To measure the flow velocity and direction in the near field of an unmanned underwater vehicle, an optical measurement unit containing an image sensor and a phosphor-integrated pillar that mimics the neuromasts of a fish was constructed. To analyze pillar movement, which changes with fluid flow, fluorescence image analysis was conducted. To analyze the flow velocity, mean force analysis, which could determine the relationship between the light intensity of a fluorescence image and an external force, and length-force analysis, which could determine the distance between the center points of two fluorescence images, were employed. Additionally, angle analysis that can determine the angles at which pixels of a digital image change was selected to analyze the direction of fluid flow. The flow velocity analysis results showed a high correlation of 0.977 between the external force and the light intensity of the fluorescence image, and in the case of direction analysis, omnidirectional movement could be analyzed. Through this study, we confirmed the effectiveness of optical flow sensors equipped with phosphor-integrated pillars.
The response of the freshwater microalga, Chlorella vulgaris, to heavy metal stress was examined based on chlorophyll fluorescence analysis to assess the toxic effects of heavy metals in freshwater ecosystems. When toxic effects were analyzed using regular chlorophyll fluorescence analysis, photosystem II activity($F_v/F_m$) decreased significantly when exposed to $Cu^{2+}$ and $Hg^{2+}$ for 12 h, and decreased in the order of $Hg^{2+}>Cu^{2+}>Cd^{2+}>Ni^{2+}$ when exposed for 24h. The effective photochemical quantum yield(${\phi}{\prime}_{PSII}$), chlorophyll fluorescence decrease ratio($R_{Fd}$), minimal fluorescence yield($F_o$), and non-photochemical quenching(NPQ), but not photochemical quenching(qP), responded sensitively to $Hg^{2+}$, $Cu^{2+}$, and $Cd^{2+}$. These results suggest that $F_v/F_m$, as well as ${\phi}{\prime}_{PSII}$, $R_{Fd}$, $F_o$, and NPQ could be used to assess the effects of heavy metal ions in freshwater ecosystems. However, because many types of heavy metal ions and toxic compounds co-occur under natural conditions, it is difficult to assess heavy metal toxicity in freshwater ecosystems. When Chlorella was exposed to heavy metal ions for 12 or 24h, $F_v/F_m$ and maximal fluorescence yield($F_m$) changed in response to $Hg^{2+}$ and $Cu^{2+}$ based on image analysis. However, assessing quantitatively the toxic effects of several heavy metal ions is challenging.
Optical methods such as UV and fluorescence spectrophotometers can be applied not only in the qualitative analysis of dissolved organic matter (DOM), but also in real-time quantitative DOM monitoring for wastewater and natural water. In this study, we measure the UV254 and fluorescence excitation emission spectra for a sewage treatment plant influent and effluent, and river water before and after sewage effluent flows into the river to examine the composition and origin of DOM. In addition, a correlation analysis between quantified DOM characteristics and dissolved organic carbon (DOC) was conducted. Based on the fluorescence excitation emission spectra analysis, it was confirmed that the protein-type tryptophan-like DOM was the dominant substance in the influent, and that the organic matter exhibited relatively more humic properties after biological treatment. However, DOM in river water showed the fluorescence characteristics of terrestrial humic-like and algal tyrosine-like (protein-like) organic matter. In addition, a correlation analysis was conducted between the DOC and optical indices such as UV254, the fluorescence intensity of protein-like and humic-like organic matter, then DOC prediction models were suggested for wastewater and river monitoring during non-rainfall and rainfall events. This study provides basic information that can improve the understanding of the contribution of DOC concentration by DOM components, and can be used for organic carbon concentration management in wastewater and natural water.

In this paper, laser-induced fluorescence properties of four plastics were characterized through spectrometer analysis for real-time microplastic counting. Recently, environmental problems related to microplastics have emerged. In order to detect microplastics, analysis methods such as FT-IR and Raman are used. However, they have the disadvantages of being time-consuming and requiring a pretreatment process. In most plastic products on the market, 10% to 30% of plasticizers and reinforcing agents are added. Therefore, most microplastics present in seawater and freshwater emit fluorescence signals by 270 nm UV light source regardless of their type due to their molecular structure due to additives. Real-time microplastics counting is possible more easily by using the proposed laser-induced fluorescence detection method because of the fluorescence expression characteristic of 340 nm that appears due to the plasticizer of plastics.
In order to improve the transfection efficiency of CHO/dhfr- cells using cationic lipid, optimal concentrations of the cationic lipid($LipofectAmine^{TM}$) and DNA(pEGFP-C1) need to be determined. The use of green fluorescent protein(GFP) gene as a reporter gene facilitated the quantification of transfection efficiency. The green fluorescence intensity of each cell transfected at various lipid-DNA concentrations was measured using fluorescence-activated cell sorter(FACS) analysis. A combination of $2.0{\mu}L$ cationic lipid and 0.4{$\mu}g$ DNA in a well resulted in the highest trasfection efficiency. Taken together, the method using FACS analysis of GFP is simple and fast, facilitating the optimization of transfection.

This study suggests a new fluorescence microscope to observe micro-samples within fluorophore in a variety of biomedical fields including the fluorescence analysis of a biochip, such as a DNA micro-array. A fluorescence microscope is a device for irradiating light onto a micro-object, executing an excitation and fluorescence emission process. In this study, it adopts a total internal reflection fluorescence(TIRF) method to excite a whole micro-sample substrate different from an existing way which uses an evanescent wave resulting from a total internal reflection on the micro-sample surface. Suggested TIRF microscope can reduce optical noise and obtain images with higher sensitivity thus obtain precise information about the density, quantity, location, etc. of a flurophore, and can simultaneously process separate images even when plurality of fluorophores having different excitation and fluorescent wavelength ranges is distributed, thus easily obtain information about the fluorophores.

The characteristics of soft clays is very important for the land development plan. This study is to investigate correlations between the engineering properties and the characteristics of clay minerals of the disturbed clay samples obtained from Sihwa area. This study included X-Ray Diffraction Analysis, X-Ray Fluorescence Spectrometer Analysis, Scanning Electron Microscopy Analysis and Energy Dispersive X-Ray Spectrometer Analysis. The correlations between the clay mineral properties and the laboratory and field testing results were investigated.
A comparison was made of two detection methods(UV absorbence detection and fluorescence detection with pre-column derivatization, with trifluoroacetic acid) coupled with HPLC for the simultaneous determination of aflatoxin $B_1, B_2, G_1$ and $G_2$. A good separation of the four aflatoxins was achieved on a reversed-phase $C_{18}$ column (30 cm x 3.9 mm) with methanol-acetonitrile-water(20+20+60) for absorbence detection or acetonitrile-water(25+75) for fluorescence detection at the flow rate of 1.0 ml/min. The calibration graphs were linear over the ranges 100 ppb-1 ppm for $B_1/G_1$ and 30~300 ppb for $B_2/G_1$ with absorbence detection, and 1~500 ppb for $B_1/G_1$ and 0.3~150 ppb for $B_2/G_2$ with fluorescence detection. The correlation coefficients were greater than 0.94 and 0.99 for absorbance detection and for fluorescence detection, respectively. The detection limit was 100 ng for $B_1/G_1$ and 30 ng for $B_2/G_2$ with absorbence detection, and 1 ng for $B_1/G_1$ and 0.3 ng for $B_2/G_2$ with fluorescence detection. Recovery rates of aflatoxin $B_1, B_2, G_1$ and $G_2$ added to yeast-extract sucrose broth medium were 66.6%, 59.4%, 67.5% and 59.2%, respectively, for absorbence detection and 82.9%, 71.5%, 80.0% and 69.3%, respectively, for fluorescence detection. The four aflatoxins in culture medium were quantitatively detected by the two methods. The aflatoxins in the rice sample were not detected the absorbence detection method, but were below 10 ppb using the fluorescence detection method. Analysis of aflatoxins by both the absorbence and fluorescence methods coupled with HPLC showed acceptable linearity and good recovery. The absorbence detection was less timeconsuming and safer for treatment. The fluorescence detection was more elective and sensitive though elevated $B_1$ and $G_1$ contents were determined from the TFA-induced conversion of $B_1$ to $B_{2a}$ and $G_1$ to $G_{2a}$.
As an intracellular degradation system, autophagy is an essential and defensive cellular program required for cell survival and cellular metabolic homeostasis in response to various stresses, such as nutrient deprivation and the accumulation of damaged organelles. In general, autophagy flux consists of four steps: (1) initiation (formation of phagophore), (2) maturation and completion of autophagosome, (3) fusion of autophagosomes with lysosomes (formation of autolysosome), and (4) degradation of intravesicular components within autolysosomes. The number of genes and reagents that modulate autophagy is increasing. Investigation of their effect on autophagy flux is critical to understanding the roles of autophagy in many physiological and pathological processes. In this review, we summarize and discuss ways to analyze autophagy flux quantitatively and qualitatively with the use of imaging tools. The suggested imaging method can help estimate whether each modulator is an inhibitor or a promoter of autophagy and elucidate the mode of action of specific genes and reagents on autophagy processes.
In this paper, we propose a skin lesion detection to develop the system of fluorescence image analysis to identify the fluorescence of topical methyl aminolevulinate(MAL) idduced PpIX in patients with BCC accurately. By fluorescence image analysis we define the border between tumo and tumor-free areas on fluorescence image after topical application of MAL ointment. We excised both the tumor and peri-tumoral areas widely from the 10 patients with BCC, and divided tissue samples into 3 area, such as tumor area, suspected tumor area, tumor-free area, respectively. Our proposed method migt play a role as an adjunctive tool to define the border between tumor and tumor-free areas for Mohs' micrographic surgery.
