Accurate Metabolic Flux Analysis through Data Reconciliation of Isotope Balance-Based Data

  • Various techniques and strategies have been developed for the identification of intracellular metabolic conditions, and among them, isotope balance-based flux analysis with gas chromatography/mass spectrometry (GC/ MS) has recently become popular. Even though isotope balance-based flux analysis allows a more accurate estimation of intracellular fluxes, its application has been restricted to relatively small metabolic systems because of the limited number of measurable metabolites. In this paper, a strategy for incorporating isotope balance-based flux data obtained for a small network into metabolic flux analysis was examined as a feasible alternative allowing more accurate quantification of intracellular flux distribution in a large metabolic system. To impose GC/MS based data into a large metabolic network and obtain optimum flux distribution profile, data reconciliation procedure was applied. As a result, metabolic flux values of 308 intracellular reactions could be estimated from 29 GC/ MS based fluxes with higher accuracy.

  • A heat-flux meter has been designed and manufactured to measure the heat flux from the flame. A calibration method of the heat-flux meter by a calibration furnace has also been proposed. The k-type (Chromel-Alumel) thermocouple material has been used as the material for the beat-flux meter. The electormotive force (e.m.f.) from the K-type thermocouple is shown to be linearly proportional to the heat flux absorbed. The characteristics of the heat-flux meter become better as the radius of heat absorbing disk becomes larger and its thickness thinner.

  • SRM is designed to meet operating standards such as low cost, simple magnetic structure, a desired operating speed range, high efficiency, high performance, and good matching for DC power. The magnetic flux of SRM is independent of its direction to develop a torque and it allows the flexible characteristics of the magnetic structure for SRM. In this paper, SRM can widely classify two types, Radial-Flux SRM and Axial-Flux SRM, according to the flux direction. Radial-Flux SRM includes Conventional, Segmented stator and rotor, and Double stator SRM, etc. and Axial-Flux SRM includes C-core stator and the Axial-airgap SRM. This paper is subjected the basic characteristics to select the best of the magnetic structure of SRM in the appropriate application by the classification of SRM.

Comparative Study of Flux Regulation Methods for Hybrid Permanent Magnet Axial Field Flux-switching Memory Machines

  • This research comparatively studies three kinds of flux regulation methods, namely, stored capacitor discharge pulse (SCDP), constant current source pulse (CCSP), and quantitative flux regulation pulse (QFRP), which are used for hybrid permanent magnet (PM) axial field flux-switching memory machines (HPM-AFFSMMs). Through an analysis of the operation principle and the series hybrid PM flux regulation mechanism of the objective machine, the circuit topologies and flux regulation process of these flux regulation methods are addressed in detail. On the basis of a simulation, the flux regulation characteristics of the researched machine during the magnetization and demagnetization processes are comparatively evaluated. Then, machine performance, including back EMF, direct and quadrature axis inductances, and magnetization and demagnetization characteristics, is quantitatively investigated. Results show that the QFRP enables the HPM-AFFSMM to achieve a less harmonic component of back EMF by approximately 7.28% and 7.97% at the magnetization and demagnetization states, respectively, and a more complete magnetization process than the SCDP and CCSP.

Transdermal Delivery of Ketoprofen and the Effect of Electroosmosis (케토프로펜의 경피전달 및 전기삼투압의 영향)

  • We investigated some important factors which affect the transdermal flux of ketoprofen, a nonsteroidal anti-inflammatory agent, as a first step to provide some basic knowledge for the development of a iontophoretic transdermal patch system. Factors such as current density, polarity, buffer (HEPES) and electrolyte concentration and pH were studied using hairless mouse skin. The effect of poly(L-lysin), which is known to affect the electro-osmotic flow through skin, on flux was also studied. Passive flux was about $20\;{\mu}g/cm^2hr$ at pH 4.0, but was negligible at pH 7.4 where all ketoprofen molecules dissolved are ionized (ketoprofen pKa=5.94). At pH 4.0, application of anodal current increased the flux further above the passive level, however anodal flux at pH 7.4 was much smaller than passive flux at pH 4.0. The application of cathodal current at pH 4.0 increased the average flux to $30-40\;{\mu}g/cm^2hr$, depending on the current density applied. At pH 7.4, cathodal flux was only about $5\;{\mu}g/cm^2hr$. Decrease in buffer and electrolyte concentration increased this cathodal flux about 10 fold. However decrease in HEPES buffer concentration 100 fold did not affect the flux. Anodal flux of acetaminophen was much larger than cathodal flux, indicating that electroosmotic flow can be playing an important role in the flux. Poly(L-lysin) increased the cathodal flux at pH 7.4. These results provide some important insights into the mechanism of transdermal flux of ketoprofen and the role of electroosmotic flow.

Theoretical Analysis of Magnetic Flux Density Distribution in an Electro-Magnetic Chuck

  • The distribution of magnetic flux density of electro-magnetic chucks may clarify the clamping characteristics, which is strongly related to the machining efficiency and machining accuracy in surface grinding machine. Therefore the distribution of the normal and the tangential components of magnetic flux density have been analyzed theoretically. It appears that the normal component of magnetic flux density increases and the tangential component of magnetic flux density increases as the ratio of the separator width to the pitch, e/p decreases. The results seem to increase the stability and uniformity of normal component of magnetic flux density for the decreased e/p.

Design of a Discrete Flux Observer by the Power Series Approximation

  • The power series approximation method is proposed for real time implementations of a discrete flux observer. The proposed method improves the performance of the discrete flux observer in the case of a low sampling rate and high speed range, where the simple discrete flux observer converted by the Euler method cannot estimate the actual flux precisely. The performance of discrete flux observers with different orders of approximation is compared to find out the proper order of approximation. The validity of the proposed method is verified through simulation and experiment.

Determination of Curvature Radius of Magnetic Tool Using Weighted Magnetic Flux Density in Magnetic Abrasive Polishing (자속밀도 가중치에 의한 자유곡면 자기연마 공구곡률 선정)

  • During the magnetic abrasive polishing of a curved surface, the improvement in surface roughness varies with the maximum value and distribution of magnetic flux density. Thus, in this study, the magnetic flux density on the curved surface was simulated according to curvature radii of magnetic tool. As a result of the simulation, the 14.5mm of the magnetic tool had a higher maximum magnetic flux density and it showed a large weighted magnetic flux density. The weighted magnetic flux density means the highest value for the magnetic flux density in the curvature of the magnetic tool. From the experimental verification, the better improvement in surface roughness was observed on wider area at the 14.5mm radius of the magnetic tool than other radii.

Development of High-Temperature Heat Flux Gauge for Steel Quenching (강재 급속냉각용 고온 열유속게이지 개발)

  • The present study was motivated by increasing demands on quantitative measurements of the heat flux through the water cooling and quenching process of hot steel. The local heat flux measurements are employed by a novel experimental technique that has a function of high-temperature heat flux gauge in which test block assemblies are directly used to measure the heat flux variation during water cooling and quenching of hot steel. The heat flux can be directly achieved by Fourier's law and is also compared with numerical estimation which is solved by inverse heat conduction problem (IHCP). The high-temperature heat flux gauge developed in this study can be applicable to measure cooling rate and history during the actual cooling applications of steelmaking process. In addition, the measurement uncertainty of heat flux is calculated by a quantitative uncertainty analysis which is based on the ANSI/ASME PTC 19.1-2005 standard.

Design of Linear Transverse Flux Machine for Stelzer Machine using Equivalent Magnet Circuit and FEM

  • This paper presents the new design and validation process of the linear transverse flux machine of the stelzer machine for hybrid vehicle application. A linear transverse flux machine is a novel electric machine that has higher force density and power than conventional electric machine. The process concentrates on 2-dimensional and 3-dimensional analysis using equivalent magnetic circuit method considering leakage elements and it is verified by finite element analysis. Besides the force characteristics of all axis of each direction are analyzed. The study is considered by dividing the transverse flux electric excited type and the transverse flux permanent magnet excited type. Additionally three-dimensional analysis in this machine is accomplished due to asymmetric structure with another three axes. Finally, it suggests the new design and validation process of linear transverse flux machine for stelzer machine.
