Expression of Recombinant Human Follicle-stimulating Hormone in the Chinese Hamster Ovary Cell

  • As an preliminary experiment for making transgenic animals producing human follicle stimulating hormone (hFSH), we tried to express recombinant hFSH gene in vitro. hFSH is a heterodimeric glycoprotein hormone produced in the anterior pituitary gland. The hormone is essential in the regulation of reproductive processes, such as follicular development and ovulation. Genes encoding the common gonadotrophin alpha subunit and FSH-specific beta subunit were inserted into retroviral vectors under the control of the rat beta actin promoter. Gene transfer to the Chinese hamster ovary (CHO) cells was done by infection of the retroviruses harvested from PT67 packaging cells transfected with recombinant retrovirus vector DNA. After selection with G4l8, PCR and RT-PCR analyses of the G4l8-resistant CHO cells showed successful transfer and expression of both ${\alpha}$ and ${\beta}$ fragments of the FSH gene.

  • This study was carried out to investigate the ultrastructural changes of ooplasm and follicular membrane of oocytes, obtained from 150 of 3-year-old female rainbow trout(Oncorhynchus mykiss). All data were collected from March in 1989 to February in 1990, and from August to October in 1991. The size of the nucleoli and number of the yolk granules increased as the oocyte growed. Yolk granules were deposited in the oocyte as crystalline granules. Due to the presence of large early and late maturing oocytes, their ovaires were enlarged, transparent and granular. The lattice was broken down at hydration, leaving the egg transparent. As thepercentages of fish in LPO and EMO stage increased from September to October, Mean GSI values increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in bothgonadosomatic index and macroscopic apearance. Under the natural conditions,t he ovarian follicle influences the histological development and periodical secretion of the hormones, sufficient for a oogenesis and gonadal steroid production. The electrophoretic pattern of major band in mature stage was much thicker(70∼110k dalton) than that in previtellogenic phase.

  • A observation of the ovarian development and oogenesis of Pieris rapae Linne has been carried out during metamorphosis using stereo-microscope, light microscope and electron microscope. The results obtained through this experiment are as follows: 1. The ovarian development and vitellogenesis begin at the 3-day old pupa and the 6-day old pupa respectively, and the adult ovary right after their emergence contains a few mature eggs. 2. The species described above are further observed at six different stages in oogenesis, and the results are summarized as follows. 1) Pieris rapae has polytrophic ovarioles. The cell organelles of the nurse cells are transfered to the oocyte through the ring canal at the early oogenesis. 2) At stage 2, the nuclear envelope of oocyte nucleus is less infolding than that of nurse cell nucleus. In the oocyte cytoplasm a large number of ribosomes are observed. 3) At stage 3 and 4, many micropinocytotic vesicles are observed in the oocyte cytoplasm. These vesicles are fused together to form large proteid yolks. 4) At stage 5, the vitelline membrane is laid down in the intercellular space between the follicle cells and oocyte. 5) At stage 6, the chorion is formed by the follicle cells. 6) A micropyle and a number of aeropyle are observed on the surface of a mature egg.

Establishment and Characterization of Immortalized Human Dermal Papilla Cells Expressing Human Papillomavirus 16 E6/E7

  • Primary human dermal papilla cells (HDPCs) are often preferred in studies on hair growth and regeneration. However, primary HDPCs are limited by their reduced proliferative capacity, decreased hair induction potential, and extended doubling times at higher passages. To overcome these limitations, pTARGET vectors containing human papillomavirus16 (HPV16) E6/E7 oncogenes were transfected into HDPCs and selected using G-148 to generate immortalized cells here. HPV16 E6/E7 oncogenes were efficiently transfected into primary HDPCs. Immortalized HDPC showed higher proliferative activity than primary HDPC, confirming an increased proliferation rate. Expression of p53 and pRb proteins was downregulated by E6 and E7, respectively. E6/E7 expressing HDPC cells revealed that cyclin-dependent kinase (CDK) inhibitor p21 expression was decreased, while cell cycle-related genes and proteins (CDK2 and cyclin E) and E2F family genes were upregulated. Immortalized HDPCs maintained their responsiveness to Wnt/β-catenin pathway and hair follicle formation capability, as indicated by their aggregative properties and stemness. E6/E7 immortalized HDPCs may facilitate in vitro hair growth and regeneration studies.

Developmental Competence of Intrafollicular Oocytes Derived from Preantral Follicle Culture with Different Protocols after Parthenogenetic Activation

  • This study was conducted to improve efficiency of a follicle culture system without reducing developmental competence of intrafollicular oocytes. Preantral follicles (100 to $125{\mu}m$ in diameter) of F1 hybrid (B6CBAF1) mice were cultured singly for 216 h in modified ${\alpha}$-MEM-glutamax medium, to which 2.5 IU/ml hCG and epidermal growth factor was added 16 h prior to the end of culture. Medium change was either performed three times (54 h interval), twice (72 h interval), once (108 h interval), or not at all (216 h interval). Maturation (progression to the metaphase II stage) of intrafollicular oocytes was detected from 4 days after culture in the three-times change treatment, while all treatments yielded mature oocytes from day 5 of culture. Compared with the three-times change, decreasing the change frequency to once did not reduce the capacity to begin maturation (germinal vesicle breakdown of 82 to 86%), to mature (78 to 79%) and to develop into blastocysts after parthenogenetic activation (29 to 32%). Morphological parameters were similar among these treatments. Except for the no medium change treatment, similar colony-forming activity of inner cell mass cells after culturing of blastocysts in leukemia inhibitory factor-containing medium was detected, while the morphology of the colony-forming cells deteriorated in the change-once treatment compared with the change twice or three-times. In conclusion, the efficiency of the preantral follicle culture system could be improved by reducing frequency of medium change up to a 72 h interval (three times in total 216 h culture) without decreasing developmental competence of oocytes.

  • The localization of worm antigen of Neodiplostomum seoulere was examined by immuno-electronmicroscopic observation. Not only the immunized serum of mice with crude worm extract of N. seoulene but also serum of infected mouse were reacted to the worm section. Using immunized serum as primary antibody. the gold particles were deposited on the rough endoplasmic reticulum of the cell of tribocytic organ, spermatozoa in the seminal vesicle, microvilli of the caecum and vitelline follicle. Using infected serum, gold particles were deposited only on the vitelline follicle prominently. This finding suggested that the tribocytic organ, seminal vesicle, caeca and vitelline follicles may play a role of antigen to immunized serum with crude worm extract of N. seoulense, whereas the vitelline follicle, to the infected serum.

Deficiency of Formyl Peptide Receptor 2 Retards Hair Regeneration by Modulating the Activation of Hair Follicle Stem Cells and Dermal Papilla Cells in Mice

  • Hair loss is one of the most common chronic diseases, with a detrimental effect on a patient's psychosocial life. Hair loss results from damage to the hair follicle (HF) and/or hair regeneration cycle. Various damaging factors, such as hereditary, inflammation, and aging, impair hair regeneration by inhibiting the activation of hair follicle stem cells (HFSCs) and dermal papilla cells (DPCs). Formyl peptide receptor 2 (FPR2) regulates the inflammatory response and the activity of various types of stem cells, and has recently been reported to have a protective effect on hair loss. Given that stem cell activity is the driving force for hair regeneration, we hypothesized that FPR2 influences hair regeneration by mediating HFSC activity. To prove this hypothesis, we investigated the role of FPR2 in hair regeneration using Fpr2 knockout (KO) mice. Fpr2 KO mice were found to have excessive hair loss and abnormal HF structures and skin layer construction compared to wild-type (WT) mice. The levels of Sonic hedgehog (Shh) and β-catenin, which promote HF regeneration, were significantly decreased, and the expression of bone morphogenetic protein (Bmp)2/4, an inhibitor of the anagen phase, was significantly increased in Fpr2 KO mice compared to WT mice. The proliferation of HFSCs and DPCs was significantly lower in Fpr2 KO mice than in WT mice. These findings demonstrate that FPR2 impacts signaling molecules that regulate HF regeneration, and is involved in the proliferation of HFSCs and DPCs, exerting a protective effect on hair loss.

Effects of Adrenalectomy on Sexual Gland, Thyroid Gland and Serum Components in Rats (흰쥐에 있어서 부신척출이 성선, 갑상선 및 혈청성분에 미치는 영향에 관한 연구)

  • This study aimed to determine the effects of adrenalectomy on the sexual gland, the thyroid gland and the serum components. A total of 192 Wister strain albino rats were evenly divided into 4 sexually equal groups for the comparisons. Each groups was divided into a control and a treatment group by sexuality. Each tissue of the sexual gland and the thyroid gland was microscopically examined, and at the same time body weight and serum components were also examined on the 1st, 7th, 14th, 28th, 42nd, 56th, 70th and 84th day respectively following adrenalectomy. The results obtained were as follows: 1. The body weight was sufficiently retarded in decreasingly after the adrenalectomy as compared to that of control. 2. The histological change of testis started atrophy of spermatogonia and degeneration of spermatocyte. There was a, pp.ared no cell division activity. Spermatozoa in the seminiferous tuble was not noticed but degeneration of interstitial cell was started and spermatozoa in the epididymal duct disa, pp.ared. 3. Degeneration of oocyte and follicular cell was noticed as the histological change of ovary. There was not a, pp.arance of primary follicle and corpus Iuteum but interstitial cell was proliferated. 4. For the effect of adrenalectomy on the histological change of the thyroid gland, the number of small follicle decreased and that of large follicle increased as the time passed following the adrenalectomy. And the follicular epithelial cell became squamous as typical condition of functional decreased. 5. It was shown that in the total contents of serum protein no difference with control occurred with in the 70th day for male and female adrenalectomized, respectively. But the differences in protein contents were significanly decreased on the 70th day. 6. No difference occurred in the total serum lipids on the 7th day, but they decreased significantly on the 42nd day, and (P<0.01) on the 56th, and 70th day. A tendency to decrease was noted as time elapsed following adrenalectomy. 7. Increase and decrease were intercrossed in the serum cholesterol contents between the control and th treatment groups on the 28th day. But the difference significantly decreased on the 42nd, 56th, and 70th day after adrelalectomized. The contents showed tendency to decrease as time elapsed following adrenalectomy. 8. The blood glucose contents rapidly decreased with time. The difference were significant on the 28th and 42nd day, and highly significant on the 56th and 70th day for male adrenalectomized. 9. In case of solium, there was no noticeable sexual distinction. There was slight in creasing or decreasing for the control group. But the treatment group tended continuously to decrease for male and female. 10. It was shown that potassium contents tended to increase on the 28th day for male and female, but the differences were small. 11. As for chlorine, it tended to decrease rapidly on 7th day for male and female adrenalectomized, and the tendency continued.

A Case of Trichilemmal Carcinoma in Auricle (이개에 발생한 모낭암종 1예)

  • Trichilemmal carcinoma is a rare malignant neoplasm of the hair follicle from the outer root of the hair follicle sheath. This tumor can be misleading, and a false diagnosis of a squamous cell carcinoma. We report a case of trichilemmal carcinoma with a review of literature. The patient presented with an exophytic well circumscribed nodular mass on the left auricle, which was detected 6 months ago. Histopathologically, the tumor consisted of atypical clear cells which contained abundant glycogen. The tumor cells shows lobular growth pattern with necrosis, foci of trichilemmal keratinization and peripheral pallisading. Total excision and repair with full-thickness skin graft was done with minimal surgical morbidity. The patient has been free of recurrence or metastasis for 8 months.

  • Uptake and accumulation of vitellogenin into ovary were traced by radiolabeled vitellogenin produced in the fat body. Vitellogenin reached the oocytes through the follicle cells and the intercellular space and penetrated the oocyte membrane via the endocytotic vesicles and then accumulated to the growing yolk granules.
