Analytical Characteristics of GC/MS and HPLC according to the Concentration Distribution of PAHs (PAHs 농도 분포에 따른 GC/MS와 HPLC의 분석특성에 관한 연구)

  • Objectives: The purpose of this study was to determine the best method to analyze PAHs at extremely low concentrations. To this end, 16 PAHswere analyzed simultaneously by GC/MS, HPLC/FLD and HPLC/UVD, and the analytical characteristics of HPLC and GC/MS were compared. Methods: This study was conducted by GC/MS and HPLC/FLD/UVD, and evaluated linearity, precision and detection limit. Standard solutions were prepared for 21 samples in the range of $0.00001{\sim}1.0{\mu}g/mL$ and the samples were divided into four groups. All samples were made in three sets and analysis was replicated seven times. Results: Sixteen PAHs could be simultaneously separated by HPLC and GC/MS, and the adequate equipment was HPLC/FLD. The retention times by HPLC were shorter than GC/MS, and HPLC had better separation for most PAHs than GC/MS. The peaks of naphthalene and naphthalene-D8 partially overlapped for GC/MS. HPLC/FLD had a 20-2000 times lower limit of detection than GC/MS and UVD. However FLD was not adequate for analyzing acenaphthylene because it has too low a fluorescence quantum yield to be detected. The precision of HPLC/FLD/UVD and GC/MS showed less than 20% at $0.001{\mu}g/mL$ PAHs and when the concentration was higher, the coefficient of variation was decreased. HPLC/FLD was better for the overall detection of limits. Conclusions: The results indicate that the HPLC/FLD method has good linear range, precision and a detection of limits from $0.00001{\sim}0.0001{\mu}g/mL$ for all 16 PAHs. This study contributes to providing useful data for analysis technology and can be applied to occupational exposure measurement for PAHs in workplaces.

Expression of Antioxidant Isoenzyme Genes in Rice under Salt Stress and Effects of Jasmonic Acid and ${\gamma}$-Radiation

  • Analysis of chlorophyll (Chl) fluorescence implicated treatment of 40 mM NaCl decreased maximal photochemical efficiency of photosystem II (PSII) (Fv/Fm), actual quantum yield of PSII (${\Phi}_{PSII}$), and photochemical quenching (qP) in rice, but increased non-photochemical quenching (NPQ). Decreases in Fv/Fm, ${\Phi}_{PSII}$, and qP were significantly alleviated by $30\;{\mu}M$ jasmonic acid (JA), while NPQ increase was enhanced. Transcription levels of antioxidant isoenzyme genes were differentially modulated by NaCl treatment. Expression of cCuZn-SOD2 gene increased, while those of cAPXb, CATb, and CATc genes decreased. JA prevented salt-induced decrease of pCuZn-SOD gene expression, but caused greater decrease in mRNA levels of cAPXa and Chl_tAPX genes. Investigation of vacuolar $Na^+/H^+$ exchanger (NHX2) and 1-pyrroline-5-carboxylate synthetase (P5CS) gene expressions revealed transcription level of NHX2 gene was increased by JA, regardless of NaCl presence, while that of P5CS gene slightly increased only in co-presence of JA and NaCl. Unlike JA, ${\gamma}$-radiation rarely affected expressions of antioxidant isoenzyme, NHX2, and P5CS genes, except for increase in mRNA level of Chl_tAPX and decrease in that of pCuZn-SOD. These results demonstrate enhanced salt-tolerance in JA-treated rice seedlings may be partly due to high transcription levels of pCuZn-SOD, NHX2, and P5CS genes under salt stress.

Photodissociation Dynamics of Allyl Alcohol in UV: The Exit Channel Barrier for OH Production

  • Photodissociation dynamics of allyl alcohol ($H_2C$=CH-$CH_2OH$) has been investigated at 205 - 213 nm along the UV absorption band by measuring rotationally-resolved laser-induced fluorescence spectra of OH radicals. Observed energy partitioning of the available energy among products at all photon energies investigated was similar and the barrier energy for OH production is about 574.7 kJ/mol from the OH yield measurements. The potential energy surfaces for the $S_0$, $T_1$, and $S_1$ excited states along the dissociation coordinate were obtained by ab initio quantum chemical calculations. The observed energy partitioning was successfully modeled by the "barrier-impulsive model" with the reverse barrier and the geometry obtained by the calculated potential energy surfaces. The dissociation takes place on the $T_1$ excited state potential energy surface with an energy barrier in the exit channel and a large portion of the photon energy is distributed in the internal degrees of freedom of the polyatomic products.

  • The luminescence of N-methyllutidone is studied in ethanol matrix at $77^{\circ}C$K. No fluorescence is observed but a strong phosphorescence with the quantum yield of 0.1 and the lifetime of 0.2 sec is recorded. An unusually high triplet energy of 85.1 kcal/mole is determined for the compound from the O-O band of phosphorescence. The cis ${\leftrightarrow}$ trans photoisomerization of high triplet energy olefins such as 2-hexene and trans-1,4-dichlorobutene-2 is efficiently sensitized by N-methyllutidone substantiating the high triplet energy of the compound. The negative polarization of O-O band reveals the emitting triplet state to be $({\pi},{\pi}^*)^3$ state. Alkaline metal salts such as lithium chloride enhances the phosphorescence intensity through cation-N-methyllutidone coordination widening the gap between $({\pi},{\pi}^*)^3$and $(n,{\pi}^*)^3$ states.

Effect of Calcium Chloride (CaCl2) on the Characteristics of Photosynthetic Apparatus, Stomatal Conductance, and Fluorescence Image of the Leaves of Cornus kousa (염화칼슘 처리가 산딸나무 잎의 광합성 기구, 기공전도도 및 형광이미지 특성에 미치는 영향)

  • Deicing salt is used to melt snow and ice on the road for traffic safety during the winter season, which accumulates in the roadside vegetation and induces visible injuries. The damage may accelerate particularly when it coincides with early spring leaf out. In order to better understand the response mechanisms, C. kousa (3-year-old) was irrigated twice prior to leaf bud in a rhizosphere with solutions of 0.5, 1.0, and 3.0% calcium chloride ($CaCl_2$) concentration, that were made by using an industrial $CaCl_2$ reagent practical deicing material in Seoul. Physiological traits of the mature leaves were progressively reduced by $CaCl_2$ treatment, resulting in reductions of total chlorophyll contents, chlorophyll a:b, photosynthetic rate, quantum yield, stomatal conductance, $F_V/F_M$, and NPQ. On the contrary, light compensation point and dark respiration were increased at high $CaCl_2$ concentration. A decrease in intercellular $CO_2$ concentration by stomatal closure first resulted in a reduced photosynthetic rate and then was accompanied by low substance metabolic rates and photochemical damage. Based on the reduction of physiological activities at all treatments ($CaCl_2$ 0.5%, 1.0%, and 3.0%), C. kousa was determined as one of the sensitive species to $CaCl_2$.

Changes in Growth and Physiological Characteristics of Iris laevigata Fisch. by Shading Treatment (차광처리가 제비붓꽃의 생장 및 생리적 특성에 미치는 영향)

  • In this study, we investigated the growth and physiological responses of Iris laevigata Fisch. to shading treatments in order to suggest optimal light conditions for ex-situ conservation of the northern lineage plants. For this purpose, a control plant receiving full sunlight and different shading treatments (50%, 75%, 95%) were installed, and leaf mass per area, chlorophyll content and fluorescence response, and photosynthetic characteristics were investigated. I. laevigata developed leaves with higher photosynthetic efficiency to adapt to lower light intensity as shading levels increased. Chlorophyll content increased with increasing shading levels, and leaf mass per area decreased with increasing leaf area. The chlorophyll fluorescence responses Fv/Fm and NPQ did not change with shading, and the activity of the carbon fixation system did not differ between treatments. I. laevigata exhibited a light-saturation point equivalent to that of sun plants and maintained photosynthetic capacity similar to that of controls up to 75% shading. The apparent quantum yield of I. laevigata decreased significantly at 95% shading, indicating adaptation to lower light conditions. It seems that the photosynthetic capacity of I. laevigata decreases when grown under 95% shading level compared to full sunlight, and it is judged that the longer the light is restricted by continuous shading, the more unfavorable the growth will be.

Application of Chlorophyll Fluorescence Parameters for the Detection of Water Stress Ranges in Grafted Watermelon Seedlings (수박접목묘의 건조스트레스 범위 탐지를 위한 엽록소형광 지수의 적용)

  • This study was carried out to quantify the drought stress in grafted watermelon seedlings non-destructively by using chlorophyll fluorescence (CF) imaging technique rather than the visual judgment. Six-day old watermelon seedlings were grown under uniform irrigation for 3 days, and then given drought stress. Afterward, the sensor for the measurement of water content in plug tray cell unit was used to classify the drought-stress level into nine groups from D1 (53.0%, sufficient moisture state) to D9 (15.7%, extremely dry stress), and the 16 CF parameters were measured. In addition, re-irrigation was performed on the drought stressed seedlings(D5 - D9) to determine the growth and photosynthesis recovery level, which was not confirmed by visual judgment. The kinetic curve patterns of CF in three different drought stressed seedling groups were found to be different for the early detection of drought stress. All the 16 CF parameters decreased continuously with exposure to drought stress and drastically decreased from D5 (32.1%) where the visual judgment was possible. The fluorescence decline ratio (Rfd_Lss) started to decrease from the initial drought stress level (D5 - D6), and the Maximum PSII quantum yield (Fv/Fm) was significantly decreased in the later extreme drought stress range (D7 - D9) by re-irrigation recovery test. Thus, Rfd_Lss and Fv/Fm parameters were finally selected as potent indicators of growth and photosynthesis recovery in the initial and later stages of drought stress. Also, to the differences in the numerical values of the individual chlorophyll fluorescence parameters, the drought stress level was intuitively confirmed through the image. These results indicate that Rfd and Fv/Fm can be considered as potential CF parameters for the detection of low and extremely high drought stress, respectively. Furthermore, Fv/Fm can be considered as the best CF parameters for recovery at re-irrigation.

Physiological Responses of Bupleurum latissimum Nakai, Endangered Plants to Changes in Light Environment (광환경조절에 따른 멸종위기식물 섬시호의 생리적 반응)

  • This study was conducted to investigate the physiological responses of Bupleurum latissimum, endangered plants by light condition. We investigated photosynthetic parameters, chlorophyll contents and chlorophyll fluorescence under different shading treatments (Shaded 50%, 70%, 90% and non-treated). Results showed that net apparent quantum yield (AQY) and chlorophyll contents were significantly increased with elevating shading level. However, light compensation point (LCP) and dark respiration ($R_d$) were shown the opposite trend. Especially, non-treated exhibited photoinhibition such as reduction of chlorophyll contents and maximum photosynthesis rate ($Pn_{max}$) also variation trend of stomatal conductance ($g_s$), and transpiration rate (E) were decreased to prevent water loss. Photosynthetic rate ($P_{Nmax}$) and photochemical efficiency (Fv/Fm) of 90% treatment showed a drastic reduction in July. This implies that photosynthetic activity will be sharply decreased with a long period of low light intensity. The 50% treatment showed relatively higher photosynthetic activity than other treated. This result suggested that growth and physiology of B. latissimum adapted to 50% of full sunlight.

Physiological Response and Growth Performance of Parasenecio firmus under Different Shading Treatments (차광처리에 따른 병풍쌈의 생리반응 및 생장특성)

  • This study was conducted to investigate the chlorophyll contents, photosynthetic characteristics, chlorophyll fluorescence, and growth performance of Parasenecio firmus under changing light environment. Parasenecio firmus was grown under non-treated (full sunlight) and three different shading conditions (88~93%, 65~75% and 45%~55% of full sunlight) for the experiment. Total chlorophyll content, photochemical efficiency (Fv/Fm), T/R ratio, specific leaf area (SLA), leaf area ratio (LAR), and leaf weight ratio (LWR) were increased with increasing shading level, but decreased dark respiration. Therefore, light absorption and light utilization efficiency were improved under the low intensity light. Plants under 65~75% of full sunlight had best maximum photosynthetic rate and net apparent quantum yield in May. On the other hand, the non-treated plants had lower maximum photosynthetic rate, photochemical efficiency, and chlorophyll content than the treated ones. Parasenecio firmus considered to be a sciophyte, is fairly sensitive to high intensity light. If 88-93% of full sunlight lasts for a long period, photosynthetic capacity will be sharply decreased, though limiting light. These results suggest that growth of Parasenecio firmus adapted to 65~75% of full sunlight.

The responses of Growth and Physiological traits of Acer triflorum on Calcium Chloride ($CaCl_2$) Concentration (염화칼슘 농도에 따른 복자기의 생장 및 생리적 반응 특성)

  • To prevent freezing of the road by fallen snow, Calcium chloride($CaCl_2$) as a deicer is used to very often and it can be harmful to roadside trees. This study was conducted to investigate the effects of Calcium chloride($CaCl_2$) as a deicer on growth and physiological traits of Acer triflorum according to different concentration of $CaCl_2$. We measured growth, chlorophyll contents, gas exchangement characteristics, chlorophyll fluorescence and mineral nutrition concentration in plant and soil. The experimental group was composed of four treatments including 0mM(control), 9mM(0.5 %), 18mM(1.0 %), 54mM(3.0 %). Before germinating new shoot, the dissolution of $CaCl_2$ was irrigated twice interval of a week. At 30 days after treatment, all treatments decreased total cholorophyll content, photosynthetic rate, transpiration rate, stomatal conductance and photochemical efficiency($F_v/F_m$) with increasing concentration of $CaCl_2$ and especially, they significantly reduced in 3.0 % treatment. In contrast, chlorophyll a/b ratio increased with an increase of $CaCl_2$ concentration and water use efficiency increased in 1.0 % and 3.0 % treatments. At 50 days after treatment, all treatments were decreased in chl a, chl b, total chlorophyll content, carotenoid content, photosynthetic capacity, photochemical efficiency($F_v/F_m$) and quantum yield of photosystem II(${\Phi}_{PSII}$) compared with control and 3.0 % treatments were withered. $Ca^{2+}$ and $Cl^-$ were accumulated in leaves and soil, which inhibited water absorption and electron transport and it caused the reduction of height growth rate more than 50 %. Although there was a little difference according to time and $CaCl_2$ concentration, all treatments decreased in growth rate and physiological activity slowed down. As time passed, these results got worse. Therefore we need to take a measure earlier in order to minimize damage of trees.
