황기의 저밀도지질단백질 (LDL)산화에 미치는 영향 (Effect of Astragali Radix on Low Density Lipoprotein Oxidation)

  • The root of Astragalus membranaceus Bunge (Leguminosae), which has been used for the treatment of hypertension, chronic hepatitis, duodenal ulcers, chronic nephritis and promotion of immunity in folk remedies. Several lines of evidence indicate that oxidative modification of low-density lipoprotein (Ox-LDL) may play an important role in atherogenesis. Hence, the role of antioxidants in the prevention of LDL oxidation needs to be determined. To investigate the antioxidant activity. we determined the MeOH ex. and fractions of Astragali Radix on the inhibition of LDL oxidation. The CH$_2$C1$_2$ and EtOAc orations inhibited the oxidative modification of LDL by a decrease in the lipid peroxide content and the electrophoretic mobility of LDL. Calycosin-7-0-$\beta$-D -glucoside which was isolated from EtOAc fraction inhibits the oxidative modification of LDL.

  • Cyperus rotundus L. (Cyperaceae) has been used as an analgesic and antiiflammatory agent and in the treatment of menstrual disorder in folk remedies. Cyperi Rhizomata, processed and unprocessed, were extracted with methanol and fractionated with petroleum ether, chloroform. Butanol, water. The effect of unprocessed Cyperi Rhizoma and processed Cyperi Rhizoma on $CCl_4$ induced lipid peroxidation and hepatotoxity have been tested in rats. BuOH. Water fractions of unprocessed Cyperi Rhizome enhanced the inhibition of antilipid peroxidative effects in liver lipid. In chemical parameters obtained from serum analysis. Butanol fraction of unprocessed Cyperi Rhizoma showed significant decrease in hepatotoxicity. In result, unprocessed Cyperi rhizoma has significant antilipid peroxidative effect and hepatoprotective activity.

  • Symplocarpus renifolius, a folk medicinal herb has been used for treatment of hypertension, rheumatis, tetanus, ringworm, cough and epilepsy in north and middle part of Korea. The extracts from the root of S. renifolius were evaluated for antibacterial, antihypertensive and analgesic activities. The crude extract of the root of S. renifolius showed antibacterial activity against Gram(+) bacteria and dose dependantly decrease the blood pressure of SHR. The chloroform extract of the roots of S. renifolius was also exibits analgesic action in mice.

  • Saururus chinensis Baill (Saururaceae) is a perennial plant that has been used in the treatment of edema, jaundice and gonorrhea in Korean folk medicine. This study was carried out to investigate the inhibitive effects of Saururus chinensis Baill (SCB) on lipid metabolism in Sprague-DaweIy rat(SD-rat) accutely exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD). After 7 days from TCDD(1$\mu$g/kg) injection, SCB(200mg/kg) was administered into rats intraperitoneally for 4 weeks. (omitted)

  • Ternstroemia japonica (Theaceae) is widely distributed in Korea, Japan, Taiwan and China. The tree is a useful source of lumber, dye and horticulture. Its fruits have been used as folk medicine in Japan for the treatment of chest pain or numbness. Previously, we have isolated saponins and jacaranone derivatives from the fruits. In our continuous study on the same plant, the leaves of Ternstroemia japonica were extracted with MeOH and the MeOH extract was fractionated with solvents. The n-BuOH soluble fraction was separated by repeated column chromatographies on silica gel and Sephadex LH-20, and further purified by reversed phase HPLC. (omitted)

  • Styrax japonica Sieb. et Zucc. (Styracaceae) is a deciduous tree growing in Korea, Japan, and China. The pericarps of this plant have been used as a folk medicine for treatment of cough. Jegosaponins, deacyl jegosaponins and benzofurans have been reported from the fruits and seeds of this plant, and these compounds have been shown antisweet and cytotoxic activities. As a part of a research aimed at the discovery of biological active compounds from plant sources, we have studied a chemical constituent of the stem-bark of S. japonica. (omitted)

  • As part of our systematic study for Korean Compositae plants, we have studied Saussurea nutans, collected at Gangwon Province on August 2001. Phytochemical studies on various species of genus Saussurea have resulted in the isolation sesquiterpene, triterpene and flavonoid$\^$1)/. S. nutans has been used for the treatment of rheumatic arthritis and dysmenorrhea in the Chinese folk medicine$\^$2)/. However, chemical constituents of this plant have not been reported until now. The MeOH extract of the aerial parts of this source was solvent fractionated into n-hexane, methylene chloride, ethyl acetate and BuOH soluble portions. (omitted)

  • Hibiscus sabdariffa L., a tropical beverage material, is used commonly as in folk medicine against hypertension, pyrexia, inflammation, liver disorders, and obesity. However, the mechanism by which Hibiscus sabdariffa L. modulates adipogenic differentiation is remained to be elusive. This report was designed to investigate the inhibitory effect of Hibiscus extract on insulin signaling pathway during adipocyte differentiation in 3T3-L1 preadipocytes. 3T3-L1 preadipocytes were differentiated with isobutylmethylxanthine, dexamethasone, and insulin (MDI) and followed by the addition of Hibiscus extract. Treatment with Hibiscus resulted in a decrease of lipid droplet accumulation, which was suppressed by PI-3 kinase inhibitor wortmannin in 3T3-L1 preadipocytes. Also, Hibiscus extract markedly attenuated the mRNA expression of adipogenic transcriptional factor PPAR${\gamma}$ and adipogenic hormon Leptin during adipogenesis. However, it did not affect the expression of adiponectin in 3T3-L1 preadipocytes differentiated with MDI mixture. Furthermore, Adipogenic differentiation by MDI mixture increased the phosphorylation and expression of PI3-Kinase and Akt in 3T3 preadipocytes, which was markedly suppressed by Hibiscus extract treatment. Taken together, our results suggest that Hibiscus extract suppressed the adipogenic differentiation of 3T3 preadipocytes through activation of PI3-Kinase and Akt signaling pathway.

황기의 COX-2 활성 억제 효과 (Inhibitory effect of Astragali Radix on COX-2 activity)

  • The root of Astragalus membranaceus Bunge (Leguminosae), which has been used for the treatment of hypertension, chronic hepatitis, duodenal ulcers, chronic nephritis and promotion of immunity in folk remedies. Cyclooxygenase (COX-2) is responsible for the production of large amounts of proinflammatory prostaglandins (PGs) at the inflammatory site. Thus, a logical approach to the treatment of inflammatory disease should involve the inhibitors of COX-2. To develop new COX-2 inhibitors from natural products, Astragali Radix was screened by inhibiting prostaglandin $E_2(PGE_2)$ generation in the culture medium using enzyme immunometric assay. Two isoflavone glycosides, $7,2'-dihydroxy-3',4'-dimethoxyisoflavan-7-O-{\beta}-D-glucoside$ and $calycosin-7-O-{\beta}-D-glucoside$ isolated from Astragali Radix inhibited COX-2 activity.

박물관에서의 저산소 농도를 이용한 살충 기술 (Anoxia Techniques to Eradicate Insects for Conservation of Cultural Properties in Museums)

  • 박물관에서 질소나 아르곤과 같은 불활성 가스를 사용한 저산소 농도 살충처리법은 메틸브로마이드나 유독성 살충제의 대체법으로써 성공적으로 사용되어 왔다. 저산소 농도 살충법의 살충 효과는 해충의 종, 산소 농도, 온도, 습도, 가스의 종류 등에 따라 영향을 받으며, 박물관에서의 문화재 보존 환경(온도 $15{\sim}25^{\circ}C$, 습도 40~60%)에서 산소 농도를 0.03% 이하로 유지함으로써 1개월 이내에 가장 내성이 강한 문화재 해충의 살충이 가능하다. 그리고 살충 처리 대상 문화재의 수량이나 크기에 따라 소형 중형 백, 대형 텐트, 버블, 챔버 등 다양한 시스템을 적용할 수 있다.
