을 살펴 본 결과, 저농도에서는 세포의 억제(desmutagenicity) 특성을, 0.75-1.0 mg/plate의 비교적 고농도에서는 세포내 억제특성을 나타내었다.
