The Effects of Supplementation of Ascidian Tunic Shell into Laying Hen Diet on Egg Quality (산란계 사료에서 우렁쉥이껍질 첨가가 계란 품질에 미치는 영향)

  • This study was conducted to evaluate the feeding value of ascidian tunic shell the effects of its dietary supplementation on laying performance, egg-yolk pigmentation, egg-shell strength and egg taurine content. A total of 168 brown layers at the age of 29wks in commercial cage were fed for 4 wks with 7 different diets containing ascidian tunic shel1(AST) at varying levels of 0$\sim$5% Dm or 0% AST with 100ppm carophyll red. No differences were found in egg production and weight among the treatments indicating that ascidian tunic shell did not adversely affect the laying performances. Adding the ascidian tunic shell to the diets increased egg-yolk pigmentation compared to the control and resulted in simillar or better effect on egg-yolk pigmentation compared to 100ppm carophyll red. The data suggest that ascidian tunic shell may be used as feed ingredients in layer diet enrichment of egg-yolk pigmentation in the place of carophyll red(chemical pigment). Specific gravity and breaking strength of egg shell were significantly increased by the adding ascidian tunic shell to the diet, suggesting that ascidian tunic shell may be used as feed ingredients for increasing egg shell strength. Also taurine content of egg was significantly increased with increasing supplementation of ascidian tunic shell to the diet(p<0.05). Therefore, ascidian tunic shell may be used as feed ingredients in laying hen diet to improve egg quality such as egg-yolk pigmentation, egg-shell strength and egg taurine enrichment.

Effect of Monascus-fermentation on the Content of GABA and Free Amino Acids in Soybean (홍국균 발효가 콩의 GABA와 유리아미노산 함량에 미치는 영향)

  • The changes in the contents of GABA ($\gamma$-aminobutyric acid) and free amino acids (FAA) were analysed by HPLC in soybean fermented with Monascus pilosus IFO 480, which showed the highest GABA-producing ability among the five different strains. The significant increase (p<0.05) of GABA was observed in Monascusfermented soybean (MFS) with an average 4.5-fold increase of FAA. The contents of GABA (78.5 mg /100 g dry weight, dw) and FAA (5458.5 mg /100 g dw) were enhanced in MFS compared with unfermented soybean (31.5 mg /100 g dw, 1634.0 mg/100 g dw, respectively) during 30 days fermentation at $30^{\circ}C$. Acidic amino acids (i.e., glutamic acid and aspartic acid) in FAA files were found in large quantities (837.0 mg /100 g dw, 580.5 mg /100 g dw, respectively). Moreover, the sum of essential amino acids of MFS (1681.5 mg /100 g dw) at 20 days fermentation increased by 3.6 times compared with that of the control (469.0 mg /100 g dw). The results indicate that Monascus-fermentation have great potential for the enrichment of GABA and free amino acids in soybean.

Screening of the liver, serum, and urine of piglets fed zearalenone using a NMR-based metabolomic approach

  • Zearalenone (ZEN), a mycotoxin produced by Fusarium in food and feed, causes serious damage to the health of humans and livestock. Therefore, we compared the metabolomic profiles in the liver, serum, and urine of piglets fed a ZEN-contaminated diet using proton nuclear magnetic resonance ($^1H-NMR$) spectroscopy. The spectra from the three different samples, treated with ZEN concentrations of 0.8 mg/kg for 4 weeks, were aligned and identified using MATLAB. The aligned data were subjected to discriminating analysis using multivariate statistical analysis and a web server for metabolite set enrichment analysis. The ZEN-exposed groups were almost separated in the three different samples. Metabolic analysis showed that 28, 29, and 20 metabolites were profiled in the liver, serum, and urine, respectively. The discriminating analysis showed that the alanine, arginine, choline, and glucose concentrations were increased in the liver. Phenylalanine and tyrosine metabolites showed high concentrations in serum, whereas valine showed a low concentration. In addition, the formate levels were increased in the ZEN-treated urine. For the integrated analysis, glucose, lactate, taurine, glycine, alanine, glutamate, glutamine, and creatine from orthogonal partial least squares discriminant analysis (OPLS-DA) were potential compounds for the discriminating analysis. In conclusion, our findings suggest that potential biomarker compounds can provide a better understanding on how ZEN contaminated feed in swine affects the liver, serum, and urine.

Gene Co-expression Analysis to Characterize Genes Related to Marbling Trait in Hanwoo (Korean) Cattle

  • Marbling (intramuscular fat) is an important trait that affects meat quality and is a casual factor determining the price of beef in the Korean beef market. It is a complex trait and has many biological pathways related to muscle and fat. There is a need to identify functional modules or genes related to marbling traits and investigate their relationships through a weighted gene co-expression network analysis based on the system level. Therefore, we investigated the co-expression relationships of genes related to the 'marbling score' trait and systemically analyzed the network topology in Hanwoo (Korean cattle). As a result, we determined 3 modules (gene groups) that showed statistically significant results for marbling score. In particular, one module (denoted as red) has a statistically significant result for marbling score (p = 0.008) and intramuscular fat (p = 0.02) and water capacity (p = 0.006). From functional enrichment and relationship analysis of the red module, the pathway hub genes (IL6, CHRNE, RB1, INHBA and NPPA) have a direct interaction relationship and share the biological functions related to fat or muscle, such as adipogenesis or muscle growth. This is the first gene network study with m.logissimus in Hanwoo to observe co-expression patterns in divergent marbling phenotypes. It may provide insights into the functional mechanisms of the marbling trait.

Growth of Larval Rockfish Sebastes schlegeli Fed Moina macrocopa (물벼룩(Moina macrocopa) 급여가 조피볼락(Sebastes schlegeli) 자어의 성장에 미치는 영향)

  • Several our studies have focused recently on the mass production of the freshwater Cladoceran Moina macrocopa which can substitute Artemia nauplii for the culture of larval marin fish. A 6 weeks experiment was conducted to investigate the effects of enrichment on the fatty acid composition of Moina macrocopa through feeding Schizochytrium sp. containing highly unsaturated fatty acids and to study the impacts of n-3-HUFA enriched Moina on improving survival rate and fatty acid compostion of larval rockfish Sebastes schlegeli. After feeding for 6 weeks, the Moina-fed fry resulted in a higher survival rate of 99.2% compared to the Artemia-fed fry 12.8%. In addition, the Moina-fed fry had the fast growth rate 45.6mm compare to the Artemia-fed fry 25.7 mm at the end of the experiment. The Moina-fed fry showed significantly higher level of 16.47% DHA than their Artemia-fed fry counterparts of the level of 3.97% with respect to DHA. PL, the cell membrane components in living food organisms, constituted 63.8% of the Moina, which was significantly higher than in the 40.1% of the Artemia. The present study indicate that Moina macrocopa can be used as Artemia substitute and improving the survival rate rockfish larvae through enchriment Schizochytrium sp.

Plant Molecular Farming Using Oleosin Partitioning Technology in Oilseeds

  • Plant seed oil-bodies or oleosomes ate the repository of the neutral lipid stored in seeds. These organelles in many oilseeds may comprise half of the total cellular volume. Oleosomes are surrounded by a half-unit membrane of phospholipid into which are embedded proteins called oleosins. Oleosins are present at high density on the oil-body surface and after storage proteins comprise the most abundant proteins in oilseeds. Oleosins are specifically targeted and anchored to oil-bodies after co-translation on the ER. It has been shown that the amino-acid sequences responsible for this unique targeting reside primarily in the central hydrophobic tore of the oleosin polypeptide. In addition, a signal-like sequence is found near the junction of the hydrophobic domain and ann N-terminal hydrophilic / amphipathic domain. This "signal" which is uncleaved is also essential for correct targeting. Oil-bodies and their associated oleosins may be recovered by floatation centrifugation of aqueous seed extracts. This simple partitioning step results in a dramatic enrichment for oleosins in the oil-body fraction. In the light of these properties, we reasoned that it would be feasible to create fusion proteins on oil-bodies comprising oleosins and an additional valuable protein of pharmaceutical or industrial interest. It was further postulated that if these proteins were displayed on the outer surface of oil-bodies, it would be possible to release them from the purified oil-bodies using chemical or proteolytic cleavage. This could result in a simple means of recovering high-value protein from seeds at a significant (i.e. commercial) scale. This procedure has been successfully reduced to practice for a wide variety of proteins of therapeutic, industrial and food no. The utillity of the method will be discussed using a blood anticoagulant, hirudin, and industrial enzymes as key examples.

Taurine Enrichment of Eggs with Feather Meal and Pyridoxine

  • The effects of dietary supplementation of feather meal (FM) and pyridoxine ($B_6$) on the taurine content of egg yolk and performance of laying hens were investigated. A feeding trial was conducted in nine hundred 31-wk-old $Hy-Line^{\circledR}$ Brown layers over 4 wk. The hens received 6 dietary treatments: Control, FM 3% supplemented diet (FM 3%), FM 3%+$B_6$ supplemented diet (FM 3%+$B_6$), FM 6% supplemented diet (FM 6%), FM 6%+$B_6$ supplemented diet (FM 6%+$B_6$), and synthetic taurine 0.25% supplemented diet (Taurine). Parameters of production were significantly (p<0.05) affected by treatments. The egg production of hens fed FM 3% was the highest and hens fed FM diets were more productive than the Taurine and Control groups. The egg weights of the Taurine group were significantly lower than those of the FM 3% and FM 6% groups, but not significantly different from those of other treatments. The feed intake of the Control group was highest among all groups. The feed conversion ratio of the Control group was higher than in groups receiving other treatments of which FM 6% was the lowest. The broken egg production of the Taurine group was highest, while that of the Control group was lowest among treatments. The taurine content of egg yolk was significantly (p<0.01) increased by supplementation of taurine (64.7%), FM 6%+$B_6$ (57%), FM 3%+$B_6$ (32.1%), and FM 6% (16.6%) over a 4 wk average. Sensory evaluation data of the Taurine group showed the highest score in all of the sensory attributes and those of other treatments were not significantly (p<0.05) different. In conclusion, taurine can be enriched in egg yolk by supplementation of 6% FM and $B_6$, as well as 0.25% synthetic taurine.

Isolation and Characterization of 3,4-Dichloroaniline Degrading Bacteria from a Sandbank (갯벌에서 분리한 3,4-Dichloroaniline 분해 미생물의 특성)

  • The compound 3,4-dichloroaniline (DCA) is an aromatic amine used as an intermediate product in the synthesis of herbicides, azo-dyes and harmaceuticals. It is also a degradation product of some herbicides (diuron, propanil, and linuron) and of trichlorocarbanilide, a chemical used as active agent in the cosmetic industry. 3,4-DCA, however, is considered potential pollutants due to their toxic and recalcitrant properties to humans and other species. A bacterium capable of growth on 3,4-DCA was isolated by dilution method from 3,4-DCA-containing enrichment culture. Finally, a strain, YM-14, capable of degrading efficiently 3,4-DCA was isolated from a sandbank. The isolated strain, YM-14 was identified to be Arthrobacter sp.. Fifty ppm 3,4-DCA in 1/10 LB media was completely degraded by the growth of Arthrobacter sp. YM-14 for 12 h at $30^{\circ}C$. The isolated strain is capable of growth on 3,4-DCA as sole carbon source and also able to degrade other chloroaniline compounds. Also, the isolated strain showed high level of catechol 1,2-dioxygenase activity by 3,4-DCA exposure. The catechol 1,2-dioxygenase was supposed to be ones of the important factors for 3,4-DCA degradation.

Production and Properties of Hemicellulases by a Cellulosimicrobium sp. Isolate (Cellulosimicrobium sp. 분리균의 Hemicellulases 생산성과 효소특성)

  • A bacterial strain capable of hydrolyzing xylan and locust bean gum (LBG) was isolated from farm soil by enrichment culture using mixture of palm kernel meal (PKM) and wheat bran as carbon source. Nucleotide sequence of 16S rDNA amplified from the isolate YB-1107 showed high similarity with those of genus Cellulosimicrobium strains. Xylanase productivity was increased when the Cellulosimicrobium sp. YB-1107 was grown in the presence of wheat bran or oat spelt xylan, while mannanase productivity was increased drastically when grown in the presence of PKM or LBG. Particularly, maximum mannanase and xylanase activities were obtained in the culture filtrate of media containing 0.7% PKM or 1% wheat bran, respectively. Both enzyme activities were produced at stationary growth phase. Mannanase from the culture filtrate showed the highest activity at $55^{\circ}C$ and pH 6.5. Xylanase activity was optimal at $65^{\circ}C$ and pH 5.5. The predominant products resulting from the mannanase or xylanase hydrolysis were oligosaccharides for LBG or xylan, respectively. In addition, the enzymes could hydrolyze wheat bran and rice bran into oligosaccharides.

Development and characterization of nine microsatellite loci from the Korean hare (Lepus coreanus) and genetic diversity in South Korea

  • The Korean hare, Lepus coreanus, is an important mammal in ecosystem food chains, and is distributed across the entire Korean peninsula and northeastern China. Polymorphic microsatellite loci were developed using the biotinenrichment technique for use in population genetics studies. Five trinucleotide and four dinucleotide microsatellite loci were selected and tested on 22 Korean hare specimens collected from Gangwon Province and Gyeongsangbuk Province in South Korea. The number of alleles across the two sampling regions ranged from three to nine with a mean of 6.1. Mean observed and expected heterozygosities and polymorphic information content were 0.540, 0.627 and 0.579, respectively. Only one locus, Lc06, showed departure from Hardy-Weinberg equilibrium after applying the Bonferroni correction. Four microsatellites, Lc01, Lc03, Lc12, and Lc19, satisfied the criteria to serve as a core set of markers recommended for population genetics studies. These new microsatellite markers will be widely applicable to future genetic studies for management and conservation of the Korean hare and related species, including assessment of the genetic diversity and population structure of L. coreanus.
