Quality Characteristics of Yakju (a Traditional Korean Beverage) after Addition of Different Tissues of Opuntia ficus indica from Shinan, Korea (신안산 손바닥선인장의 첨가비율 및 부위별에 따른 약주의 품질특성)

  • Fermentation characteristics of yakju prepared with addition of Opuntia ficus indica were examined, with respect to the nature and concentration of additional added materials. The pH level began to decrease after the secondary brewing stage and the total acid content increased during fermentation. The level of reducing sugars in yakju prepared with Opuntia ficus indica increased at the first brewing stage and then slowly decreased after 4 days of fermentation. The ethanol content of supplemented yakju rapidly increased during the initial 6 days of fermentation, to a maximum content of 17.1% after 14 days. The free sugar content was higher in yakju fermented with Opuntia ficus indica stem compared with fruit. The level of organic acids increased as the amount of Opuntia ficus indica material increased. Organic acid level increased during fermentation and lactic acid was the main organic acid in yakju fermented with Opuntia ficus indica. Sixteen volatile compounds were found by GC-MS in supplemented yakju. The most prominent volatile component was iso-amyl alcohol, followed by butyl alcohol and methyl esters. The DPPH radical-scavenging activity of yakju fermented with Opuntia ficus indica stem was higher than when yakju was prepared with fruit. Sensory scores of yakju fermented with 20% (w/v) fruit and 10% (w/v) stem were greater than those of yakju prepared by other treatments.

Effects of Medicinal Plant Extract on the Change of Cerebral Hemodynamic in Rats (약용식물 추출물이 흰쥐의 뇌혈류학적 변화에 미치는 영향)

  • As an attempt to develop new functional health beverage by using medicinal herb, we investigated the effect of medicinal plant extract (MPE) on mean arterial blood pressure (MABP) and regional cerebral blood flow (rCBF) of rats. The changes of MABP and rCBF were determined by LDF methods. LDF allows for real time, noninvasive, continuous recordings of local CBF. MABP in MPE treated rats showed significant change of MPE 1.0 and 10.0 mg/kg. MPE i.v. administration showed significant increase of rCBF in a dose-dependent manner. Propranolol pretreated MABP showed significant change in the increase of MPE. rCBF of propranolol pretreated rats showed significant change from the i.v. injection concentration of 1.0 and 10.0 mg/kg. The ischemia/reperfusion induced oxidative stress may have contributed to cerebral damage in rats, and the present study provides clear evidence for the beneficial effect of MPE on ischemia induced brain injury. Also, the action mechanism in elevation effect of MPE on rCBF might be concerned with the role of $\beta$-adrenoceptor. The exact component and mechanism remains for the future study.

  • Soong-Neung is a Korean traditional beverage served after meals and is made from Soong-Neung producing rice (Noo-Roon-Bap) which is cooked and toasted rice produced on the bottom of the container during the rice cooking process. In order to study the chemical changes occurring in Soong-Neung producing rice with temperature$(20{\sim}220^{\circ}C)$ during the cooking process, thermal analysis, total sugars, total nitrogen, reducing sugars, water soluble nitrogen, total acid, carbonyl content, phenolic compounds were determined. Thermal analysis showed that decrease of weight and endothermic reaction caused by evaporation of water in the sample appeared at $95{\sim}130^{\circ}C.$ The production of volatile compounds increased gradually beginning at $130^{\circ}C$, however, those compounds increased markedly at $160^{\circ}C$ and above. Maximum absorption of ultraviolet spectra of an aqueous distillate occurred at about $273m{\mu}$. Organoleptic analysis showed that an acceptable flavor was produced in the temperature range of $125{\sim}155^{\circ}C.$

Conformance of Caffeine Content and Labeling of Ready-to-drink Coffee (Ready-to-drink (RTD) 커피 음료의 카페인 함량과 표시사항의 적합성)

  • This study monitored the caffeine content of ready-to-drink coffee and verified the appropriateness of the labeling. The caffeine content was analyzed using HPLC. The average caffeine content of cold brew coffee was 0.31-1.04 mg/mL, with an average of 0.55 mg/mL. The average content of product was 147.27 mg/bottle, and taking into account the recommended daily intake, an adult can consume 2.7 bottles. Americano coffee was 0.15-0.38 mg/mL, with an average of 0.28 mg/mL. The average content of product was 110.42 mg/bottle, and considering the recommended daily intake, an adult can consume 3.6 bottles. The caffeine content of decaffeinated cold brew coffee was 5.14 mg/bottle and compared to Americano coffee, more than 95% of the caffeine was removed. In addition, we verified the tolerance level of the total caffeine content in ready-to-drink coffee, and none of them exceeded 120%, signifying that all commercial products were effectively managed.

The Sensory Characteristics and Estimation of Shelf-life by $Q_10$ Values with Mixtures of High-Intensity Sweeteners for Beverage during Storage (청량음료용 고강도 감미료의 혼용 비율에 따른 관능적 특성 및 저장기간 산정)

  • This study investigated the sensory characteristics of mixtures of high-intensity sweeteners for beverage and estimated the shelf life during storage. Sensory characteristics of mixtures of sweeteners (Aspartame/ Acesulfame-K, Aspartame/Sucralose and Acesulfame-K/Stevioside) were evaluated in aqueous (ranging from 90:10 to 50:50) and citrate buffer (ranging from 90:10 to 50:50) solutions. Significant synergistic effects were found in Aspartame/Acesulfame-K and Aspartame/Sucralose mixtures. No significant differences were found in other taste attributes (astringency, bitterness, metallic taste etc.). Aspartame/Acesulfame-K 5:5 solution showed the most acceptable sensory attributes. $Q_10$ values of Aspartame and Acesulfame-K mixture in citrate buffer (ranging from 90:10 to 50:50) solution were calculated from the temperature data (between $40^{\circ}C\;and\;50^{\circ}C$) determined by HPLC. $Q_10$ values were in the range of 2.01-2.25. Their shelf lives were calculated to be lengthened with increasing Acesulfame-K mixture ratio. Their shelf lives in Aspartame/Acesulfame-K 5:5 citrate buffer solution estimated at $20^{\circ}C\;and\;30^{\circ}C$ were 178 days and 88 days, respectively.

Effects of Tea Fungus/Kombucha Beverage on Growth, Serum Protein Level and Enzyme Activities in Male Rats (발효홍차가 흰쥐의 성장률, 혈청 단백질농도 및 효소활성에 미치는 영향)

  • The current study was undertaken to determine the prolinged effects of fasting-refecding on the lipoprotein lipase (LPL) activity and lipogenesis of adipose tissus in male Sprague-Dawley rals and to investigate the effects of various degrecs of food restriction during refeeding on the LPL activity and lipogenesis. The control group (n=5) was fed ad libitum and killed in the fed state at the beginning of the experiment. All rats except conteol group were fasted for 2 days (n=50). Five rats were killed at the end of fasting and others (n=45) were refed either ad libitum (ad libitum group) or mildly restricted (20% food restricted group), or esverely restricted diet (40% food restricted group). Rats were killed on the day of 7th, 14th, and 21st of refeeding. Lipogenesis was determined by the amount of glucose converted to the total lipid. Body weight and epididymal adipose tissue weight returned to control states by 5 days in ad libitym group and by 14 days in 20% food restricted group. As expected, in 40% food restriction during refeeding weight and epididymal adipose tissue weight did not rcturn to control states until day 21. On day 21 after refeeding, the serum total cholesterol concentration of ad libitum group was significantly (p<0.05) higher than that of control group. The serum HDL-C concentration of 40% food restricted group during refeeding was significantly (p<0.05) higher than that of control group. However, there were no significant dif-ferences in serum HDL-C/total cholesterol (TC) ratio and trigluceride concentration among the groups. Fasting for 2 days decreased lipogenesis and LPL activity (p<0.05). On day 21 after refecding, the lipogencsis of ad libitum group was significantly (p<0.05) lower than that of control group. The lipogenesis of 40% food restricted group during refeeding was significantly (p<0.05) higher than that of conrtol group. Ad libitum group and 20% food restricted group during refecding allowed heparin-releasable (HR) LPL or total extractable (TE) LPL activity to return to control states. 40% food restricted group during refeeding delayed the return of HR-LOL or TE-LPL activity to return to control states until day 21 of refeeding. These results suggest that food restriction during refeeding can partially or completely prevent the overshoot of LPL activity, and this may influence the rate of lipid accumulation in adipose tissue during refeeding.

  • In order to observe the K contents, Korean common foods, especially processed foods were analyzed by Atomic Absorption Spectrophotometer. The results obtained were summarized as followed; (I) The K contents of instant noodle(ramyon) was 105-180 mg/100g and K ontents of their soup powder was 500-1000 mg/100g. (2) The K contents of potato products and bean products were less than those of original foods. So it was recongnized that the K contents might reduced during the food process. (3) The K contents of meats was 130-475 mg/I00g and those of meats products was relatively low, 80-260 mg/100g. (4) The K contents of carbonate beverage was very low but that of instant coffed was very high, 2400-2600 mg/100g. (5) There was no Statistical difference between the results of wet ash method and dry ash method in the K contents of all food groups.

Effect of Steam Distillates Prepared from Herbal Medicines on Immunostimulating Activity (생약으로부터 조제된 수증기 증류물의 면역활성)

  • Of hot- water extracts prepared from 30 kinds of herbal medicines, Acanthopanax senticosus (75.6% inhibition of control), Atractylodes macrocephale (71.3%), Panax ginseng (70.0%), Glycyrrhiza uralensis (66.3%) and Angelica acutiloba (63.1%) showed the potent tumor metastasis inhibition activity against colon 26-M3.1 lung carcinoma at 2.5 mg/kg body weight, whereas the other extracts had a little activity, except for Pueraria thunbergiana (58.6%) and C. leticulata (54.9%) having the intermediate activity. We also found that Citrus leticulata (1.80-fold of control), A. macrocephale (1.73-fold), A. senticosus and G. uralensis (1.64-fold) enhanced on Peyer's patch cells mediated-hematopoietic response at 100 $\mu\textrm{g}$/mL. In addition, these active herbal medicines were prepared into steam distillates to improve the food rheology as beverage, and to remove the inactive components. Among these steam distillates, A. macrocephale, G. uralensis and A. senticosus showed the significant tumor metastasis inhibition activity at 2.5 mg/kg body weight (58.7%, 50.3% and 41.9%, respectively), and A. macrocephale had the potent activity even at 0,25 mg/kg body weight (49.7%). In treatments of steam distillates with Peyer's patch cells, A. macrocephale and A. senticosus significantly increased the bone marrow cell proliferation even at 10 $\mu\textrm{g}$/mL (1.49- and 1.28-fold of control). Although steam distillates had lower activity than hot-water extracts, herbal medicines, such as A. macrocephale and A. senticosus, showed the high immunostimulating activity in hot-water extracts as well as steam distillates. Therefore, these results assumed the possibility that steam distillates from herbal medicines might be utilized to food industry for beverage.

Optimization of γ-Aminobutyric Acid Production Using Lactobacillus brevis spp. in Darae Sap (Lactobacillus brevis 균주를 이용한 다래 수액에서의 감마아미노뷰티르산 (γ-Aminobutyric Acid) 생산 최적화)

  • This study was performed to increase the production of ${\gamma}$-aminobutyric acid (GABA) by lactic acid bacteria (Lactobacillus brevis CFM11) and manufacture an optimum medium using the sap from Darae (Actinidia arguta). The concentration of GABA in the fermented sap was determined using GABase enzymatic assay. The isolated L. brevis CFM11 produced $605.67{\mu}g/mL$ GABA after incubation for 24 hours at $37^{\circ}C$ in broth. The sap was fermented by L. brevis CFM11 under optimum conditions of $32^{\circ}C$ for 48 hours with 40% rice bran extract, 1.0% sucrose, 3.0% soytone, 0.2% magnesium sulfate, and 0.2% MSG. The fermented sap produced a concentration of $1366.13{\mu}g/mL$ GABA. These results demonstrate that fermenting Darae sap using L. brevis CFM11 can produce a fermented sap beverage with increased GABA content.

Effect of Oral Sport Beverages with Medicinal Herbs Added on Short-term Recovery from Exercise-induced Fatigue (한의약소재 스포츠음료수 섭취가 운동-유발성 피로의 단시간 회복에 미치는 영향)

  • Objectives : Ginseng Research Group in Korea Food Research Institute developed Saeng Mac San (KFRI-2)and Je Ho Tang (KFRI-3) with their sensory factors more acceptable. And we examined their effects on the short-term recovery capacity for cycling exercise (EX) maintained to all-out. Methods : Seven healthy young subjects (aged $24.0{\pm}2.1yr$) were volunteered at this double blind test. Each of KFRI-2, 3, a commercial sport beverage and control (CON) was offered randomly on a series of EX protocol including 65% VO2max-90min EX (D-ride). 1h-recovery and 85% VO2max EX to all-out (P-ride) under the control of their heart rate (HR) and rating perception of exertion (RPE). Blood samples were collected before D-ride, 30, 60 and 90min in D-ride, 30 and 60min in the recovery period and each 10min in P-ride. Plasma analysis items were glucose, insulin, cortisol (CORT), testosterone (TEST), free fatty acid (FFA), $Na^+$, Cl-and $K^+$. The collected data (Means${\pm}$SE) were analysed by two-way ANOVA and statistically significant differences between treatments (p<0.05) by LSD.; the significant level in FFA, $Na^+$, Cl-and $Na^+$ was p<0.01 Results : At 30min during recovery. plasma glucose level in KFRI-3 was significantly higher than CON, and also insulin in KFRI-3 was than CON and KFRI-2. FFA in KFRI-3 was significantly lower than CON during recovery. $Na^+$ in KFRI-3 significantly higher than CON at 90min in D-ride, and also KFRI-2 was at 60min during recovery. However CORT, TEST, Cl-and $Na^+$ in treated beverages were not significant. KFRI-2, 3 elevated the time for P-ride more than CON did. Conclusions : KFRI-2, 3 elevated the time for P-ride about 12% more than CON did. It is based on rapid recovery of plasma glucose level and inhibition of lipolysis during recovery.
